Quality Characteristics and Antioxidant Activity of Mulberry Leaf Tea Fermented by Monascus pilosus (Monascus pilosus로 발효시킨 뽕잎차의 품질특성과 항산화능)

  • This study was carried out to investigate the contents of monacolin K and citrinin, along with the sensory quality and antioxidant activity of mulberry leaf tea fermented by $Monascus$ $pilsous$ (FMM). Total monacolin K content of FMM was 0.058%, but citrinin was not detected. Redness of brewed FMM was remarkably higher than that of unfermented mulberry leaf tea (UFM). In sensory evaluation of brewed FMM, while astringent taste and savory taste were lower, flavor, color, and overall acceptability were significantly higher than those of UFM. Total polyphenol contents of UFM and FMM were 83.1 and 23.61 mg/g (dry basis), total flavonoid contents of UFM and FMM were 17.96 and 3.99 mg/g (dry basis), respectively. Xanthine oxidase (XO) inhibitory activity and superoxide dismutase (SOD)-like activity of FMM were lower than those of UFM. Electron-donating ability and ferric-reducing antioxidant power of FMM were slightly lower than those of UFM. However, the antioxidant activities of FMM per polyphenol content were markedly higher than those of UFM. These results suggest that FMM may scavenge excessive reactive oxygen spices (ROS) via inhibition of XO and SOD-like activity. Furthermore, FMM demonstrated relatively higher acceptability and antioxidant ability along with functionality of $Hongguk$ (red yeast rice), and therefore could be utilized to prevent various ROS-induced diseases.

Effects of Dietary Supplementation of Mulberry Leaves and Silkworm Excreta Extracts on Weight Performance, Blood Characteristics and Cecal Microflorae of Chicks (뽕잎과 잠분 추출물 급여가 육계의 생산성, 혈액 성상 및 맹장 내 미생물군에 미치는 영향)

  • The object of this study was to evaluate the effects of dietary supplementation of mulberry leaves and silkworm excreta ethanol extracts on weight performances, blood characteristics, cecal microflorae of chickens. Two hundred forty male broiler chicks(Ross) were fed diets for five weeks containing 0.1%(MLA) and 1%(MLB) of mulberry leaves ethanol extracts, and 0.1%(SEA) and 1%(SEB) of silkworm excreta ethanol extracts. Weight performance did show no significant difference in all test groups which were fed with supplementation of mulberry leaves and silkworm excreta ethanol extracts. They showed better weight gain and feed conversion than the negative control group which was fed only with forage without antibiotics. ABTS(2'-azine-bis[3-ethylbenzothiazoline-6-sulfonic acid]) test was conducted to investigate free radical scavenging activity of blood in tested groups. ABTS scavenging activities of tested groups were higher than control groups in significant level, though there was no significant difference(P = 0.396). Specifically, MLB group showed the highest scavenging activity. Blood-level concentration of MDA, which is an indicator of lipid peroxidation, was also decreased in tested groups and the lowest level was observed in SEA(P = 0.001). As storage time increased at $4^{\circ}C$, muscle-level MDA concentrations of all tested groups were generally increased and significant difference was obsereved between tested groups and controls in total increase of MDA concentration($P=4.417{\times}10^{-3}$). In cecal microflorae, SEA and SEB showed decreased total microbe population compared to NC($P=6.462{\times}10^{-5}$) and even to PC. Supplementation of mulberry leave and silkworm excreta ethanol extract did show a similar inhibition effect against Salmonella sp., furthermore, MLB did enhanced the growth of Lactobacillus sp.($P=3.636{\times}10^{-7}$). In summary, ethanol extract of silkworm excreta may be a potential alternative of antibiotics for chicks. In addition, both of ethanol extracts supplementation to broiler chicks would be very useful not only to improve antioxidant effect of blood but also to suppress lipid peroxidation without any loss of weight performance in poultry farming.

The Effects of Cudrania tricupidata Tea Leaves on the Blood Glucose and Serum Lipids Profiles of Streptozotocin-Induced Hyperglycemic Rats (꾸지뽕잎차 첨가 식이가 Streptozotocin으로 유발한 고혈당 흰쥐의 혈당 및 혈청지질 함량에 미치는 영향)

  • The effects of pan-fired (PM) and fermented (FM) Cudrania tricupidata tea leaves on $\alpha$-glucosidase inhibitory activity, oral glucose tolerance, blood glucose levels and serum lipids profiles in streptozotocin (STZ)-induced hyperglycemic rats were investigated. The $\alpha$-glucosidase inhibitory activity of FM ethanol extracts (20 mg/mL) was higher (92.5%) than that of raw dried leaves (RM) (69.1%) and PM (54.6%). In addition, the results of a glucose tolerance test revealed that the glucose levels of hyperglycemic rats that were fed PM and FM ethanol extracts and then orally administered glucose began to decrease after 60 minutes, but recovered after 120 minutes. However, the blood glucose levels in the hyperglycemic control group did not begin to decrease for 360 minutes. Additionally, the results of animal experiments that were conducted over five weeks to compare the dietary effects of PM and FM following hyperglycemic induction to the effects on the hyperglycemic control group (DM) were as follows: The body weight gain and FER of the treated rats were $12.9{\sim}16.9%$ higher than those of the DM group, whereas the amounts of feed and water intake by the treated rats were $6.8{\sim}10.1%$ lower. Additionally, the levels of blood glucose and serum fructosamine decreased by $27.3{\sim}39.8%$ and $6.7{\sim}20.0%$, respectively, in the treated rats. Moreover, the serum triglyceride, total cholesterol and LDL-cholesterol concentrations in the treated rats were $24.9{\sim}27.1%$, $15.9{\sim}17.4%$ and $33.8{\sim}38.4%$ lower, respectively. Finally, the HDL-cholesterol contents were $20.5{\sim}24.8%$ higher in the treated rats than in the control group. The above results suggest that PM and FM exerts an anti-hyperglycemic effect that occurs due to the inhibition of $\alpha$-glucosidase activity as well as via prevention and/or inhibition of changes in the serum lipid profile. In addition, the results of this study revealed that the synthetic anti-hyperglycemic effect of FM was greater than that of PM. However, further detailed studies are needed to confirm these results.

Study on the Residual Tobacco poison to Cocoon Crops according to the Tobacco Cultivation of the Mulching System (개양 Mulching 담배 재배에 따른 잔류독성 기간에 관한 연구)

  • In autumn season residual period of nicotine is observed in the larval duration, weight of molting larvae at the various stages, percentage of delayed molting larvae, and survival rate up to the 3rd stage from the 1st season to the 3rd season and up to the stage for four seasons, respectively. Mulberry tree is planted in pot from June 18, 1973 to August 4, 1973 and placed every other row between tobacco variety of Hicks cultivated in mulching system and control is prepared in the Sericultural Experiment Station, placing in the every ridge of mulberry field with the four seasons ; 5 day isolation (1st season), 15 day isolation (2nd season), 25 day isolation (3rd season), and 50 day isolation (4th season). 1) 1st and 3nd instar larvae fed with the tobacco stained mulberry leaf has longer larval duration for the four seasons, as compared with those of the control, increasing 12% in index with 28 hours, 17% in index with 37 hours, 23% in index with 50 hours, 17% in index with 43 hours from the 1st season to the fourth season and the 3rd to the 4th instar larvae, increasing 18% in index with 55 hours. 2) 2nd instar molting larvae, 3rd instar molting larvae from the 1st to the 4th season and 4th instar molting larvae at the 4th season decrease their larval weight respectively, as compared with the control. 3) Percentage of delayed molting larvae from the 3rd stage to the 4th stage goes up higher than that of the control in the End larval stage at the 3rd to the 4th season, in the 3rd larval stage at the 1st, the End, and the 4th season, and in the 4th larval stage at the 4th season, respectively. 4) Survival rates in the 2nd larval stage at the 2nd season to the 4th season, in the 3rd larval stage at the 1st to the 4th season, and in the 4th larval stage at the 4th season respectively are higher than that in the control.

Mulberry Growth Promotion by Nitrogen Application under Abnormally Wet and Cool Weather Conditions (하추기 이상 저온하에서 뽕나무 발육부진요인과 추비에 의한 생육증진)

  • Abnormally cool and wet weather conditions during the summer of 1993 offered an opportunity to evaluate the effects of abnormal weather conditions on mulberry growth and to develop cultivation practicies to reduce leaf yield loss under the similar abnormal weather conditions. Different methods of nutrient supplementation were evaluated in Suwon and Kongju. Nitrogen was supplemented by foliar spray of urea(1.7%) or composite chemical fertilizer Jamsibiryo #8, or by the application of ammonium sulfate(60kg/ha) to the soil. During the period between the late June and the early September which corresponds to the mulberry growing season after summer pruning, mean temperature was 1.4$^{\circ}C$ lower and precipitation 83mm higher than normal year. This weather condition in 1993 caused reduction in leaf yield by 16.4% than common year. Soil nitrogen content decreased due to higher precipitation. Leaf yield loss was reduced by the supplementation of nitrogen to the soil. Leaf nitrogen content increased with the foliar urea spray and nitrogen supplementation to the soil.

  • The study was carried out to investigate the effect of ethrel (2-chloroethylphosophonic acid, 39%) on the graftages quality. Ethrel was sprayed 5 days interval from October 10 to October 30 at 6 places (Suwon (I), (II), Chungju, Kongju, Wanju, Jangsong). Spraying conentration was 1, 000 ppm (400 times dilution). 1. Spray took 3.3 hrs and 500 m of ethrel on 10a eara. 2. The passed rate of graftages decreased 5-8% on the treatments earlier spray than on October 20. 3. New shoot length and number of shoot of the ethrel treatments were the same as the natural leaf falling.

광식성 누에품종 육성

  • 1-4령 저염인공사료 공동육+5령 뽕잎육(농가개별육)$\longrightarrow$일주일 양잠에 적합한 광식성 누에품종육성에 필요한 기초지식을 얻기 위해 광식성유전자의 검색, 유전현상, 특성, 광식성계통 작출, 실용광식성 누에 품종육성방법 등에 대하여 소고하였다. 1. 광식성 유전자의 검색은 의잠을 사용하는 것이 가장 성력적이고 편리하며, 또한 섭식성을 정확히 파악할 수 있다. 2. 광식성 누에의 LP-1 사료에 대한 섭식성은 사육온도, 광조건, 발육단계 및 절식에 의하여 크게 영향 받지 않는다. 3. 광식성 유전자는 열성형질이며 1개의 주유전자와 수개의 수식유전자로 구성되어 있는 것으로 추찰된다. 4. LP-1 인공사료에 첨가한 morin 및 n-hexacosanol의 누에 섭식촉진효과는 미소하였으나, 상엽분말 첨가로 인한 누에 섭식촉진효과는 매우 양호하였다. 5. 광식성누에의 섭식성은 어분 및 효모에 의해 영향받지 않았다. 6. 섭식저해물질인 brucin 및 salicine은 정도의 차는 있으나 광식성누에의 섭식성을 저해하였다. 7. 광식성 및 비광광성누에의 LP-1 인공사료에 대한 섭식성의 차이는 누에의 두부에 있는 감각기관의 형태적 차이 때문이 아니라 기능적 차이에 기인하는 것으로 추찰된다. 8. 광식성 육성소재창출은 광식성유전자의 도입육종에 의해 가능하다. 9. 실용 광식성 누에품종육성을 위해 광식성검정에 적합한 인공사료를 사용하고, 실용형질이 우수하면서도 광식성유전자를 가진 계통을 선발, 교잡 또는 계통육성법에 의해 실용형질 및 섭식성을 개량하여 육종 목표를 달성하는 것이 최선책으로 생각된다.

Comparison of Leaf Productivity in Various Densities of Plantation with Mulberry Saplings or Grafts (접목묘 및 접목원묘의 뽕밭 식재밀도별 생산성 비교)

  • Profitability was analysed in the fields of various planting densities from 1,666 trees/10a to 4,166 with mulberry grafts or conventional saplings under the consideration of leaf yield and quality. Leaf yield per tree decreased with the higher planting densities. Seasonal and total leaf yield per area, however, increased by 20 to 63% as a mean for 3 years in the densities of 2,083~4,166 trees per 10a than in the conventional density of 1,666 trees per 10a. The increase in leaf yield per area was not so high in the densities over 2,083. Leaf yield in the sapling plots was a little higher than that in the graft plots. Topping of shoot tip affected neither on the branch length nor on yield. Yield was higher in the planting spacing with single raws than in that with double raws. Quality of leaves in the densities from 2,083 to 3,333 trees per 10a was relatively good judged based on the results of the pupation rate and cocoon yield and quality.

Study on the Effects of the Phosphate and Potassium Fertilizer Amount on the Mulberry Yields (숙전에서의 인산및 가리시비량이 뽕잎수량에 미치는 영향)

  • The effect of phosphorus and potassium on the leaf yielding from the mulberry field was surveyed, designing four application levels of potassium and phosphorus respectively; Po, P$^1$/$_2$ and P$_1$and P$_2$ for phosphorus, K$^1$/$_3$, K$^2$/$_3$, K$_1$and K$_2$for potassium after amount of nitrogen was constant at 25kg/l0a. The standard application of three elements is at 25-11-15kg/l0a. No application only showed significance among the treatments in the 3rd year autumn. However there is no significance, ever decreasing 2.7∼5.9% of leaf yielding in application of potassium and 6.4∼11.5% of leaf yielding in application of phosphorus. There is also no increase of leaf yielding even with application of double quantity of phosphorus and potassium. Potassium in soil was considerably fluctuated, while fluctuation of phosphorus is very low. With these results, it was evident that the conventional application level for phosphorus (11kg/l0a) is reasonable and for potassium may be cut down to 5∼10kg/l0a without showing any significance for the successive three years.

Study on Antioxidant Potency of Green Tea by DPPH Method (DPPH 방법을 통한 녹차의 항산화 활성에 대한 연구)

  • The present study was conducted to compare antioxidant activity of green teas, fermented teas and other related common teas by examining radical scavenging activity using DPPH (2,2 diphenyl l-picryl hydrazyl). Scavenging activity ($SC_{50}$/) of epigallocatechin gallate (EGCG) for 0.1 mM DPPH radical was 5.5 $\mu$M or 4.2 mg/L by weight, then catechin, 14 $\mu$M or 2.5 mg/L and vitamin C, 22 $\mu$M or 3.9 mg/L, respectively. Kyokuro tea (okro) powder of 24.2 mg/L or green tea powder of 25.2 mg/L was used to reach $SC_{50}$/ for 0.1 mM DPPH. One serving of 2 g green tea provides antioxidant activity equivalent to 109∼147 mg EGCG, 145∼185 mg catechin or 131∼168 mg vitamin C. Teas from the first harvest had the highest radical scavenging activity when compared with later harvest green teas grown in the same region, but there is virtually no difference by the harvest time. A Chinese green tea, Dragon well had the highest antioxidant activity among other green teas tested providing antioxidant capacity equivalent to 168 mg EGCG or 188 mg vitamin C per 2 g serving, but partially fermented Chinese teas had much lower antioxidant activity than any green tea tested. Black tea which is fully fermented showed as strong antioxidant activity as green teas (76.3 mg vs 86.7∼67.6 mg per tea bag). One tea bag of green teas from market provided antioxidant capacity equivalent to 52∼86 mg EGCG, 70∼105 mg catechin or 63-96 mg vitamin C. Teas made of persimmon leaf, pine needle, mulberry leaf had comparatively low anti-oxidant activity equivalent to 2.5∼4.8 mg EGCG or 15∼21 mg vitamin C per teabag. The third brewed green tea still had enough antioxidant activity, while tea from tea bag brewed for 3 min or 5 min did not have any difference in their antioxidant activity. More systemic studies are needed to clarify the relationship between tea catechins and antioxidant capacity focusing on how growing, harvest time, fermentation and other processes can influence on this.
