Immunohistochemical study of the endocrine cells of the gastrointestinal mucosa of the African clawed toad, Xenopus laevis (아프리카 발톱두꺼비의 소화관내분비세포에 관한 면역조직화학적 연구)

  • The gastrointestinal endocrine cells of the African clawed toad, Xenopus laevis have been investigated immunohistochemically using the avidin-biotin method. Seven antisera were tested and three endocrine cell types immunoreacted with antisera to neurotensin, GRP and substance P. A moderate number of neurotensin-immunoreactive cells were weakly reacted in the small intestine. GRP-immunoreactive cells were mainly situated among the upper portion in the fundic glands, and the basal portion in the pyloric glands. These cells were oval and round in shape. On the other hand, in the intestine they were thin spindly cells with the epithelium. Substance P-immunoreactive cells were observed in among intestinal epithelium. However, no secretin-, motilin-, M-Enk- and PYY-immunoreactive cells were found in the GIT of the African clawed toads.

Calretinin-Containing Neurons in the Deeper Layers of the Hamster Superior Colliculus (햄스터 상구의 deeper layers에서 calretinin이 함유 신경세포)

  • Calcium-binding protein calretinin is thought to play important roles in calcium buffering. Recently, we reported on the distribution, morphology of calretinin-immunoreactive (IR) neurons and the effects of eye enucleation on the immunoreactivity of calretinin in the superficial layers of the hamster superior colliculus (SC). In the present study, we describe the distributions and types of labeled cells and effects of enucleation in the deeper layers by immunocytochemistry. We also compare this labeling to that of GABA, the major inhibitory neurotransmitter in the central nervous system. In contrast to the superficial layers, the deeper layers contained many calretinin-IR neurons which formed two tiers. The first tier, which was very distinctive, was found within the intermediate gray layer. The second tier was found in the deep gray layer. Labeled neurons varied dramatically in morphology and included vertical fusiform, stellate, round/oval, and horizontal neurons. In contrast to the superficial layers, enucleation appeared to have no effect on the distribution of calretinin immunoreactivity in the deeper layers. Two-color immunofluorescence revealed that none of calretinin-IR neurons were labeled with an antibody to GABA. The present results demonstrate that calretinin identifies unique neuronal sublaminar organizations in the hamster SC. The present results also demonstrate that none of the calretinin-IR neurons in the hamster SC is GABAergic interneurons. As many calretinin-IR cells are GABAergic interneurons in most other brain areas, this phenomenon in hamster SC is exceptional.

  • In this study, we carried out immunostaining and immunogold labeling with antibodies to serotonin and somatostatin to examine the characteristics and functions of the neurons that secrete neurotransmitters in optic lobes of Todarodes pacificus and Octopus minor. As a result of immunostaining with anti-somatostatin, the nerve cells of Todarodes pacificus reacted as similar to the anti-serotonin, but in Octopus minor, only large cells in the outer granule cell layer reacted positively. In the immunogold labeling with anti-serotonin, the nerve cells in the inner grande cell layer and medulla of Todarodes pacificus reacted strongly, 30 gold particles being labeled per $0.5{\mu}m^2$ of the cytoplasm. However, in Octopus minor, only 17 gold particles were labeled, which stated a weak reaction. On the other hand, in the anti-somatostatin case, the nerve cells in the outer and inner granule cell layers and medulla of Todarodes pacificus showed strong reaction, 30 gold particles being labeled per $0.5{\mu}m^2$ of the cytoplasm while the nerve cells in the outer granule cell layer of Octopus minor reacted weakly, about 3 gold particles being labeled per the equivalent area. As a result of immunostaining and immunogold labeling with two types of antibodies to each part of the optic lobes, we found that the reactive nerve cells were distributed differently in the two species. In particular, the degree of reactivity to the immunostaining and immunogold labeling appeared stronger in Todarodes pacificus than in Octopus minor.

Immunohistochemical study of the pancreatic endocrine cells in the ICR mice (ICR 마우스 췌장 내분비세포에 대한 면역조직화학적 연구)

  • The regional distribution and relative frequency of the pancreatic endocrine cells in the ICR mouse were studied by immunohistochemical (PAP) method using four types of specific antisera against insulin, glucagon, somatostatin and human pancreatic polypeptide (PP). The pancreas of mice could be divided into three portions; pancreatic islets, exocrine and pancreatic ducts. Pancreatic islets, furthermore, were subdivided into three regions (central, mantle and peripheral region) according to their located types of immunoreactive cells. In the pancreatic islet portions, insulin-immunoreactive cells were located in the central and mantle regions but most of somatostatin-, glucagon- and PP-immunoreactive cells were detected in the mantle and peripheral regions with various frequencies. In addition, PP-immunoreactive cells were also found in the central regions of pancreatic islets of ICR mouse. In the exocrine portions, all four types of immunoreactive cells were demonstrated in the ICR mouse. In the pancreatic duct portions, insulin- and glucagon-immunoreactive cells were situated in the epithelial lining of ICR mouse with a few and rare frequencies, respectively. In addition, rare PP-immunoreactive cells were also demonstrated in the subepithelial regions of the pancreatic duct. However, no somatostatin-immunoreactive cells were demonstrated.

  • 임신 10일부터 20일까지 흰쥐 태반형성과정에서 heat shock protein 27(HSP27) 분포를 면역조직화학적으로 조사하였다 임신 10일 태반에 자궁간막 탈락막세포. 영양막세포 및 거대영양막세포가 찰찰되었다. 이후 탈락막세포수는 감소하고 영양막세포는 증가하는데 특히 임신 14일에 현저하며 영양막은 해면영양막과 미로영양막으로 구분되었다 거대영양 막세포수의 변화는 현저하지 않았다. 자궁간막 탈락막에서 HSP27은 임신 10일 및 12일에 탈락막세포의 세포질 또는 핵에 반응이 나타나고 특히 핵에 반응하는 세포수가 많았으며 임신 14일 이후 탈락막 세포수 감소와 함께 반응이 나타나지 않았다 거대영양막세포는 임신 10일부터 일부 세포의 세포질 또는 핵에 반응이 관찰되나 임신 18일부터는 반응이 나타나지 않았다 영양막의 HSP27은 임신 10 및 12일에 일부 세포에서만 세포질 또는 핵에 반응이 관찰되나, 임신 14일부터 반응세포수 증가와 함께 대부분 핵에 반응을 나타내고 반응정도는 임신 16일 및 18일에 가장 현저하였으며, 미로영양막에서 반응세포수가 더 많았다.

An immunohistochemical study of endocrine cells in the gut of the Prussian carp, Carassius auratus (붕어(Carassius auratus) 장 내분비세포에 관한 면역조직화학적 연구)

  • The regional distribution and relative frequency of neurohormonal peptides-producing cells were demonstrated in the gut of the stomachless teleost, the Prussian carp, Carassius auratus Linnaeus, using 10 types of specific antisera raised against mammalian regulatory peptides. The gut of the Prussian carp was divided into five portions from proximal to distal (Segments I~V). Most of immunoreactive cells in the epithelial lining portion, between epithelial cells, were generally spherical or spindle shape having long cytoplasmic process that reached to the lumen (open typed cell) while cells showing round in shape (close typed cell) were found in the basal portions of epithelial lining occasionally. Somatostatin-, cholecystokinin (CCK)-8- and pancreatic polypeptide (PP)- immunoreactive cells were observed in this study. However, no serotonin-, glucagon-, chromogranin A-, secretin-, vasoactive intestinal peptide (VIP)-, substance P- and bombesin-immunoreactive cells were found. Somatostatin-immunoreactive cells were restricted to most proximal segments of the gut (Segment I) with rare frequency and CCK-8-immunoreactive cells were demonstrated in the proximal segments of the gut (Segments I and II) with a few to rare frequencies. In addition, pancreatic polypeptide-immunoreactive cells were demonstrated in the proximal to middle segments (Segments I~III) with moderated to rare frequencies. In conclusion, the distribution and relative frequency of these immunoreactive cells are well corresponded to the previous reports in stomachless teleost but somewhat peculiar patterns are also detected.

  • The regional distribution and relative frequency of endocrine cells in the GIT of the snakehead, Channa(Oph icephalus) argus were studied immunohistochemically. Five kinds of endocrine cells and one kind of nerve cell were identified in this study. A few numbers of secretin-immunoreactive cells were restricted to the cardia and fundus of the stomach. Moti-lin- and GRP-immunoreactive cells were rare in the epithelium of the distal and proximal intestines. PYY-immunoreactive cells were found in a few number in the pyloric caeca and were rare in the distal intestine. Substance P-immunoreactive cells were distributed relatively numerous from the fundus to the distal intetsine. Also, their nerve cells were detected in occurrence in the lamina propria in the distal intestine. No neurotensin-, met-Enk- and GIP-immunoreactive cells were found in the GIT of the snakehead.

  • The GEP endocrine cells of the African clawed toad, Xenopus laevis, were studied immunohistochemically. Five kinds of the endocrine cells were identified in this study A moderated number of 5-HT-immunoreactive cells were detected throughout the gastro intestinal tract, being almost uniform frequency. Gas/CCK -immunoreactive cells were restricted to the basal portion of the pyloric gland and among the duodenal mucosa. A rare glucagon-immunoreactive cells were weakly reacted in the fundic region of the stomach and observed in the exocrine portions of the pancreas. Somatostatine-immunoreactive cells were distributed throughout the gastrointestinal tract with except for the rectum, and not only the periphery of the islets but also the exocrine portions in the pancreas. No CGs- and insulin-immunoreactive cells were found in the gastrointestinal tract, whereas in the pancreas, the later was seen in the central region of the islets and the exocrine portions.

Tyrosine Hydroxylase - Immunoreactive Dopaminergic Neurons in the OX Retina (Ox retina내 tyrosine - hydroxylase 면역 반응되는 dopaminergic neuron에 대하여)

  • The morphology of dopaminergic neurons in the adult ox retina was studied. The dopaminergic neurons were identified using antibody immunocytochemistry. The great majority of tyrosine hydroxylase - immunoreactive neurons were located at the innermost border of the inner nuclear layer. The processes were monostratified and ran laterally within layer 1 of the inner plexiform layer. The second major population of tyrosine hydroxylase - immunoreactive neurons was displaced amacrine cells. The processes of displaced tyrosine hydroxylase - immunoreactive amacrine cells were also located within layer 1 of the inner plexiform layer. Some processes of a few neurons were located in the outer plexiform layer. A very low density of neurons had additional bands of tyrosine hydroxylase - immunoreactive processes in the middle and deep layers of the inner plexiform layer. The processes of dopaminergic neurons widely extended radially and formed large, moderately branched dendritic fields. These processes occasionally had varicosities but did not have "dendritic rings". These results indicate that dopaminergic cells make up specific neuronal cell types in the ox retina.

  • The regional distribution and relative frequencies of gastrointestinal endocrine cells were studied immunohistochemically in the GIT of the cat-shark. Five kinds of endocrine cells were identified in this study. Mostly these cells were of open types, except for glucagon-, BPP- and somatostatin-immunoreactive cells in the stomach which seemed to be of closed type. 5-HT-immunoreactive cells were detected throughout the GIT, and were more freguent than of the other regions. Glucagon-and BPP-immunoreactive cells were demonstrated in the entire GIT except for the duodenum an the rectum, and occurred in various frequencies. A few numbers of Gas/CCK-immunoreactive cells were restricted to the duodenum. Somatostatin-immunoreactive cells were distributed in the whole GIT, and were a few in numbers in the stomach and rare in the intestine, respectly. These results suggest that the pattern of the regional distribution is rather similar to that reported fur previously, but relative number was less frequent than that of other species.
