Functional Defect and Its Possible Mechanism of Diabetic Cardiomyopathy (당뇨성 심근질환에서의 근장그물 기능이상과 그 작용기전)

  • Oxidative modification of cellular proteins and lipids may play a role in the development of diabetic complications. Diabetic cardiomyopathy has been suggested to be caused by the intracellular $Ca^{2+}$ overload in the myocardium, which is partly due to the defect of calcium transport of the cardiac sarcoplasmic reticulum (SR). In the present study, the possible mechanism of the functional defect of cardiac SR in diabetic rats was studied. Both of the maximal $Ca^{2+}$ uptake and the affinity for $Ca^{2+}$ were decreased in the diabetic rat SR in comparison with the control. To investigate whether the functional defect of the cardiac SR in streptozotocin-induced diabetic rat is associated with the oxidative changes of cardiac SR proteins, the carbonyl group content and glycohemoglobin levels were determined. The increase in carbonyl group content of cardiac SR (2.30 nmols/mg protein, DM; 1.78, control) and in glycohemoglobin level $(13{\sim}17%,\;DM;\;3{\sim}5%,\;control)$ were observed in the diabetics. The extent of increase in calcium transport by phospholamban phosphorylation was greater in the diabetic cardiac SR membranes than that in the control. The phosphorylation levels of phospholamban, as determined by SDS-PAGE and autoradiography with $[{\gamma}^{32}P]ATP$, were increased in diabetic cardiac SR. These results suggest that the impaired cardiac SR function in diabetic rat could be a consequence of the less-phosphorylation of phospholamban in the basal state, which is partly due to the depleted norepinephrine stores in the heart. Furthermore, the oxidative damages in cardiac SR membranes might be one of the additional factors leading to the diabetic cardiomyopathy.

Effects of Calcium Injection and Chitosan Coating on Shelf Life and Quality of Pork (칼슘 주입과 키토산 코팅 처리가 돈육의 저장성 및 품질에 미치는 영향)

  • Effects of calcium and chitosan treatments on storage and quality of pork were investigated. Porks were coated with 1% each of 30 and 120 kDa chitosans in 3% gelatin solution. Self-life and antioxidation increased significantly in all samples. In contrast with non-treated group, pH of the chitosan-treated group was very stable. Moisture content and water activity (Aw) were similar among all samples. Water-holding capacity decreased slightly, whereas cooking loss increased, during storage in all samples. Value of redness increased in chitosan-coated pork. Tenderness of pork injected with calcium was the highest among all samples. These results showed that preservation and quality of pork were improved by the treatment of calcium and 30 kDa of chitosan.

Influences of Divalent Cations and Membrane Phosphorylation Inhibitors on $Na^+-Ca^{++}$ Exchange in Synaptosomes (이가 양이온과 세포막 인산화 반응의 억제제가 Synaptosome에서의 소듐-칼슘 교환이동에 미치는 영향)

  • Verapamil, tetrodotoxin and tetraethylammonium chloride in the stated amount did not affect the $Na^{++}$ induced $Ca^{++}$ release. $Cd^{++}$ and $Zn^{++}$ significantly inhibited the $Na^{++}$ induced $Ca^{++}$ release. $Mn^{++}$ also inhibited $Na^+-Ca^{++}$ exchange. $Cd^{++}$ inhibited $Na^+-Ca^{++}$ exchange noncompetitively with an apparent inhibition constant (Ki) of $100\;{\mu}M$. $Cd^{++}$ caused loss of sulfhydryl group, whereas $Zn^{++}$ did not show any significant effect. $Cd^{++}$ and $Zn^{++}$ effectively inhibited $Na^+-Ca^{++}$ ATPase and slightly inhibited $Ca^{++}-Mg^{++}$ ATPase. Carbonyl cyanide chlorophenylhydrazone, 2,4-dinitrophenol and sodium arsenate stimulated the $Na^{++}$ induced $Ca^{++}$ release. Dibucaine and oligomycin slightly inhibited it. The results suggest that the $Na^+-Ca^{++}$ exchange on the synaptosomal plasma membrane may be not accomplished by ion channels. The $Na^+-Ca^{++}$ exchange is sensitively inhibited by $Cd^{++}$ and this transport process appears to be partially regulated by sulfhydryl groups of the synaptosomal plasma membrane. It is also postulated that $Na^+-Ca^{++}$ exchange is suppressed during the phosphorylation reaction of protein component on the neuronal membrane.

Effect of Agitation and Additive on the Vaterite Contents of Precipitated Calcium Carbonate from Oyster Shell Waste (폐 굴껍질 이용 침강성 탄산칼슘 제조에서 교반속도와 첨가제가 Vaterite 함유량에 미치는 영향)

  • An experiment was conducted to produce vaterite-type precipitated calcium carbonate from waste oyster shells in order to use them as recyclable resources. Calcined oyster shells containing calcium oxide as their main component were prepared at a temperature of 800℃ for 24 h. The oyster shells were dissolved in nitric acid or hydrochloric acid solution to make 0.1 M calcium nitrate or calcium chloride aqueous solution, and a carbonation reaction was performed using a 0.1 M sodium carbonate aqueous solution under various experimental conditions, which included varying the amount of aspatic acid additive, the amount of NH4OH added, the reaction time, the reaction temperature, the stirring speed, and the type of dissolved acid. The XRD, SEM, and size distributions were analyzed and the vaterite content was calculated. Spherical precipitated calcium carbonate with a vaterite content of 95.9% was synthesized by adding 0.1 mol aspatic acid/1 mol CaO and 2 cm3 of NH4OH, and reacting for 1 h at 25℃ while stirring at 600 rpm. The average particle diameter was found to be 12.11 ㎛. Calcium carbonate contatining high vaterite is used as high value added calcium carbonate for medical, food, inke additiver, etc.

Synthesis and Shape Control Calcium Hydroxide Fine Powders by Hydration of Calcium Oxide (산화칼슘의 수화에 의한 수산화칼슘 미분말의 합성과 형상제어)

  • Calcium hydroxide fine powders were synthesized by hydration of calcium oxide, and the shapes of powders obtained were examined for each synthethic condition. When distilled water was used as a solvent, irregular and agglomerated submicron powders were obtained, and it was impossible to control of the shapes. In methanol-added solutions, hexagonal plate-like particles were obtained, but addition of ethanol had no effect. However on the occasion that substituted ethylene glycol for ethanol of 5 vol%, hexagonal plate-like powders were obtained. The shapes of powders synthesized in acetic acid and salicylic acid solutions were hexagonal platelike, and were spherical and very fine in citric acid and oxalic acid solutions, respectively. But in some solutions, calcium salts were precipitated by the reaction between calcium and acid added. And the size of powders were very fine using ultrasonic vibration instead of mechanical agitation.

Pretreatment sterilization of garlic and ginger using antimicrobial agents and blanching (데치기 및 초고압 처리를 이용한 항균처리 마늘 및 생강의 살균 효과 연구)

  • The sterilization rates were investigated to develop effective pretreatment methods of garlic and ginger using antimicrobial agents and blanching. Antibacterial components in chopped garlic and ginger, such as allicin, ginger, and shogaol, exhibited an antibacterial effect that reduced bacteria by a factor of $10^1(CFU/g)$. The total number of bacteria in garlic and ginger was decreased by $10^1(CFU/g)$ after individually soaking in 1% (w/w) citric acid, calcium oxide and 200 ppm sodium hypochlorite solution for 1 h, respectively. When chopped garlic and ginger were immersed in 1% (w/w) calcium oxide solution for 1 h, the bactericidal effect was increased by $10^4-fold$. Total cells with thermoduric bacteria exhibited a $10^3-10^4-fold$ reduction after heating for 30-40 s at $90-95^{\circ}C$. The combination of soaking in 200 ppm sodium hypochlorite solution and blanching at $95^{\circ}C$, 40 s showed $10^1-fold$ decrease in total number of cells compared to blanching in whole ginger.

Physicochemical properties and biological activity of three-year-old and seven-year-old Platycodon grandiflorum extracts (3년근 도라지와 7년근 도라지의 이화학 특성 및 생리활성)

  • This study was designed to evaluate the physicochemical properties and biological activities (antioxidant activity and antimicrobial activities) of three and seven-year-old Platycodon grandiflorum extracts. Three and seven-year-old Platycodon grandiflorum contained crude saponins, free amino acids and minerals. Water extracts of the three and seven-year-old plants were prepared using reflux extraction methods. The total polyphenol contents (TPC), total flavonoid contents (TFC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and antimicrobial effects of the extracts were determined. The seven-year-old Platycodon grandiflorum extract had higher TPC ($5.08{\pm}0.07TAEmg/g$) and TFC ($3.80{\pm}0.07QUEmg/g$). DPPH radical scavenging activity ($IC_{50}$ value: $288{\pm}3.88{\mu}g/mL$) and ABTS radical scavenging activity ($IC_{50}$ value: $568{\pm}2.09{\mu}g/mL$). The three and seven-year-old Platycodon grandiflorum extracts exhibited a strong antimicrobial effect against three kinds of bronchus disease-inducing bacteria; the seven-year-old Platycodon grandiflorum extracts showed a stronger antimicrobial effect than the three-year-old Platycodon grandiflorum extracts.

Efficacy of antibacterial treatments of fresh ginseng (Panax ginseng C.A. Meyer) (항균제 처리에 따른 수삼의 미생물 저감화 효과)

  • Fresh, washed ginseng can be contaminated with microorganism loads as high as 6.5 log CFU/g for total bacteria and 4.3 log CFU/g for mold. The goal of this study was to test eight antibacterial agents on ginseng. Immersing fresh ginseng washed in 1% (w/w) sodium citrate, sodium diacetate, sodium acetate, citric acid, and sodium lactate solution for 1 h resulted in a bactericidal effect of 31.0-97.5% for total bacteria. Among the organic acids, sodium citrate had the best antibacterial effect, with total bacteria reduced from 6.5 log to 4.9 log CFU/g. A 1% (w/w) vitamin B1 lauryl sulfate solution with surfactant function by hydrophilic and hydrophobic sites can reduce 2.7 log CFU/g (99.8% inactivation) on total bacteria. In the 1% (w/w) calcium oxide solution, total bacteria were reduced by 3 log, showing an excellent inactivation effect of 99.9%. Calcium oxide is a highly useful material for inactivation of microorganisms in fresh ginseng.

Purification and Crystallization of the Recombinant Catalytic Subunit of Pyruvate Dehydrogenase Phosphatase (Pyruvate Dehydrogenase Phosphatase의 Catalytic Subunit의 분리정제 및 결정화)

  • Pyruvate dehydrogenase phosphatase (PDP) is a mitochondrial protein serine/threonine phosphatase that catalyzes the dephosphorylation and concomitant reactivation of the pyruvate dehydrogenase component of the pyruvate dehydrogenase complex (PDC). PDP consists of a catalytic subunit (PDPc, Mr 52,600) and regulatory subunit (PDPr, Mr 95,600). In the presence of $Ca^{2+}$, PDPc binds to the dihydrolipoamide acetyltransferase (E2) component of the pyruvate dehydrogenase complex in proximity to its substrate, the phosphorylated E1 component, thereby increasing the rate of dephosphorylation. PDPc possesses and intrinsic $Ca^{2+}$ binding site and a second $Ca^{2+}$ site is generated in the presence of E2. Using the unique interaction, highly pure PDPc was produced by the GSH-Sepharose-GST-L2 matrix with a specific activity of approx. 1000 U/mg and a yield of about 80%.

Effects of Amitriptyline and Imipramine on Superoxide Generation, Myeloperoxidase Release, Leukotriene $B_4$ in Human Neutrophils (Amitriptyline과 Imipramine이 호중구에서의 Superoxide 생성, Myeloperoxidase 유리, Leukotriene $B_4$생성과 칼슘 동원에 나타내는 영향)

  • A number of tricyclic antidepressants appear to have inhibitory action on calmodulin. Although amitriptyline, imipramine and doxepine have been shown to inhibit calcium uptake, oxidative phosphorylation and ATPase activities, effects of amitriptyline, imipramine and doxepine on functional responses of human neutrophils have not been elucidated. In this study, effects amitriptyline, imipramine and doxepine on superoxide and hydrogen peroxide generation, myeloperoxidase release, leukocriene B4 formation and intracellular calcium level were investigated. Superoxide and hydrogen peroxide production in heat aggregated IgG-activated neutrophils were inhibited by amitriptyline, imipramine and doxepine. EDTA, EGTA, verapamil and bepredil inhibited heat aggregated IgG-induced superoxide production. Chlorpromazine, trifluoperazine, staurosporine and H-7 also inhibited it. PMA-induced superoxide production was inhibited by amitriptyline, imipramine, doxepine, chlorpromazine and H-7. Amitriptyline, imipramine, chlorpromazine and trifluoperazine inhibited the myeloperoxidase release by heat aggregated IgG. Productions of $LTB_4$, and 5-HETE in heat aggregated IgG-activated neutrophils were inhibited by amitriptyline, imipramine and doxepine. In neutrophils, elevation of intracellular calcium induced by heat aggregated IgG was inhibited by amitriptyline, imipramine, doxepine, chlorpromazine and EGTA, while verapamil slightly inhibited increase of intracellular calcium and H-7 did not inhibit it. These results suggest that the inhibitory effect of amitriptyline, imipramine and doxepine on respiratory burst, myeloperoxidase release and LTB4 production in heat aggregated IgG-activated neutrophils appears to be ascribed to the inhibition of calcium mobilization, calmodulin and protein kinase C.
