Isolation and Identification of Klebsiella oxytoca C302 and Its Degradation of Aromatic Hydrocarbons (Klebsiella oxytoca C302의 분리 동정 및 방향족 탄화수소물질의 분해특성)

  • A bacterial isolate capable of degrading benzoate was selected from wastewater of Yocheon industrial complex and examined its biochemical characteristics and fatty acid composition. The isolate was identified as Klebsiella oxytoca strain C302. The strain C3O2 degraded catechol, protocatechuate, and 4-hydroxybenzoate as well as benzoate. The strain grew on and degraded 0.5 to 1.0 mM catechol most actively in MM2 medium at pH 7.0 and $30^{\circ}C$.

Optimization of Peptide Production from Leg Meat of Yeonsan Ogae by High Hydrostatic Pressure and Protein Hydrolytic Enzyme and Its Characteristic Analysis (고압처리와 단백질 분해효소를 이용한 연산오계 다리육 펩타이드 생산 최적화 및 특성 분석)

  • The purpose of this research was the optimization of protein hydrolysate production using a commercial enzyme bromelain 1200 derived from the leg of Yeonsan Ogae by response surface methodology. Yeonsan Ogae has long been known as supporting health and high efficacy treatment. In recent days, as the efficacy of functional peptides becomes more known, optimization of oligopeptide production and its characteristics from Ogae leg meat has been performed. Response surface methodology was performed for optimization of enzyme hydrolysis. The process was varied in pressure (30 to 100 MPa), time (1 to 3 h), and substrate concentration (10 to 30%). The degree of hydrolysis, amino acids, and molecular weight of products were analyzed. The optimum conditions were determined to be a pressure of 100 Mpa, time of 3 h, and substrate concentration of 20%. Under optimized conditions, degree of hydrolysis was 34.10%. The average molecular weight of protein hydrolysates was less than 1,000 Da. Major amino acids were leucine, lysine, alanine, glutamic acid, and phenylalanine.

  • Yeonsan Ogae has been known as supporting health and high efficacy of treatment. In recent days, as the efficacy of functional peptides has known, the optimization of oligo peptides production and its characteristics from Ogae viscera has been performed. Response surface method was used to perform the optimizaion of enzyme hydrolysis. The range of processes was temperature (40, 50 and $60^{\circ}C$), pH(6.0, 7.0 and 8.0), and enzyme(1, 2 and 3%). The degree of hydrolysis, amono acids, molecular weight of products were analyzed. The optimum process of enzyme hydrolysis were determined as temperature $58^{\circ}C$, pH 7.5, and enzyme concetration 3%. At optimum conditions, the degree of hydrolysis after 2 h reaction was 75-80%. The total amino acids of amino acid and were 386.15 mg/100 g and 155.26 mg/100 g, respectively. The molecular weight of products by using Maldi-TOF was ranged from 300 to 1,000 Da.

Cloning a Mannanase 26AT Gene from Paenibacillus woosongensis and Characterization of the Gene Product (Paenibacillus woosongensis으로부터 Mannanase 26AT 유전자의 클로닝과 유전자 산물의 분석)

  • An open reading frame coding for mannanase predicted from the partial genomic sequence of Paenibacillus woosongensis was cloned into Escherichia coli by polymerase chain reaction amplification, and completely sequenced. This mannanase gene, designated man26AT, consisted of 3,162 nucleotides encoding a polypeptide of 1,053 amino acid residues. Based on the deduced amino acid sequence, Man26AT was identified as a modular enzyme, which included a catalytic domain belonging to the glycosyl hydrolase family 26 and two carbohydrate-binding modules, CBM27 and CBM11. The amino acid sequence of Man26AT was homologous to that of several putative mannanases, with identity of 81% for P. ihumii and identity of less than 57% for other strains of Paenibacillus. A cell-free extract of recombinant E. coli carrying the man26AT gene showed maximal mannanase activity at $55^{\circ}C$ and pH 5.5. The enzyme retained above 80% of maximal activity after preincubation for 1 h at $50^{\circ}C$. Man26AT was comparably active on locust bean gum (LBG), galactomanan, and kojac glucomannan, whereas it did not exhibit activity on carboxymethylcellulose, xylan, or para-nitrophenyl-${\beta}$-mannopyranoside. The common end products liberated from mannooligosaccharides, including mannotriose, mannotetraose, mannopentaose, and mannohexaose, or LBG by Man26AT were mannose, mannobiose, and mannotriose. Mannooligosacchrides larger than mannotriose were found in enzymatic hydrolyzates of LBG and guar gum, respectively. However, Man26AT was unable to hydrolyze mannobiose. Man26AT was intracellularly degraded into at least three active proteins with different molecular masses by zymogram.

  • 본 연구는 근원에 따른 용존 유기물의 특성을 평가하기 위해 하천수, 호소수, 하수 2차 처리 유출수의 자연수 및 Aldrich, Wako 사에서 공급된 휴믹산을 대상으로 분자량 분포에 따른 물리 화학적인 수질 특성, 생분해도, 소독, 부산물의 생성 등을 고찰하였다. SUVA를 이용해서 자연수의 생분해 가능성을 평가한 결과 호소수, 하수, 하천수 순으로 예측되었으며 이는, 실제 생분해도 실험 결과와 유사하였다 생분해 반응 중 저분자 영역대의 용존 유기물은 점차 감소하였고 반면에 고분자 영역대의 용존 유기물은 증가하였다. SMP는 전체 용존 유기물의 0.7-5.5%정도 관찰되었으며 고분자 물질을 많이 함유한 시료에서 높게 나타났다. THM 생성은 대체로 고분자 물질이 높을수록 증가하였으며 THMFP도 Wako 휴믹산을 제외한 샘플에서 이와 유사한 경향을 나타냈다.

  • 생체막의 주요 구성성분인 인지질의 2번 위치에 결합한 불포화지방산은 각종 전이금속이나 각종 활성산소들의 공격을 쉽게 받아 지질과산화반응이 일어나서 생체에 유독한 화합물을 생성하게 된다. 생체는 이러한 기구의 해독을 위하여 크게 2가지 방어기전을 갖고 있다. 즉 Vitamin- C, $\alpha$-tocopherol, flavonoid, SOD, catalase 등과 같이 생성된 활성산소를 제거시키는 기구와. 활성산소에 의해 생성된 과산화물을 제거시키는 기구로 glutathione peroxidase (GPX)가 알려졌으며 GPX에 의해 독성이 낮은 수산화물까지 환원시키는 기구가 보고되었다. 그러나 인지질의 과산화물 그대로는 GPX의 기질이 쥘수 없으므로, 산화된 지방산을 절단하는 효소에 대한 기구의 해석이 요구되고 있다. 최근 여러질병에 관련되어 있는 인지질 2번위치의 지방산을 분해하는 phospholipase $A_2$ (PLA$_2$)가 과산화지질의 분해에 관여한다는 주장이 제기되었다. 따라서 본 연구에서는 rat liver microsome에 $CCl_4$투여로 일어나는 과산화반응에 있어서 PLA$_2$의 역할을 규명하기 위하여 본 실험을 행하였다.

  • Separation and identification of the polycyclic aromatic compounds (PAC) from pyrolysis products of cellulose and lignin were performed using a combination of acid-base solvent partitioning and silicic acid column chromatography with fused-silica capillary column gas chromatography/ mass spectrometry. Sixteen PAC were positively identifited by retention indices and mass spectra data. Both materials produced the same kinds of PAC. But lignin produced much more PAC than cellulose. Almost no highly carcinogenic heterocyclic PAC containing nitrogen and sulfur were produced.

Gas Chromatography를 이용한 식물성 유지의 지방산의 정량 정성 및 산화안전성 평가

  • 본 연구에서는 현재 우리나라에서 식용으로 많이 쓰이는 식물성 유지인 콩기름, 올리브유, 고추씨기름을 Gas Chromatography를 이용해 각 유지별 지방산의 정성, 정량하였으며 이들 유지의 산화 안정성 평가를 위하여 산화적 반응의 기작의 생성물인 휘발성 분해 산물인 Pentanal 생성을 비교하여 분석하였다. 1. Soybean oil의 지방산 조성은 불포화 지방산은 87.79%, 포화지방산은 12.21%를 차지했으며 Olive oil의 지방산 조성은 불포화 지방산은 82.77%, 포화지방산은 17.23%를 차지했다. Hot pepper oil의 지방산 조성은 불포화 지방산은 85.42%, 포화지방산은 14.58%를 차지했다. 2. 콩기름, 올리브기름, 고추씨기름의 pentane의 함량은 43.4, 30.4, 19.7nL로 측정되었다. 따라서 고추씨기름이 다른 유지에 비해 산화에 대하여 안정함을 알 수 있다. 이상의 결과로부터 실험에 선택된 콩기름, 올리브유, 고추씨기름에는 포화 지방산보다 불포화 지방산의 함량이 많은 것을 알 수 있었고, 특히 체내에서 합성이 불가능해 식품으로 섭취해야만 하는 필수 지방산의 함량이 높아 체내 생체 활동에 유용한 것으로 판단되었다. 산패도 비교 평가에서는 hot pepper seed oil이 olive oil이나 soybean oil보다 낮은 산패도를 나타내어 다른 유지보다 산화 안정성이 뛰어난 것으로 나타났다.

  • Physicochemical properties of starches from the rices of Akibare, Samgang and Mahatma and hardness of the cooked rice were examined Water binding capacity and amylose content were heigher in Samgang. Peak viscosity and breakdown were heigher in order of Akibare, Samgang and Mahatma, while consistency and set back were reverse order. Hydrolytic patterns of three starches with 2.2N HCl at $35^{\circ}C$ showed two distinct stages. Hydrolysis extent of Mahatma starch was lower than those of Akibare and Samgang starches. The relative crystallinities of these starches were heigher in order of Mahatma, Akibare and Samgang. Mahatma was more resistant to heat and acid treatments, lower in water binding capacity and harder when it was cooked.
