Occurrence of Fungal Contamination in Ginseng Sprout and Mycotoxigenic Potential (새싹삼의 곰팡이 발생과 독소생성능)

  • In order to investigate frequency of fungal contamination in ginseng sprout, we collected 18 types of retail ginseng sprouts and analyzed them. Overall frequency of fungal contamination ranged from 113.3 to 174.1% with the highest occurrence of Penicillium spp. Fungal detection rate was significantly higher in moss than in stem, leaf and root of ginseng sprout. Penicillium spp. occurred in leaf and stem with the highest incidence and Fusarium spp., in root. Among Penicillium spp. and Fusarium spp., P. olsonii and F. oxysporum were dominant, respectively. Nine Fusarium species, Aspergillus westerdijkiae, Aspergillus flavus, and 11 Penicillium species were identified by phylogenetic analysis. PCR screening of mycotoxigenic potential revealed that 19 out of 25 isolates tested were positive for respective mycotoxin biosynthetic gene. Two 2 A. flavus and 11 A. westerdijkiae isolates produced varying amount of aflatoxin or ochratoxin A in czapek yeast extract brothsome of which showed high levels of mycotoxin production. These results suggests a need for continuous monitoring and management program to control fungal contamination in the ginseng sprout production chain.

  • 인간은 소리를 통해 많은 정보를 얻을 수 있다. 누가 어떤 말을 하는지 뿐만 아니라, 상황에 따라서는 소리가 발화된 환경 또한 시각적인 정보 없이 유추할 수 있다. 이러한 판단을 내리기까지, 인간은 경험을 통해 스스로 학습하는 과정을 거친다. 이와 같은 학습 과정에 생물의 사고과정을 모방하여 복잡한 상관관계를 추론하는 인공지능형 알고리즘을 적용하면, 인간의 두뇌가 경험을 통해 학습하고 판단하던 역할을 기계적으로도 모방할 수 있게 된다. 본고에서는 음향이 발화된 환경의 정보를 나타낼 수 있는 파라미터들에 대해 알아보고, 그 파라미터들을 지능형 알고리즘을 이용해 도출해내는 과정과 기법들을 소개한다.

  • 본 논문에서는 새로운 형태의 혼합형 5자유도 마이크로 매니퓰레이터를 제안하고 개발하고자 한다. 제안된 기구는 평면형 2자유도 XY-stage와 공간형 3자유도 병렬 기구로 구성된다. 3자유도 병렬기구는 3개의 직렬체인으로 이루어져 있으며, 공간상에서 두 방향의 회전 움직임과 Z축 방향으로의 병진운동을 생성한다. 모의실험을 통하여 개발된 기구의 작업공간과 출력단에서의 정밀도 해석을 수행한다. 개발된 기구는 조작 대상 물체의 미세 위치제어를 필요로 하는 반도체 및 기계, 생물, 의료분야 등에 적용 될 수 있다.

  • 쌍생아 402명의 지문형을 弓狀, 左蹄狀, 右蹄狀, 過狀으로 분류하고, 가마의 頭上위치를 左在, 中在, 右在의 3型으로, 가마의 수를 1,2로 구분하여 지문형과 관련성을 조사한 바, 지문형에 있어서는 過狀紋이 과반수를 다음은 右蹄紋, 弓狀紋의 차례로 그 출현빈도가 적었고, 가마의 유형별 빈도의 다소는 右在型, 左在型, 中在型의 차례로 나타났다. 過狀紋의 百分率은 흑인, 소련인, 미국인, 일본인 보다 현저히 높았고, 여자보다 남자가 높은 것은 위의 나라들과 같은 경향을 나타내었다. 지문의 過狀型과 가마의 右在型은 각각 보다 높은 빈도를 나타내었으므로 연관유전 여부를 밝히는 한 단서가 된다.

Production of Mycelia and Water Soluble Polysaccharides from Submerged Culture of Lentinus lepideus in Bioreactor (생물반응기를 이용한 잣버섯(Lentinus lepideus)의 균사체 및 수용성 다당체 생산특성)

  • The mushroom Lentinus lepideus was used to produce mycelial as well as soluble polysaccharides in bioreactor cultures. To determine optimal submerged culture conditions, both growth characteristics and water soluble polysaccharides production were compared among four different types of bioreactor and culture conditions. For the production of mycelial biomass, the following bioreactors were proven to be effective in decreasing order: an external-loop type air-lift bioreactor (ETAB; 7g/l), a balloon type air bubble bioreactor (BTBB; 6.2g/l), a stirrer type bioreactor (STB; 6g/l), and a column type air bubble bioreactor (CTBB; 5g/l). Maxiaml production of water soluble exopolysaccharides (EPS; 0.62g/l) and endopolysaccharides (PPS; 7.7%) could also be obtained from BTBB. The mycelial biomass increased with increase in glucose concentration from 15g/l to 75g/l in the media. In contrast, PPS contents in the cells decreased with increase in glucose concentration in the media, showing the highest PPS content (7%) at 15g/l. Among different medium feeding types, fed-batch culture based on concentration control in media (10g/l) produced higher mycelia than fed-batch culture based on volume control of media (5.8g/l) or batch culture (3.4g/l). EPS production was also higher in fed-batch culture based on medium concentration control than that in other feeding types.

  • In order to develop facility for complex farming with aquaculture and vegetable nutrient culture and to investigate growth of tilapia and water quality in the channel type aquaculture system, these experiments were carried out. When tilapia(Tilapia nilotica) was reared in the channel type aquaculture system, Quality of culture water and growth of tilapia were normal. And the growth of tilapia in the channel type and round type aquaculture system was much the same. Channel type aquaculture facilities was applicable to the dual culture system for aquaculture and vegetable culture. Channel type integrated system was composed of aquaculture and sandculture bed, and it was thought that suitable size of the system was about 1.4m(width) $\times$ 1m(height) $\times$ 20m(length).

Development and Evaluation of Criterion-Referenced Performance Assessment Items Based on the 7th National Science Curriculum -Subject Unit of Reproduction and Biological Accumulation- (제7차 교육과정에 근거한 준거지향적 수행평가 문항의 개발과 평가 -고등학교 과학 "생식"과 "생물 농축" 단원을 중심으로-)

  • In recent years, there has been an increased emphasis on performance assessment to evaluate students' abilities. Our nation has introduced a change in testing and assessment. Additional work on the efficacy, reliability, and comparability in order to develop the performance assessment item has been needed in the enforcement of the 7th National Science Curriculum. Also, criteria for professional and technical standards has been needed to be developed. The purpose of this study was to draw out various key concepts and to develop achievement standards, assessment standards and performance assessment items based on the 7th National Science Curriculum on the subject matter of reproduction(chapter 13) and biological accumulation(chapter 17). And also, this study examined the validity of completed performance assessment items based on classical test theory and polytomous item response theory. Twelve key concepts in chapter 13(reproduction) and four from chapter 17(biological accumulation) were abstracted. Twenty-six achievement standards in chapter 13(reproduction), and nine in chapter 17(biological accumulation) were developed. The achievement standards were determined in terms of knowledge(K), process skill(P) and attitude(A). Twenty-five assessment standards in chapter 13(reproduction) and nine in chapter 17(biological accumulation) were developed. Based on the developed achievement standards and assessment standards, twenty-two performance assessment items(seventeen open-ended questions, three essays, and two portfolios) with concrete grading criteria were developed. Eight open-ended items were applied to 240 10th graders to evaluate reliabilities of the test which consisted of four items per each chapter. The results would be suggested that the applied items were valid for performance assessment because item difficulties and item discriminations were proper. There was not much differences in item discrimination between interpretation from classical test theory and that from polytomous item response theory. However, there were some differences in item difficulties between the interpretations of two theories because the characteristics of examinees were reflected in classical test theory.

Characterization of Chinese Cabbage Phospholipase D by a Multistirring Batch System Bioreactor (다중 교반형 생물반응기에 의한 배추 Phospholipase D의 특성연구)

  • Phospholipase D catalyzes the phosphatidohydrolysis and transphosphatidylation of phospholipid in the biological systems. In this study we were partially purified phospholipase D from Chinese cabbage and the characterization of the enzyme was carried out in a multistirring batch system bioreactor. The enzyme showed optimum activity at pH ,5.6, highest activity at 37$^{\circ}C$ and Ca2+ is important for the enzyme activity. Optimum concentrations of Ca2+ for phosphatidohydrolysis was 20 mM and for transphosphatidylation was 40 mM, respectively. Some organic solvents such as diethylether, isopropylether and butylacetate were activated the enzyme activity. On the other hand, EDTA, Ba2+, Mn2+ and Zn2+ showed inhibitory effect on the enzyme activity. The base acceptors in transphosphatidylation by the Chinese cabbage phospholipase D were tested. Various poly-and monohydroxy alcohols were found to be active.

Conical Diffuser Design and Hydraulic Performance Characteristics in Bioreactor Using Empirical and Numerical Methods (원뿔형 산기관 설계와 생물반응조에서 수력학적 운전특성에 관한 실험 및 해석)

  • In this study, we develop a highly efficient conical-air diffuser that generates fine bubble. By inserting a sufficient number of aerotropic microorganisms with dissolved oxygen from an air diffuser and minimizing the air-channel blockages within the air diffuser, we expect to improve the efficiency and durability of the decomposition process for organic waste. To upgrade the conventional air diffuser, we perform experiments and numerical analysis to develop a conical-type that generates fine bubble, and which is free from nozzle blockage. We complement the air-diffuser design by numerically analyzing the internal air-flow pattern within the diffuser. Then, by applying the diffuser to a mockup bioreactor, we experimentally and numerically study the bubble behavior observed in the diffuser and the 2-phase fluid flow in the bioreactor. The results obtained include statistics of the cord length and increased velocity, and we investigate the mechanisms of the fluid-flow characteristics including bubble clouds. Throughout the study, we systemize the design procedures for the design of efficient air diffusers, and we visualize the fluid-flow patterns caused by bubble generation within the mockup bioreactor. These results will provide a meaningful basis for further study as well as the detection of oxygen transfer and fluid-flow characteristics in real-scale bio-reactors using sets of air diffusers.

The Biological Data Converter based on BSML for Sharing Information (정보 공유를 위한 BSML 기반의 생물학 데이터 변환기)

  • 현재 생물학 연구실에서 시퀀싱 실험을 통해 생성되거나 또는 공개용 생물 데이터베이스로부터 획득된 유전체 및 단백질 정보는 각각 이질적인 데이터형식을 사용하고 있다. 이 때문에, 생물정보를 분석하여 상호간의 정보를 효율적으로 사용하기 위해서는 공통된 형식의 데이터 표준화작업이 필수적이다. 그리고 이러한 이질적 데이터 형식에 대한 표준화 연구의 미비로 인하여 플랫 파일간의 정보공유에 어려움을 겪고 있다. 따라서, 이 논문에서는 다양한 유전체 및 단백질 정보를 관리.공유하기 위해 이질적인 포맷간의 맵핑 과정을 통하여 BSML(Bioinformatic Sequence Markup Language) 형태로 변환하고, 이를 객체관계형 데이터베이스(Object Relational DataBase)에 저장하는 시스템을 개발하였다. 그리고, 개발된 시스템은 생물정보 데이터의 표준화를 위해 개발된 XML(Extend Markup Language) 기반의 BSML을 이용함으로써 효율적으로 생물학 데이터들 간의 정보를 공유할 수 있으며, 개인 생물학 데이터베이스 구축이나 다양한 생물학적 데이터를 통합 관리하는 시스템에서 유용하게 쓰일 수 있다.
