Ecotoxicity Evaluation of PFCs using Marine Invertebrate, Sea Urchin (Mesocentrotus nudus) (둥근성게(Mesocentrotus nudus)를 이용한 과불화화합물의 생태독성평가)

  • In this study, the toxic effects of PFOA and PFOS potassium salt on Mesocentrotus nudus using 10 min-fertilization rate and 48 h-normal embryogenesis were confirmed through the calculation of toxicity values such as Non-observed effective concentration, Low-observed effective concentration, and 50% of effective concentration. The case of 10 min-fertilization rate and 48 h-normal embryogenesis showed the concentration-dependent reduction pattern when exposed to PFOA and PFOS potassium salt, in tested concentration, respectively. The EC50 values of 10 min-fertilization rates for PFOA and PFOS potassium salt were 1346.43 mg/l and 536.18 mg/l, respectively, and the EC50 values of 48 h-normal embryogenesis were 42.67 mg/l and 17.81 mg/l, respectively. Both toxicity test methods showed high toxicity sensitivity to PFOS potassium salt. Recent studies have shown that the concentration of PFOA and PFOS in the marine environment has continuously decreased, and it is not enough to show acute toxicity to sea urchin. However, PFOA and PFOS have a very long half-life and can accumulate throughout the life of marine life, so it is still observed at a high concentration in shellfish. Therefore, a study on chronic toxicity through the whole-life cycle of marine organisms in coastal environments should be needed.

Spawning and Early Growth of the Sea Urchin, Strongylocentrotus intermedius (북쪽말똥성게, Strongylocentrotus intermedius의 산란유발 및 초기성장)

  • Spawning induction and early growth of the sea urchin, Strongylocentrotus intermedius were studied with the purpose of artificial seedling production. Gonadosomatic index(GSI) of the sea urchin showed the highest value in October, and rapidly decreased in December. It means that October and November is the peak of spawning season of the sea urchin in the latitude. Spawning induction by injection of potassium chloride solution in October has showed 44.0~100.0% reaction rate, and were produced 6,300$\times$10$^4$ eggs. Spawned eggs have shown the fertilization rate of 92.3~98.2% and the hatching rate of 78.2~87.0%. The metamorphosis of larvae after hatching in the seawater temperature of 13.7~17.1$^{\circ}C$ resulted in early eight-armed larvae in 13 days and late eight-armed larvae in 20 days. The collection of progenies was possible in 24~25 days after hatching and collection rate was 18.5~26.1% (mean 22.3%). Test diameter immediately after collection had a mean 350 ${\mu}{\textrm}{m}$. Survival rate and test diameter of juvenile sea urchin after collection were 58.5%, 1.32 mm in 30 days, 27.7%, 3.82 mm in 92 days and 15.6%, 11.70 mm in 181 days, respectively.

Toxic effects of antifouling agents (diuron and irgarol) on fertilization and normal embryogenesis rates in the sea urchin (Mesocentrotus nudus) (둥근성게(Mesocentrotus nudus)의 수정 및 배아 발생률에 미치는 신방오도료(Diuron, Irgarol)의 독성영향)

  • Toxic assessment of antifouling agents (diuron and irgarol) was conducted using the fertilization and the normal embryogenesis rates of the sea urchin, Mesocentrotus nudus. Bioassessment began with male and female reproductive cell induction. White or cream-colored male gametes(sperm) and yellow or orange-colored female gametes (eggs) were acquired and fully washed, separately. Then, the fertilization and normal embryogenesis rates were measured after 10 min and 48 h of exposure to the toxicants, respectively. The fertilization and embryo development rates were greater than 90% in the control, validating the suitability of both endpoints. The normal embryogenesis rates were significantly decreased with increasing concentrations of diuron and irgarol, but no changes in the fertilization rates were observed in concentrations ranging from 0 to 40 mg L-1. The EC50 values of diuron and irgarol for the normal embryogenesis rates were 20.07 mg L-1 and 22.45 mg L-1, respectively. The no observed effect concentrations (NOEC) were <1.25 mg L-1 and the lowest observed effect concentrations (LOEC) were 1.25 mg L-1 and 2.5 mg L-1, respectively. From these results, concentrations of diuron and irgarol over 1.25 mg L-1 and 2.5 mg L-1, respectively, can be considered to have toxic effects on invertebrates, including M. nudus. The ecotoxicological bioassay in this study using the noted fertilization and normal embryogenesis rates of M. nudus can be used as baseline data for the continued establishment of environmental quality standards for the effects of antifouling agents(especially diuron and irgarol) in a marine environment.

Toxicity Assessment of Ocean Dumping Wastes Using Fertilization and Embryo Development Rates in the Sea Urchin (Hemicentrotus pulcherrimus) (말똥성게 (Hemicentrotus pulcherrimus)의 수정 및 배 발생률을 이용한 해양배출 폐기물의 독성평가)

Spawning was induced by injecting 1 mL of 0.5 M KCl into coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. Experiments were began within 30 min the collection of both gametes. The fertilization and embryo development rates test were performed for 10 min and 64 h after fertilization, respectively. The fertilization and embryo development rates in the control condition (not including ocean dumping wastes sludge elutriate) were greater than 90%, but suddenly decreased with increasing of ocean dumping waste sludge elutriate concentrations. The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste ($EC_{50}$=4.37; $EC_{50}$=1.76), urban sewage ($EC_{50}$=5.79; $EC_{50}$=2.00) and food waste ($EC_{50}$=7.68; $EC_{50}$=2.16), respectively. The NOEC (<3.13) and LOEC (3.13) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest that biological assay using the fertilization and embryo development rates of H. pulcherrimus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

  • Effects of salinity and standard toxic metals on fertilization and embryo development rates were investigated in the sea urchin, Strongylocentrotus nudus. Spawning was induced by injecting 1ml of 0.5 M KCl into the coelomic cavity. The fertilization and embryo development rates were below 20% when salinity was 25 psu or lower, but were above 90% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition (not including Cu and Cd) were greater than 90%, but decreased with a high negative correlation (r) of 0.89 and 0.91 with the increasing of Cu and Cd concentrations, respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of S. nudus are between 30 and 35 psu, and the biological assays of fertilization and embryo development rates using S. nudus are useful methods for the ecological toxicity test of marine pollution elements.

Spawning was induced by injecting $1{\sim}2$ mL of 0.5 M KCl into the coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. The fertilization rate was below 30% when salinity was 20 psu and lower, but was almost above 90% when salinity was 25 psu and higher. The embryo development rate was below 60% when salinity was 25 psu and lower, but was above 80% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition(not including Cu and Cd) were greater than 90%, but decreased a high negative correlation with the increasing of Cu(r=-0.80, r=-0.78) and Cd(r=-0.90, r=-0.82) concentrations, respectively. The fertilization and embryo development rates were significantly inhibited in the addition of Cu($EC_{50}$=17.30 ppb, $EC_{50}$=10.32 ppb) and Cd($EC_{50}$=364.57 ppb, $EC_{50}$=244.04 ppb), respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of H. pulcherrimus are above 25 psu and 30 psu, respectively, and the biological assays of fertilization and embryo development rates using H. pulcherrimus are useful methods for the ecological toxicity test of marine pollution elements.

The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste($EC_{50}$=5.76; $EC_{50}$=4.53), urban sewage($EC_{50}$=9.82; $EC_{50}$=9.67) and food waste($EC_{50}$=3.90; $EC_{50}$=3.27), respectively. The NOEC(>3.13%) and LOEC(3.13%) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest biological assay using the fertilization and embryo development rates of S. nudus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.

Toxic effects of new anti-fouling agents (diuron and irgarol) on the embryogenesis and developmental delay of sea urchin, Hemicentrotus pulcherrimus (신규방오도료물질(Diuron, Irgarol)이 말똥성게(Hemicentrotus pulcherrimus)의 배아발생과 지연에 미치는 독성영향)

  • The aim of this study was to define the toxic effects of diuron and irgarol, which are new-antifouling agents, on the fertilization rate and normal embryogenesis rate in the sea urchin, Hemicentrotus pulcherrimus. In addition, the study was intended to confirm the hindrance of development in sea urchins. The fertilization rate of H. pulcherrimus was not decreased by the tested concentrations. However, the normal embryogenesis rate was decreased in a concentration-dependent manner. The 50% effective concentrations (EC50) of normal embryogenesis rate were 7.12 mg L-1 and 2.31 mg L-1, respectively. As the embryos developed into pluteus larvae, after 18 h of exposure to diuron and irgarol at EC50, development of the early gastrula stage was delayed, and significant developmental delays were observed after 24 h. After this, continuous developmental delays were observed in the process leading to the early gastrular, gastrular, early pluteus, and pluteus stages. Therefore, the toxic effects of diuron and irgarol on sea urchins were attributed to the delay in the developmental processes in the early life stages. Diuron and irgarol are highly persistent in the environment and have known-well toxic effects on various marine organisms including invertebrates, as shown in this study. Therefore, it is urgent to establish an environmental protection strategy to prevent the pollution of and preserve the marine environment.

Calmodulin of Olive Flounder Paralichthys olivaceus : Cloning and Expression Analysis

  • Calmodulin (CaM) is a $Ca^{2+}$-binding protein essential for biological functions mediated through $Ca^{2+}$-dependent mechanism. A cDNA clone for CaM was isolated from a cDNA library of olive flounder Paralichthys olivaceus. The CaM cDNA concists of 782 bp and encodes a polypeptide of 149 amino acids with four $Ca^{2+}$-binding motifs EF-hands (EF-I, EF-II, EF-III, and EF-IV). The deduced amino acid sequence of CaM shows 97-100% amino acid sequence identity to other CaM sequences. Semi-quantitative PCR analysis revealed that the CaM transcription was began during early development and the CaM mRNA is expressed highly in brain and intestine, and moderately in kidney, gill, and eye of healthy olive flounder. Taken together, CaM may be necessary for early olive flounder development and that it may have a part in homeostasis.
