Effects of Apigenin, an Antioxidant, on the Bioavailability and Pharmacokinetics of Etoposide (항산화제인 아피제닌이 에토포시드의 생체이용률 및 약동학에 미치는 영향)

  • 에토포시드와 아피제닌의 약동학적 상호작용 연구를 위하여 아피제닌 (0.4, 2.0 또는 8 mg/kg)과 에토포시드의 경구(6 mg/kg) 및 정맥 (2 mg/kg) 투여 하여 본 연구를 실시하였다. 아피제닌이 cytochrome P450 (CYP) 3A4 활성과 P-glycoprotein (P-gp)의 활성에 미치는 영향도 평가하였다. 아피제닌의 CYP3A4의 50% 효소활성억제는 $1.8{\mu}M$ 이었다. 아피제닌은 MCF-7/ADR 세포의 로다마인-123 세포 축적을 증가 시키므로 P-gp를 억제시켰다. 아피제닌은 에토포시드의 혈장곡선하면적과 최고혈장농도 (AUC and $C_{max}$)를 유의성 있게 증가시켰으나, 에토포시드의 최고혈장농도 도달시간 ($T_{max}$)과 생물학적 반감기 ($t_{1/2}$)에는 영향을 미치지 않았다. 따라서, 아피제닌 존재하에 에토포시드의 절대적생체이용률 (AB)은 대조군과 비교하여 유의성있게 증가되었다. 경구투여시와는 대조적으로, 아피제닌은 정맥 내로 투여된 에토포시드에서는 약동학적 파라미터에 어떤 영향도 미치지 않았다. 따라서 아피제닌이 에토포시드의 생체이용률을 증가시킨 것은 아피제닌이 소장과 간장에서 CYP3A4을 억제 및 소장에서 P-gp를 억제 시켰기 때문으로 사료된다.

  • 척추동물의 초기 발생에 관여하는 물질로 알려져 있는 retinoic acid(RA)를 Xenopus 배(embryo)에 처리한 결과, 체절의 형성에 이상 발생이 나타났다 즉 낭배 초기에 RA에 노출된 개체의 체절은 대조군의 화살머리 모양((((() 대신 기등 모양(1111)을 형성하고 있었으며, 고농도일수록 직선에 가까운 모습을 하고 있었다. 신경배 시기에 노출된 개체의 체절은 정상적인 분절이 이루어지지 않았으며, 이들 이상 체질의 위치는 처리 시기에 따라 다르게 나타났으나 농도에는 영향을 받지 않았다. 그러나 분절 과정중 일어나는 체절 세포들의 회전은 RA에 의해 억제되지 않았다. 이와 같은 결과는 체절의 형성에 분절 이전의 세포의 행동을 주도하는 프로그램과 분절을 주도하는 프로그램이 분리되어 있음을 암시한다. 한 가지 특이한 사항은 열처리 등으로 유발된 혼란이 수 개의 체절에 국한되어 나타나는 점과는 달리 RA의 처리는 어느 지점 이 후부터 모든 체절에 혼란을 야기시킨다는 점이다. 이와 같은 연속적인 분절의 흔란 현상은 RA가 다른 저해제에 의해 영향을 받는 유전자의 활동을 조절하는 전 단계의 유전자에 영향을 주기 때문인 것으로 사료된다. 또한 신경배 시기에 RA에 노출된 배의 a-cardiac actin은 정상적으로 발현되는데 비해 낭배 초기에 RA에 노출된 배는 감소된 양의 a-cardiac actin myNA의 축적을 보였다. 그러나 EFI-u은 RA의 처리 시기와는 무관하게 정상적으로 발현되었다.

Biosorption of Pb and Cd by Indigenous Bacteria Isolated from Soil Contaminated with Oil and Heavy Metals (유류와 중금속으로 오염된 토양에서 분리한 미생물의 Pb와 Cd 생물흡착 특성)

  • Indigenous bacterium which shows a tolerance to high metal toxicity was isolated from soil concomitantly contaminated with oil and heavy metals. The characteristics of the bacterium for Pb and Cd biosorption was investigated under the various experimental conditions such as bacterial growth phase, the initial metal concentration, the input biomass amount, temperature and pH. The Langmuir adsorption isotherm modeling was described to know the capacity and intensity of biosorption. The low initial concentration of heavy metals and high biomass has a maximum heavy metal removal efficiency, but biosorption capacity of Pb and Cd has different values. Biosorption efficiency was highest in the end of the microbial growth stage and under pH 5~9 condition, but was less affected by temperature variation of 25~$35^{\circ}C$. The maximum biosorption capacity for Pb and Cd was 62.11 and 192.31 mg/g, respectively and each $R^2$ was calculated as 0.71 and 0.98 by applying Langmuir isothermal adsorption equation. Biosorption for Cd was considered as monomolecular adsorption to single layer on the surface of cells, whereas biosorption for Pb was considered as accumulation process into the cell by the microbial metabolism and precipitation reaction with anion of bacteria.

Toxicity assessment of food additive(E171) in aquatic environments (식품첨가물 E171이 수생물에 미치는 독성 평가)

  • E171, a mixture of titanium dioxide, has been widely used as a food additive due to its whitening effect and low toxicity. However, it has been proven that E171 is no longer safe for public health. So far, there are insufficient studies on the toxic effects of E171 on organisms especially using standardized test methods. In this study, toxicity assessments of E171 to two aquatic species, water flea (Daphnia magna) and zebrafish (Danio rerio), were performed using modified standardized test methods based on the physicochemical properties of E171. The hydrodynamic diameter, polydispersity index, and turbiscan stability index (TSI) were measured to ensure the dispersion stability of E171 in exposure media during the test period. The EC50 for immobilization of water flea was 141.7 mg L-1 while zebrafish was not affected until 100 mg L-1 of E171. Measurements of reactive oxygen species (ROS) and antioxidant enzyme activities confirmed that E171 induced oxidative stress, leading to the activation of superoxide dismutase and catalase in both water flea and zebrafish, although the expression of antioxidant enzyme genes differed between species. These results suggested the potential risk of E171 to aquatic organisms and provided toxicological insights into the impacts of E171 on the environment.

Development of One-Step Immuno-Chromatography Assay System for Salmonella typhimurium (Immuno-Chromatography 방법을 이용한 식중독균(Salmonella typhimurium) 1단계 분석시스템의 개발)

  • One-step immuno-chromatography assay system for heat-killed Salmonella typhimurium antigens was developed. Three major components used were a glass fiber membrane (placed at the bottom of the system) with an antibody (specific to the analyse, detection antibody)-gold conjugate deposited in a dry state on the surface, a nitrocellulose membrane (middle) with an antibody (also, specific to the analyse but recognized different epitome: capture antibody) and anti-detection antibody immobilized in spatially separated areas, and a cellulose membrane (top) as absorption pad. These membranes were partially superimposed such that a wicking of aqueous solution containing sample can continuously take place through membranes. Variables that affected the system performance were the concentration of capture antibody, the location on the membrane, inert protein used for blocking of the membrane and for carrying the sample, and the concentration of the gold conjugate. Under optimal conditions, within 15 minutes after absorption of a sample solution from the bottom of the system antigen-antibody complexes of sandwich type were formed on the membrane surface area with immobilized capture antibody and a color signal was generated in proportion to the analyse concentration. The minimum do tection limit of the analyse was $1{\times}106$ Salmonella cells/mL.

  • Recombinant Saccharomyces cerevisiae BZ-pJ containing the gene coding for metallothionein, a metalbinding protein was grown in the medium with high cadmium concentrations to study the characteristics of growth and cadmium uptake. High concentrations of cadmium reduced cell growth and final cell density and increased the lag phase periods of the recombinant yeast. Addition of 10 mg $Cd^{2+}$/L to the growth medium remarkably decreased a lag period and enhanced the specific cadmium uptake to 52.6 mg $Cd^{2+}$/g dry cell. The effect of copper addition was further investigated in the medium of 680 mg Cd2+/L. An increase in copper concentration from 11.0 to 33.3 mg/L enhanced the specific cadmium uptake from 17.0 to 42.0 mg Cd2+/g dry cell.

  • A transformat Alcaligence eutrophus GA5 harboring phbC gene from A. latus was cultivated for production of Poly(3-hydroxybutyrate-4-hydroxybutyrate)[P(3HB-4HB)] containing high molar fraction of 4-hydroxybutyrate(4HB)] containing high molar fraction of 4-hydroxybutyrate(4HB). Transformation did not influenced significantly on total cell growth, on total cell growth, concentration, and content of P(3HB-4HB), however, significantly influenced on 4HB molar fraction in P(3HB-4HB) increasing from 12.3 to 23.5 mol% after 48 h cultivation in two-stage using 1.0%(W/V) of ${\gamma}$-butyrolactone as a precursor compare to parent strain. Above increment may be due to the accelerated polymerization between 3HB and 4HB converted from precusor compound by amplified phbC gene. Citrate increased remarkbly total cell mass and P(3HB-4HB) concentration, but did not influenced on the molar fraction of 4HB, meanwhile, magnesium ion influenced on P(3HB-4HB) concentration and 4HB molar fraction significantly. The two-stage cultivation method was modified, in such a way minimizing P(3HB) accumulated inside of cell grown at first-stage, consquently, 26.3% of P(3HB-4HB) containing 61.0 mol% of 4HB fraction was obtained after 72hr. Furthermore, semi-homopolymeric P(4HB) containing 92.0 mol% of 4Hb was obtained, and its structure was confirmed by $^1$H-NMR.

The Cadmium Biosorption Mechanism in Gram Negative Bacteria, Serratia marcescens (Gram 음성 세균인 Serratia marcescens에 의한 카드뮴 흡착 기작)

  • Serratia marcescens, an enterobacterium of gram-negative bacteria, is characterized by resistance of the admium. Cadmium sensitive PM strain did not grow in the medium at cadmium concentration of 50 ppm. PA strain was induced to accommodate to cadmium by cultivating the mother strain (PC strain) in the medium with 50 ppm cadmium. As compared with PC and PM strains, PA strain revealed the excellent growth in cadmium media and accumulated four to five times higher cadmium concentration in cell than other strains. PA strain accumulated 23% of cadmium in cells when cultured in medium treated with 100 ppm cadmium and this cadmium was more accumulated in cytosol fractions than membrane fractions. Analysis by TEM indicated that cadmium was concentrated as a form of granule in cytosol. In protein patterns of cell after the treatment of cadmium, two inducible proteins (28 KDa and 64 KDa) and one reducible protein (45 KDa) were detected by SDS-PAGE. By Atomic Absorption Spectrophotometer, the amounts of cadmium attached to inducible proteins of 28 KDa and 64 KDa were 318.28 ㎍ and 325.37 ㎍ per gram of protein, respectively. It is assumed that these inducible proteins play an important role in the mechanism of cadmium accumulation in cells. A plasmid of 23Kbp was found in S. marcescens. The ability of resistance to cadmium in plasmid was confirmed by curing experiments.

Effects of Paper Bag Coated Calcium on the Calcium Concentration, Lenticel Development, and Quality in 'Chuhwangbae' Pear Fruits (칼슘이 코팅된 봉지 괘대가 '추황배' 과실의 칼슘함량, 과점 발달 및 과실품질에 미치는 영향)

  • In a pear fruit 'Chuhwangbae' was investigated the effect of the calcium-coated double paper bag on the physical properties of the paper bag, micro meteorological phenomena, and calcium contents in quality of fruit. The calcium-coated paper bag, compared with official paper bags, did not give any effect on light transmission ratio and tensile strength. The change of the inside relative humidity of the paper bag was a little compared with conventional paper bags, but there was no difference in temperature. The contents of the accumulated calcium of the pericarp was remarkably greater than conventional paper bags during the period of 65 days to 160 days after the full bloom, but the flesh remarkably increased at 160 days. The calcium content per concentration of calcium coating greatly increased in 12% of yellow double paper bags and 9, 12% of newspaper double paper bags in case of the pericarp, and in 3% of yellow double paper bags and 6, 9, 12% of newspaper double paper bags in case of the pericarp. As a result of treatment of a radioactive isotope, the amount of accumulated calcium in the pericarp continued until 60 hours after treatment, but there was no difference in the calcium amount between the flesh and no-treatment pericarp. As to the hardness of fruits at the time of harvest, there was no difference in the concentration in case of a yellow double bags. But newspaper double paper bags 6, 12% was significantly difference. Soluble solid remarkably increased in yellow double paper bags 6, 9% and yellow double paper bags 3, 6, 9%. Also, it did not effect on changes of the pericarp, fruit weight and the color of the pericarp.

Bioaccumulation and Expressions of Stress Response Genes in Benthic Oligochaete Worm Tubifex tubifex to Exposure of Cadmium-spiked Sediment (카드뮴 (Cd) 노출 퇴적물에 따른 실지렁이 Tubifex tubifex의 체내 축적과 스트레스 반응 유전자 발현)

  • Cadmium (Cd), a heavy metal found in the aquatic environment, accumulates in organisms through the food chain. In the study, we investigated the survival rates, measurement of body Cd levels, and expression analysis of the stress response genes (Heat shock protein 70: HSP70 and Heat shock protein 60: HSP60) and antioxidant enzyme Glutathione S-Transferases (GST) on benthic oligochaete worm Tubifex tubifex exposed three concentrations of Cd, to analyze the bioaccumulation and changes of stress gene expressions to exposure toxicity of the Cd-spiked sediment. Survival rates of T. tubifex exposed to the Cdspiked sediment were 93% at 0.4 mg kg-1 Cd, 96% at 1.87 mg kg-1 Cd, and 93% at 6.09 mg kg-1 Cd for 10 days. Cd concentration in the body of T. tubifex was higher than that in the sediment. After Cd exposures for 10 days, the body Cd levels were 18.4 mg kg-1, 13.06 mg kg-1, and 79.11 mg kg-1 at exposed three concentrations of Cd, respectively. Upregulation of HSP70 gene expression was observed at all concentrations of exposed Cd as a time-dependent manner, whereas transcriptional expression of the HSP60 gene increased as a timedependent manner in T. tubifex exposed to the relative high concentration (6.09 mg kg-1) of Cd. However, GST gene expression increased on day 1 at all concentrations after Cd exposures, and then downregulated until 10 days. These results indicate to ecotoxicological and molecular effects in benthic oligochaete worm T. tubifex to Cd-spiked sediment and provide the basic information for the utilization of environmental toxicity assessment using the T. tubifex as a aquatic pollution indicator species.
