Phytoremediation of Soils Contaminated with Heavy Metal by Hosta longipes in Urban Shade (도시 내 음지 중금속 오염지에 대한 비비추의 식물정화 효과)

  • Hosta longipes is one of the most popular ornamental perennials in use in Korea today, and is mainly used as a groundcover plant in urban shaded places. In this study, the pytoremediation effect of Hosta longipes was tested using four concentrations (Control, 100, 250 and $500mg{\cdot}kg^{-1}$) of Cd, Pb and Zn in soil. The plants were planted in $300mm{\times}200mm{\times}250mm$ drainless-containers, which were filled with a artificial amended soil for 7 months. The results showed that the contents of heavy metals cadmium and lead in the shoot of Hosta longipes increased with increasing heavy metal concentration levels exception of zinc. The amount of zinc, cadmium, and lead accumulated in roots were increased with heavy metal concentration levels up in soil. The shoot/root ratios(TF; translocation factor) values were found to be more than 80% of total Zn, Cd, and Pb take up by Hosta longipes. These results indicated that root is the major part for accumulation of heavy metal. The removal contents of zinc, cadmium, and lead increased significantly with the increasing heavy metal concentration in the soil, which was planted with Hosta longipes. The heavy metal concentration accumulation in plant/soil ratios (BF; bioaccumulation factor) values for three metals were found to be more than 30% in cadmium and lead, but lower in zinc treatments. The different responses of Hosta longipes suggest that in heavy metal contaminated soils the plant adsorbs available metals depending on the concentration soils in which they are present. Therefore, this species can be an efficient phytoremediator for soils contaminated with cadmium and lead in urban shaded places.

  • Aluminum metal process in surface micromachining enables to fabricate Al electrodes or Al structures, which improve electrical characteristics by reducing contact- and line-resistance or makes the whole process to be simple by using oxide as sacrificial layer. However, it is not possible to use conventional sacrificial layer etching process, because HF solution attacks aluminum as well as sacrificial oxide. The mixed solution of BHF and glycerine as an alternative shows the adequate properties to meet with this end. The exact etching properties, however, are sensitively depends on the geometry of the released structure, because the most etching process of sacrificial layer proceeds to the lateral direction in narrow space. Also, the surface roughness of aluminum affects to the etching characteristics. This paper reports experimental results on the effect of microstructure and surface roughness of aluminum to the etching properties. Considering these effects, we propose the optimized etching condition, which can be used practically for the fabrication of aluminum electrodes and microstructures by using standard surface micromachining process without modification or additional process.

  • The Mireuksaji stone pagoda in Iksan is the largest stone pagoda existing in Eastern Asia. It was assumed that originally it had been established in the shape of nine-storied pagoda but as time went by only six-storied pagoda remained partially due to collapsing, repair and reconstruction. According to the reference, we can't make sure when its modification happened. The form that the pagoda is having now, was modified with concrete by the Japanese during the 1910s. The materials mixed in concrete were mixture of Portland cement, all sorts of stone, sand, and a little bit of new building stone, additive and compound. And also these materials were applied to cultural assets without any experiment at the time of 1910s as maintenance, which are still used recently. To prevent the change of its shape, the west side, south side and the north side which is partially destructed was rebuilt and reinforced with concrete and some of the deformed parts were also filled. The amounts of concrete used were about 200 tons. Such method had prevented the pagoda from destructing, however, by choosing a wrong repair method, its surface of the stone has secondary contaminants and precipitation caused by concrete. This kind of contamination speeds up the weathering which accelerates the aging mechanism of the stone to make it even harder to revive the absence of historical nature. Therefore, we are to find the best cleaning method to remove the secondary hazard contaminants.

Homology Modeling and Active Sites of PolyMG-specific Alginate Lyase from Stenotrophomonas maltophilia KJ-2 (Stenotrophomonas maltophilia KJ-2 균주로부터 얻은 PolyMG-specific 알긴산분해효소의 상동성 모델링 및 활성자리 연구)

  • Alginates are linear acidic polysaccharides composed with (1-4)-linked ${\alpha}$-L-guluronic acid and ${\beta}$-Dmannuronic acid. Alginate can be degraded by diverse alginate lyases, which cleave the alginate using a ${\beta}$-elimination reaction and produce unsaturated uronate oligomers. A gene for a polyMG-specific alginate lyase possessing a novel structure was previously identified and cloned from Stenotrophomonas maltophilia KJ-2. Homology modeling of KJ-2 polyMG-specific alginate lyase showed it belongs to the PL6 family, whereas three Azotobacter vinelandii polyMG lyases belong to the PL7 family of polysaccharide lyases. From $^1H$-NMR spectra data, KJ-2 polyMG lyase preferably degraded the M-${\beta}$(1-4)-G glycosidic bond than the G-${\alpha}$(1-4)-M glycosidic bond. Seventeen mutants were made by site-directed mutagenesis, and alginate lyase activity was analyzed. Lys220Ala, Arg241Ala, Arg241Lys, and Arg265Ala lost alginate lyase activity completely. Arg155Ala, Gly303Glu, and Tyr304Phe also lost the activity by 60.7-80.1%. These results show that Arg155, Lys220, Arg241, Arg265, Gly303, and Tyr304 are important residues for catalytic activity and substrate binding.

Rapid Gas Chromatographic Screening of Dairy Products for Volatile and Nonvolatile Organic Acids (기체크로마토그래피법에 의한 유제품내 휘발성 및 비휘발성 유기산의 신속한 스크리닝)

  • A rapid gas chromatographic (GC) profiling method was applied to dairy products (milk and cheeses) for the simultaneous analysis of volatile and nonvolatile organic acids. Cheese samples were first made into aqueous samples by dissolving in water. The aqueous samples were then extracted with organic solvents after the acidification and NaCI saturation. The organic layers (diethyl ether : hexane= 1 : 1) were extracted with $NaHCO_3$ saturated solution with subsequent solid-phase extraction of the aqueous phases using Chromosorb P column/diethyl ether followed by triethylamine treatment. The resulting triethylammonium salts of acids were directly converted into volatile tert.-butyldimethylsilyl derivatives, which were analyzed by dual-capillary column GC, and GC-mass spectrometry. From milk and four cheese samples studied, 31 organic acids including 21 fatty acids and other hydroxy and dioic acids were tentatively identified. The amounts of the fatty acids were different among the kinds of cheese and thus the simplified retention index (RI) spectra of organic acids were useful for the visual pattern recognition of each sample, when the Direct Comparision method between cheese and a blind sample were attempted, it was quickly recognized to be a gouda cheese with the 999 ppt match quality value.

  • A Corynebacterium diphtheriae iron-repressible gene dirA, that was homologous to TSA of Saccharomyces cerevisiae and AhpC subunit of Salmonella typhimurium alkyl hydroperoxide reductase, was amplified with PCR and expressed in E. coli. The DirA purified from the transformed E. coli crude extracts prevented the inactivation of enzyme caused by metal-catalyzed oxidation (MCO) system containing thiols but not by ascorbate/Fe$^{3+}$/$O_2$ MCO system. The DirA concentration, which inhibited the inactivation of glutamine synthetase by 50% (IC$_{50}$) against MCO system, was 0.12 mg/ml. The multimeric forms of DirA were converted to the monomeric form in SDS-PAGE under the thioredoxin system comprised of NADPH, Saccharomyces cerevisiae thioredoxin reductase, and thioredoxin. Also, DirA showed thioredoxin dependent peroxidase activity. All of these results were consistent with the characteristics of a thiol specific antioxidant (TSA) protein having two conserved cysteine residues.

  • Street Trees is an important object for urban environment improvement. Especially the height of the trees needs to be precisely measured as a factor that greatly influences the removal of air pollutants in the Urban Street Canyons. In this study, we extracted the height of the tree based on the stereo image using the precisely adjusted UAV Images of the target area. The adjustment of UAV image was applied photogrammetric SfM (Structure from motion) based on the collinear condition. We measured the height of the trees on the Street Canyon using stereoscopic vision on stereo plotting system. We also acquired the height of the building adjacent to the street trees and the average height of the road surface was calculated for accurate measurement of the height of each object. Through the visual analysis with the plotting operation system, it was possible to measure height of the tree and to calculate the relative height difference value with building quickly. This means that the height of buildings and trees can be calculated without making a 3D point cloud of UAV and it has the advantage of being able to utilize non-experts. In the future, further studies for semiautomatic/automation of this technique should be performed. The development and research of these technologies is expected to help to understand the current status of environmental policies and roadside trees in urban areas.

Effect of pH and Temperature on the Adsorption of Heavy Metals in Acid Mine Drainage (AMD) Onto Coal Mine Drainage Sludge (CMDS) (탄광슬러지를 이용한 금속광산 산성배수 처리 시 pH및 온도의 영향)

  • In this study, the effect of pH and temperature on the adsorption behavior of acid mine drainage (AMD) on coal mine drainage sludge (CMDS) has been investigated during the treatment of coal mine drainage (CMD) by electrical purification method. The pH$_{zero\;point\;charge}$ (pH$_{zpc}$) of CMDS was 5. The removal ratio of copper, zinc, cadmium, iron were increased according to the increase of pH value. The adsorption amount of copper showed 0.64 mg g$^{-1}$ sludge. It was independent of pH value. The adsorption amount of the other metals showed l.l times when pH was 3. The adsorption amount of chromium was a little bit increased at the pH value higher than 7 due to a small amount of the chromium was eluted as $Cr(OH)_6^{3-}$. The amount of metals' absorption were decreased according to temperature was increase at pH value was 3. The selectivity order was Cd>Fe > Zn > Cu. The amount of absorption showed q$_{max}$ Cu 2.747 mg g$^{-1}$ andZn 2.525 mg g$^{-1}$ when pH value higher than 5. It was independent of temperature.

  • 하천유량자료는 이수, 치수, 수질관리 등의 목적으로 널리 사용되기 때문에 여러 가지 수문관측 자료 중 가장 중요하다고 할 수 있다. 그러나 우리나라의 유량자료는 여러 가지 한계를 가지고 있어서 수문자료로서 제대로 사용되지 못하고 있는 실정이다. 특히 홍수기 부자측정 방법에 의해 산정된 유량자료는 측정 여건, 방법, 기기 등의 한계로 인해 그 정확도가 더욱 낮다. 홍수기 부자측정 방법에 의한 유량자료의 정확도 향상을 위해서는 현장 유량측정의 정확도를 향상시키는 것이 일차적으로 필요하지만, 측정된 자료를 과학적이고 체계적인 계산과정을 통하여 유량으로 환산하는 것도 매우 중요하다. 국내의 경우 일반적으로 여름에 집중호우가 빈발하고 경사가 급한 산지하천이 많다. 그래서 홍수시 하천의 유속이 매우 빠르고 하천수내에 부유물이 많이 함유되어 있다. 이러한 요인들로 의해 대부분 홍수시 유속계를 이용한 유량측정이 불가하여 대안으로 부자를 이용하여 측정하고 있다. 그 결과 평저수시 유속계 이용시에 비해 측정 및 산정과정에서 매우 큰 오차가 발생하고 있다. 이와 같이 국내의 경우 홍수시 유량측정을 위해 부자에 전적으로 의존하는 현실임에 불구하고 부자를 이용한 유속측정 및 유량산정에 대한 연구는 매우 미흡하였다. 외국의 경우도 부자 측정에 대한 방법론이 ISO 748과 일본수문관측에 간략하게 설명되어 있고 USGS와 WMO에서는 거의 내용을 다루고 있지 않고 있다. 현재 우리나라의 경우는 ISO 748을 일부 참조하고 대부분 일본수문관측 기준에 준해 측정을 하고 있다. 자연하천임을 감안하면, 부자에 의한 유속 측정시 발생할 수 있는 여러 오차들의 경우 적절한 구간의 선택, 충분한 측선수의 확보 등과 같은 측정기준의 개선을 통하여 상당부분 제거가 가능하다. 그러나 부자를 이용해 측정된 성과를 신뢰도 높은 유량으로 산정하기 위해서는 정확한 측정과 더불어 과학적이고 표준화된 유량산정 기준과 절차가 필요하다. 본 연구에서 분석된 결과에 의하면 부자유선 모임, 홍수터 유속 미측정, 기준 흘수 미적용 등과 같은 측정 자체의 문제점을 제외하면, 부자측정 방법에 의한 유량산정시 가장 큰 오차원인은 홍수시 측정된 유속측선의 위치와 홍수 전후로 측정된 횡단면상의 위치가 일치하지 않는 점과, 대부분 두 측정 구간의 평균값을 대푯값으로 사용한다는 점이다. 본 연구는 다년간의 유량 측정 및 검증 경험과 자료를 토대로 현장에서 부자를 이용하여 측정된 측정성과를 정확도 높은 유량자료로 산정하는데 있어서의 문제점을 도출하고, 이로 인해 발생하는 오차를 추정하여 그 개선방안을 제시해 보고자한다. 더불어 보다 정확한 유량 산정을 위한 기준과 범주를 제시하고자 한다.

A screening of Alzheimer's disease using basis synthesis by singular value decomposition from Raman spectra of platelet (혈소판 라만 스펙트럼에서 특이값 분해에 의한 기저 합성을 통한 알츠하이머병 검출)

  • In this paper, we proposed a method to screening of Alzheimer's disease (AD) from Raman spectra of platelet with synthesis of basis spectra using singular value decomposition (SVD). Raman spectra of platelet from AD transgenic mice are preprocessed with denoising, removal background and normalization method. The column vectors of each data matrix consist of Raman spectrum of AD and normal (NR). The matrix is factorized using SVD algorithm and then the basis spectra of AD and NR are determined by 12 column vectors of each matrix. The classification process is completed by select the class that minimized the root-mean-square error between the validation spectrum and the linear synthesized spectrum of the basis spectra. According to the experiments involving 278 Raman spectra, the proposed method gave about 97.6% classification rate, which is better performance about 6.1% than multi-layer perceptron (MLP) with extracted features using principle components analysis (PCA). The results show that the basis spectra using SVD is well suited for the diagnosis of AD by Raman spectra from platelet.
