Screening and Characterization of Drosophila Development Mutants Using Single P[en-lacZ] Element Mutagenesis (Drosophila single P[en-lacZ] element mutagenesis를 이용한 발생 관련 돌연변이체 작성)

  • Single P[en-lacZ] element including 5.7 kb of engrailed upstream sequences and the E. coli lacZ fusion gene, localized on 48A in rxyho25 strain was transposed to different sites in the Drosophila genome by the jumpstart technique. From 3315 individual genetic crosses, 113 new insertion lines carrying P[en-lacZ] inserted at different sites were obtained. $\beta$-Galactosidase expression in larval tissues of 113 insertion lines were detected by X-gal staining. & among 113 lines have been indentified to be for recessive lethal mutations. Among 7 lines, the #1119 line being lethal during embryogenesis was examined about the ${\beta}$$-Galactosidase expression, nuclear behavior and cellularization pattern during embryogenesis. The P[en-lacZ] insertion lines obtained in this study could be utilized for studying structure and function of the Drosophila development-related genes.

Comparison of Mitochondria-related Conserved Genes in Eukaryotes and Prokaryotes (진핵생물과 원핵생물의 미토콘드리아 관련 보존적 유전자 비교)

  • Sixty-two conserved orthologous groups (OGs) of proteins, in 63 prokaryotes and seven eukaryotes were analyzed to identify essential proteins in the mitochondria of eukaryotes, and their counterparts in prokaryotes. Twenty OGs were common in eukaryotic mitochondria, and all were translation related. Encephalitozoon cuniculi, an obligate parasitic eukaryote, shares no common mitochondrial OGs with the other 69 organisms. Seventeen conserved OGs were mitochondria related in the 69 organisms. Mitochondria related- and nonrelated-OGs were divided into prokaryotic genomes (p<0.001, paired t-test) unlike eukaryotic genomes in the distance value analysis. The most commonly conserved mitochondria-related OG was COG0048-KOG1750 (ribosomal small subunit S12), whereas it was COG0100-KOG0407 (ribosomal small subunit S11) in nonrelated OGs. These results could be applied in scientific research to determine phylogenetic relationships and in areas such as drug development.

  • Abnormal distribution of the enteric nerves such as adrenergic, cholinergic and peptidergic nerves may cause the functional obstruction in Hirschsprung's disease (HD). Although the sustained contraction of the aganglionic segment is the main pathophysiology of HD, the etiology and pathogenesis is not thoroughly understood, With the recent progress of molecular biology and genetics,a more detailed approach to the pathogenesis of the HD can be undertaken. In this review, the roles of the nitric oxide, nitric oxide synthase and interstitial cells of Cajal on smooth muscle relaxation, the effects of extracellular matrix, cell adhesion molecules, neurotrophic factors on the migration and maturation of the neural crest cells are described. In the section of genetic factors, familial occurrences, association of chromosomal abnormalities, RET gene, glial cell line-derived neurotrophic factor gene, endothelin-3 gene and endothelin-B receptor gene and their r elationships to HD is briefly reviewed.

Anticancer Effect of Activated Natural Killer Cells on Human Colorectal Tumor (결장암에 대한 활성 자연살해세포의 항암효능)

  • Colorectal cancer is one of the most common alimentary malignancies. In this study, the antitumor activity of activated human natural killer (NK) cells against human colorectal cancer was evaluated in vivo. Human NK cells are the key contributors of innate immune response and the effective functions of these cells are enhanced by cytokines. Human peripheral blood mononuclear cells (PBMC) were cultured with interleukin-2 (IL-2)-containing medium for 14 days and resulted in enriched NK cell population. The resulting populations of the cells comprised 7% $CD3^+CD4^+$ cells, 25% $CD3^+CD8^+$ cells, 13% $CD3^-CD8^+$ cells, 4% $CD3^+$CD16/$CD56^+$ cells, 39% $CD3^+$CD16/$CD56^-$ cells, and 52% $CD3^-$CD16/$CD56^+$ cells. Tumor necrosis factor alpha (TNF-$\alpha$), interferon gamma (IFN-$\gamma$), IL-2, IL-4, and IL-5 transcripts of the activated NK cells were confirmed by RT-PCR. In addition, activated NK cells at doses of 2.5, 5 and 10 million cells per mouse inhibited 10%, 34% and 47% of SW620-induced tumor growth in nude mouse xenograft assays, respectively. This study suggests that NK cell-based immunotherapy may be used as an adoptive immunotherapy for colorectal cancer patients.

Anti-Inflammatory Effects of Fusion-Fermented Aralia continentalis Radix (fACR) on THP-1 cells (THP-1 세포에서 융합 발효 독활의 항염증 효과)

  • This study was to demonstrate the anti-inflammation effect using extracts derived from fusion-fermentation to add the function of Araliae Continentalis Radix (fACR). Since there are limitations to the use of ACR, available strains were selected through fermentation using lactobacillus strains, and the increases in total amount of polyphenols and amino acids was confirmed through comparison with Hot Water Extract of ACR. To determine the anti-inflammatory effect of the fACR was measure cytokine inflammation associated with arthritis, the arthritis when cartilage destruction is accompanied mainly MMP-9 activity was confirmed to evaluate the inhibition effect. These results show that fermentation using lactobacillus increases major biological activities and inflammatory response-restraining effects of ACR. This study is expected to be a basis for studying the preventive effect of fACR on arthritis.

  • Study on spermatid and spermatozoon of a Korean endemic freshwater fish, Microphysogobio yaluensis, has been carried out by transmission electron microscope. Spermiogenesis involves conspicuous modifications intercelluar movement such as centrioles and mitochondria migration, nuclear depression and structural changes for instance chroatin condensation, excess of cytoplasm. The mature spermatozoa are similar to those of other cyprinids as follows: a spherical nucleus with a shallow nuclear fossa, a short midpiece containing several mitochondria and encircling the basal body of the flagellum. However there are some differences in the orientation of the centrioles,the number of the mitochondria and distribution of vesicles from other cyprinids. It could be concluded that the M. yaluensis spermiogenesis belongs to type 2, and its mature spermatozoon is charaterized by a unique feature which may provide a useful systematic character.

  • Recently, the trial and success of malicious cyber attacks has been increased rapidly with spreading of Internet and the activation of a internet shopping mall and the supply of an online internet, so it is expected to make a problem more and more. Currently, the general security system based on Internet couldn't cope with the attack properly, if ever, other regular systems have depended on common softwares to cope with the attack. In this paper, we propose the positive selection mechanism and negative selection mechanism of T-cell, which is the biological distributed autonomous system, to develop the self/non-self recognition algorithm, the anomalous behavior detection algorithm, and AIS (Artificial Immune System) that is easy to be concrete on the artificial system. The proposed algorithm can cope with new intrusion as well as existing one to intrusion detection system in the network environment.

  • Macrophage Migration Inhibitory Factor (MIF) are well-defined role as unique cytokine and critical mediator in acute and chronic inflammatory diseases, autoimmune diseases. In this study, we isolated and characterized a full-length of MIF cDNA from the abalone (Haliotis discus hannai). The full-length cDNA of abMIF was of 1264 bp, consisting of a 5'-terminal UTR of 143 bp, an open reading frame of 360 bp and a 3-terminal UTR of 761 bp. The abalone MIF cDNA encodes a 119-amino acid polypeptide with a calculated molecular mass of 13.4 kDa and isoelectric point of 9.07. Multiple alignments and phylogenetic analysis with the deduced abalone MIF protein and showed strong homology with disk abalone (Haliotis discusdiscus). The deduced amino acid sequence of abMIF exhibited homology with other reported MIFs, such as 80%, with that of other disk abalone H. discus discus MIF gene. Quantitative real-time PCR (qRT-PCR) analysis indicated that abMIF was highly expression observed in hapatopacreas, intestine, foot, and gonad of normal conditioned abalone. Even though AbMIF mRNA level in hemocytes was low under the normal condition, it was sharply up-regulated and reached the maximum at 6 h post-infection with Vibrio parahaemolyticus, and then decreased at 24 h post-infection. This result indicates that abMIF plays an important role in responding in the innate immune system.

Effects of Silibinin and Resveratrol on Degradation of $I{\kappa}B$ and Translocation of NF-${\kappa}B$ p65 Induced by Tumor Necrosis Factor-${\alpha}$ in Cultured Airway Epithelial Cells (배양된 기도 상피세포에서 종양괴사인자에 의한 $I{\kappa}B$의 분해와 NF-${\kappa}B$ p65의 핵으로의 이동에 미치는 실리비닌과 레스베라트롤의 영향)

  • We examined whether silibinin and resveratrol affect airway mucin production, degradation of $I{\kappa}B$ and translocation of NF-${\kappa}B$ p65 induced by TNF-${\alpha}$ in NCI-H292 cells. Cells were pretreated with each agent for 30 min and then stimulated with TNF-${\alpha}$ for 24 h or the indicated periods. The two compounds suppressed TNF-${\alpha}$-induced airway mucin production, degradation of $I{\kappa}B$ and translocation of NF-${\kappa}B$ p65. This result suggests that silibinin and resveratrol can regulate the production of mucin induced by TNF-${\alpha}$ through the inactivation of NF-${\kappa}B$ pathway in airway epithelial cells.

Characterization of an Apple Polygalacturonase-inhibiting Protein (PGIP) from Apple Fruits (사과 과실로부터 분리한 polygalacturonase-inhibiting protein(PGIP)의 생화학적 특성)

  • An apple polygalacturonase-inhibiting protein (PGIP), that specifically inhibited endopolygalacturonase (PG, EC 3.2.1.15) from Botryosphaeria dothidea, was purified from B. dothidea infected apple (Malus domestica cv. Fuji) fruits. The apple PGIP was a mixed-type inhibitor of PG from B. dothidea. Optimal temperature for the maximum enzyme activity was $40^{\circ}C$, and optimum pH of the purified PGIP was pH 5.0. PGIP was stable up to temperature of $60^{\circ}C$ and was completely suppressed after heating at $70^{\circ}C$ for 10 min, PGIP was stable at pH between 4 and 8. Inhibition of PG by PGIP was reduced by $K^+$, $Cu^{2+}$, $Mg^{2+}$, $Ca^{2+}$ and $Zn^{2+}$ metal ion, sodium dodecyl sulfate (SDS) and 1,2-diaminocyclohexane tetra acetate (CDTA).
