  • Pollen microspores isolated from peony anthers were cultured by agarose embedding method in the MS medium with 2,4-D(1mg/l) or phenylacetic acid(1, 10, 100mg/l), and without plant hormone. It was observed that pollen microspores cultured on hormone-free medium were directly developed into embryos. Callus formation was enhanced from microspores which were cultured on medium supplemented with 1mg/l PAA. Embryos were also formed from the calli transferred into the hormone-free medium.

  • Regeneration and growth of bulblets from bulblet-derived bulb-scale segment of Lilium longiflorum (cv Georgia) were investigated. Bulblets were initiated on bulb scales taken from bulblets on MS medium containing 0.05 mg/L 2,4D with 3% sucrose or 0.02 mg/L 2,4D with 9% sucrose. Benzyladenine promoted the differentiation of bulblets but inhibited the growth of differentiating bulblets. The growth of bulblet was promoted by supplying 1/2 strength 1/2 NH$_4$NO$_3$ concentration in MS medium containing 12% sucrose.

Selection of Suitable Plant Growth Regulators for Augmenting Resistance to Waterlogging Stress in Soybean Plants (Glycine max L.) (콩 침수 스트레스에 대한 식물생장조절물질 처리 효과)

  • This research was conducted to evaluate methods of enhancing the waterlogging resistance of soybean plant. Thus, we applied seven types of plant growth regulators (PGRs) to soybean plants and exposed them to waterlogged conditions for a total of 14 days. To evaluate stress resistance, we monitored plant growth characteristics data such as height, chlorophyll content, and chlorophyll fluorescence for 28 days after the initial waterlogging (14 days under waterlogging conditions and 14 days after waterlogging). According to the results, plant height was significantly increased by gibberellin A4 ($GA_4$) treatment compared to the control treatment and waterlogging-only treatment. However, we could not detect plant height owing to plant death when we applied abscisic acid (ABA). Except for $GA_4$ and ABA treatments, plant heights slightly decreased in all treatments compared to the waterlogging-only treatment. The chlorophyll content and chlorophyll fluorescence showed a similar tendency among PGR treatments. The chlorophyll content and chlorophyll fluorescence were significantly increased by ethephon and kinetin treatments 28 days after waterlogging compared to the waterlogging-only treatment. Consequently, kinetin and ethephon treatments induced more resistant phenotypes in soybean plants during or after exposure to waterlogging conditions.

Effects of Growth Regulators on Adventitious Root Growth and Eleutherosides and Chlorogenic Acid Accumulation in Air Lift Bioreactor Cultures of Eleutherococcus koreanum (생물반응기 배양에서 생장조절제에 따른 섬오갈피 부정근 증식 및 Eleutheroside와 Chlorogenic Acid 생산)

  • The influence of different growth regulators on biomass of adventitious roots and secondary metabolites accumulation in bioreactor cultures of Eleutherococcus koreanum was studied. The maximum growth of adventitious roots was observed in the presence of $2.0\;\cal{mg/L}$ IBA (6.7 g DW/L). However the productivity of eleutheroside E was not significantly different among IBA levels. High level of thidiazuron (TDZ) efficiently increased both root growth and secondary metabolites production. Especially when $0.1\;\cal{mg/L}$ TDZ was combined with $3.0\;\cal{mg/L}$ IBA, the highest eleutheroside E accumulation ($3,327{\mu}g/g $DW) was observed. When the same TDZ level was treated in combination with $5.0\;\cal{mg/L}$ IBA, both chlorogrnic acid and eleutheroside B were accumulated to maximum level. In contrast, zeatin remarkably suppressed the accumulation of both eleutheroside B and chlorogenic acid.

Growth Inhibition Effect of Environment-friendly Agricultural Materials in Botrytis cinerea In Vitro (친환경 유기농자재의 잿빛곰팡이병 병원균의 생장 억제 효과)

  • Inhibition effects on spore germination and mycelia growth for gray mold (Botrytis cinerea) were investigated in vitro using environment-friendly agricultural materials as well as environment-friendly pesticides. The inhibition effect on mycelia growth of gray mold is the highest when the gray mold mycelia were treated with a pesticide (commercial name: Koreayeok, Jihabudea KM, Sootingtan, Sootingstar) that contains a mixture of Bacillus subtilis, resulting in 100% inhibition of the mycelia growth. Meanwhile, the range of less than 20% inhibition effects on the growth of gray mold mycelia was observed with other commercial agricultural materials. The significant inhibition effects on spore germination of gray mold fungus were shown in vitro with two water dispersible pesticides containing sulfur [BTB (97.7%) and SulfurStar (92.3%)], respectively. These in vitro results of inhibiting of the spore germination and mycelia growth together cannot found. It remains to be determined whether the selected environment-friendly agricultural materials in effective control of gray mold in vitro can be used to control gray mold in field.

Height Control of Pot Chrysanthemum according to Daminozide Applications at Different Planting Period in C-Channel mat Irrigation System (C-형강 매트 저면관수 시스템을 이용한 분화 국화 재배 시 정식시기와 Daminozide 처리에 따른 초장 조절)

  • Effect of daminozide concentration and application methods, in combination with different planting period, on the growth of chrysanthemum was investigated for small pot-plant production. Daminozide, a plant growth regulator widely used for plant height control, was applied by drench or sub-application. For pot chrysanthemums of first planting time, plant height was reduced over $4,000mg{\cdot}L^{-1}$, when daminozide was drenched directly into the pot medium. However, $5,000mg{\cdot}L^{-1}$ of daminozide drench affected plant height reduction for secondarily planted pot chrysanthemum. Analysis of variance revealed the plant height was affected by planting time. For sub-application experiment of daminozide, it showed that the daminozide affected the number of flower buds formation and fresh and dry weight. These results suggest that planting period affected growth retardation of chrysanthemum. Therefore, application of growth retardant in combination with planting time and application methods may provide more efficient growth control for pot-chrysanthemum production.

The Role of Nitric Oxide on the Growth Regulation of Chinese Cabbage (Brassica campestris L.) Primary Leaves (배추 (Brassica campestris L.) 제 1엽의 생장조절에 대한 Nitric Oxide의 역할)

  • The possible role of nitric oxide (NO)-induced cell division was investigated to explain the physiologycal effects of a NO donor, sodium nitroprusside (SNP) on the growth of primary leaves in chinese cabbage seedling plants. Exogenous treatment of SNP to chinese cabbage plants for 8 days at different concentrations (0, 200, 500 and 1000 ${\mu}M$) affected the leaf growth in a concentration-dependent manner, showing a maximum growth at $200\;{\mu}M$. In accordance with leaf growth responses, the chlorophyll and soluble protein contents increased strongly to 142% and 134% of control at $200\;{\mu}M$ SNP, respectively. However, a very little decrease in chlorophyll and a 13%> decrease in protein were observed at $1000\;{\mu}M$ SNP. In addition, the content of DNA and RNA also increased maximumly to 142% and 139% of the control at $200\;{\mu}M$ SNP, respectively, whereas they decreased to 80% and 84% of the control at $1000\;{\mu}M$ SNP. With respect to the development of enzymes related to cell wall synthesis, $200\;{\mu}M$ SNP led to the maximum activities in both phenylalanine ammonia-lyase (212% of the control) and guaiacol peroxidase (134% of the control). However, the activities of both enzymes were not modified significantly at $1000\;{\mu}M$ SNP. In conclusion, these results suggest that the enhancement of leaf growth in chinese cabbage plants by SNP at the effective concentration was probably due to the NO ability in the induction of cell division.

Effects of UV-B and Growth Inhibitor on Physiological Changes and Antioxidat Enzyme Activities in Fruit Vegetable Plug Seedlings (UV-B와 생장억제 처리가 과채류 플러그묘의 생리 변화 및 항산화 효소의 활성에 미치는 영향)

  • The physiological changes and antioxidant enzyme activities of plug seedlings irradiated with 4 kJ${\cdot}m^{-2}{\cdot}d^{-1}$ UV-B or sprayed growth inhibitors of 50 ${\cdot}L^{-1}$ diniconazole and 500 mg${\cdot}L^{-1}$ hexaconazole for retarding the overgrowth of cucumber, tomato and hot pepper seedlings in summer season were investigated. Photosynthetic rate just after UV-B irradiation was significantly low but recovered to the level of non-treated seedlings 10 days after IV-B irradiation. And photosynthetic rate of chemical-sprayed seedlings was slightly lower in tomatoes but not different in cucumbers and peppers, compared with non-treated seedlings. Cholrophyll contens in the leaves showed no difference among treatments. Anthocyanin contents in the leaves of UV-B-irradiated seedlings were higher and no difference inchmical-sprayed seedlings, compared with non-treated seedlings. the activites of SOD(superoside dismutase), CAT )catalase), and POD(perosidase) of UV-B-irradated seedlings were higher thatn those of the non-irradiated seedlings just after UV-B irradiation. However. 10 days after UV-B irradiation, the activities were decreased to half of the value just after UV-B irradiation Whereasd the activies of chemical-sprayed seedlings were slightly higher than those of non-treated ones but lower than those of UV-B-irradiated one.

In Vitro Shoot Tip Culture of Pear 'Niitaka' as Related to Tree Vigor Sampling Time and Plant Growth Regulators (배 '신고'의 경정배양에 있어서 모수의 수세, 경정의 채취시기 및 생장조절제의 영향)

  • In vitro shoot tip culture technique was established in pear (Pyrus pyrifolia 'Niitaka') as related to tree vigor, sampling time, and plant growth regulators and sucrose supplemented to medium. Shoot tips excised in June from the tree having medium-vigor developed good shoots. BA (1.0 and 2.0 mg/L) without NAA produced shoots suitable for proliferation, and NAA supplemented to medium resulted in poor shoot growth and excessive callus formation. BA of 2.0 mg/L combined with 0.01 mg/L NAA provided shoots suitable for rooting and sucrose of 30 g/L was recommended for proliferation medium. A fourth strength MS medium supplemented with 0.1 mg/L NAA produced plantlets in good quality of root number and root length.
