Study on the elution of biostimulant for in-situ bioremediation of contaminated coastal sediment (오염된 연안저질의 현장생물정화를 위한 미생물활성촉진제의 용출특성 연구)

  • A study on the elution characteristics of biostimulating agents (sulfate and nitrate) from biostimulants which are used for in-situ bioremediation for the coastal sediment contaminated with organic matter was performed. The biostimulating agents were mixed with the coastal sediment, and then massed the mixture into ball. Two kinds of ball type biostimulant were prepared by coating the ball surface with two different polymers, cellulose acetate and polysulfone. A granular type biostimulant (GTB) was also prepared by impregnating a granular activated carbon in the biostimulating agent solution. The image of scanning electron microscopy for the biostimulant coated with cellulose acetate (CAB) showed that the inner side of the coating layer consisted of irregular and bigger size of pores, and the surface layer had tight structure like beehive. For the biostimulant coated with polyfulfone (PSB), the whole coating layer had a fine structure without pore. The elution rate of the biostimulating agents for the CAB was higher than that for the PSB, and the elution rate for the GTB was considerably higher than that for the PSB in distilled water as well as in sea water. The elution rate of the biostimulating agents in turbulent water flow was about 3 times higher than that in standing water, and the elution rate of nitrate was higher than that of sulfate from the stimulating agents.

Physiological Activities of Hot Water Extract from Pine Bud (Pinus densiflora) (솔순 열수 추출물의 생리활성)

  • This research was performed to determine the antioxidant activity, nitrite scavenging activity, and its inhibitory activity on angiotensin converting enzyme (ACE), xanthine oxidase, $\alpha$-glucosidase, and elastase of hot water extract from pine bud (WPB). Antioxidant activity of WPB was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity. DPPH radical scavenging activity and SOD-like activity of WPB were remarkably increased in a dose-dependent manner, and were about 71.4 and 85.4% at 2 mg/mL, respectively. The xanthine oxidase and ACE inhibitory activities were about 70.9 and 51.9% at 2 mg/mL of WPB, respectively. Nitrite scavenging activity of WPB was about 59.1, 53.8, and 39.5% on pH 1.2, 3.0, and 6.0 at 2 mg/mL, respectively. The WPB also showed elastase and $\alpha$-glucosidase inhibitory effects. These results revealed that pine bud have strong antioxidant activity and positive effects on the inhibition of xanthine oxidase, ACE, and elastase.

Biological Activities of Specialized Crops in Namhae (남해군 특용작물의 생리활성)

  • This study was conducted to investigate the antioxidation and biological activities of 5 kinds of specialized crops (asparagus, aloe, curcumin, backnyuncho and garlic) cultivated in Namhae. All crops were freeze dried and then extracted with 10 folds of $60^{\circ}C$ hot-water, for 8 hours. Collected twice extracted water extract were also freeze dried. Dried extract samples were diluted with deionized water for examination. The total phenolics content was the highest observed in asparagus extract to $23.79{\pm}0.33mg/g$ and total flavonoids of aloe extract was the highest content $17.10{\pm}0.05mg/g$. In all samples, DPPH, ABTs and NO radical scavenging activity were depends on the concentration of the extract tended to increase. ABTs radical scavenging activity was the highest conducted $96.10{\pm}0.06%$ effect examined at $2,000{\mu}g/mL$ in asparagus hot-water extract. NO radical scavenging activity of hot-water extract from curcumin was the highest $250{\mu}g/mL$ concentration observed in $45.04{\pm}1.39%$. In all sample, tyrosinase activity was significantly inhibited with increasing of hot-water extract concentration. As a result, ${\alpha}$-glucosidase inhibition activity was the highest in hot-water extract from backnyuncho on $31.38{\pm}0.94%$ at $250{\mu}g/mL$.

Physiological activities of leaf extract of Lonicera morrowii A.Gray, a plant native to Ulleungdo (울릉도 자생식물인 섬괴불나무(Lonicera morrowii A.Gray) 잎 추출물의 생리활성)

  • This study was performed to investigate total phenolic contents, anti-oxidant, biological disease-related enzyme inhibitory, and anti-microbial effects of extracts of Lonicera morrowii leaves prepared with water and 40% ethanol. Anti-oxidative activities of the extracts increased in a dose-dependent manner and were very high even at low phenolic concentration. At phenolic concentrations ranging between 50 and 200 ㎍/mL, the water and ethanol extracts inhibited 39.34-76.35 and 47.53-99.83% of xanthine oxidase activity, 30.21-79.06, and 59.40-87.14% of angiotensin converting enzyme activity, and 59.81-80.35 and 36.06-63.58% of α-glucosidase activity, respectively. All inhibitory effect were increased in a dose-dependent manner. In the paper disc agar diffusion assay against seven microbes, L. morrowii extracts had anti-bacteria activities against Helicobacter pylori, Propionibacterium acne but no effect against other bacteria. These results indicate that the extracts from L. morrowii leaves have the possibility to be developed as a physiologically functional source for prevention of adult diseases.

Physiological Activities of Hot Water Extract and Solvent Fractions of Pleurotus ferulea (아위버섯 열수추출물 및 유기용매 분획물의 생리활성 효과)

  • To investigate the physiological activities of hot water extract and solvent fractions isolated from Pleurotus ferulea, antioxidative, fibrinolytic, thrombin inhibitory, and ${\alpha}$-glucosidase inhibitory activities were examined. Pleurotus ferulea, hot water extract was fractionated into hexane, chloroform, ethyl acetate, butanol and water fraction. Each of these was assayed individually. The antioxidative activities of ethyl acetate and butanol fractions were 86.79% and 87.82%, respectively. Using the fibrin plate method, only the ethyl acetate fraction showed a plasmin activity of 0.08 units/ml. Thrombin inhibitory activities of chloroform and ethyl acetate fractions were 74.90% and 71.08%, respectively. In the ${\alpha}$-glucosidase inhibitory activity test, butanol fraction showed the highest activity at 49.67%. From the above results, we anticipate that solvent fractions of Pleurotus ferulea can be used as a materials for the development of biofunctional foods for cardiovascular diseases.

  • 효소에 의한 펩티드합성법은 합성반응의 개발초기에는 용매로서 침전계를 이용하여 아미노산 혹은 펩티드기질에서 펩티드생성의 방향으로 평형화하는 방법을 많이 이용하였지만 최근에는 물에 섞이는 유기용매들을 수용액과 함께 사용하여도 합성반응이 이루어진다는 사실이 밝혀졌다. 하지만, 효소를 공업적으로 이용할 때 값이 너무 비싸고 합성물과의 분리, 정제에 어려움이 있다. 이러한 문제를 해결하기 위하여 효소의 고정화법이 널리 이용되고 있다. 한편 protease에 의한 펩티드의 역합성은 단백질공학의 한 수법인 반합성으로서 주목을 끌고 있다. DNA의 조작기술이 오늘날과 같이 발전하기 이전에는 단백질합성의 공학적기법은 이 반합성과 화학적수식으로 한정되었다. 본고에서는 protease를 이용한 생리활성펩티드의 합성에 필요한 protease의 종류, 이들의 펩티드합성작용기구, 합성설계조건 및 유용 생리활성펩티드의 합성 예를 소개하였다.

  • 지구상에서 생합성된 천연물은 완급의 차이는 있겠으나 궁극적으로는 CO$_{2}$ H$_{2}$O, NH$_{3}$ 등으로 다시 산화분해된다. 이 분해과정은 미생물의 효소반응으로 이루어지며 생물권의 항상성도 일차적으로는 미생물의 이 산화분해작용으로 유지된다고 볼 수 있다. 미생물이 영위하는 이러한 효소반응을 생리활성물질의 공업적합성에 이용한 초기의 예로는 부현피질호르몬의 합성과정에서 Rhizopus nigricans를 사용한 수산화반응과 ascorbic acid 합성과정에서 Acetobacter xylinum을 사용한 탈수반응이 유명하며 이러한 반응은 아직도 이용되고있다. 특히 지난 십수년간의 응용미생물학의 발전은 대단한 것이며 여러가지 항생물질, 당질, amino산, 핵산관련물질들이 생리활성물질과 함께 발효법으로 생산되고 있다. 그러나 이글에서 다루는 미생반응은 발효과정에서 생산되는 미생물자체의 일차적 대사산물 (primary metabolite)이나 이차적대사산물 (secondary metabolite)를 대상으로 한것이 아니며 어디까지나 외부에서 공급되는 화학물질에 대한 균체의 효소반응산물을 목적물로 하고있다. 또한 근래 활발히 이용되는 균체고정및 효소고정법을 이용한 효소공학적수법도 제외하고 배양과정의 균체 또는 배양후의 군체를 이용한 미생물반응에 한정코저 한다.

Physiological Activities of Agaricus bisporus Extracts as Affected by Solvents (용매에 따른 양송이 추출물의 생리활성 효과)

  • Physiological activities of Agaricus bisporus extracts were examined. Nitrite-scavenging activity, superoxide dismutase (SOD)-like activity and electron-donating ability (EDA) of Agaricus bisporus extracted with water, 50%, and 100% ethanol were measured. Nitrite- scavenging activities at pH 1.2 were the most effective in water and 50% ethanol extracts from brown and white Agaricus bisporus samples. SOD-like activities of water and 50% ethanol extracts of both samples were $19.02{\sim}55.42%$ lower than those of 1.0% and 0.1% L-ascorbate solutions. SOD-like activities of brown Agaricus bisporus extracts were higher than those of white Agaricus bisporus extracts, and water extracts of the samples were the highest. EDAs of brown Agaricus bisporus extracts were higher $(40.35{\sim}66.54%)$ than those of white Agaricus bisporus extracts, while those of both extracts were lower than 1.0% and 0.1% L-ascorbate solutions. The results will be useful for understanding the physiological activities of Agaricus bisporus extracts.

Physiological Activities of Ginkgo biloba Sarcotesta Extract with Heat Treatment (열처리에 따른 은행 외종피 추출물의 생리활성)

  • This study was performed to investigate the physiological activities of Ginkgo biloba sarcotesta extracts before and after heat treatment. G. biloba sarcotesta was heated at $130^{\circ}C$ for 2 h and extracted with water, 70% ethanol and 80% methanol. ABTS and DPPH radical scavenging activities increased after heating in the water (14.95 mg AAE/g and 7.36 mg TE/g) and ethanol extracts (12.20 mg AAE/g and 6.23 mg TE/g). ${\alpha}$-Glucosidase inhibitory activity decreased after heating in all but the water extract. Angiotensin converting enzyme I inhibitory activities decreased after heating in all extracts. Nitric oxide production inhibitory activity increased from 12.40~44.55% of the raw sample to 40.76~72.39% of the heated sample at a concentration of $200{\mu}g/mL$. Lipid accumulation inhibitory activities were similar before and after heat treatment. The highest antiproliferative effects on MCF-7 human breast cancer cell lines were observed in 80% methanol extract in the heated sample. Cell viability at concentrations of 25, 50, 100, and $200{\mu}g/mL$ measured 34.88, 17.58, 8.44 and 10.48%, respectively. From the results, the antioxidant and antiproliferative activities of G. biloba sarcotesta extracts increased with heat treatment, and research on the identification of the structure for the active compounds are needed in further studies.

Biological activities of Hovenia dulsis $T_{HUNB}$ (헛개나무의 생리활성 탐색)

  • Four different parts of Hovenia dulcis $T_{HUNB}$; fruit, bark, vessel area, fruit coat were extracted with water and ethanol. The ethanol extracts of bark, fruit coat and fruit were fractionized into diethyl ether, chloroform and aqueous partitions. Ethanol extract of fruit coat increased the activity of cathepsin B up to 55 %, which can enhance the alcohol dehydration in the liver. The ethanol extracts was more effective than water extracts against the growth of Hep3B, MCF7. The ethanol extracts of bark (0.5mg/ml) inhibited 90% the growth of MCF7. Each extracts and fractions (0.5mg/ml) did not show considerable cytotoxicity on HEL299. In overall, most of the fractions had similar effects to ethanol extracts; however, diethyl ether and chloroform fractions had higher bioactivity than ethanol extracts, but aqueous fraction.
