  • This study was designed to investigate the hypoglycemic effects of Benincasa hispida (Wax gourd) in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in the male rats by intravenous injection of STZ at a dose of 45 mg/kg dissolved in citrate buffer. The diabetic animals then had plasma glucose concentration of above 300mg/dl. The experimental groups were divided into five groups; normal, STZ-control and three Wax gourd groups (5%, 10% and 20% intake groups). Normal and STZ-control groups were fed on a AIN-93 diet and experimental groups were fed a AIN-93 diet with the Wax gourd powder (5%, 10% and 20%/kg diet) for 4 weeks. The body weight, diet intake and feed efficiency ratio (FER) were monitored. The blood glucose and cholesterol levels were determined everyweek. After 4 weeks, the rats were sacrificed and the levels of glucose, insulin, cholesterol, HDL-cholesterol, triglyceride and free fatty acids in plasma and levels of glycogen in liver and muscle were analyzed. Diabetic rats showed the lower weight gain compared to the normal rats. The weight gain and feed efficiency ratios in 15 and 20% Wax gourd groups were higher than in STZ-control group. The plasma glucose levels were significantly lower in all Wax gourd groups than in STZ-control group. The plasma insulin levels in diabetic groups were not significantly different compared to the normal group, but the level of 20% Wax gourd group was higher than other diabetic groups. The experimental diabetic groups showed the higher levels of muscle glycogen compared to STZ-control group. The lower levels of plasma cholesterol were noticed in 20% Wax gourd group throughout the experimental period. The plasma level of triglyceride was elevated in STZ-diabetic control and the levels were slightly decreased in Wax gourd groups. Rats of 10% Wax gourd group showed the lower levels of plasma free fatty acids. It is suggested, from the results, that the possibility of therapeutic or preventive use of wax gourd to the diabetes mellitus.

  • The effect of $Cd^2+$ on chlorophyll content, water potentials, ion transport, photosynthesis, stomatal apertures and $Cd^2+$ accumulation of organs in Commelina communis was investigated. 3-weeks old Commelina. communis was transferred to and grown in Hoagland solution in the presence or absence of 5 mM $Cd^2+$ for 4 days. $Cd^2+$ was accumulated in all parts of the organs including leaves, roots and stem. The proximity from the root and the age of leaf were significant factors responsible for the distribution of cadmium. Most of $Cd^2+$ was accumulated in the first leaf which was the nearest from the root. $Cd^2+$) accumulation in the leaves led to significant reductions in a series of physiological metabolism. $Cd^2+$ reduced total chlorophyll content up to 70%, and changed chlorophyll a/b ratio to 2. $Cd^2+$ also reduced about 20% of water potential. The treatment of $Cd^2+$ showed about 60% inhibition of photosynthetic activity when measured at various light intensity (100-1,000 $\mu$mol $Em^-2s^-1$). Similar effect was found in terms of stomatal conductance. Therefore, it could be concluded that the treatment of $Cd^2+$ decrease or block various physiological activities. • We studied oocyte steroidogenesis in the blackfin flounder Glyptocephalus stelleri as a region-specific species, in the East Sea of Korea during the spawning season. Maturing oocytes (0.76, 0.82, 0.88, and 0.91 mm in oocyte diameter) were incubated in vitro in the presence of [$^3H$] $17{\alpha}$-hydroxyprogesterone ($[^3H]17{\alpha}$-OHP) as a precursor. Steroid metabolites were extracted from the incubated medium and oocytes, and the extracts were separated and identified by thin-layer chromatography (TLC), high-performance liquid chromatography (HPLC) and gas chromatographymass spectrometry (GC/MS). The major metabolites produced from $[^3H]17{\alpha}$-OHP were androgens [androstenedione (A4) and testosterone (T)] and estrogens [$17{\beta}$-estradiol (E2) and estrone (E1)] and progestins [$17{\alpha},20{\alpha}$-dihydroxy-4-pregen-3-one ($17{\alpha}20{\alpha}P$) and $17{\alpha}20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$)] in maturing oocytes. The metabolic rate of $17{\alpha}20{\beta}$ was elevated (29.04%) in oocytes measuring 0.88 mm (nucleus migration stage following the induction of germinal vesicle breakdown), but was very low in oocytes measuring 0.76, 0.82, and 0.91 mm (0.42, 0.67, and 2.62%, respectively). From these results, we suggest that $17{\alpha}20{\beta}P$ acts as a maturation-inducing steroid in the blackfin flounder.

  • The purpose of this study was to investigate the effect of butanol (BuOH) fraction of Alisma canaliculatum (Ac) and/or selenium (Se) treatment on glycogen level, lipid metabolism and lipid peroxidation in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were assigned to one of the five groups: normal, STZ-control, and three experimental groups (Ac group, Ac-Se group, and Se group). Diabetes was experimentally induced by intravenous administration of 45 mg/kg of STZ in citrate buffer. The BuOH fraction of Ac (400 mg/kg bw) was orally administered for 3 weeks. The Se group were fed a AIN-93 recommended diet mixed with Na$_2$SeO$_3$ (2 mg/kg diet). The liver glycogen level of Ac and Ac-Se groups were significantly higher, when compared with the STZ-control groups. The muscle glycogen level was not significantly differ among all groups. The levels of liver triglyceride were higher in Ac-Se group than the STZ-control group. Pancreas protein levels were significantly increased in Ac-Se group than STZ-control group. The concentration of liver malondialdehyde (MDA) was significantly decreased in Ac and Se groups and decreased in Ac-Se group. Administration of BuOH fraction of Alisma canaliculatum and selenium supplementation increased the liver glycogen and triglyceride levels, and reduced peroxidative liver damage in STZ induced diabetic rats. These results suggest that treatment with a BuOH fraction of Alisma canaliculatum in combination with selenium has no synergistic antioxidative effect. Selenium supplementation may lead a decrease MDA of liver in diabetic rats.

  • The present experiments aimed to investigate the metabolism of calcium during oocyte maturation in rat. The concentration of free calcium and calmodulin in oocytes was measured respectively by using of fluo-3/AM and FITC with microscope fluorescence spectrometer. The ultrastructural localization of calcium precipitates in oocytes was observed with the transmission electron microscope. Cumulus-free immature oocytes(GV-oocyte) were cultured in vitro through 15 hours. The free calcium concentration in GV oocyte was $55.9{\pm}3.5nM$. In calcium-containing medium, the free calcium concentration was increased in germinal vesicle breakdown(GVBD) oocyte($64.2{\pm}7.3nM$). In normal medium after calcium chelator treatment ($10{\mu}M$ BAPTA/AM), the free calcium contents were slightly lower than those in control group. In calcium-free medium, the free calcium content was drastically increased in GVBD($72.7{\pm}3.4nM$) and metaphase I - anaphase I ($88.0{\pm}3.4nM$) oocyte. In maturation rate of oocytes, GVBD rate was high in control group($82.9{\pm}6.55%$) and calcium chelator treatment group($91.2{\pm}4.4%$), but in calcium-free medium group, it was low and then the oocyte was degenerated without polar body formation. Relative content of calmodulin in oocyte was significantly(P<0.001) increased in metaphase I - anaphase I than in GV and GVBD oocyte. The calcium precipitates were observed in mitochondria and cytoplasm of GV oocyte but that were not observed in mitochondria of GVBD and metaphase I - anaphase I oocyte. And then the calcium precipitates reappeared in mitochondria of metaphase II oocyte. The above results indicate that changes in free calcium and calmodulin concentration of oocyte occur according to the maturational stages and the extracellular calcium is required during oocyte maturation. Also change of calcium localization in oocyte occurs according to the maturational stages.

  • Changes in levels of phenolic compounds such as total phenols, flavonols and anthocyanins in Macrophoma-infected apples were studied at various developmental stages of apple fruits. The amounts of total phenols in apple fruit flesh and peel drastically decreased as apples became mature. Apple rot resulted in concentration of total phenols somewhat lower than those of healthy apple flesh. The decline in amount of total phenols was distinct in infected fruit flesh of the cultivar Fuji, which was more susceptible to Macrophoma sp. than the cultivar Miller. Higher amounts of total phenols were found in infected than in heathy fruit peel. In the case of the cultivar Miller, increased accumulation of total phenols was pronounced in infected peel. Apple rot resulted in concentrations of flavonols much higher than those of healthy apples. In particular, the drastically increased accumulation of flavonols was detected in infected peel at the first collection on 10 July, when the cultivars tested were completely resistant to Macrophoma sp. Production of anthocyanins was increased considerably by apple rot: anthocyanins in infected fruits of the cultivar Miller increased markedly as compared with their concentration from healthy fruits. These results suggest that the altered phenolic metabolism in apple fruits may be associated with the development of apple rot.

  • A rice OsLeuD gene for small subunit of 3-isopropylmalate isomerase (IPMI) (EC 4.2.1.33) has been isolated. OsLeuD gene is located on 109.3 cM of chromosome 2. OsLeuD gene was expressed abundantly in metabolically active organs including leaves and developing seeds, indicating that OsLeuD gene expression is developmentally regulated. The cDNA of OsLeuD gene was coded for 257 amino acids which showed 58% and 48% homology to small subunits of IPMI in OsLeuD genes of cyanobacteria and green sulfur bacteria, respectively. The molecular character of OsLeuD is closely related to those of photosynthetic bacteria rather than those of eukaryotes including fungi and yeast. This suggests that OsLeuD gene in chromosomal genome of plants may possibly be originated from chloroplast genome.

  • The changes in the quantitative distribution and in cytoplasmic granules of tongue mast cells and duodenal enterochromaffin cells in male albino rats were observed following oral administration of 40mg/kg body wt. isonicotinic acid hydraside (INH) and 20mg/kg body wt. pyridoxine. The results obtained are summarized as follows: 1. INH administered-rat showed a marked decrease in the number of mast cells, caused by leakage of cytoplasmic granules, while pyridoxine-rat showed increased the number of mast cells. 2. Similarly, INH-rat showed a marked decrease in the number of enterochromaffin cells. In the case of pyridoxine-rat, however, the number of enterochromaffin cells increased compared with that of the controls. 3. In view of the fact that a large dose of INH was harmful to the formation of mast cells and enterochromaffin cells. And considering that a moderate dose of pyridoxine stimulated the formation of the two kinds of cells and the amounts of cytoplasmic granules, it was concluded that pyridoxine might be concerned with the metabolism of secretory products, 5-Hydroxytryptamine.

  • In de novo biosynthesis of the extracellulor enzymes-proteinsaes, alpha and gluc-amylases during the synchronized differentiation of Aspergillus niger in submerged culture and surface liquid culture were investigated. Gluc-amylase was synthesized in the stage of presporulation in which phialide formation is involved. Proteinase was synthesized both in the stages of conidiophore formation and presporulation. Alpha-amylase was synthesized during presporulation and sporulation stages, the activity of enzyme lasted for seven days on surface liquid culture. It seemed that the synthesis was occured in de novo partly repressed by the catabolite, and its nature was found to be constitutive since it is produced in non-starch medium. Polyacrylamide gel electrophoresis have shown that presporulating and sporulating body produced diverse types of the proteins whereas the earlier stages of vegetative body showed simpler profiles. The uptake of C-14 uracil into RNA and C-14 glutamate into protein were shown to be vigorous in presporulating body rather than those in sporulating body. Coincidence of alpha-amylase biosynthesis in de novo and sporulation may be significant in the study of differentiation in which gene expression is involved.
