Preparation and Characteristics of Bread by Medicinal Herb Composites with Immunostimulating Activity (면역활성을 가진 생약복합물을 이용한 빵의 제조 및 특성)

  • In this study, the breads with medicinal herbs (MH) composites showing immunostimulating activity were prepared and their characteristics were examined. Fourteen kinds of medicinal herbs were extracted with hot water and divided into 3 groups (MH-1, MH-2, MH-3) based on their contents. All groups showed immunostimulating activity in terms of macrophage phagocytosis, nitrite production, cytostatic activity and cytokine production. In the preparation of breads containing MH extracts of various contents (0, 30, 50, 70, and 100%), there was no significant difference among dough pHs of all groups after first fermentation, but loaf volume was significantly (p<0.05) increased in 70% added group while decreased in 30%, 50%, and 100% added groups compared to the control. The "a" and "b" values of bread crumb increased with the content of MH extracts while "L" value decreased, but these values of bread crust were similar to the control group. Most improvements in hardness, adhesiveness, gumminess and chewiness of bread were noticed by the addition of 70% MH extracts, but those of springiness, cohesiveness and resilience were mostly by the 50% addition ones. Through the sensory evaluation, it was revealed that mouth feeling, taste and overall preference decreased at breads containing 70% and 100% extracts, although appearance and crumb texture were not significantly (p<0.05) different among all groups.

Immunostimulatory and Anti-Obesity Activity of Lonicera insularis Nakai Extracts in Mouse Macrophages RAW264.7 Cells and Mouse Adipocytes 3T3-L1 Cells (섬괴불나무(Lonicera insularis Nakai) 추출물의 면역자극 및 항비만 활성)

  • In this study, we investigated in vitro immuno-stimulatory and anti-obesity activity of fruit (LIF), leaves (LIL) and stems (LIS) from Lonicera insularis Nakai in mouse macrophages RAW264.7 cells and mouse pre-adipocytes 3T3-L1 cells. LIF, LIL and LIS increased the production of immunostimulatory factors such as nitric oxide (NO), inducible nitric oxide synthase (iNOS), interleukin-1β (IL-1β), cyclooxygenase-2 (COX-2), tumor necrosis factor-α (TNF-α) and activated phagocytosis in RAW264.7 cells. Inhibition of toll-like receptor 2/4 (TLR2/4) partly blocked LIF, LIL and LIS mediated production of immunostimulatory factors. In addition, inhibition of mitogen-activated protein kinases (MAPK) signaling attenuated the production of immunostimulatory factors induced by LIF, LIL and LIS. Based on these results of this study, LIF, LIL and LIS is thought to activate macrophages the production of immunostimulatory factors and phagocytosis through toll-like receptor 2/4 (TLR2/4) and MAPKs signaling pathway. In anti-obesity study, LIF reduced the lipid accumulation in 3T3-L1 cells. LIF increased the protein phosphorylation expressions such as AMP-activated protein kinase (AMPK), hormone sensitive lipase (HSL), adipose triglyceride lipase (ATGL) related to the lipolysis of the adipocytes. In addition, LIF increased the expression of proteins involved in energy metabolism and brown adipose tissues differentiation such as peroxisome proliferator-activated receptor gamma coativator 1α (PGC-1α) and PR domain-containing16 (PRDM16). These results suggest that LIF is involved in lipid accumulation inhibition through expressing the proteins such as lipolysis and differentiation of white adipocytes to brown adipocytes.

Effect of Herbs on Vaccine Efficacy to Olive flounder, Paralichthys olivaceus (생약제 첨가사료가 넙치의 백신효과에 미치는 영향)

  • In this study we investigated the effect of vaccine efficacy and immune activities by feeding additives several natural herbal remedies extracts of Sweet Wormwood (Artemisia annua), Galla Rhois, Oriental raisin tree (Havenia Dulicis) and 6-mixed herb to Oilve flounder, Paralichthys olivaceus. Each group of fish fed with the herbal remedies added feed and basic formula during the seven weeks and vaccinated with Edwardsiella tarda Formalin killed vaccine after 4 weeks feeding. Relative Percentage Survival values (RPS) in the group was assessed by the challenge with E.tarda. All groups with the herbal remedies added feed enhanced growth rate, but there were not significantly different in lysozyme activity and agglutination titer. In a challenge experiment with E.tarda, RPS in the all groups feeding additives natural herbal remedies was higher than that of the control group. These results suggest that the natural herbal remedies extracts of Sweet Wormwood (Artemisia annua), Galla Rhois, Oriental raisin tree (Havenia Dulicis) and 6-mixed herb would be effective to enhance efficacy of vaccination to Olive flounder.

Effect of Storage Temperature and Keeping Materials on Storability and Quality of Chinese Yam (貯藏溫度와 充塡材料가 마의 貯藏과 品質에 미치는 影響)

  • The experiment was done to clarify the effect of keeping material and storage temperature on weight and quality of Chinese yam (Dioscorea opposita) tuber. After the yam tubers were placed into the plastic boxes filled with different keeping materials [polyethylene (PE) film, hull, soil, sand, vermiculite], they were stored under different storage temperature(room, cold) from Oct. 15 to Mar. 15 when all the characters related to the tubers were measured. Soil or PE film as keeping materials was the lowest sound tuber rate when stored at room or cold temperature, respectively, while vermiculite was the highest in both storage temperature. When PE film and vermiculite in both storage temperatures were used as keeping materials, tuber weight were less reduced than the others. Brightness of chromaticity and moisture content were lower in room temperature storage than in cold temperature storage although the characteristics related to marketability were not affected by storage temperature. PE film had greater brightness and value 'a' of chromaticity but lower its 'b' value in the latter temperature than in the former temperature. Vermiculite, however, did the reverse result in comparison with PE film.

  • Three antioxidants were isolated: (-)-epigallocatechin and (-)-epigallo catechin gallate from Theae Folium generally called green tea, and gallic acid from Moutan Cortex. They inhibited $Fe^{3+}-ADP/ascorbate-induced$ lipid peroxidation in rat liver microsomal fraction, showing $IC_{50}$, the concentration of antioxidants which is required to inhibit 50% of the peroxidation for microsomal membranes at 1 mg protein/ml, to be $0.2\;{\mu}g/ml$, $0.4\;{\mu}g/ml$ and $2.5\;{\mu}g/ml$, respectively.

  • On the purpose of development of antioxidative compounds from natural sources, 38 plants known to have antioxidant activity have been examined concerning DPPH radical scavenging activity. Among 13 plants exhibiting the activity, the seed of Carthamus tinctorius L, was selected as resources to search for active compounds due to rareness of study. The seed of the plant has been used as edible oil or preventive and remedial drugs for osteoporosis, arthritis. To reveal the principal component manifesting the antioxidant activity, the MeOH extracts was successively solvent-fractionated with EtOAc, n-BuOH and water. In order to isolate active component from the EtOAc fractions, application of silica gel column chromatographies and activity tests were repeated for a active component to be isolated. Its chmical structure was determined to be N-feruloylserotonin, a conjugated serotonin compound, by the interpretation of spectral data, NMR. MS and the adaptation of chemical reactions.

Ecological Characteristics and Distribution of Native Trillium tschonoskii in Ulleung Island (울릉도 자생 큰연령초의 분포와 자생지의 생태적 특성)

  • Trillium tschonoskii Maxim. is a kind of 64 endangered plant species designated by the Ministry of Environment in Korea. It is very a rare native plant throughout the country. Therefore, this study was conducted to investigate the native T. tschonoskii in Ulleung Island for their distribution and ecological characteristics. They were mostly distributed valley, mid-slope of the mountain from 186 to 458 m altitude. They had a little low light conditions as they faced west, north west, south west, south-south west, north-north east and east-east north. They were provided with slightly humid condition. Aerial humidities of these quadrates were variable as 21.5-71%. They were located on the variable slopes of 5 to 43 degrees. Soil humidity was slightly dry. Native sites were semi-shaded because tree layer such as Tilia insularis, Zelkova serrata, okamotoanum etc.and sub-tall tree layer such as Ligustrum foliosum, Comellia japonica, Tilia insularis, Callicarpa japonica and so on were grown near by. Woody plants such as A. Okamotoanum, Sorbus commixta, Fagus japonica var. multinervis etc. were frequently observed. These environmental conditions were suitable that T. tschonoskii grows up naturally because of high Percentage as 52.78% of blooming individuals in this study. These native sites were classified with three communities, T. tschonoskii-Majanthemum dilatatum community, T. tschonoskii-Anthriscus sylvestris community and T. tschonoskii-Hepatica maxima community in accordance with altitude and aerial humidity. The best way ffr the preservation of T. tschonoskii habitats and maintainance of its recent population is maintained in recent environmental condition and ecological condition without any management by human-being.

Studies on the Processing of Crude Drugs(VII) -On the Constituents and Biological Activities of Gardeniae Fructus by Processing- (한약의 수치에 관한 연구(제 7보) -치자의 수치에 의한 성분변화 및 생리활성-)

  • We have conducted to characterize the physico-chemical change and pharmacological transformation of traditional herbal medicines by means of processing. Processed Gardeniae Fructus was prepared by heating of fruit of Gardenia jasminoides(GF) for $30{\sim}50\;minute$ in the roster designed for herb processing. The contents of drying loss, water extract, diluted ethanol extract, ether extract and geniposide in non-processed GF and processed GF were examined. The contents of drying loss, water extract and geniposide in processed GF showed a decrease as compared with those of non-processed GF, however the contents of dilute ethanol and ether extract showed a increase as compared with those of non-processed GF. The rate of decrease/increase of those index were in proportion to heating time. And, biological activities of methanol extract of non-processed GF and processed GF were investigated. DPPH scavenging effects and inhibitory effect of xanthine oxidate and hemolysis of processed GF exhibited more effective than those of non-processed GF in vitro. Accelerating effect of large intestinal transport and purgative action of non- processed GF were discriminated by processing of GF. Methanol extracts of non-pro- cessed GF and processed GF showed the protective effects against the hepatotoxicity induced by ${\alpha}-naphthylisothiocyanate$ in rats. These results suggested that the transformation of biological activities of GF by means of processing may be due to the physico-chemical change of the constituents in GF by heating.

Hypoglycemic Activity of Opuntia ficus-indica var.sabotan on Alloxan- or Streptozotocin-Induced Diabetic Mice (Alloxan 및 Streptozotocin 유도 당뇨모델 동물에서 손바닥 선인장의 혈당강하 효과)

  • Hypoglycemic activity of Opuntia ficus-indica var. sabotan on alloxan or streptozotocin-induced diabetic mice was investigated. Fructus and folium of Opuntia ficus-indica vars. sabotan inhibited intestinal ${\alpha}-glucosidase$ of rats as well as glucose elevation in blood of normal mice, Opuntia ficus-indica var. sabotan exhibited inhibitory activities on streptozotocin-induced diabetic mice rather than on alloxan-induced diabetic mice loaded with maltose and sucrose. Its folium was more effective than its fructus. These results suggest that Opuntia ficus-indica var. sabotan might be effective on diabetic mellitus.

  • Arachidonic acid(AA), which is stored in membrane glycerophospholipids, is liberated by phospholipase $A_2(PLA_2)$ enzymes and is sequentially converted to cyclooxygenase (COX) and lipoxygenase (LOX) then to various bioactive prostaglandins (PGs,) and leukotrienes (LTs). In order to find the specific inhibitors of AA metabolism enzymes such as $PLA_2$, COX-2, 5-LO and lyso PAF acetyltransferase. 195 Korean indigenous plant extracts were evaluated for their inhibitory activity on $PGD_2,\;LTC_4$ production from cytokine-induced mouse bone marrow-derived mast cells (BMMC) and arachidonic acid released from phospholipid and PAF production from lyso PAF. From this screening procedure, methanol extract of eight plants such as Saururus chinensis, Aster tataricus, Chrysanthemum cinerariaefolium, Reynoutria japonica, Disocorea nipponica, Epimedium koreanum, impatiens textori, Veronica rotunda var. subintegra were found to inhibit production of inflammatory mediators in vitro assay system.
