Radiation Damage Mechanism of the Small Intestine's Absorptive Epithelium and Radioprotection Effect after Propolis Abdominal Cavity Injection (프로폴리스 복강 주사 후 소장 흡수상피조직의 방사선 손상 기전과 방어효과)

  • As Small Intestine Absorptive Epithelium Cells are surrounded by mucus polysaccharide and lymphocytes and mitochondria, they are sensitive to radiation energy. Damaged cells lead to a deficiency of nutrients and the imbalance of electrolyte metabolism, which in turn can becomes a major cause of an intestine tract death. This research observed ultra structures after injecting propolis into the abdominal cavity in order to reveal the radiation damage mechanism and radioprotection effect of intestine absorptive epithelium cells. The result of this research's observation found that stenosis occurred in the small intestine in some tissues 20 days after 5Gy irradiation, their surface turned black, and their elasticity dropped. Through observation with an optical microscope, it was found that the size of the goblet cells decreased, while the paneth granulate cells atrophied and were vacuolated. Observation with an transmission electron microscope(TEM) revealed that while microvill and lysosome were normally observed in jejunum tissues, mitochondria membrane was damaged and uneven surfaces were formed on lymphocytes. The membrane of absorptive epithelium cells hypertrophied in tissues of the ileum, and vacuole was observed. However, the observation after injecting propolis into the abdominal cavity found that mitochondria damage dropped dramatically, and radioprotection effects were to some extent confirmed, considering that glycocalyx of villi was clear, and M cells could be observed.

Protective Effects of 5-Androstendiol (5-AED) on Radiation-induced Intestinal Injury (방사선에 의한 장점막 손상에 대한 5-Androstenediol의 보호효과)

  • Purpose: We examined the radioprotective effects of 5-androstendiol (5-AED), a natural hormone produced in the reticularis of the adrenal cortex, as a result of intestinal damage in gamma-irradiated C3H/HeN mice. Materials and Methods: Thirty mice (C3H/HeN) were divided into three groups; 1) non-irradiated control group, 2) irradiated group, and 3) 5-AED-treated group prior to irradiation. Next, 5-AED (50 mg/kg per body weight) was subcutaneously injected 24 hours before irradiation. The mice were whole-body irradiated with 10 Gy for the histological examination of jejunal crypt survival and the determination of the villus morphology including crypt depth, crypt size, number of villi, villus height, and length of basal lamina, as well as 5 Gy for the detection of apoptosis. Results: The 5-AED pre-treated group significantly increased the survival of the jejunal crypt, compared to irradiation controls (p<0.05 vs. irradiation controls at 3.5 days after 10 Gy). The evaluation of morphological changes revealed that the administration of 5-AED reduced the radiation-induced intestinal damages such as villus shortening and increased length of the basal lamina of enterocytes (p<0.05 vs irradiation controls on 3.5 day after 10 Gy, respectively). The administration of 5-AED decreased the radiation-induced apoptosis in the intestinal crypt, with no significant difference between the vehicle and 5-AED at 12 hours after 5 Gy. Conclusion: The results of this study suggest that the administration of 5-AED has a protective effect on intestinal damage induced by $\gamma$-irradiation. In turn, these results suggest that 5-AED could be a useful candidate for radioprotection against intestinal mucosal injury following irradiation.

Quantitative Analysis of Small Intestinal Mucosa Using Morphometry in Cow's Milk-Sensitive Enteropathy (우유 과민성 장병증(cow's milk-sensitive enteropathy)에서 소장 생검조직의 형태학적 계측을 이용한 정량적 분석)

  • Purpose: To make objective standards of small intestinal mucosal changes in cow's milk-sensitive enteropathy (CMSE) we analyzed histological changes of endoscopic duodenal mucosa biopsy specimens from normal children and patients of CMSE. Methods: We review the medical records of patients who had been admitted and diagnosed as CMSE by means of gastrofiberscopic duodenal mucosal biopsy following cow's milk challenge and withdrawal. Thirteen babies with CMSE, ranging from 14 days to 56 days of age, were studied. Five non-CMSE patients were used as control, ranging from 22 days to 72 days of age. The morphometric parameters under study were villous height, crypt zone depth, ratio of villous height to crypt zone depth, total mucosal thickness and length of surface epithelium by using H & E stained specimens under the drawing apparatus attached microscope. In addition, the numbers of lymphocytes in the epithelium and eosinophil cells in the lamina propria and epithelium were measured. Results: In the duodenal mucosal biopsy specimens in CMSE we found partial and subtotal villous atrophy with an increased number of interepithelial lymphocytes. The mean villous height($135{\pm}59\;{\mu}m$), ratio of villous height to crypt zone depth ($0.46{\pm}0.28$), total mucosal thickness ($499{\pm}56\;{\mu}m$), length of surface epithelium of small intestinal mucosa ($889{\pm}231\;{\mu}m$) in CMSE was significantly decreased compared with the control (p<0.05). The mean crypt zone depth ($311{\pm}65\;{\mu}m$) was significantly greater than the control ($188{\pm}24\;{\mu}m$)(p<0.05). Infiltration of interepithelial lymphocytes ($34.1{\pm}10.5$) were significantly greater than the control ($13.6{\pm}3.6$)(p<0.05). The number of eosinophil cells in both lamina propria and epithelium was no significant differences between groups (p>0.05). The small intestinal mucosa in treated CMSE showed much improved enteropathy of villous height, crypt zone depth, interepithelial lymphocytes compared with the control as well as untreated CMSE. Conclusion: Quantitation of mucosal dimensions confirmed the presence of CMSE. It seems to be a limitation in the capacity of crypt cells to compensate for the loss of villous epithelium in CMSE. Specimens obtained by gastrofiberscopic duodenal mucosal biopsy were suitable for morphometric diagnosis of CMSE. Improvement of CMSE also can be confirmed histologically after the therapy of protein hydrolysate.

Studies on the Efficacy of the Newly Developed Automatic Liquid Feeder for 7-day Old Early Weaned Piglets (7일령 조기 이유자돈에 대한 액상사료 자동급이기 이용효과)

  • This study was conducted to test the efficacy of the automatic liquid feeder(designated as NLRI) developed by National Livestock Research Institute, Korea for 7-day old early-weaning piglets. The other automatic liquid feeder imported from USA(designated as IALF) was used for a comparative purpose. A total of sixty piglets of 7 days of age were allotted to three treatments. The control group of 20 piglets was raised by their own sows until 21 days of age, while others were shifted to automatic liquid feeders of either NLRI or IALF on 7 days of age and reared during 14 days post-weaning. After then, all the piglets were fed nutritionally identical mash diets until 70 days of age. Compared with control group, body weight of piglets reared on automatic liquid feeders were lighter(P〈0.05) at 21 days of age [5.55kg (control) vs 4.97 (NLRI) and 4.98kg (IALF)], while heavier(P〈0.05) at 70 days of age(24.82kg vs 30.17 and 29.42kg). The results indicated that pigs reared on liquid feeding showed higher(P〈0.05) average daily gain than control pigs during the whole experimental period [346.7g (control) vs 425.8 (NLRI) and 416.1g (IALF)], while no difference was found in feed/gain (1.67 vs 1.78 and 1.84). There was no difference in growth performance and incidence of diarrhea between two automatic liquid feeders, NLRI and IALF. Compared with control group, intestinal villi tended to be shorter in liquid feeding group during the first week, but were recovered within two weeks. The results suggest that the automatic liquid feeder newly developed by National Livestock Research Institute, Korea can be successfully used for rearing young piglets weaned at very early age.

Effects of Single Cells of Carrot and Radish on the Fecal Excretion Properties, Mineral Absorption Rate and Structure of Small Intestine and Colon in Rats (당근과 무의 단세포물 섭취가 흰쥐의 배변특성, 무기질 흡수율 및 소장과 대장의 구조에 미치는 영향)

  • The effects of single cells of carrot or radish on the fecal excretion, mineral absorption rate and structure of small intestine and colon were investigated in rats fed 5% single cell diet for 4 weeks. Carrot contained 28.76% of total dietary fiber and 21.45% of insoluble dietary fiber, and radish contained 23.14% of total dietary fiber and 16.77% of insoluble dietary fiber on a dry weight basis. Total dietary fiber contents of the single cell were 44.68% for carrot, 48.0% for radish. Absorption rates of magnesium were higher in the carrot and radish single cell groups than the other groups. Cellulose significantly increased fecal weight and weight of small intestine. The consumption of cellulose and single cells decreased digestibility of lipid. The length of colon were longer in the carrot and the radish group than the other groups. Scanning electron microscopy studies showed that small intestine microvilli with leaf-shaped were seen in cellulose and single cell fed groups. These results suggest that the diet containing 5% single cells of carrot or radish increases the digestibility of dietary fiber, weight gain, and fecal output and shorten the gastrointestinal transit time.

A method of assisting small intestine capsule endoscopic lesion examination using artificial neural network (인공신경망을 이용한 소장 캡슐 내시경 병변 검사 보조 방법)

  • Human organs in the body have a complex structure, and in particular, the small intestine is about 7m long, so endoscopy is not easy and the risk of endoscopy is high. Currently, the test is performed with a capsule endoscope, and the test time is very long. The doctor connects the removed storage device to the computer to store the patient's capsule endoscope image and reads it using a program, but the capsule endoscope test results in a long image length, which takes a lot of time to read. In addition, in the case of the small intestine, there are many curves due to villi, so the occlusion area or light and shade of the image are clearly visible during the examination, and there may be cases where lesions and abnormal signs are missed during the examination. In this paper, we provide a method of assisting small intestine capsule endoscopic lesion examination using artificial neural networks to shorten the doctor's image reading time and improve diagnostic reliability.

A Study on the Change of the IEL Number in the Intestine of Rats Infected with Echinostoma hotense (Echinostoma hortense를 실험 감염시킨 흰쥐 장상피에서 림프구 (IEL)증감에 관한 연구)

  • The relationship between the intestinal histopathology and number and position of intraepithelial lymphocytes (IEL) was observed chronologically in the small intestine of rats experimentally infected with Echinostoma hortense. Sprague-Dawley rats were orally infected with 200 metacercariae obtained from Misgurnus anguillicaudatus. The rats each were sacrificed on the week 1, 2, 4, 6, 8 post-infection (PI) and samples of the intestine in the part of duodenum and jejunum were taken. The samples were stained with Hematoxylin-eosin and Giemsa. The intestinal histopathology was the severest after the week 1 PI and characterized by villous atrophy, crypt hyperplasia and decrease of villus/crypt(v/c) ratio, which continued until the week 8 Pl. The number of IEL dramatically decreased during the week 1 PI, but increased gradually thereafter with a slight decrease on the week 8 PI. In control rats, the great majority of the IEL were located at the basal region of the epithelium. During the early stage of infection, however, we found a considerable proportion of IEL to moved to the intermediate or apical regions of the epithelium. From the above results, it is sugested that the change of IEL number and position during the course of E. hortense infection should be closely related to the progression and recovery of the intestinal histopathology.

The effect of reinfection with Neodiplostomum seoulensis on the histopathology and activities of brush border membrane bound enzymes in the rat small intestine (서울주걱흡충의 재감염이 흰쥐 소장의 조직병리 및 미소융모막효소 활성도에 미치는 영향)

  • Neodiplostomum seoulensis, one of the human intestinal trematodes , was reinfected to albino rats, and worm recovery rates, histopathology and activity changes of the intestinal brush border membrane bound enzymes were observed. The experimental groups were three: uninfected, primary infection and reinfection. The worm recovery rate in the reinfection group was much lower than in the primary infection group 14 days after infection. The duodenal histopathology showed villous atrophy during the first and second week in the primary infection group. In the reinfection group, however. villous changes occurred as early as 3 days after the infection, and the lesion was found healed 7 days after infection. The activities of alkaline phosphatase and sucrase in the duodenum of primary infection rats decreased nearly half of the controls 2 weeks after infection, whereas the activities were unchanged in the reinfection group. However, no changes in the activities were observed in the proximal jejunum between the experimental groups. These findings suggested that a secondary infection of N. seouLensis in rats should make less damage on the intestinal mucosa than a primary infection. Expression patterns of proliferating cell nuclear antigen in the small intestine of mice infected with Metagonimus yokogawai and Metagonimus Miyata type (요코가와흡충 및 미야타흡충 감염 마우스 소장의 PCNA발현 양상)

  • Genus Metasonimus has been a subject of taxonomic debates for several years. In morphological aspects, M. vokogawci has been thought to have three subtypes, namely Yokogawa type (M. yokogawai in strict sense) , Miyata type and Koga type. But dif- ferences in the intestinal pathology induced by these subtypes have not been studied yet. In this study we compared the pathological reactions inducted by M. yokoBnwai and Metcgonimus Miyata type using proliferating cell nuclear antigen (PCNA) index. Metacercariae (Mc) of M. yokogcuani were collected by artificial digestion of Plecoslossw oLtiveLis and Mc of Metngonimus Miyata type were collected from Zacco pEntvpus. Three hundreds Mc of each species were infected orally to ICR mice. The mice were sacrificed at 3, 6. 10. 16 and 23 days after infection and the small intestines were resected into three portions (proximal. middle, and distal) . Immunohistochemical staining for PCNA was done using PC-10 (DAKO-PCNA, CA, USA) The PCNA indices in M. Wokosnwci infected group on the 6th and 23rd day after infection were lower than in the control and Miyata type infect ed groups (p < 0.05) from all of the three intestinal regions. On the other hand, the control group and Ifetngonimus Miyata type infected group did not make any differences in PCNA indices. The villus/crypt WIC) ratio was also decreased significantly in M. Wokogcwni infected mice but not in Metagonimus Miyata type infected ones. It is suggested that M. wokogawci induce villous atrophy through a decrease in the cell proliferation at the crypt. The results of this study suggested that M. WokoBnwai induce more serious intestinal pathology than Metcsonimuf Miyata type.

Occurrence of Clostridium perfringens according to Raising Periods in Broilers (육계의 사육 일령별 Clostridium perfringens 발생 분석)

  • The objective of this study was to investigate occurrence patterns of Clostridium perfringens on different raising periods in broilers. In different raising periods, we investigated the change in the gross lesion and microscopic histological findings of the mucose of the small intestine, colony forming unit (CFU) and the types C. perfringens with PCR assay. According to the gross lesions on the mucose of small intestine with 10-days-old broilers, the non-antibiotic group showed a higher value (0.6) than the antibiotic group (0.0). Whereas 20-days-old broilers with, the antibiotic treatment had a slightly lower value (1.0) than the non-antibiotic group (1.3). In the histological examination on the villi of the small intestine, there was no damage of the villi of the small intestine with 1-day-old broilers in both groups; however, the non-antibiotic group showed a higher value (0.4) than the antibiotic group (0.0) with 10-days-old broilers. In the non-antibiotic group, the CFU of C. perfringens of the fecal samples from the small intestine increased from 10 days of raising broilers and rapidly increase after 20 and 30 days of raising broilers. There was no detection of C. perfringens types with PCR assy in 1-day-old broilers, but we found C. perfringens type A in 10-, 20- and 30-days-old broilers. Although it is possible to raise healthy broilers by using antibiotics, the addition of antibiotics to concentrate feed is prohibited for public health. The results of this study would contribute to proper feeding management through the careful use of antibiotics.
