Production of Host-specific Toxin by Alternaria mali and its Biological Activity (사과점무늬낙엽병균(Alternaria mali)이 생성(生成)하는 기주특이적(寄主特異的) 독소(毒素)와 그의 생물활성(生物活性))

  • Pathogenic isolates of Alternaria mali produced host-specific toxin(AM-toxin) in liquid culture. The toxin was also released by germinating spores of the fungus. The physiological event of apple leaves induced by germinating spores was an increased loss of electrolytes from susceptible leaves. This reaction was evident soon after spore inoculation, indicating that the leakage was caused by AM-toxin from germinating spores. Typical symptoms were developed only in susceptible leaves of apple within 48hr after inoculation with pathogenic spores. Similar symptoms occurred on susceptible leaves when non-pathogenic isolates plus AM-toxin were used.

Quality Evaluation of Basalt Aggregates from JEJU Island (제주산 현무암의 콘크리트용 골재 사용을 위한 품질 특성 평가)

  • This study was carried out to assess the suitability in terms of the standards of material quality of basalt aggregates from JEJU Island as a source for concrete aggregate. Quality assessments on the basalt aggregates were performed to assess the soundness of coarse aggregates using sodium sulfate solution, aggregate crushing test, and Los Angeles abrasion test. In addition, XRD, XRF, porosity, and compressive and tensile strength tests were performed to analyze the chemical components and the mechanical properties. In general, the mechanical properties of basalt aggregates from some areas did not meet the Korea Standards (KS), but the levels of compressive and tensile strength were higher than those of granite, andesite, and sandstone of other regions.

Distribution of Electrically Conductive Sedimentary Layer in Jeju Island Derived from Magnetotelluric Measurements (MT 탐사자료를 이용한 제주도 지역의 전도성 퇴적층 분포 연구)

  • We investigate the spatial distribution of highly conductive layer using the one-dimensional inversions of the new magnetotelluric (MT) measurements obtained at the mid-mountain (400 ~ 900 m in elevation) western area of Jeju Island and the previous MT data over Jeju Island, Korea. The conductive layer indicates the sedimentary layer comprised of Seoguipo Fomation and U Formation. There is a definite positive correlation between the top of conductive layer and the earth surface in elevation. On the contrary, the bottom of conductive layer has a negative correlation with the surface elevation. In other words, the conductive layer has a shape of convex lens, which is thickest in the central part. The basement beneath the conductive layer could be concave in the central part of Jeju Island. A kriging considering the correlation between the layer boundary and the surface elevation provides a reliable geoelectric structure model of Jeju Island. However, further studies, i.e. three-dimensional modeling and interpretation integrated with other geophysical or logging data, are required to reveal the possible presence of three-dimensional conductive body near the subsurface vent of Mt. Halla and the causes of the bias in the depths of layer estimated from MT and core log data.

Time-based Expression Networks of Genes Related to Cold Stress in Brassica rapa ssp. pekinensis (배추의 저온 스트레스 처리 시간대별 발현 유전자 네트워크 분석)

  • Plants can respond and adapt to cold stress through regulation of gene expression in various biochemical and physiological processes. Cold stress triggers decreased rates of metabolism, modification of cell walls, and loss of membrane function. Hence, this study was conducted to construct coexpression networks for time-based expression pattern analysis of genes related to cold stress in Chinese cabbage (Brassica rapa ssp. pekinensis). B. rapa cold stress networks were constructed with 2,030 nodes, 20,235 edges, and 34 connected components. The analysis suggests that similar genes responding to cold stress may also regulate development of Chinese cabbage. Using this network model, it is surmised that cold tolerance is strongly related to activation of chitinase antifreeze proteins by WRKY transcription factors and salicylic acid signaling, and to regulation of stomatal movement and starch metabolic processes for systemic acquired resistance in Chinese cabbage. Moreover, within 48 h, cold stress triggered transition from vegetative to reproductive phase and meristematic phase transition. In this study, we demonstrated that this network model could be used to precisely predict the functions of cold resistance genes in Chinese cabbage.

Study on the surface modification of zirconia with hydrophilic silanes (친수성기를 가진 실란을 이용한 지르코니아의 표면의 개질 연구)

  • Since microzirconia has excellent thermal and mechanical properties with high chemical and electrical resistance, it can be used in various fields. When the surface of zirconia becomes hydrophilic, its dispersibility in water will be improved as well as the resistance to most hydrophobic contaminants will be increased. In this study, we investigated the introduction of a hydrophilic groups on the microzircornia surface through hydrolysis and condensation reactions with two different silanes containing hydrophilic functional groups, such as ${\gamma}$-aminopropyltrimethoxysilane (APS) and ${\gamma}$-ureidopropyltrimethoxysilane (UPS) at different pH and concentration conditions. A covalent bond formation between the surface hydroxyl groups of zirconia and that of hydrolyzed silanes was confirmed by ninhydrin test and FT-IR spectroscopy. However, the presence of Si on the surfaces of both silane modified microzirconias was unable to detect by SEM/EDS technique. In addition, particle size analysis results provide that the size of microzirconia was changed to smaller or bigger than that of original zirconia due to crushing and aggregation during the modification process. The water dispersibility was improved for only APS modifed zirconia (AS-2 and AS-3) under neutral pH condition, but the water dispersibility and stability for all cases of 0.5~2% UPS modifed zirconia (US series) were much improved.

Estimation on the Durability of High-Strength Concrete using Metakaolin (Metakaolin 혼합 고강도 콘크리트의 내구특성 예측)

  • Metakaolin is a cementitious material for producing high-strength concrete. This material is now used as substitute for silica-fume. In this paper, we did the mechanical and durability test such as compressive/tensile/flexural strength test, chloride ion diffusion, chemical attack and repeated freezing and thawing, carbonation test. In the mechanical tests, 10~15% for binder is optimum substitute rate. And, in the chloride ion diffusion test, according to the increase of substitute of metakaolin & silica-fume for binder, the diffusion coefficient was more reduced. In the chemical attack test, by the filler effect of fine powder such metakaolin and silica-fume, the resistance is more excellent than normal concrete. In the other durability test, the concrete using metakaolin also compared with those of silica-fume substitute concrete. Through these tests, we recognized that metakaolin can be used as a substitute for silica-fume.

Development of the Gene Therapy Vector for Targeting Ovarian Cancer Cells through ErbB Receptors (ErbB 수용체를 이용한 난소암세포 표적 유전자치료 벡터의 개발)

  • Inefficiency of in vivo gene transfer using currently available vectors reflects a major hurdle in cancer gene therapy. Both viral and non-viral approaches have been described to improve gene transfer efficiency but suffer from a number of limitations. Here we tested an adenovirus carrying the small peptide ligand derived from heregulin${\beta}$ EGF-like domain onto fiber, the adenoviral capsid protein, to deliver transgene to ovarian cancer cells which overexpress ErbB, the cognate receptors for heregulin. The attachement of 53 amino acids to fiber didn't affect on the fiber's trimer structure that is critical for the viral entry to cells. The fiber-modified adenovirus can mediate entry and expression of a ${\beta}$-galactosidase into cancer cells in an increased efficiency compared the unmodified adenovirus. Particularly, the gene transfer efficiency was improved up to 5 times in OVCAR3 cells, an ovarian cancer cell line. Such transduction systems hold promise for delivering genes to ErbB receptor overexpressing cancer cells, and could be used for future cancer gene therapy.

  • Pseudohypoparathyroidism (PHP) is a disorder characterized by hypocalcemia and hyperphosphatemia due to end organ resistance to parathyroid hormone. PHP is caused by the deficiency of the α-subunit of the stimulatory G protein encoded by the GNAS gene, and this defect arises from genetic or imprinting disturbances. Sporadic PHP 1b shows two or more methylation defects of upstream of GNAS gene and some of them lead to loss of maternal GNAS imprints, therefore, only paternally derived GNAS gene is expressed. Here, we report a 10 year 9 month old boy presented with intermittent tetany who was finally diagnosed with PHP 1b caused by paternal uniparental disomy of chromosome 20q.

Corrosion mitigation of photovoltaic ribbon using a sacrificial anode (희생양극을 이용한 태양광 리본의 부식 저감)

  • Degradation is commonly observed in field-aged PV modules due to corrosion of the photovoltaic ribbon. The reduced performance is caused by a loss of fill factor due to the high series resistance in the PV ribbon. This study aimed to mitigate the degradation by corrosion using five sacrificial anodes - Al, Zn and their alloys - to identify the most effective material to mitigate the corrosion of the PV ribbon. The corrosion behavior of the five sacrificial anode materials were examined by open circuit potential measurements, potentiodynamic polarization tests, and galvanic current density and potential measurements using a zero resistance ammeter. Immersion tests for 120 hours were also conducted using materials and damp heat test tests were performed for 1500 hours using 4 cell mini modules. The Al-3Mg and Al-3Zn-1Mg sacrificial anodes had a low corrosion rate and reduced drop in power, making then suitable for long-term use.

A Study on the Effect of Improving Permeability by Injecting a Soil Remediation Agent in the In-situ Remediation Method Using Plasma Blasting, Pneumatic Fracturing, and Vacuum Suction Method (플라즈마 블라스팅, 공압파쇄, 진공추출이 활용된 지중 토양정화공법의 정화제 주입에 따른 투수성 개선 연구)

  • A stratum with a complex composition and a distributed low-permeability soil layer is difficult to remediate quickly because the soil remediation does not proceed easily. For efficient purification, the permeability should be improved and the soil remediation agent (H2O2) should be injected into the contaminated section to make sufficient contact with the TPH (Total petroleum hydrocarbons). This study analyzed a method for crack formation and effective delivery of the soil remediation agent based on pneumatic fracturing, plasma blasting, and vacuum suction (the PPV method) and compared its improvement effect relative to chemical oxidation. A demonstration test confirmed the effective delivery of the soil remediation agent to a site contaminated with TPH. The injection amount and injection time were monitored to calculate the delivery characteristics and the range of influence, and electrical resistivity surveying qualitatively confirmed changes in the underground environment. Permeability tests also evaluated and compared the permeability changes for each method. The amount of soil remediation agent injected was increased by about 4.74 to 7.48 times in the experimental group (PPV method) compared with the control group (chemical oxidation); the PPV method allowed injection rates per unit time (L/min) about 5.00 to 7.54 times quicker than the control method. Electrical resistivity measurements assessed that in the PPV method, the diffusion of H2O22 and other fluids to the surface soil layer reduced the low resistivity change ratio: the horizontal change ratio between the injection well and the extraction well decreased the resistivity by about 1.12 to 2.38 times. Quantitative evaluation of hydraulic conductivity at the end of the test found that the control group had 21.1% of the original hydraulic conductivity and the experimental group retained 81.3% of the initial value, close to the initial permeability coefficient. Calculated radii of influence based on the survey results showed that the results of the PPV method were improved by 220% on average compared with those of the control group.
