Resistance to Northern Root-knot Nematode, Meloidogyne hapla, in Peony Strains (당근뿌리혹선충에 대한 작약계통의 저항성 검정)

  • This study was carried out to screen 11 peony varieties collected for resistance to northern root-knot nematode, Meloidogyne hapla. The larval population of Meloidogyne halpa to the Uisongjakyak, Taebaekjakyak, Doseo, EP368, EP512 peony were detected high denisties with individuals of 98-173 per 300 ml soil, but Sabeol, Yongsu, Kimchon, Pl23 peony showed low number of larvar with 21-60. The root rot were lowered in Yongsu, Kimchon peony than Uisongjakyak, Taebaekjakyak, EP368, EP512. It seems to involved with densities of 2nd juvernile population in that rot rate of root was increased as increase of 2nd stage larvae. Two peony, Uisongjakyak, Taebaekjakyak were susceptable to M. hapla with high egg mass formation which showed 49,28 in each root. And Yongsu, Doseo, EP368, EP512 were Mid resistance with 1-15 egg mass of each root. Whereas Sabeol, Kimchon, Pl23, EP337, EP425 were found to be resistant to M, hapla with few egg mass formation of 4-7 each root.

Preparation and Evaluation of Polymer Microspheres Containing Broussonetia Kazinoki Root Extract (닥나무 뿌리 추출물을 함유하는 고분자 마이크로입자 제조 및 평가)

  • This study demonstrate that polymeric microspheres composed of poly (ethylene adipate) (PEA) and poly (methyl methacrylate) (PMMA) can encapsulate and remarkably stabilize Broussonetia kazinoki root extract. We compared the long-term stability and the activity of Broussonetia kazinoki root extract in polymeric microspheres fabricated with different polymer ratio of PEA and PMMA. PMMA was incorporated to the PEA microsphere in order to reinforce the physical strength of microsphere, and there was no noticeable negative effect on the activity of Broussonetia kazinoki root extract. Optical microscope (OM), polarized microscope (PM), and scanning electron microscope (SEM) results showed that PMMA incorporated microspheres were very spherical and had smoothsurface. On the other hand, PEA microspheres showed relatively irregular morphology due to the low physical strength of microspheres. Moreover, the mushroom tyrosinase activities were measured for testing the inhibitory activity of Broussonetia kazinoki root extract encapsulated in polymeric microspheres, and these microspheres showed the effective suppression of mushroom tyrosinase activity. Consequently, polymeric microspheres produced in this study may be beneficial for the research of improving stability and protecting labile substances incorporated into the polymeric microspheres.

Study on Keratinocyte Differentiation and Skin Barrier Function of Adeonphorae Radix Root Extracts (Adenophorae Radix 뿌리 추출물에 의한 Keratinocyte의 분화 및 피부장벽 기능에 대한 연구)

  • We have studied on the keratinocytes differentiation and skin barrier function using Adenophorae radix (A. radix) root extract, which was known to contain triterpenoid, saponin and starch. A. radix root extracts showed the $PPAR{\alpha}$ expression level of Wy-14,643 $0.5-1.0{\mu}M$ in CV-1 cells. The cornified envelop formation (CE) of human keratinocyte cell line (HaCaT) and normal human keratinocyte (NHK) showed a statistically significant increased compared to the control. When HaCaT cells were treated with A. radix root extract, transglutaminase (TGase-1) was significantly increased. As a result of clinical study of the simple cosmetic formulation containing A. radix root extract for about 2 weeks, TEWL values were significantly decreased and water contents were increased. The ceramides, which were obtained from the inner forearm, were also significantly increased statistically. We suggest that the A. radix root extract can be used as a preventive and therapeutic agent for skin diseases such as dry skin and atopy.

Physiological Activities of Ethanol Extracts from Different Parts of Allium hookeri (삼채부위별 에탄올 추출물의 생리활성탐색)

  • Biological compounds such as crude saponin and total polyphenol were determined in Allium hookeri roots and leaves. In addition, activities of DPPH radical scavenging activity, hydroxyl radical scavenging activity, ${\alpha}$-amylase inhibition activity and nitrite scavenging were detected in concentrates of 70% ethanol extracts of Allium hookeri roots and leaves. The crude saponin content of the roots and leaves were 4.28 mg/g, and 4.17 mg/g, respectively. Total polyphenol and total flavonoid content was higher in leaves than roots. Furthermore the DPPH radical scavenging activity and hydroxyl radical scavenging activities of leaves was higher than roots. For ${\alpha}$-glucosidase inhibitory activity, 70% ethanol extracts of Allium hookeri roots and leaves showed 60% activity inhibition at a 10 mg/mL concentration. Nitrite scavenging abilities under acidic conditions were most effective for Allium hookeri leaves extracts. These results suggested that Allium hookeri leaves might be used as the components of health functional food.

Comparative Anatomy of the Secondary Xylem in the Root and Stem of Some Korean Lauraceae (수종(數種)의 한국산(韓國産) 녹나무과(科) 식물(植物)에 있어서 뿌리와 줄기의 이기목부(二期木部)의 비교해부(比較解剖))

  • A comparative anatomy between the secondary xylem in the root and the stem of some Korean Lauraceae, including 6 genera and 7 species, was carried out in this study. The results are as follows; diameter and length of vessel element and fiber are wider and longer in the root than the stem. The angle of end wall of vessel element is more inclined in the root than the stem. Also more number of bar on the perforation plate of vessel element in root wood is found. From the anatomical characters of root and stem wood in some Lauraceous species, it is suggested that the wood of the root is phylogenetically mote primitive than that of the stem, except the diameter of vessel element.

Ectomycorrhizal Fungal Diversity on Abies korea and Taxus cuspidata at Two Altitudes in Mt. Halla (고도에 따른 한라산 구상나무와 주목의 외생균근균 다양성 비교)

  • In this study, the community structures of the ectomycorrhizal (ECM) fungi in the roots of Abies koreana and Taxus cuspidata were investigated at different altitudes of Mt. Halla. We identified the collected ECM root tips based on morphological characteristics and phylogenetic analysis through sequencing of the rDNA ITS regions. From the roots of A. koreana and T. cuspidata, 11 species and 12 species were identified, respectively. The Shannon's index and species evenness and abundance of the ECM fungi were higher in the higher than lower regions, regardless of host plant species, however, the number of ECM root tips showed the opposite pattern. The community similarity among the ECM fungi in A. koreana was significantly higher than that among the ECM fungi in T. cuspidata or than that between A. koreana and T. cuspidata (p < 0.05). These results could be useful for the conservation and management of the habitat of A. koreana, which is threatened with extinction due to increasing ambient air temperature.

  • IOT middleware is required to play a pivotal role in interpreting, managing, and controlling data information of Internet devices (sensors, etc.). In particular, the root industry has different process flows for different industries, and there are various data processing requirements for each company. Therefore, a general purpose IOT middleware is needed to accommodate this. The IOT middleware structure proposed by this paper is a plug-in that can be used as an engine part for middleware basic processes such as communication, data collection, processing and service linkage, We propose a flexible and effective smart process for root industry. In addition, we propose a method to strengthen prevention and security against tampering, deodorization, etc. through encryption of network data between middleware plug - in and related service layer. We propose a system that will be developed as an IOT middleware platform that is specialized in the root industry so that it can be extended in various network protocols such as MQTT, COAP, XAMP.

  • The present researches were carried out to classify the species of ginseng by electrophoretic methods with isozyme patterns of LAP. esterase, GOT, phosphatase, peroxidase and proteins. All variants of Korean and Japanese ginseng had identical band patterns of the investigated enzymes in roots as well as in seeds. However, American ginseng had different patterns from those of Korean or Japanese.

Occurrence of Meloidogyne incognita Infecting Resistant Cultivars and Development of an Efficient Screening Method for Resistant Tomato to the Mi-virulent Nematode (뿌리혹선충 저항성 토마토를 감염하는 Meloidogyne incognita의 발생 및 이 선충을 이용한 효율적인 저항성 검정법 확립)

  • Root-knot symptoms were found on a commercial tomato cultivar carrying Mi, a resistance gene to root-knot nematodes including Meloidogyne incognita, M. arenaria, and M. javanica in 2012 at Buyeo, Chungnam Province in Korea. The isolate was identified as M. incognita based on molecular analyses using two species-specific primer sets. Pathogenicity of the isolate on one susceptible and three resistant tomato cultivars to the root-knot nematodes was tested. The nematode isolate showed strong pathogenicity on all the tested cultivars at all tested incubation temperatures. In addition, resistance degree of 33 commercial tomato cultivars, 8 susceptible and 25 resistant cultivars to root-knot nematodes, was also tested. Plants were determined as resistant when they suppressed the nematode reproduction. All the cultivars demonstrated strong susceptibility to the nematode regardless of resistance of the tomato cultivars. To our knowledge, this is the first report on the occurrence of Mi infecting M. incognita isolate in Korea. On the other hand, to construct an efficient screening method for selecting resistant breeding source to the nematode isolate, root-knot development of M. incognita on four tomato cultivars according to several conditions such as inoculum concentration, plant growth stage, and incubation period after transplant was investigated. Reproduction of the nematode on all the tested cultivars according to inoculum concentration increased in a dose-dependent manner. Except for inoculum concentration, there was no significant difference in reproduction level of the cultivars according to the other tested conditions. On the basis of the results, we suggest an efficient screening method for new resistant tomato to the nematode isolate.

Vegetation Strucure of Hwangjeong Wetland around Geumho River (금호강 황정 습지의 식생 구조)

  • Vegetation structure of the vascular plants was investigated from April 2003 to August 2003 in Hwangjeong wetland around Geumho River, Yeongcheon-si, Gyeongsangbuk-do, Korea. Actual vegetation of Hwangjeong wetland largely can be classified by the floristic composition and the physiognomy into 12 communities; Ambrosia artemisiifolia var. elatior, Humulus japonicus, Miscanthus sacchariflorus, Phragmites japonica, Zizania latifolia-Miscanthus sacchariflorus, Zizania latifolia-Nymphoides peltata, Miscanthus sacchariflorus-Phragmites japonica, Phragmites communis-Phragmites japonica, Phragmites japonica-Salix gracilistyla, Salix koreensis-Salix glandulosa, Salix nipponica-Salix koreensis, and Phragmites japonica-Zizania latifolia. Among them, the distribution area of the Phragmites japonica community was the largest as 49.46 ha(11.03%). The dominant vegetation type was Phragmites japonica community and Ambrosia artemisiifolia var. elatior community based on the phytosociological method, and Phragmites japonica community was classified into two subcommunities; Nymphoides peltata subcommunity and Salix glandulosa subcommunity. Differential species of Phragmites japonica community were Phragmites japonica, Miscanthus sacchariflorus, Persicaria thunbergii, Oenanthe javanica, Leersia oryzoides var. japonica, and Rorippa indica; differential species of Ambrosia artemisiifolia var. elatior community were Ambrosia artemisiifolia var. elatior, Setaria glauca, Commelina communis, Cyperus orthostachyus, Digitaria sanguinalis, Xanthium strumarium, Erigeron annuus, Erigeron canadensis, Kummerowia striata, Trifolium repens, and Medicago sativa; differential species of Nymphoides peltata subcommunity were Nymphoides peltata, Zizania latifolia, Scirpus tabernaemontani, and Eleocharis mamillata var. cyclocarpa; differential species of Salix glandulosa subcommunity were Salix glandulosa, Salix koreensis, and Salix gracilistyla. It was expected that Hwangjeong wetland is worthy of conservation contributed purifying water pollution, giving habitats of many lifes, and providing beautiful scenes of Geumho River.
