Monitoring for the Resistance of Strobilurin Fungicide Against Botrytis cinerea Causing Gray Mold Disease (Strobilurin계 살균제에 대한 잿빛곰팡이병균 Botrytis cinerea의 저항성 검정)

  • This study was conducted to investigate the resistance of Botrytis cinerea to azoxystrobin, which belongs to strobilurin fungicides. The sensitivity of B. cinerea isolates, which were collected from infected pepper, strawberry, cucumber and tomato by a single spore isolation, to the fungicide was tested through a agar dilution method on PDA amended with fungicides and $100{\mu}g\;mL^{-1}$ of salicylhydroxamic acid (SHAM). All isolates of B. cinerea tested in this study were classified as a sensitive and a resistant group by $1.0{\mu}g\;mL^{-1}$ of $EC_{50}$ value to azoxystrobin. While the sensitive isolates accounted for 46.5% of B. cinerea population, the resistant ones did for 53.5%. According to the regions isolating B. cinerea, the highest isolation frequency was showed as 81.1% in Chungnam among the all. Among 4 host plants as pepper, strawberry, cucumber and tomato, the highest isolation frequency was obtained in strawberry, while the lowest was done in pepper. The isolate resistant to azoxystrobin showed the cross resistance to other fungicides included into strobilurins as kresoxim-methyl and trifloxystrobin. In spite of an excellent efficacy of strobilurins, it should be taken care to use them in the field, because of the high risk in the fields.

The control effect of some fungicides against cucumber sclerotinia rot and the sensitivity of sclerotinia isolates to fungicides (오이 균핵병에 대한 몇 가지 살균제의 방제 효과와 살균제에 대한 균핵병균의 감수성 정도 조사)

  • As Sclerotinia sclerotiorum causing cucumber sclerotinia rot was the fastest in the mycelial growth at $25^{\circ}C$, its pathogenicity was strong at the same temperature among several temperatures. All the isolates of Sclerotinia sclerotiorum showed a strong pathogenicity against cucumber fruits, which was confirmed by a disk assay and a wound assay. A wound assay was superior to a disk assay to develop the assay system for assessing the fungicidal activity of several fungicides against Sclerotinia sclerotiorum. In a disk assay, it was very difficult to assess the fungicidal activity, because the pathogenicity of isolates used in the experiment was very strong. At 500 and $3.0{\mu}g/mL$, the activity of dichloflouanid and the mixture of carbendazim and diethofencarb against cucumber sclerotinia rot was 14.3 and 42.3%, respectively, by using a disk assay. However, at same concentration two fungicides showed the high controlling activity as 100 and 92.5%, through a wound assay in a laboratory. Also, the activity of two fungicides was good against cucumber sclerotinia rot in the greenhouse where cucumber plants were cultivated in the field, showing the control value as 91.1 and 82.9% at 100 and $825{\mu}g/mL$, respectively. All the isolates of Sclerotinia sclerotiorum from cucumber fruits sampled in the polyvinyl house were subjected to monitoring for the resistance to 7 fungicides. The $EC_{50}$ value of 7 fungicides was as follows: fenhexamid; $0.13{\mu}g/mL$, procymidon and iprodione; 0.18 and $0.24{\mu}g/mL$, carbendazim and the mixture of carbendazim and diethofencarb; 0.13과 $0.05{\mu}g/mL$, iminoctadine and dichlofluanid; 1.94 and $8.95{\mu}g/mL$. Ultimately it was not found that resistant isolates of Sclerotinia sclerotiorum were appeared in the field.

Difference of Susceptibility on Bentgrass Cultivars to Pink Snow Mold Caused by Microdochium nivale (벤트그라스 품종이 Microdochium nivale에 의한 자주색설부병에 대한 감수성 차이)

  • The susceptibility of cultivars of three bentgrass species (creeping, colonial, and velvet bentgrass) was evaluated on detached leaves assays with pink snow mold 9 isolates caused by Microdochum nivale in Petri dishes and whole plants under controlled conditions. The pink snow mold isolates obtained from infected turfgrasses on golf courses in Wisconsin were tested on response of fungicides and temperature. Detached leaf assay and susceptibility of bentgrass cultivars were evaluated with potted adult seeding during 80 days. Nine isolates were susceptible to two fungicides and were significantly different among isolates. Mycelial growth was varied in response of temperatures among isolates. There were significant differences in development and colonization of the fungus on detached leaf assay among bentgrass species include culvitars. There were significant differences on whole plants in disease severities among the three bentgrass species, particularly between tetraploids (creeping and colonial) and diploid (velvet) species, and among cultivars within each species, indicating that there are varying levels of susceptibility in species and cultivars to M. nivale. This study could be applied to evaluate the susceptibility of bentgrass to pink snow mold and also to predict a prospective evaluation of bentgrass cultivars to pink snow mold in fields in a breeding program.

Sensitivity of Sclerotium cepivorum Causing Garlic White Rot Isolated from Taeahn, Seosan and Goheung Areas to Fungicides (서산, 태안 및 고흥 지역에서 마늘 흑색썩음병을 일으키는 Sclerotium cepivorum의 살균제 감수성 조사)

  • To investigate the sensitivity of Sclerotium cepivorum causing garlic white rot to 5 fungicides, including prochloraz, tebuconazole, flutolanil, iminoctadine and isoprothiolane, 147 isolates isolated from infected garlics from 2008 to 2009 through a single sclerotium isolation were screened. While each mean value of $EC_{50}$ (effective concentration reducing mycelial growth by 50%) of S. cepivorum isolates collected in 2008 to each fungicide was 0.054, 0.012, 23.189, 0.901, and $21.362{\mu}g\;mL^{-1}$, that of 2009 isolates were 0.030, 0.020, 10.367, 1.684, $33.406{\mu}g\;mL^{-1}$. There was a difference in mean value of $EC_{50}$ of S. cepivorum according to regions isolated. $EC_{50}$ values of S. cepivorum isolated in Goheung to flutolanil and isoprothiolane were 14.468 and $24.653{\mu}g\;mL^{-1}$, respectively, which was lower than those of Seosan and Taeahn. Isolates from Taeahn showed the lowest $EC_{50}$ value to prochloraz as $0.008{\mu}g\;mL^{-1}$. In addition, we could not find any resistant isolates to fungicides tested. The $EC_{50}$ values in this study will be used in a fungicide resistance monitoring program to determine whether shifts in sensitivity to fungicides included into different groups are occurring in S. cepivorum populations.

Establishment of Baseline Sensitivity of Phytophthora capsici Causing Pepper Phytophthora Blight to Carboxylic Acid Amide Fungicides (Carboxylic acid amide계 살균제에 대한 고추 역병균 Phytophthora capsici의 감수성 기준 설정)

  • Baseline sensitivity to benthiavalicarb, iprovalicarb and dimethomorph included into carboxylic acid amide (CAA) group was evaluated in 180 isolates of Phytophthora capsici over 4 years from 2005 to 2008. $EC_{50}$ (effective concentration inhibiting mycelial growth by 50%) value of benthiavalicarb ranged from $0.015{\mu}g\;mL^{-1}$ to $0.049{\mu}g\;mL^{-1}$ with a mean of $0.033{\mu}g\;mL^{-1}$. The mean values of $EC_{50}$ of iprovalicarb and dimethomorph were 0.411 (0.197 - 0.556) ${\mu}g\;mL^{-1}$ and 0.271 (0.101 - 0.798) ${\mu}g\;mL^{-1}$, respectively. Although there was no increasing tendency in $EC_{50}$of benthiavalicarb and iprovalicarb during 4 years, $EC_{50}$ of dimethomorph was increased gradually by laps of time. There was no cross-resistance between each fungicide used in this study and metalaxyl. Among fungicides included into CAA group, there was a positive correlation between benthiavalicarb and iprovalicarb, and between dimethomorp and mandipropamid.

Monitoring for the Resistance of Botrytis cinerea Causing Gray Mold Against Mepanipyrim (Mepanipyrim에 대한 잿빛곰팡이병균의 저항성 검정)

  • With 86 isolates of Botrytis cinerea obtained from diseased fruits of tomato, strawberry and cucumber, it was conducted to detect the fungicide resistance of B. cinerea against mepanipyrim through an agar dilution method. FGA medium was used for monitoring the resistance. Among 86 isolates of B. cinerea, resistant isolates were composed by 23.3%, which was different according to regions isolating the pathogen. In accordance with isolation region, the isolation frequency of resistant isolates was as follows; that of Gyeonggi, Gyeongnam/Pusan and chungnam was 28.6%, 33.3% and 12.8%, respectively. The resistant isolates of B. cinerea showed the cross resistance between mepanipyrim and pyrimethanil. Also the pathogenicity of B. cinerea resistant to mepanipyrim was high as like that of sensitive isolates. Because of them, it should be necessary to manage the spraying system of mepanipyrim and the monitoring for the fungicide resistance.

The Effectiveness of a New Systemic Fungicide EL-291 for the Control of Rice Blast Disease (새로운 침투성살균제 EL291의 벼 도열병 방제효과)

  • Experiments were carried out to determine the effectiveness of a new systemic fungicide EL-291 (5-Methyl-1, 2,4-triazolo (3,4-b) benzothiazole) for the control of rice blast disease in greenhouse and paddy field. The efficiency of EL-291 was much greater when applied before inoculation than when applied after inoculation. Kasugamin and Benlate were most effective as eradicants. For control of leaf blast, effectiveness of EL-291 was not significantly different than either Kasugamin or Benlate. However, EL-291 was considered more economical and reliable than either Kasugamin or Benlate. EL-291 required only a single foliar application or a transplant root soak, whereas two applications of the other fungicides were required. EL-191 was also more effective against panicle blast when applied only once, compared with two applications of Kasugamin or Benlate. The highest riceyields were obtained in plots treated with EL-29l.

Decomposition Characteristics of Fungicides(Benomyl) using a Design of Experiment(DOE) in an E-beam Process and Acute Toxicity Assessment (전자빔 공정에서 실험계획법을 이용한 살균제 Benomyl의 제거특성 및 독성평가)

  • We investigated and estimated at the characteristics of decomposition and mineralization of benomyl using a design of experiment(DOE) based on the general factorial design in an E-beam process, and also the main factors(variables) with benomyl concentration(X$_1$) and E-beam irradiation(X$_2$) which consisted of 5 levels in each factor was set up to estimate the prediction model and the optimization conditions. At frist, the benomyl in all treatment combinations except 17 and 18 trials was almost degraded and the difference in the decomposition of benomyl in the 3 blocks was not significant(p > 0.05, one-way ANOVA). However, the % of benomyl mineralization was 46%(block 1), 36.7%(block 2) and 22%(block 3) and showed the significant difference of the % that between each block(p < 0.05). The linear regression equations of benomyl mineralization in each block were also estimated as followed; block 1(Y$_1$ = 0.024X$_1$ + 34.1(R$^2$ = 0.929)), block 2(Y$_2$ = 0.026X$_2$ + 23.1(R$^2$ = 0.976)) and block 3(Y$_3$ = 0.034X$_3$ + 6.2(R$^2$ = 0.98)). The normality of benomyl mineralization obtained from Anderson-Darling test in all treatment conditions was satisfied(p > 0.05). The results of prediction model and optimization point using the canonical analysis in order to obtain the optimal operation conditions were Y = 39.96 - 9.36X$_1$ + 0.03X$_2$ - 10.67X$_1{^2}$ - 0.001X$_2{^2}$ + 0.011X$_1$X$_2$(R$^2$ = 96.3%, Adjusted R$^2$ = 94.8%) and 57.3% at 0.55 mg/L and 950 Gy, respectively. A Microtox test using V. fischeri showed that the toxicity, expressed as the inhibition(%), was reduced almost completely after an E-beam irradiation, whereas the inhibition(%) for 0.5 mg/L, 1 mg/L and 1.5 mg/L was 10.25%, 20.14% and 26.2% in the initial reactions in the absence of an E-beam illumination.

Identification of Quantitative Trait Loci Associated with Anthracnose Resistance in Chili Pepper (Capsicum spp.) (고추 탄저병 저항성 관련 양적형질 유전자좌 분석)

  • Pepper ($Capsicum$ spp.) anthracnose caused by $Colletotrichum$ $acutatum$ is a destructive disease susceptible to areas where chili peppers are grown. $Capsicum$ $baccatum$ var. $pendulum$ (Cbp) is resistant to anthracnose and has actively been used for interspecific hybridization for the introgression of resistance gene(s) into cultivated chili peppers. The goals of this study were to determine the inheritance of resistance to anthracnose within $Capsicum$ $baccatum$ and to map quantitative trait loci (QTLs) for the anthracnose resistance. A genetic mapping population consisting of 126 $F_2$ plants derived from a cross between $Capsicum$ $baccatum$ var. $pendulum$ (resistant) and $Capsicum$ $baccatum$ 'Golden-aji' (susceptible) was used for linkage mapping. The linkage map was constructed with 52 SSRs, 175 AFLPs, and 100 SRAPs covering 1,896cM, with an average interval marker distance of 4.0cM. Based on this map, the number, location, and effect of QTLs for anthracnose resistance were studied using plants inoculated in the laboratory and field. A total of 19 quantitative trait loci (2 major QTLs and 16 minor QTLs) were detected. Two QTLs ($An8.1$, $An9.1$) showed 16.4% phenotypic variations for anthracnose resistance after wounding inoculation. In addition, five minor QTL loci ($An7.3$, $An7.4$, $An4.1$, $An3.1$, $An3.2$) showed a total of 60.73% phenotypic variations of anthracnose resistance in the field test. Several significant QTLs were also detected and their reproducibility was confirmed under different inoculation conditions. These QTLs are now being confirmed with different breeding populations. Markers tightly linked to the QTLs that are reliable under different environmental conditions will help to determine the success of marker-assisted selection for anthracnose -resistant breeding programs in chili pepper.

Fungicide Resistance of Gibberella fujikuroi Isolates Causing Rice Bakanae Disease and Their Progeny Isolates (벼키다리병균(Gibberella fujikuroi) 균주 및 교배 후대균주의 살균제에 대한 저항성)

  • A total of 25 isolates of Fusarium fujikuroi were obtained from diseased rice plants in Korea from 2006 to 2007 to assess their resistance against fungicides prochloraz and benomyl + thiram. Minimal inhibitory concentration (MIC) values of F. fujikuroi isolates were examined by agar dilution method. Most of the isolates were sensitive to the fungicides. Out of 25 isolates, six were resistant to prochloraz and three to benomyl + thiram. In addition, the isolates CF245, CF249 and CF337 showed resistant to both fungicides. The progenies ($F_1$ isolates) obtained through two different crosses between sensitive parental isolates(CF202, CF232 and CF179) and resistant parental isolate (CF337) were evaluated for their mycelial growth at different temperatures and resistance against fungicides. Mycelial growth rate of $F_1$ isolates originated from CF202 $\times$ CF232 was similar to the parental isolates. However mycelial growth rate of $F_1$ isolates originated from CF179 $\times$ CF337 was faster than their parent isolates. In case of prochloraz, distribution ratio of sensitivity(S) to resistance(R) against to the fungicide of $F_1$ isolates originated from CF202 $\times$ CF232 and CF179 $\times$ CF337 was 86 : 14 and 78 : 22, respectively. In case of benomyl+thiram, all the $F_1$ isolates originated from CF202 $\times$ CF232 were sensitive to the fungicide, however ratio of sensitivity(S) to resistance(R) against to the fungicide of $F_1$ isolates originated from CF179 $\times$ CF337 was 35 : 65.
