Analysis of two-stage Continuous Culture System by Transient Response of Single-stage Continuous Culture System (일단 연속 생물반응기의 과도상태 거동을 이용한 이단 연속 생물반응기의 해석)

  • Two-stage continuous culture system has been studied intensively to maximize the productivity of a cloned gene product in unstable recombinant microorganism. As an effort to optimize the two-stage process, transient behavior of the second-stage was studied theoretically as well as experimentally using Escherichia coli Kl2$\delta$Hl$\delta$trp. A mathematical model describing the transient response to a step change in dilution rate was developed based on the assumption that the adaptation rate of cell growth is proportional to the available growth potential, which is defined as the difference in dilution rates between before and after shift-up. The kinetic parameters appearing in the model equations were the dimensionless step increase in growth rate($\alpha$) and the adaptation rate constant(k). These parameters were evaluated for various dilution rates and temperatures by washout method. This relatively simple adaptation model could predict the specific growth rate of the second-stage successfully. Advantage and disadvantage of the proposed model are also discussed.

  • In the studies of characterization of crystal prototoxin isolated from Bacillus thuringinensis var. alesti and its bioassay to the silkworm larvae, Bombyx mori L., the following results were obtained. 1. The complete lysis of bacteria grown on the nutrient agar at 30$^{\circ}C$ in an incubator took 30 days, but the period could be reduced by a half in a devised broth media in a fermentation jar. 2. The protein toxin extracted directly from a mixture of crystals and spores without separation of crystals and spores was pure as same as the protein toxin extracted from only crysals separated. 3. By SLS polyacrylamide gel electrophoresis of prototoxin released from crystals in alkali (pH 9.5) three different proteins in molecular weights of 120,000, 87,000, 74,000 were separated. 4. In the bioassay of the toxin to the silkworm larvae, LD$\sub$50/ per gram of the larvae in the 4th instar was 0.080 $\mu\textrm{g}$.

Experimental study of Nutrient Removal by Endogeneous Nitrate Respiration (ENR) Mechanism in domestic wastewater (질산성질소의 내생탈질기작을 이용한 하수내 영양소 제거에 관한 실험적 연구)

  • The purpose of this study is to develop the efficient nutrient removal process and to verify operation and design parameters in domestic sewage. Endogenous nitrate respiration (ENR) was used for denitrification of nitrate in return sludge without additional organic carbon source. ENR reactor before the anaerobic tank enable to reduce nitrate below 3 mg/L and increase phosphate release at anaerobic reaction. Primary effluent during pilot scale plant were shown as TCOD/TP ratio of 40~60 and TCOD/TKN ratio of 5~7. Effluent concentrations were 10 to 12mg/L as TN and 1mg/L as TP respectively. In lab scale experiments endogenous denitrification rate of ENR reactor ranges from 0.042 to $0.057gNO_3-N/gMv.d.$ $SP_{rel}/SCOD_{rm}$ was shown as from 0.13 to 0.17 in anaerobic reaction. These kinetic parameters are expected to be available for BNR(Biological Nutrient Removal) plant design and ENR reaction is available for nutrient removal in low strength wastewater.

Characteristics of Lecithin-adsorbed Magnetic Nanoparticle and Biocompatibility of Its Fluid (Lecithin이 흡착된 나노 자성입자의 특성과 그 자성유체의 생체 친화성)

  • Magnetic nanoparticles were prepared by thermal decomposition and adsorbed with lecithin by applying ultrasonic. The size and saturation magnetization of magnetic nanoparticles were observed with different lecithin concentration, and the maximum tolerated dose (HTD) and toxicity of magnetic fluid was investigated through a biological test. The thickness of lecithin-adsorption layer increased non-linearly with increasing amounts of added lecithin, and the desirable adsorption amount was observed in the lecithin concentration of 20%(w/v). The dispersibility and magnetic properties of lecithin-adsorbed magnetic nanoparticles were most excellent when the ultrasonic exposure time was 1.5h. Also, the maximum tolerated concentration with best cell viability was $32{\mu}g/ml$ in vitro test, and lecithin-adsorbed magnetic fluids improved the biocompatibility by 1.2 times compared with bare magnetite fluids in vivo.

  • In the development of biomaterials as a substitute of hard tissues, the biocompatibility and osteoconductivity of the biomaterial are considered to be one of the most significant considerations. These biological properties of a material can be greatly improved by the modification of the surface properties by the depositing calcium phosphate thin films on the material since calcium phosphate films possess similar chemical compositions to hard tissues. The success of a material as a biomaterial will be determined by the interaction of the surface of the material with the adhesion molecules which induce cellular adhesion and biological responses of the adherent cells. Depending on the adsorption mechanisms and adsorbed conformation of the adhesion molecules on the surface of the biomaterial, cellular responses, such as adhesion, proliferation and differentiation of osteoblast cells, can be promoted or restricted. It has been reported that materials of which surfaces were modified with thin films of calcium phosphate appeared to be more osteoconductive. Rapid formations of bone nodule in addition to higher differentiations of osteoblast have been observed on the calcium phosphate thin films.

Purification and Characterization of Glutathione Peroxidase Isolated from Rat Erythrocyte and Histochemical Study of its Localization in Liver of White Rat (흰쥐 적혈구에 있는 Glutathione Peroxidase의 순화 및 성질과 간에서의 용작부위에 대한 조직화학적 연구)

  • A glutathione peroxidase from white rat (Wistar strain)erythrocytes was partially purified and characterized. In addition, localization of this enzyme in the liver was studied by histochemical method. A glutathione peroxidase was purified approximately 33.5-folds by ammonium sulfate precipitation, Sephadex filtration column and DEAE-Sephadex column chromatography. The optimum temperature of the crude glutathione peroxidase was $40^\\circC$, and the optimum pH was 7.5. This crude glutathione peroxidase was most stable at $30^\\circC$ and the values of Km and Vmax were calculated to be 8.5mM and 15.6 $\\mu$moles/min for glutathione, and 40 $\\mu$M and 10.5 $\\mu$moles/min for hydrogen peroxide, respectively. The molecular weight of this enzyme was estimated by Sephadex G-200 gel filtration to be approximately 90, 000. By electron microscopic examination, histochemical reaction products were microbodies that were prominent in the peripheral parts of the lobule. The reaction products exhibited round shapes, the diameter of which varied $0.2\\sim0.7 \\muM$ and their boundary membranes were not distint.

Removal Characteristic of Ammonia Nitrogen and Behavior of Nitrogen in Synthetic Wastewater Using Leclercia Adecarboxylata (Leclercia Adecarboxylata를 이용한 합성폐수의 암모니아성질소 제거특성 및 질소거동)

  • In this study, the removal characteristic of ammonia nitrogen and behavior of nitrogen was investigated using Leclercia adecarboxylata, which was derived from the culture contaminated by ammonia nitrogen of high concentration. The method of ammonia nitrogen removal was not biological nitrification and denitrification but elimination of nutrient salt with internal synthesis of microorganisms which use ammonia nitrogen as substrate. L. adecarboxylata(one of ammonia synthesis microorganisms) was highly activated and showed the most high removal efficiency in free salt condition but the removal efficiency decreased badly in salt concentration of more than 4%. About 80 mg/L of $NH_3-N$ was mostly removed within 20 hours and 500 mg/L of $NH_3-N$ showed less then removal efficiency of 50% because carbon source was not enough. However, ammonium nitrogen concentration was decreased again when the carbon source was inserted additionally thus, ammonium nitrogen removal efficiency by L. adecarboxylata, was related to amount of carbon source. pH decreased from 8.0 to 6.36 according to growth of L. adecarboxylata. Concentration of nitrite nitrogen and nitrate nitrogen did not increase and TKN concentration showed no variation while ammonia nitrogen was removed by L. adecarboxylata. In addition to, when content of protein in organic nitrogen was measured, protein was not detected at the beginning of microorganism synthesis but protein of 193.1 mg/L was detected after 48 hours. Hence, ammonium nitrogen was not decomposed as nitrate nitrogen and nitrite nitrogen but synthesized by L. adecarboxylata, which has excellent ability of nitrogen synthesis and can threat ammonia nitrogen of high concentration in wastewater.

Optimization of Production Conditions of Biosurfactant from Bacillus sp. and its Purification (Bacillus sp.에 의한 생물계면활성제의 생산 및 그의 성질)

  • A bacterium capable of emulsifying hydrocarbon, n-hexadecane, and decreasing surface tension of the culture media using oil collapsing method was isolated. The bacterium was partially identified as Bacillus sp. and named BJS-51. n-Hexadecane was the most effective carbon source for production of biosurfactant. Surface tension was decreased from 76 dyne/cm to 31 dyne/cm and CMD (critical micelle dilution) had the highest value of 5.7 at 3% n-hexadecane. Ammonium phosphate was the most effective nitrogen source, when C/N ratio was 60, surface tension and CMD were 29 dyne/cm and 9.2, respectively. Optimum pH and temperature were 7.2 and $30^{\circ}C$, respectively. Produced biosurfactant was extracted and purified using organic solvent extraction method and preparative HPLC systems. After analysis by various color reaction, this biosurfactant was identified as lipopolysaccharide. Surface tension and CMC (critical micelle concentration) of purified biosurfactant were 27 dyne/cm and 0.08 g/l, repectively. CMD was 9.2, so the yield of biosurfactant was about 0.74 g/l at the optimal conditions. The biosurfactant was very stable at wide range of $pH\;2{\sim}12$ with surface tension $29{\sim}31\;dyne/cm$ and showed $29{\sim}30\;dyne/cm$ of surface tension after heat treatment at $100^{\circ}C$ for 60 min.

Classification of Antimicrobial Peptides among the Innate Immune Modulators (선천성 면역조절자인 항생펩타이드 분류)

  • Multidrug-resistant super bacterial, fungal, viral, and parasitic infections are major health threaten pathogens. However, to overcome the present healthcare situation, among the leading alternatives to current drugs are antimicrobial peptides (AMPs), which are abundantly produced via various species in nature. AMPs, small host defense proteins, are in charge of the innate immunity for the protection of multicellular organisms such as fish, amphibian, reptile, plants and animals from infection. The number of AMPs identified per year has increased steadily since the 1980s. Over 2,000 natural AMPs from bacteria, protozoa, fungi, plants, and animals have been listed into the antimicrobial peptide database (APD). The majority of these AMPs (>86%) possess 11–50 amino acids with a net charge from 0 to +7 and hydrophobic percentages between 31–70%. This report classified AMP into several categories including biological source, biological functions, peptide properties, covalent bonding pattern, and 3D structure. AMP functions not only antimicrobial activity but facilitates cell biological activity such as chemotatic activity. In addition, fibroblastic reticular cell (FRC) originated from mouse lymph node stroma induced the expression of AMP in inflammatory condition. AMP induced from FRC contained whey acidic protein (WAP) domain. It suggests that the classification of AMP will be done by protein domain.

Optimization of Cell Concentration and Dilution Rate in Cell Recycled Ethanol Fermentation (세포재순환 에탄올 발효에서 세포농도와 희석률의 최적화)

  • The rheological characteristics of the ethanol fermentation broth were pseudoplastic when the yeast concentration was above 150g/L. From the viewpoint of rheological properties, the cell concentration below 150g/L was recommended for ethanol fermentation. Since the cell floc was formed at the cell concentration of 100 g/L, yeast cells were not much plugged in the pores of the membrane. The cell concentration above 100g/L was desirable when considering the permeability of the membrane. Since ethanol productivity was the highest when the cell concentration was 130 g/L in cell recycled ethanol fermentation. The optimal dilution rate was determined at 1.3 h-1 at constant cell mass of 130g/L. At this dilution rate, the ethanol productivity and glucose conversion ratio ware 80 g/L$\cdot$h and 0.94, respectively.
