  • Cell density was gradually decreased as the dilution rate was increased under chemostat cultivation of HeLa cells. Maxium cell density was maintained at the dilution rate of 0.020 (1/h) which was far less than the wash-out rate of 0.050 (1/h), Maxium cell productivity of 2 (mL of cells/L/h)was obtained at the dilution rate of 0.030 (1/h) by showing the culture also required maintenance period at low dilution rates, whose result meant the deviation of continuous culture theroy. Methods of indirectly measuring cell density have been introduced to represent mammalian cell growth, which are packed cell volume and oxygen uptake rate, and these values showed good linear relationship with actual cell density by having correlation factor of 0.90. Theoretical maximum oxygen yield, $Y_{O2}^{max}$ and maintenance oxygen consumption rate, m$_{O2}$, were estimated as 4.1$\times$10$^5$ (cells/mmole $O_2$) and 10.71$\times$10$^{-9}$ (mmole $O_2$/cells/h) by employing oxygen yield model.

  • Gross photosynthetic rats of leaves of hydroponically grown cucumber plants(Cucumis sativus L. cv. Guwoosalichungjang) were measured under various conditions of photosynthetic photon flux(PPF), ambient $CO_2$ concentration, air temperature and leaf nitrogen contents. Light compensation point of leaf photosynthesis appeared to be in the range of 10~20$\mu$mol.m$^{-2}$ .s$^{-1}$ and light saturation point be above 1000$\mu$mol.m$^{-2}$ .s$^{-1}$ . Gross photosynthetic rates increased persistently and asymptotically as air temperature rose from 12$^{\circ}C$ to 32$^{\circ}C$. However, there were only small differences in gross photosynthetic rates in the range of 24-32$^{\circ}C$, so that the range seemed to be optimal for photosynthesis of cucumber plants at the condition of $CO_2$ concentration of 400$\mu$mol.mol$^{-1}$ and PPF of around 400$\mu$mol.m$^{-2}$ .s$^{-1}$ . $CO_2$ compensation point of leaf photosynthesis appeared to be in the range of 20-40$\mu$mol.mol$^{-1}$ and $CO_2$ saturation point be above 1200$\mu$mol.mol$^{-1}$ . Gross photosynthetic rates increased sigmoidally as leaf nitrogen content increased. These environmental factors interacted synergistically to enhance gross photosynthetic rate, so that the rate increased multiplicatively s level of one factor increased progressively with higher levels of he other factors. Mathematical models wer developed to estimate the gross photosynthetic rate in accordance with the variations of these environmental factors. These modes can be used not only to explain he variation of growth or yield of cucumber plants under different environmental conditions but also as building blocks of plant growth model or expert system of cucumber plants.

  • This study was carried out to evaluate growth characteristics of Kimchi cabbage cultivated in various highland areas, and to create a predicting model for the production of highland Kimchi cabbage based on the growth parameters and climatic elements. Regression model for the estimation of head weight was designed with non-destructive measured growth variables (NDGV) such as leaf length (LL), leaf width (LW), head height (HH), head width (HW), and growing degree days (GDD), which was $y=6897.5-3.57{\times}GDD-136{\times}LW+116{\times}PH+155{\times}HH-423{\times}HW+0.28{\times}HH{\times}HW{\times}HW$, ($r^2=0.989$), and was improved by using compensation terms such as the ratio (LW estimated with GDD/measured LW ), leaf growth rate by soil moisture, and relative growth rate of leaf during drought period. In addition, we proposed Excel spreadsheet model for simulation of yield prediction of highland Kimchi cabbage. This Excel spreadsheet was composed four different sheets; growth data sheet measured at famer's field, daily average temperature data sheet for calculating GDD, soil moisture content data sheet for evaluating the soil water effect on leaf growth, and equation sheet for simulating the estimation of production. This Excel spreadsheet model can be practically used for predicting the production of highland Kimchi cabbage, which was calculated by (acreage of cultivation) ${\times}$ (number of plants) ${\times}$ (head weight estimated with growth variables and GDD) ${\times}$ (compensation terms derived relationship of GDD and growth by soil moisture) ${\times}$ (marketable head rate).

  • The purpose of this study was estimate the optimal phosphorus and calcium levels of Korean native cow manure on the growth and cast production of earthworm(Eisenia foetida). Phosphorus and calcium levels supplemented to cow manure were 0%(without earthworm), 2% and 4%, and 0.01%, 0.02%, 0.04% and 0.06% of the cow manure by weight, respectively. The maximum fresh weight and increasing rate of earthworm were obtained with 2% phosphorus level, but there was not significant differences between calcium levels. The ratios of available phosphorus to calcium contents of cast were 1.04-1.20:1 with 2%, and 1.28-1.37:1 with 4% phosphorus levels. Conversion rate of organic matter to earthworm tissue were 0.35-0.41% with 2%, and 0.25-0.40% with 4% phosphorus levels.

  • In order to investigate antimicrobial substances from natural spices, the effects of garlic juice and ginger juice on the growth of microorganisms were investigated. It was showned that garlic juice inhibited the growth of bacteria, yeasts and fungi but ginger juice did not inhibit the growth of bacteria and yeasts. As result of concentration effect of garlic juice on the growth of microorganisms, the higher concentration of garlic juice was employed, the more growth inhibition was obtained. Comparing the specific growth rate at various concentration of garlic juice, the inhibition effect on yeast appeared much higher than that on bacteria.

  • Five denitrifying bacteria, which were identified as Pseudomonas sp., were isolated by the enrichment culture technique. The most effective denitrifying bacterium was named as Pseudomonas DWS, which was cultivated at anoxic condition. The optimal growth temperature and pH were 30$^{\circ}C$ and 7-8, respectively. Cell growth almost revealed a stationary phase at 18 hours after cultivation and nitrate was degrade 99.9% during this period. Therefore, it is suggested that Pseudomonas DWS could be effectively used for the biological treatment of wastewater containing nitrogen compounds.

  • Microbial antagonism between virulent and avirulent isolates of Pseudomonas solanacearum was studied in relation to the control of bacterial wilt of tobacco. In nutrient broth media or in soil, the avirulent isolate of P. solanacearum grew faster than did the virulent one. Inhibitory effect of avirulent isolate against growth of virulent one was negligible in mixed culture of the two isolates. The disease severity of bacterial wilt was significantly reduced when the roots of cultivar BY 4 susceptible to bacterial wilt was dipped in suspension of an avirulent isolate for 6 hours prior to transplanting to the soil infested with virulent bacteria. When the seedlings of tobacco were poured with the suspension of an avirulent isolate onto the soil in pre-planting pots 24 hours before ransplanting, there was a significant reduction in disease severity in the field. However, the reduction was noticed until early July, but after middle of July, no difference between the avirulent isolate-treated and non-treated plants was found in severity of the bacterial wilt.

  • This study was undertaken to determine the effects of cultural conditions on cell growth and casein hydrolysis for cell production in order to add Micrococcus sp. LL3 as a potential agent for industrial application with aim of shortening ripening period and improving flavor. Optimum temperature for cell growth and caseinolysis was $30^{\circ}C$ and $37{\circ}C$, respectively, and optimum pH was 7.0. The enzyme remained stable up to $50^{\circ}C$. Hydrolysis patterns of casein were also observed on SDS-PAGE. Both ${\alpha}-casein$ and ${\beta}-casein$ were totally hydrolysed by enzymes from Micrococcus sp. LL3 during culture. A preferential attack on ${\beta}-casein$ was observed. Production of aminopeptidase which cleaved polypeptides was the highest in early stationary phase during cell growth.
