Antioxidant and Anticancer Activities of Water Extracts from Ceramium kondoi (비단풀 물 추출물의 항산화력 및 항암활성)

  • The present study describes the preliminary evaluation of the antioxidant activity and the cytotoxic effect of Ceramium kondoi. The antioxidant activities and cytotoxic effect of the water extracts were evaluated by total phenolic contents (TPC), DPPH radical scavenging activity (RSA), reducing power (RP), comet assay, and MTT reduction assay. TPC, DPPH RSA, and RP of the extract at the concentration of $1,000{\mu}g/mL$ was $659.2{\mu}M$, 86.0%, and 1.084, respectively, and those were concentration dependent. The $200{\mu}M\;H_2O_2-induced$DNA damage was inhibited by C. kondoi water extract in a dose dependent manner in human leukocytes. The inhibition was by 62.3, 39.8, 24.8% and 16.4% at the concentration of 5, 10, $25{\mu}g/mL$ and $50{\mu}g/mL$, respectively. Cytotoxic activity on HT-29 cells and MCF-7 cells of the C. kondoi water extract at the concentration of $10{\mu}g/mL$ was 49% and 60%, respectively. These results strongly support the possibility of C. kondoi as a source of natural functional materials.

Protective effect of Gabjubaekmok (Diospyros kaki) extract against amyloid beta (Aβ)-induced cognitive impairment in a mouse model (아밀로이드 베타(amyloid beta)로 유도된 인지장애 마우스 모델에서 갑주백목(Diospyros kaki) 추출물의 인지기능 및 뇌 신경세포 보호 효과)

  • The current study investigated the effect of Gabjubaekmok (Diospyros kaki) ethanolic extract (GEE) on $H_2O_2$-induced human neuroblastoma MC-IXC cells and amyloid beta $(A{\beta})_{1-42}$-induced ICR (Institute of Cancer Research) mice. GEE showed significant antioxidant activity that was evaluated based on ABTS, DPPH scavenging activity, and inhibition of malondialdehyde (MDA) and acetylcholinesterase activity. Further, GEE inhibited ROS production and increased cell viability in $H_2O_2$-induced MC-IXC cells. Administration of GEE ameliorated the cognitive dysfunction on $A{\beta}$-induced ICR mice as evaluated using Y-maze, passive avoidance, and Morris water maze tests. Results of ex vivo test using brain tissues showed that, GEE protected the cholinergic system and mitochondrial functions by increasing the levels of antioxidants such as ROS, mitochondrial membrane potential (MMP), and adenosine triphosphate (ATP) against $A{\beta}$-induced cognitive dysfunction. Moreover, GEE decreasd the expression levels of apoptosis-related proteins such as $TNF-{\alpha}$, p-JNK, p-tau, BAX and caspase 3. While, expression levels of p-Akt and $p-GSK3{\beta}$ increased than $A{\beta}$ group. Finally, gallic acid was identified as the main compound of GEE using high performance liquid chromatography.

Quality characteristics of muffins added with Pholiota adiposa powder (검은비늘버섯 분말을 첨가한 머핀의 품질특성)

  • This study was carried out to investigate the quality characteristics of muffins with added Pholiota adiposa powder. Muffins were prepared with different amounts of Pholiota adiposa powder (in 0, 2.5, 5, 10, and 15% ratios to the flour quantity). For analyzing the quality characteristics during the manufacturing of muffins, the pH, moisture content, dough density, height, specific volume, baking loss, texture, color, antioxidant activity, total polyphenol content, and sensory test results were determined. As the Pholiota adiposa powder content increased, the moisture content of the muffins decreased from 26.72 to 22.56%. The hardness of the muffins was increased with increasing Pholiota adiposa powder content, with the muffins with 15% Pholiota adiposa powder the hardest among all the samples. The inner color of the muffins, particularly the L and b values, decreased with increasing powder content compared to those of the control, but the a value increased. As the Pholiota adiposa powder content increased, the antioxidant activity and total polyphenol content of the muffins also increased. In the sensory evaluation, the muffins with 5% Pholiota adiposa powder showed higher values than the control and the other samples in all aspects, showing that 5% Pholiota adiposa powder could be incorporated into muffins to meet the taste and functional needs of the consumers.

Biological activities of Aster scaber extracts (참취(Aster scaber) 추출물의 생리활성)

  • The phenolic compounds extracted from Aster scaber were examined for their biological activities owing to their potential use in health and beauty food products. The phenolic content in water and 60% ethanol extracts were $11.1{\pm}0.11$ and $4.18{\pm}0.05mg/g$, respectively. The DPPH radical scavenging activities of the water and ethanol extracts were 87% and 91% at $50{\mu}g$ phenolics/mL, respectively. At the same phenolics concentration, the respective extracts showed 84% and 95% for ABTS radical decolorization activities and 95% and 97% for TBARs. The antioxidant protection factors for the water and ethanol extracts at $200{\mu}g$ phenolics/mL were 1.87 and 2.22 PF, respectively. Enzyme inhibitory activities of the water and ethanol extracts ($50{\mu}g$ phenolics/mL) were 50.8% and 69.4% on angiotensin converting enzyme, 91% and 80% on xanthine oxidase, and 24% and 89% on ${\alpha}$-amylase, respectively. The tyrosinase inhibitory activities indicating skin-whitening were 47% and 25% for the water and ethanol extracts, respectively. Anti-wrinkle effect of the water extract was relatively higher than that of the ethanol extract. These results suggest that the water and ethanol extracts of Aster scaber can be used as an ingredient in health and beauty food products.

Effect of Black Garlic and Mugwort Extracts on Lipids Profile during Restraint Stress (흑마늘과 쑥 추출물이 구속스트레스를 가한 흰쥐의 체내 지질 성분에 미치는 영향)

  • This study investigated black garlic and mugwort extracts have anti-stress activity. The antioxidant activities of extracts from black garlic (BEP), mugwort (MEP), and three mixtures (MPA, 95:5; MPB, 90:10; MPC, 85:15, w/w% for BEP and MEP, respectively) were tested in vitro. DPPH and ABTS radical scavenging activities for the mixtures (MPA, MPB and MPC) were significantly elevated in a dose-dependent manner by the amount of mugwort extract. A restraint stress was imposed on six groups of Sprague-Dawley rats supplemented with an AIN-93 diet (RSC) or one of five kinds of hot water extract drinks containing (black garlic, RS1; mugwort, RS2; and mixtures of black garlic : mugwort at 95:5, RS3; 90:10, RS4, and a mixture of black garlic : mugwort : apple extract : xylitol=90.25:4.75:2:3, RS5; v/v%) for 4 weeks. The normal group was fed with the AIN-93 diet and not exposed to restraint stress. Food intake was higher in the group fed with garlic extract (RS1), while the body weight gain and food efficiency ratio did not significantly change. The total serum cholesterol content in the RS1 and RS2 groups was significantly lower than the RSC group (control), and the RS5 group was not significantly different compared to the RS3 group. The serum triglyceride content was significantly higher RS3~RS5 groups than RS1 and RS2 groups. In terms of HDL-C and LDL-C contents, AI and CRF in the serum were not significantly different between RS3 and RS5 groups. AST and ALP activities of RS1~RS5 groups were significantly lower than the RSC group. The liver total lipid and cholesterol contents of RS1~RS5 groups were significantly lower than RSC group, and triglyceride content was significantly lower in the RS1 group. Glycogen in the liver tissue was significantly higher in the RS2 and RS3 group compared to the RSC group. These results show that the intake of a mixture of black garlic and mugwort extracts may be effective in the alleviation of hyperlipidemia caused by restraint stress.

Effect of Dietary Intake of Salicornia herbacea L. Hot Water Extract on Anti-obesity in Diet-induced Obese Rats (함초 열수추출물의 섭취가 비만유도 흰쥐의 항비만 효과에 미치는 영향)

  • Salicornia herbacea L. is an annual herb that grows in salt marshes and salt fields along the seashores. It is also commonly used as a folk remedy in Korea. The objective of this study was to investigate the beneficial effects of Salicorrnia herbacea L. (SH) hot water extract on obesity. Five-week old male Sprague-Dawley rats (n=36) were divided into two groups and provided either a normal fat diet (11.5% fat from kcal) or a high fat diet (40.5% fat from kcal) for 6 weeks to induce obesity. Then, rats were blocked into six groups of six mice each and provided either a diet containing SH (0.5% of diet; g/kg) or a normal diet for another 6 weeks. Final body weights were significantly reduced when rats were fed SH among the high fat diet groups (HNS and HHS). Serum triglyceride concentrations significantly decreased in every group provided SH. HDL-cholesterol concentrations significantly increased in SH-fed groups among the high fat diet groups. Further, atherogenic index significantly decreased when rats were fed SH diet (HHS). There were no differences in LDL-cholesterol between the high fat diet groups, and glucose concentrations decreased when rats were fed SH diet (HNS). These results indicate that dietary intake of Salicornia herbacea L. hot water extract might have beneficial effects on obesity by reducing body weight, fat weights, and improving blood lipid profile.

Functional food activities of extracts from Pinus densiflora root (동송근(Pinus densiflora root) 추출물의 기능성식품 활성)

  • In this study, the antioxidative activity and functional food activities of water and ethanol extracts from Pinus densiflora root were examined. It was more effective to use ethanol than water when extracting phenolic compounds. The extracted phenolic compounds from Pinus densiflora root for biological activities were examined. The phenolic compounds extracted with water and 80% EtOH were $1.86{\pm}0.04mg/g$ and $6.85{\pm}0.16mg/g$, respectively. DPPH free radical scavenging activity of water and EtOH were each 86% and 85% at $100{\mu}g/mL$ phenolics, respectively. ABTS radical decolorization activity was 48% in water and 68% in EtOH at $200{\mu}g/mL$. Antioxidant Protection Factor (PF) were 1.74 PF in water and 1.96 PF in EtOH at $50{\mu}g/mL$. TBARs of water and EtOH were 93% and 98%, respectively at $100{\mu}g/mL$. The inhibition activity on xanthine oxidase was 83.7% in water extracts and 79.6% in ethanol extracts. Inhibition on xanthine oxidase of water and ethanol extracts showed a higher inhibition effect than allopurinol. The inhibition activity on ${\alpha}$-glucosidase was 14.8% in water extracts and 91.6% in ethanol extracts. The result suggests that P. densiflora root extracts may be useful as as functional food material.

Whitening effect of extracts from Matricaria chamomilla L. with B16F10 melanoma cells (B16F10 melanoma cell을 이용한 캐모마일(Matricaria chamomilla L.) 추출물의 미백 효과)

  • Matricaria chamomilla L. has been used as a bath agent in Europe because of its sterilization effect on the skin. Flowers contain terpenes, flavonoids are effective in relieving inflammation. Matricaria chamomilla L. has been reported to have various drug efficacies such as sedation, anti-diabetic effect and anti-arthritic effect, but there is little research on the scientific efficacy of whitening effect. The purpose of this study was to examine the whitening effect of Matricaria chamomilla L. extract and to investigate the mechanism of inhibition of melanogenesis. The extracts were used to determine tyrosinase inhibitory activity. The tyrosinase inhibitory activity of extracts was ineffective for water extract but in 60% ethanol extract was shown in a concentration-dependent manner. B16F10 melanoma cell was measured using a powder obtained by lyophilization of 60% ethanol extract. The toxicity was observed at a concentration of $75{\mu}g/mL$. And concentration range was selected to be at most $50{\mu}g/mL$. The effect of tyrosinase, MITF, TRP-1 and TRP-2 on the expression of melanin protein was investigated in melanoma cells of B16F10 melanoma cells. As a result, it was confirmed that as the concentration of the extract increased, the melanogenesis level decreased and the protein expression level also decreased in a concentration dependent manner. Therefore, it was concluded that Matricaria chamomilla L. extract inhibited melanogenesis in cells. Based on the above results, it is expected that it will be used as a useful basic data for industrialization of whitening functional food of Matricaria chamomilla L.

Resveratrol Ameliorates NMDA-induced Mitochondrial Injury by Enhanced Expression of Heme Oxygenase-1 in HT-22 Neuronal Cells (NMDA를 처리한 HT-22 신경세포에서 미토콘드리아 손상을 완화하는 레스베라트롤의 보호 효과와 헴 산화효소-1의 역할)

  • N-methyl-D-aspartate (NMDA) receptors have received considerable attention regarding their involvement in glutamate-induced neuronal excitotoxicity. Resveratrol has been shown to exhibit neuroprotective effects against this kind of overactivation, but the underlying cellular mechanisms are not yet clearly understood. In this study, HT-22 neuronal cells were treated with NMDA in Mg2+-free buffer and subsequently used as an experimental model of glutamate excitotoxicity to elucidate the mechanisms of resveratrol-induced neuroprotection. We found that NMDA treatment causes a drop in MTT reduction ability, disrupts inside-negative transmembrane potential of mitochondria, depletes cellular ATP levels, and stimulates intracellular ROS production. Double fluorescence imaging studies demonstrated an increased formation of mitochondrial permeability transition (MPT) pores accompanied by apoptotic cell death, while cobalt protoporphyrin and bilirubin showed protective effects against NMDA-induced mitochondrial injury. On the other hand, zinc protoporphyrin IX significantly attenuated the protective effects of resveratrol which was itself shown to enhance heme oxygenase-1 (HO-1) mRNA and protein expression levels. In cells transfected with HO-1 small interfering RNA, resveratrol failed to suppress the NMDA-induced effects on MTT reduction ability and MPT pore formation. The present study suggests that resveratrol may prevent mitochondrial injury in NMDA- treated HT-22 cells and that enhanced expression of HO-1 is involved in the underlying cellular mechanism.

Antioxidant Effect of Extracts Obtained from Four Aster Species Native to Korea (자생 쑥부쟁이속 식물 4종 추출물의 항산화 효과)

  • This study was conducted to evaluate the antioxidative potentials of 4 Aster species. Flowers and aerial parts were extracted with 80% ethanol, and antioxidative activities and bioactive substance contents were investigated. Flowers showed higher extraction yield than aerial parts of all 4 species used. Among the samples tested, Flowers of A. scaber were found to be the most effective in both DPPH and ABTS radical scavenging assay. However, $Fe^{2+}$ chelate effect of all samples were lower than EDTA. Four samples(A. scaber flowers, A. sphathulifolius aerial parts, A. koraiensis flowers, A. scaber aerial parts) out of eight showed stronger inhibitory activities against linoleic acid oxidation than BHT at 8 days. Total polyphenol and flavonoid contents of A. scaber flowers(90.46 and $75.36mg{\cdot}g^{-1}$) were the highest, and followed by A. sphathulifolius flowers.
