Growth-inhibitory Responses of Human Intestinal Bacteria to Extracts from Indian and African Plants (인도산 및 아프리카산 식물체 추출물의 장내세균에 대한 생육억제 반응)

  • Methanol extracts from 84 Indian plant samples (50 species in 31 families) and 27 African plant samples (20 species in 12 families) in vitro were tested for their growth-inhibitory activities against Bifidobacterium bifidum, Bifidobacterium longum, Lactobacillus acidophilus, Clostridium perfringens, and Escherichia coli, using a paper disc agar diffusion method under $O_2-free$ conditions. The responses varied with bacterial strain, plant species and plant part. Extracts from Cymbopogon citratus whole plants, Ocimum basilicum whole plant, Madhuca indica flowers, and Aegle marmelos leaves among Indian plant samples moderately or strongly inhibited the growth of Cl, perfringens whereas moderate growth-inhibitory activity against E. coli was obtained from extract of Indian O. basilicum whole plants. These plant extracts did not affect the growth of the lactic acid forming bacteria tested. These results may be an indication of at least one of the pharmacological actions of these tropical plants.

Inhibition Effect by Juniperus rigida S. et Z. on Organic Acids Production from Streptococcus mutans (두송실에 의한 충치균의 유기산 생성 억제효과)

  • To develop the natural antibacterial agents which don't have any toxicity against man, collected several species of medicinal plants were tested for their antibacterial activity and inhibition effect on organic acids production from Streptococcus mutans. Among the tested medicinal plants, methanol extracts and n-hexane fraction of Juniperus rigida S. et Z. had the comparatively strong activities. For example, strong antibacterial effect against S. mutans were showed in methanol extracts, n-hexane and chloroform fractions. Organic acids production were reduced to 89.3 and 90.8% of control in methanol extracts and n-hexane fraction treated concentration of 1 mg/ml, respectively.

Antioxidant Activity and Nitrite Scavenging Ability of Olive Leaf (Olea europaea L.) Fractions (올리브 잎 분획물의 항산화기능과 아질산염 소거능력 평가)

  • In this study, the antioxidant activities and nitrite scavenging abilities of olive leaf fractions acquired from plants cultivated in Australia (Olea europaea L. var. Picual) and Spain (Olea europaea L. var. Hojiblanca) were evaluated. Oleuropein was found to be the major phenolic compound in the leaves, with the butanol fractions presenting the highest contents. Antioxidant activity was evaluated in terms of superoxide dismutase (SOD)-like activity, 1,1-diphenyl-2-picryl hydroxyl (DPPH) radical scavenging activity, and the inhibitory effect on the auto-oxidation rate of linoleic acid. The SOD-like activities of the olive leaf extracts ranged from 0 to 36.8%. DPPH radical scavenging activity was highest in the ethanol extract of the Australian cultivated olive leaves. Finally, the chloroform fractions of the extracts showed inhibitory effects on the auto-oxidation rate of linoleic acid as well as nitrite scavenging ability.

  • Inhibition effects of alanine and methionine on the corrosion of aluminum were investigated in artificial sea water. Based on the low coverage of alanine and methionine we suggested that alanine and methionine adsorption process in aluminum surface is Langmuir isotherm and the carboxyl ion of amino acids seems to be adsorbed on Al.

Effect of Asarum sieboldii Extracts on the Growth, Acid Production, Adhesion, and Insoluble Glucan Synthesis of Streptococcus mutans (세신 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성억제에 미치는 영향)

  • Dental caries are the most commonly occurring chronic diseases in the dental field. Because of increasing sugar consumption and extension of average human life, these diseases are widely found all over the world as the most typical cause for a person to lose a tooth. Therefore, the development of more effective, substantial and safe preventive agents against dental caries is strongly required. Streptococcus mutans is known as the causative bacterial playing the most important role informing plaque and it is being noticed as major causative bacteria of dental caries. The present study was designed to investigate the effect of Asarum sieboldii Miquel(Aristolochiaceae) extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans(S. mutans). Both methanol and aqueous extracts showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 100, 1,000 and 2,000 μg/ml compared to the control group(p<0.05 - p<0.01). The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration of 10 μg/ml and complete inhibition was observed at the concentration of 2,000 μg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol and aqueous extracts showed more than 70% inhibition over the concentration of 1,000 μg/ml. Hence, we conclude that Asarum sieboldii might be a candidate of anticaries agent.

Effects of Heat Treatment of Chitooligosaccharide and Water-Soluble Chitosan on the Changes in Oligosaccharide Content and Anti-Rotaviral Activity (키토올리고당과 수용성 키토산의 열처리가 올리고당 함량의 변화와 항 로타바이러스성에 미치는 영향)

  • Chitosan is the deacetylated product of chitin. Chitosan and its derivatives have many properties that make them attractive for a wide variety of health applications. This study was conducted to investigate change of oligosaccharide content and antiviral effect on rotavirus of chitooligosaccharide and water soluble chitosan after heat treatment. The quantitative analysis of oligosaccharide using colorimetry showed that oligosaccharide contents in water soluble chitosan and chitooligosaccharide were decreased from 62.67% to 60.45% and from 59.48% to 54.31%, respectively, after heating. The inhibitory effect of chitosan derivatives on MA-104 cell infected with human rotavirus(HRV) measured using AEC staining method. The inhibition level of 0.125% water-soluble chitosan against cell infection by human rotavirus was 91.98 3.09% in HRV S2 and was 89.92 1.68% in HRV Wa. But, chitooligosaccharide had not shown inhibitory effect against cell infection by HRV. It considered that most oligosaccharide of chitooligosaccharides consist of oligomer of lower polymerization degree. Heat treatment of water soluble chitosan and chitooligosaccharide did not influence their antiviral effects on rotavirus.

Therapeutic Effects of Binsosan(檳蘇散) on Adjuvant-induced Rheumatoid Rats (Freund's complete adjuvant로 유발시킨 rat 류마티스성 관절염에 대한 빈소산의 치료 효과)

  • 실험목적 : 빈소산은 11가지 생약으로 구성된 복합 한약 처방으로 관절염을 포함한 다양한 염증성 질환의 치료제로 사용되어 왔으나, 관절염에 대한 직접적인 효력평가는 찾아 보기 힘들다. 따라서 본 실험에서는 빈소산 추출물이 Freund's complete adjuvant (FCA)로 유발된 rat 류마티스성 관절염에 미치는 치료 효과를 dexamethasone (15mg/kg, 복강 투여) 의 효과와 비교 평가하였다. 실험방법 : 류마티스성 관절염은 FCA (10mg in 1ml paraffin oil 0.1ml/rats)를 좌측 후지에 피내 투여하여 유발하였다. 실험동물은 Wistar 랫트를 사용하였고, FCA 투여 14일 후 유사한 무릎관절 둘레를 나타내는 류마티스성 관절염 유발 rat와 정상 rat 및 실험군을 그룹당 9마리씩 나누었다. 실험동물은 100 또는 200mg/kg의 빈소산 추출물을 FCA 투여 14일 후부터 14일간 경구 투여하였으며, dexamethasone은 15mg/kg 농도로 복강 투여한 다음, 희생하여, 체중, 연골내 collagen 함량 및 chondroitin sulphate, heparin sulphate 및 hyaluronic acid와 같은 뼈내 glycosaminoglycan 함량의 변화를 각각 관찰하였다. 실험결과는 항염 효과가 이미 입증되어 있는 dexamethasone 15mg/kg 복강 투여군과 비교하였다. 결과 : FCA 투여는 현저한 체중, 연골내 collagen 함량 및 chondroitin sulphate, heparin sulphate 및 hyaluronic acid와 같은 뼈내 glycosaminoglycan 함량의 감소와 함께 유발 관절 둘레 및 조직내 prostaglandin $E_2$의 증가와 같은 전형적인 류마티스성 염증을 초래하였으나, 이러한 류마티스성 관절염 소견은 dexamethasone 및 모든 용량의 빈소산 추출물 투여에 의해 현저히 억제되었으며, 특히 빈소산 투여군에서는 투여 용량 의존적인 감소가 인정되었다. 결론 : 이상에서 빈소산 추출물은 투여 용량 의존적인 prostaglandin $E_2$ 억제를 매개하여 FCA 유발 류마티스성 관절염에 대한 치료 효과를 나타내는 것으로 관찰되었다. 따라서 새로운 관절염에 대한 치료제로서 개발 가능성이 있을 것으로 판단된다. 한편 빈소산 추출물은 주로 prostaglandin $E_2$ 억제작용에 의해 항염 효과를 나타내는 것으로 관찰되었으나, 금후 다른 작용기전에 대한 연구와 빈소산의 구성성분 중 유효 성분 규명을 위한 실험이 수행되어야 할 것으로 판단된다.

Comparative Study of Corrosion Inhibition in Acidic and Neutral Chloride Media by Some Amino Acids (염산과 NaCl 수용액에서 알루미늄의 부식에 미치는 아미노산의 부식억제효과)

  • Inhibition effects of alanine (Ala), histidine (His), methionine (Met) on the corrosion of aluminum were investigated in deaerated 0.5 M HCl and NaCl solution. In HCl solution the inhibition efficiency for the aluminum corrosion depended on the cathodic inhibition, and the inhibition efficiency was increased in the order of Met$10^{-4\;}M$ the adsorption process can be explained by Langmuir isotherm, however, in the case of higher concentration by Temkin logarithmic isotherm due to the interaction between the adsorbed molecules.

Studies on Microbial Reduction of Chicken Carcasses (계육가공에 있어서 미생물증식억제)

  • Chlorine alone was not effective in reducing the numbers of microorganisms on the chicken carcasses. The chlorine in addition to succinic acid, in which the carcasses were immersed for 30 minutes succeeded in reducing the numbers of microorganisms on the chicken skin. Chicken drumsticks treated with 200 ppm NaOCl plus 0.5% succinic acid stored at 5$^{\circ}C$ showed a little reduction in microbial quantity for approximately 2 days, but the microbial load increased thereafter to the point of indicating organoleptic spoilage in approximately 7 days. This method extended the shelf life of chicken by 2 days.
