Patterns of Uptake and Removal by Processing Types of Triazole Fungicides in Onion (Triazole계 살균제의 양파 흡수이행 및 조리 형태별 소실 양상)

  • Uptake of the triazole fungicides, fluquinconazole and tetraconazole from shoot part of onion was assessed by determining residual amounts of applied fungicides in edible and shoot parts of onion after the foliar application. Combined product of fluquinconazole and tetraconazole (14:7, v/v) as a 21% active ingredient of suspended emulsion formulation was diluted at ratio of 500 and 200 times and sprayed on the shoot part of onion after sealing its root part with absorbent paper. At 10 days after the pesticide application, fluquinconazole residue in the shoot part was the greatest as 5.2 mg/kg at 200 times-dilution treatment, while tetraconazole residue in this part was the smallest as 1.2 mg/kg at 500 times-dilution treatment. On the other hand, the pesticide residues in edible parts of onion at all the treatments were less than limits of detection, 0.01 mg/kg. However, fluquinconazole residues in the edible part of onion divided into three groups such as 1st, 2nd, and 3rd layers were detected at concentrations of 0.04 or 0.24 mg/kg, and these results show the different distribution of pesticides in onion depending on divided layers. In addition, chopped onions were soaked in pesticide solutions prepared with dilution of 1,000 times, cooked using three food processing types such as boiling, stir frying, and pickling, and the pesticide residues in them were analyzed. The analyzed results showed the largest pesticide dissipation in onion followed boiling process (76.9~92.6%).

Establishment of Pre-Harvest Residue Limit(PHRL) of the Fungicide Amisulbrom during Cultivation of Winter-Grown Cabbage (엇갈이배추 재배기간 중 살균제 Amisulbrom의 생산단계 잔류허용기준 설정)

  • BACKGROUND: Supervised residue trials were conducted to establish pre-harvest residue limit(PHRL), a criterion to ensure the safety of the pesticide residue in the crop harvest, of amisulbrom for winter-grown cabbage in two fields. Following to application of amisulbrom on the crop, time-course study was carried out to obtain the amisulbrom dissipation of statistical significance which enabled to calculate the predicted values of PHRL. METHOD AND RESULTS: During cultivation under greenhouse condition, samples of winter-grown cabbage were collected at 0, 1, 3, 5, 7 and 10 days after amisulbrom application, and subjected to residue analysis. Analytical method was validated by recoveries ranging 93.7~100.0% as well as limit of quantitation(LOQ) of 0.04 mg/kg. Amisulbrom residues in winter-grown cabbage gradually decreased as time elapsed. The dissipation rate of the residue would be affected by intrinsic degradation along with dilution by the cabbage growth. The decay pattern was well fitted by the simple first-order kinetics. CONCLUSION: Biological half-lives of amisulbrom in winter-grown cabbage ranged 3.7~4.1 days in two field conditions. Based on the regression of amisulbrom dissipation, PHRLs of amisulbrom in winter-grown cabbage were recommended as 8.86~9.47 and 4.21~4.35 mg/kg for 10 and 5 days before harvest, respectively.

Improvement of Market Quality of Minimally Processed Horticultural Products Using Ozone (오존수를 활용한 부분 가공 원예산물의 청정화 및 유통성 증진)

  • The bacterial density of partially processed horticultural crops in the commercial market was relatively high probably due to improper handing or processing. Between crops, the bacterial density was low in peeled garlic and ranged above $10^8cfu{\cdot}g^{-1}fw$ in other crops. Especially in brake fern, the bacterial density was the highest and non food materials such as packing strips were found. There was difference in the effects of ozone-treated water washing treatment between crops. In root crops such as burdock and lotus root, ozone showed positive effect on preventing discoloration as well as lowing bacterial density resulting in the increase of marketing period. Any positive effect was not found when sliced crops were treated over 5 min at $0.3mg{\cdot}L^{-1}$ of dissolved ozone. In lettuce slices, leaf tissues were severely discolored and rapidly soften at $0.3mg{\cdot}L^{-1}$ for 2 min dipping. Ozone was highly effective on inhibiting bacterium propagation and off-flavor. This effect was more stronger in Fischer ligulariata than brake fern, probably due to the difference of tissue thickness. Results indicated that ozone-treated water washing had a strong potential to improve the market quality of partially processed horticultural crops including dried wild greens and sliced root crops.

Dissipation Pattern of a Fungicide Mandipropamid in Korean Cabbage at Different Harvest Times under Greenhouse Conditions (시설재배 엇갈이배추 중 살균제 Mandipropamid의 수확시기별 잔류 특성)

  • Supervised residue trials for mandipropamid in Korean cabbage(Brassica campestris L.) were conducted to establish its pre-harvest residue limit (PHRL), a criterion to ensure the safety of the terminal pesticide residue during cabbage production. Tissues of Korean cabbage were collected at 0, 1, 3, 5, 7, and 10 days after mandipropamid application and subjected to residue analysis. The analytical method was validated by recoveries ranging from 88.2-92.2% at two levels (0.4 and $2.0mg{\cdot}kg^{-1}$), and a limit of quantitation (LOQ) of $0.04mg{\cdot}kg^{-1}$. Mandipropamid residues in Korean cabbage gradually decreased over time. The dissipation rate of the residue would be affected by intrinsic degradation of the compound along with dilution resulting from the fast growth of Korean cabbage. The decay pattern was well fitted by simple first-order kinetics. Biological half-lives of mandipropamid in Korean cabbage ranged from 3.9-4.0 days in two field conditions. Calculated by the regression curve of mandipropamid dissipation, the PHRLs of mandipropamid in Korean cabbage were recommended as 11.07-12.19 and $5.76-6.05mg{\cdot}kg^{-1}$ for 10 and 5 days prior to harvest, respectively.

Qualities of cocoon and Cocoon filament of the Precocious Trimolting Silkworm, Bombyx mori, induced by Trifumin (Imidazole계 살균제 Trifumin에 의한 조수거3면충의 견사질)

  • In order to investigate the possibility of fine-size cocoon production by the use of imidazole compound, Trifumin, the cocoon and cocoon filament qualities, dietary efficiency and fibroin component were examined. cocoon size, cocoon weight and cocoon shell weight of trimolter were smaller than those of tetramolter. Cocoon shell weight of trimolter was 58% and 26% of that of tetramolter in the 3rd instar treated larvae and in the 4th instar treated larvae, respectively. Amount of cocoon shell produced per 1g dry matter ingested was 94% and 85% of that of tetramolter in the 3rd instar treated larvae and in the 4th instar larvae, respectively. The effects of Trifumin on the reeling tests were different according to the stage of the treatment. When Trifumin was treated at the 3rd instar larvae, reelability of cocoon filament, amount of cocoon filament, size of cocoon filament, reelability percentage and raw silk percentage of cocoon were 100%, 60%, 60%, 97% and 90%, respectively. When Trifumin was treated at the 4th imstar larvae, most of reelability tests were inferior to those of the 3rd instar treated larvae, but reelavility percentage was increased 13%. The Trifumin treatment did not affect the electrophoretic patterns of cocoon fibroin.

In vitro selection of fungicides for control of leaf blight of sweet persimmon tree caused by Pestalotiopsis theae (In vitro에서 단감나무 둥근갈색무늬병 방제를 위한 살균제 선발)

  • In vitro experiments, several fungicides including prochloraz, tebuconazole, fluazinam, fludioxonil, and iminoctadine-triacetate showed more than 85% inhibition of mycelial growth of Pestalotiopsis theae (SP-3). Dichlofluanid and chlorothalonil inhibited mycelial growth at the rate of 10 and 33%, however benomyl did not inhibit mycelial growth of the fungus. Minimum inhibitory concentration(MICs) of iminictadine-triacetate on the mycelial growth of SP-3 isolate was $10{\mu}g/m{\ell}$, but that of P. theae $\underline{MAFF}$ 752002 and P. longiseta $\underline{MAFF}$ 752001 was $1{\mu}g/m{\ell}$. MIC of benomyl, chlorothalonil, dichlorofluanid was $1,000{\mu}g/m{\ell}$, and that of fludioxonil, fluazinam, tebuconazole was $10{\mu}g/m{\ell}$. Conidial germination was inhibited more than 80% in tile medium which contained $1{\mu}g/m{\ell}$ of prochloraz, tebuconazole, fluazinam, fludioxonil, and minoctadine-triacetate. Control values of benomyl, chlorothalonil and dichlofluanid were 40, 60 and 30%, respectively. The controlling effect of iminoctadine-triacetate ($10{\mu}g/m{\ell}$) aganist P. theae (SP-3) in leaf disc test was more than 93%, but benomyl and dichlofluanid could not control leaf blight disease caused by P. theae (SP-3). Tebuconazole, fluazinam, fludioxonil, and iminoctadine-triacetate showed more than 94% of protective and curative effect of leaf blight of sweet persimmon, while, benomyl and dichlofluanid did not show any control effect of the disease.

Establishment of Analytical Method for Pencycuron in Representative Agricultural Commodities by High-Performance Liquid Chromatography (대표 농산물 중 살균제 Pencycuron의 HPLC 정밀 잔류분석법 개발)

  • The single residue analytical method was developed for determining fungicide pencycuron residues in various agricultural commodities with high-performance liquid chromatography (HPLC). Pencycuron residue was extracted with acetone from representative crops such as Korean cabbage, apple, brown rice and green pepper. After ethyl acetate/n-hexane partition and subsequent clean-up with silica gel chromatography, pencycuron residue was quantified by reversed phase HPLC with UV detection at 240 nm. The suspected residue of pencycuron was confirmed using selected-ion monitoring (SIM) LC/mass spectrometry (MS). Instrumental limit of quantitation (ILOQ) and method LOQ (MLOQ) were set at 2 ng and 0.02 mg/kg, respectively. Overall recoveries of pencycuron from different crop samples fortified at three levels (MLOQ, 10MLOQ, 100MLOQ) were 72~108%. This proposed method could be useful as official analytical method for quantification of pencycuron residues in agricultural commodities.

Residue Analysis to Establish an Index for the Safety Use of Propineb (Propineb의 안전사용(安全使用) 기준(基準) 설정(設定)을 위한 잔류분석(殘留分析))

  • Residue of Propineb in Sesame(Sesamum indicum L.) seed was determined to establish an index for the safety use of Propineb to Sesame. Evaluation was made on residual concentration of Propineb in Sesame seed as a function of application frequency and date when the mixed formulations of Propineb(56%), protectant fungicide, and Oxadixyl(8%), contact fungicide, were sprayed into Sesame leaves. Level of Propineb treatment was $0.028g/m^2$ with various combinations of application time from three to sixty days before harvest. Results are summarized as followings. 1. Recovery percentages of Propineb from Sesame seed were ranged from 84 to 96, and the minimum detectable limit of Propineb with the method employed in this experiment was 0.03mg/kg. 2. Residues of Propineb in Sesame seed were in the ranges of 0.14 to 1.38mg/kg, varying with frequency and date of Propineb application. 3. Residues of Propineb increased as increasing application frequency of Propineb or as being application date closer to harvest time. 4. Residue of Propineb in Sesame seed was decreased with time, showing to be fitted to the first-order kinetics. 5. Residues of Propineb in Sesame seen were, irresepective of treatments, lower than 2mg/kg, the Maximum Residue Limit(MRL) established by FAO/WHO. 6. Half-life of Propineb determined in this experiment was ranged from 12 to 16 days.

Establishment of Rice Bakanae Disease Management Using Slightly Acidic Hypochlorous Acid Water (미산성 차아염소산수를 이용한 벼키다리 병 방제)

  • Rice bakanae is a typical seed-borne and seed-transmitted disease caused by infection by Fusarium fujikuroi. Seed disinfection using chemical fungicides (such as benomyl and prochloraz) is most effective in controlling the disease, but the emergence of fungicide-resistant strains has recently been increasing. Slightly acidic hypochlorous acid water (SAHW) is a safe and environmentally friendly disinfectant that has a potent and broad spectrum of antimicrobial activity against viruses, bacteria, and fungi. In this study, we aimed to investigate the effectiveness of SAHW against F. fujikuroi strains, including chemical fungicide-resistant strains, as an alternative to conventional chemical fungicides in the management of bakanae disease. SAHW showed strong but similar levels of antifungal activity among the F. fujikuroi strains with a minimum inhibitory concentration (MIC90) of 5±2.5 ppm of free available chlorine (FAC). In addition, F. fujikuroi cells lost viability completely within 5 min of SAHW treatment due to the lethal damage to cell integrity. When the rice seeds infected by F. fujikuroi were treated with SAHW containing 20±10 ppm of FAC for 12 hr, the efficiencies of seed disinfection and disease control were 95-98% and 90.1-92.6%, respectively. Altogether, our data suggest that SAHW is an effective compound for controlling rice bakanae disease.

Simultaneous determination for fungicide prochloraz and its metabolites in animal commodities with GC-ECD after hydrolysis (가수분해 후 GC-ECD를 이용한 축산물 중 살균제 Prochloraz 및 그의 대사물의 동시분석)

  • The analytical method was established for simultaneous determination of fungicide prochloraz and its metabolites in several animal commodities using gas chromatography (GC) coupled with electron capture detector (ECD). Samples including beef meat, pork meat, chicken meat, milk, and egg were hydrolyzed with pyridine hydrochloride which converts prochloraz and its metabolites to 2,4,6-trichlorophenol (2,4,6-TCP) because residue definition for prochloraz was 'sum of prochloraz and its metabolites containing the 2,4,6-trichlorophenol moiety, expressed as prochloraz', for compliance with MRLs from animal commodities. Therefore, residual prochloraz was extracted with acetone, decomposed to 2,4,6-TCP, partitioned with dichloromethane, purified with aminopropyl SPE and quantified as 2,4,6-TCP with GC-ECD. The instrumental limit of quantitation and method LOQ (MLOQ) was 0.01 ㎍/mL and 0.02 mg/kg for prochloraz and 0.005 ㎍/mL and 0.01 mg/kg for 2,4,6-TCP, respectively. The linearity of the calibration curve was good with R2 >0.995 in the range of 0.005-0.2 ㎍/mL. Fortification levels of prochloraz were 0.02 mg/kg (MLOQ) and 0.2 mg/kg (10MLOQ) for recovery tests. Overall recoveries of prochloraz were >90% with <10% of coefficient variation (C.V.). This established analytical method was fully validated and could be useful for quantification of prochloraz and its metabolites in animal commodities as official analytical method.
