Microbiological Quality of Fresh-Cut Produce and Organic Vegetables (신선편의 샐러드와 유기농 채소류의 미생물학적 품질 및 식중독 미생물 오염도)

  • This study was performed to assess the microbiological quality and potential health risk of fresh-cut produce and organic vegetables sampled from supermarkets and department stores in Korea. A total of 96 samples comprised three types of fresh-cut produce (sprouts, mixed-vegetables, fruit) and three types of organic vegetables (lettuce, perilla leaf, green pepper). The samples were analyzed for total viable cell counts, coliforms, Enterobacteriaceae, Escherichia coli, Salmonella spp., Listeria monocytogenes, Vibrio parahaemolyticus, Bacillus cereus, and Staphylococcus aureus. The microbiological counts of fruit were very low. Sprouts were highly contaminated by total viable cell counts ($8.3{\pm}0.57$ log CFU/g), Enterobacteriaceae ($7.1{\pm}0.76$ log CFU/g), and coliforms ($4.9{\pm}0.40$ log MPN/g), and showed a high incidence level of B. cereus ($2.9{\pm}0.48$ log CFU/g). Of the fresh-cut produce analyzed, six (13.6%) mixed-vegetable salads were E. coli positive. S. aureus was detected in only one sprout sample and one mixed-vegetable salad, and its contamination levels were under 2 log CFU/g, which is appropriate for Korean standards (<3 log CFU/g) of fresh-cut produce. Of the organic vegetables, lettuces were highly contaminated by total viable cell counts ($6.4{\pm}0.74$ log CFU/g), Enterobacteriaceae ($5.7{\pm}0.98$ log CFU/g), and coliforms ($3.7{\pm}1.72$ log MPN/g). Two (13.6%) organic lettuce and one (7.1%) perillar leaf sample were E. coli positive, and S. aureus was detected in one lettuce and two perilla leaf samples. Salmonella spp., Vibrio parahaemolyticus, and Listeria monocytogenes were not detected in any of the fresh-cut produce or organic vegetables analyzed.

Changes in Quality Characteristics of Wild Root Vegetables during Storage (전처리 근채류의 저장과정중의 품질평가)

  • During the peeling, cutting, and shredding of wild root vegetables, the surface is exposed to air. This results in a rapid deterioration in quality and an increase in the potential of contamination by microorganisms, both of which reduce the shelf-life of wild root vegetables in retail markets. Thus, in this study, the effects of various washing treatments on the quality of wild root vegetables, including lotus root, burdock root, and bellflower root, were investigated at 10 and $24^{\circ}C$. Lotus root, burdock root and bellflower root were washed with 0.2% acetic acid (AA), 0.2% citric acid (CA), 500 ppm acidified sodium chlorite (ASC), and tap water (TW), which was used as a control, and stored at $10^{\circ}C$ and $24^{\circ}C$. The changes in total plate counts, coliform groups, polyphenol oxidase (PPO) activity, color, pH, and exterior appearance of the samples were then evaluated. The pH and initial microbial contamination levels were reduced when the root vegetables were washed with AA, CA, and ASC. In particular, initial population levels of total plate counts and coliform groups were not detected in lotus root and burdock root that had been washed with ASC and their growth was significantly (p<0.05) inhibited during storage at 10 and $24^{\circ}C$ when compared to the control (TW). In addition, the polyphenol oxidase (PPO) activities of the root vegetables washed with AA, CA and ASC were lower than that of root vegetables washed with TW. ASC was determined to be the most effective treatment for preventing microbial growth, tissue softening, and the development of browning and an unpleasant smell. At $10^{\circ}C$, the overall qualities of the wild root vegetables were maintained longer when compared to $24^{\circ}C$.

The Diagnosis of Pneumoniae Following Bone Marrow Transplantation by Bronchoscopy (골수이식후 기관지내시경을 이용한 폐렴의 진단)

  • Background : Pulmonary complications following bonemarrow transplantation (BMT) are common and associated with a high mortality rate. We investigated the yield, safety, and impact of fiberoptic bronchoscopy (FOB) for diagnosis of postBMT pneumoniae. Methods : From May 1997 to April 2000, 56 FOBs were performed in 52 post BMT patients for clinical pneumoniae. BMT patients with respiratory symptoms and/or pulmonary infiltrates had a thoracic HRCT(high resolution computed tomography) and bronchoscopic examination including BAL (bronchoalveolar lavage), TBLB (transbronchial lung biopsy), PSB (protected specimen brush). Results : The characteristics of the subjects were as follows : 37 males, 15 females, mean age of 31.3 years(l7-45), 35 sibling donor allogenic BMTs, 15 nonrelated donor allogenic BMTs, and 2 autologous BMTs. Fiftynine percent of FOBs (33 FOBs, 31 patients) were diagnostic. Isolated pathogens included the following : 12 cytomegalovirus (CMV) (21.4 %), 7 pneumocystis carinii (PC) (12.5 %), 11 CMV with PC (19.6 %), 2 Mycobacaterium tuberculosis (3.6%), and 1 streptococcus (1.8%). Most of the radiographic findings were diffuse interstitial lesions. CMV pneumoniae had mainly diffuse interstitial nodular lesion, and PC pneumoniae had diffuse, interstitial ground glass opacity(GGO). When CMV was accompanied by PC, a combined pattern of nodular and GGO was present. Of the 56 cases (23.2%), 13 died of CMV pneumoniae (n=2), PCP (n=2),mixed infection with CMV and PC (n=3), underlying GVHD (n=1), underlying leukemia progression (n=1), or respiratory failure of unknown origin (n=4). There was no major complication by bronchoscopy. Only 3 cases developed minor bleeding and 1 episode temporary hypoxemia. Conclusion : Based on our findings, CMV and PC are the major causes of postBMT pneumoniae. In addition, BAL can be considered a safe and accurate procedure for the evaluation of pulmonary complications after BMT.

  • The Seosanmaaesamjonbulsang (national treasure 84) is carved has a quite developed tectonic line in a precipitous wall of stratified rock to the vertical and horizontal directions. The main buddha and the left part of it have got the biological weathering and the efflorescence has been detected on the shoulder part of the right buddha. It has been caused by the raindrops from upper rock after the dismantling of the protection shelter in 2007. Two proper measures have been taken in order to reduce the weathering and protect it from some damages. The one is removing the contaminant by dry/wet cleaning not to get any damage on the rock. The other is the isolation by the construction of a new waterway used L-30 on the upper part of the rock not to contaminate the surface of Rock-carved triad buddha with the raindrops from upper rock. Moreover, the sunlight for buddha has been increased and good ventilation has been made by cutting down the trees around the Rock-carved triad buddha.

  • Extraction and anti-solvent crystallization were proposed to recover octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine (HMX) from pressed polymer bonded explosives(PBXs). DXC-57 and DXC-59, whose polymeric binders are Estane and HyTemp with dioctyl adipate plasticizer, respectively, were used as pressed PBX models. Estane of DXC-57 was removed by washing with tetrahydrofuran prior to extraction, which enabled the crystallization of HMX at a low degree of supersaturation, sufficient to obtain β-form HMX. Using dimethyl sulfoxide and ethanol as the extraction solvent and the anti-solvent for crystallization, respectively, HyTemp and dioctyl adipate in DXC-59 were separated from HMX. The purity of recovered β-form HMX was higher than 99 %.

스퍼터링 소스를 이용하여 빗각 증착되어진 TiN 박막의 형상 및 특성 연구

  • 빗각 증착이란 입사 증기가 기판에 수직하게 입사하는 일반적인 공정과는 다르게 증기가 기판의 수직선과 $0^{\circ}$이상의 각을 갖는 증착 방법을 의미한다. 본 연구는 공정 압력이 비교적 높은 스퍼터링 공정에서 빗각 증착을 실시하여 코팅층의 구조제어가 가능한지를 확인하였다. 본 연구에서는 조직의 치밀도 향상을 통한 특성 향상을 위해 TiN 박막을 제조함에 있어서 빗각 증착 기술을 응용하여 단층 및 다층 피막을 제조하고 그 특성을 비교하였다. 스퍼터 소스에 장착된 타겟의 크기는 6"이며, 99.5% Ti 타겟을 사용하였고, Ar 가스 분위기에서 기판으로 사용된 Si(100) 위에 코팅하였다. 기판과 타겟 간의 거리는 10 cm이며, 기판은 알코올과 아세톤으로 초음파 세척을 실시한 후 진공챔버에 장착하고 < $2.0{\times}10-5Torr$ 까지 진공배기를 실시하였다. 진공챔버가 기본 압력까지 배기되면 Ar 가스를 주입한 후 RF 파워에 약 300V의 전압을 인가하여 글로우 방전을 발생시키고 약 30분간 청정을 실시하였다. 기판의 청정이 끝난 후 다시 < $2.0{\times}10-5Torr$까지 진공배기를 한 후 Ar 가스를 주입하여 TiN 코팅을 실시하였다. 빗각 증착을 위한 기판의 회전각은 $70^{\circ}$, $80^{\circ}$$-70^{\circ}$, $-80^{\circ}$이며, TiN 박막의 총 두께는 약 $3.5{\sim}4{\mu}m$로 유지하였다. 스퍼터링을 이용한 TiN 박막의 빗각 증착 코팅을 실시하였으며, 공정조건에 따라 주상정이 자라는 모습과 기울어진 각도가 다른 구조를 갖는 박막이 제조되는 것을 확인할 수 있었다. 빗각증착을 실시하는 중에 기판 홀더에 약 -100 V의 전압을 인가하면 인가하지 않은 막에 비해 치밀한 박막이 성장한다는 사실을 확인하였다. 박막의 성능향상을 위하여 스퍼터 시스템에서 빗각 증착을 이용한 TiN 박막 형성을 실시하였다. SEM 단면 이미지에서 확인해본 결과 주상정이 자라는 형상이 공정 압력이 5 mTorr에서 2 mTorr로 낮아짐에 따라 상대적으로 치밀하면서 일정한 형태로 성장하는 것을 확인하였다. 본 연구를 통해 스퍼터링을 이용한 빗각 증착의 Structure Engineering 이 가능함을 확인하였으며 박막의 성능을 향상시키는 기술로서 응용 가능할 것으로 보인다.

Detergency improvement of hydrophilic soils in dry cleaning process (드라이클리닝 시 친수성 오구의 세탁성 향상을 위한 연구)

  • The dry cleaning effect related to the type of soil and fiber was analyzed using silicone and alcohol-based solvents(ethyl alcohol, isopropyl alcohol, acetone) that are relatively safe for the human body and environment to improve the detergency of hydrophilic soils in dry cleaning system. Based on this analysis, an effective dry cleaning method to be carried out for improving the detergency according to the type of hydrophilic soils. After dry cleaning was performed using 20 types of artificial soiled fabrics consisting of 7 types of fibers and 4 types of hydrophilic soils, the detergency was measured and the results were compared and analyzed by solvents and fiber types. The results are presented as follows; first, the detergency of hydrophilic soils using silicone solvents showed a low rate of detergency. In particular, the tannin soil showed a lower level of detergency compared to the protein soil. Second, the detergency of hydrophilic soils using silicon solvents with dry soap differed in some detergency according to the soil and fiber types. Especially, the detergency of curry soil on cotton fabric showed significant improvement. Third, the protein soil was not removed from dry cleaning using alcohol-based solvents, but the effect of dry cleaning of curry soil on both cotton and polyester fabric was substantially improved. As a result, the elimination of blood soil is more effective in silicon solvents than in alcohol-based solvents. The removal of tannin soils may improve detergency by adding dry soap to silicon solvents or by using alcohol-based solvents as alternative solvents. The use of alternative solvents such as silicon and alcohol solvents can contribute to the environmental improvement of the dry cleaning industry, which uses petroleum-based solvents. It is also expected to provide consumers with the opportunity to choose eco-friendly and efficient dry cleaning methods.

  • Laboratory experiments were conducted to the potential ability of bioremediation with bentazon such as determining the absorption, translocation, and metabolism of $^{14}C-Bentazon$ in minari after foliar applications. The absorption and translocation of $^{14}C-bentazon$ were compared when applied to foliar of minari. In foliar applications, 21% was observed in treated leaves, 66% remained in water extracts of leaf surfaces, and 13% was found in the epicuticular wax layer after 2d. Translocation of the herbicide from treated leaves to roots was very low(79 to 9%). Analysis of methanol-soluble extracts of $^{14}C$ indicated that more than 60% of the foliarapplied herbicide was metabolized in all plant sections after 2d. However, 77% or more of the bentazon was degraded in roots and shoots 2d after root absorption. The major metabolite in these experiments was an unknown compound that was less polar than bentazon and 6- and 8-hydroxy bentazon.

Skin and Eye Irritation Test of Bovine Somatotrophine-sustained Release (BST-SR) in Rabbits (토끼에게 소성장 홀몬-서방형 제형(BST-SR)의 피부 및 안점막 자극에 관한 연구)

  • According to the Established Regulations of National Institute of Safety Research, the skin-irritation test of BST-SR (Lucky Ltd.) was perfonned for seven days in New Zealand White Rabbits. During treatment periods no significant clinical symptom was observed. Significant changes such as erythema, scar tissue and edema were not shown on the applicating sites. According to Primary Irritation Index of Draize, skin irritation rate was assessed as "Zero". The eye irritation of BST-SR (Lucky Ltd.) was examined in nine New Zealand White rabbits, based on the Established Regulation of National Institute of Safety Research. 0.1 ml of test material was dropped on right eye, and after 20~30 seconds, three rabbits' eyes were cleaned with wann saline for 1 minute. Other six rabbits' eyes were left uncleaned. The untreated left eyes were negative control. The lesions of cornea, iris and conjuntiva were assessed by the Grade table (from the Regulation of National Institute of Safety Research), and it is concluded that BST-SR has no eye-irritation.

패턴 된 기판 위에 형성된 메조포러스 $TiO_2$막 형성 기구 및 미세구조 연구

  • 고효율 염료감응형 태양전지(DSSC, Dye-Sensitized Solar Cell)의 구현을 위해서 유용한 방법중 하나는 정렬된 기공 (pore)을 $TiO_2$막 내에 형성시키는 것이다. 메조포러스 (mesoporous) $TiO_2$막은 dip coating이나 spin coating과 같은 방법으로 주로 증착되고 있으며, P123이나 F127과 같은 amphiphilic triblock copolymer를 메조포러스 구조를 만들기 위한 뼈대로 사용하고 있다. 또한, 이렇게 생성된 구조에서 amphiphilic triblock copolymer는 열처리 공정을 통하여 쉽게 제거될 수 있다. 고효율 태양전지를 구현하는 또 다른 방법으로는 패턴 된 기판을 사용하는 것이다. 패턴 된 기판은 빛의 반사를 억제하여 흡수율을 높이는 역할을 한다. 그러나 패턴 된 기판 위에서 메조포러스 $TiO_2$막의 형성에 관한 연구는 부족한 실정이다. 본 연구에서는 spin coating 방법으로 패턴 된 Si (111) 기판 위에 메조포러스 $TiO_2$를 성장하고 그 미세구조를 분석하였다. 패턴 된 기판은 nanosphere lithography(NSL) 법으로 mask를 증착한 후 건식 식각 (dry etching) 공정을 통해서 제작되었으며, 마스크와 불순물 등 은 초음파 세척 등으로 제거되었다. 메조포러스 $TiO_2$막은 1-propanol, P123, titanium isopropoxide와 HCl을 섞어 만든 용액으로 1 cm${\times}$1 cm 기판 위에 3000 rpm과 4000 rpm으로 각각 증착하였으며, 5일 동안 4도에서 에이징한 후 350도에서 3시간 열처리하였다. 이렇게 형성한 메조포러스 막의 형상과 미세구조적 특성이 주사전자현미경(SEM, scanning electron microscope), X-선 회절(XRD, X-ray diffraction) 등을 이용하여 연구되었다. 특히, 증착 조건에 따른 메조포러스 $TiO_2$박막의 형성 기구에 관한 고찰이 진행되었다. 나아가, $TiO_2$박막과 패턴 사이에 형성되는 계면 구조에 관한 연구를 투과전자현미경을 이용하여 진행하였다.
