Sample Preparation for Quantitative Determination of Bisphenols in Canned Beverages Coated with Epoxy by RP-HPLC (RP-HPLC 에 의한 에폭시 통조림관 음료 중 비스페놀류의 분석을 위한 시료의 전처리)

  • A suitable method of sample treatments to minimize the analytical interferences was presented in order to determine bisphenols [bisphenol F (BPF), bisphenol A (BPA), bisphenol F diglycidyl ether (BFDGE), and bisphenol A diglycidyl ether (BADGE)] in various canned beverages coated with epoxy resin by the reversephase high performance liquid chromatography (RP-HPLC) equipped with a fluorescence detector and the gas chromatography with mass selective detection. The recovery test of bisphenols was performed using 1, 5, and 10 ${\mu}g/L$ bisphenols spiked beverages with the combined technique of the solid-phase extraction (SPE) and the liquid-phase extraction (LPE). Both BPA and BADGE showed quite adequate resolutions in HPLC-chromatograms. The recoveries of BPA obtained by LPE with diethyl ether were higher than those obtaind with methylene chloride on coffee, shikhye and fruit juice. For cola and tea, the recoveries of BPA obtaind by SPE were higher than those by LPE with diethyl ether. The recoveries of BADGE were less than those of BPA for all beverage samples treated by either SPE or LPE method. In survey of bisphenols for eighteen commercial canned beverage samples, BPA contents of coffee, cola, tea, shikhye, and fruit juice were in the range of $1.3{\sim}11.6,\;0.5{\sim}0.9,\;1.0{\sim}1.3,\;2.4{\sim}7.9$, and $3.0{\sim}3.4\;{\mu}g/L$, respectively, but there was no detection of BPA in beer sample.

내분비계 장애물질이 송사리의 생존에 미치는 영향과 홍삼의 효과에 관한 연구

  • 비스페놀 A, 노닐페놀 및 아트라진의 농도에 따른 생존율을 조사한 결과 같은 농도인 경우 노닐페놀에 노출한 실험군에서 상대적으로 낮은 생존율을 보였고, 비스페놀 A와 노닐페놀을 혼합한 실험군에 홍삼을 첨가한 경우 생존율은 상대적으로 높게 나타남을 알 수 있었다. 따라서 홍삼이 내분비계 장애물질의 독성을 부분적으로나마 억제하는 효과가 있는 것으로 사료된다.

Impact of the Sealant on Salivary and Urinary Bisphenol-A Concentration in Children (어린이에서 실란트 시술에 의한 타액 및 요 중 비스페놀-A 농도변화)

  • The purpose of this study was to quantify urinary and salivary bisphenol-A (BPA) concentrations according to sealant procedure among children. Nine students who had never treated with composite resin or sealant before, were recruited from one elementary school, Daegu, Korea from August 2013 to April 2014. Before sealant procedure, saliva and urine sample were collected. Immediately after sealant procedure saliva sample was collected and 24 hours after the procedure urine sample was collected. Creatinine was measured and adjusted to calculate urinary BPA concentration. Salivary and urinary BPA concentration after sealant procedure were $2.43{\pm}1.54{\mu}g/L$, $4.08{\pm}3.05{\mu}g/g{\cdot}$creatinine respectively, which were relatively higher than those before sealant procedure ($1.41{\pm}1.06{\mu}g/L$, $2.89{\pm}2.91{\mu}g/g{\cdot}$creatinine) but these were not statistically significant. We suggest that more large scale studies considering environmental confounders which have an effect on BPA are needed to establish the relationship between sealant exposure and BPA among children.

Vitellogenin mRNA Induction in Male Carp Treated with Bisphenol A (비스페놀A 처리 수컷 잉어에서 비텔로제닌 mRNA의 발현)

  • The estrogenic potency of bisphenol A using reverse trancriptase-PCR response of liver vitellogenin mRNA in male carp was studied. For this, six combination of primers which were synthesized on the basis of cDNA consensus region of various species, were evaluated and one pair of primers was selected as the best to show 286 bp size-transcript. By using the selected primers, vitellogenin mRNA induction in carp treated with bisphenol A was measured and the chemical showed dose-and time-dependent Induction response. From this result, it was concluded that RT-PCR technique wing the selected primers in this study can be wed to monitor the estrogenic effects exerted In carp living in Korean freshwater.

  • Laccase catalyzes the oxidation and polymerization of aromatic compounds in the presence of molecular oxygen. Studies were conducted to characterize the use of polyethylene glycol (PEG) as an additive to keep up the enzymatic stability. The enzymatic activities highly remained and bisphenol A (BPA) was rapidly converted in the presence of 5 mg/l of PEC. These effects were accomplished with PEG of molecular weight 3,350. A linear relationship was found between the quantity of BPA to be converted $(10-120\;{\mu}M)$ and the optimum dose of PEC required for greater than $95\%$ conversion. This result suggests that it is the interaction between the PEG and the reaction products. In the optimum dose of PEG, the aeration of reaction mixture neither enhanced the conversion of BPA nor retarded the inactivation of the enzyme.

  • A new simple, rapid and sensitive gas chromatographic technique for the determination of bisphenol-A in can materials, which is the major material of epoxy resin and polycarbonate polymer, is proposed. This method is characterised by derivatization of the bisphenol-A with a acylating reagent forming the acetate derivative to optimize the chromatographic property. The detection of bisphenol-A is performed based on GC/MS (gas chromatography/mass spectrometry). Several beverages were analyzed by the proposed method for the determination of bisphenol-A Bisphenol-A was assayed the range of $0.11{\sim}11.40{\mu}g/can$ from the can materials.

Screening of Peptide Sequences with Affinity to Bisphenol A by Biopanning (바이오패닝에 의한 Bisphenol A 친화성 펩타이드 서열의 탐색)

  • Bisphenol A (BPA) is a highly hazardous component to human since it is regarded as one of endocrine disruptors. For the analysis and/or removal of BPA, the searching for the specific ligand with a selective affinity to target BPA is required. In order to find the peptide moiety that specifically binds to BPA, the ultrasound-assisted biopanning was carried out with a phage-displayed peptide library expressing constrained heptamer. After six rounds of positive screening against BPA particles followed by the negative screening against the surface of eppendorf tube, the peptide sequence (CysLysSerLeuGluAsnSerTyrCys) with affinity to BPA was screened based on the order of frequency from the screened phage clones. To further verify the specificity of screened peptide sequence, the cross-binding affinity of the phage peptide toward BPA analogues such as Bisphenol S (BPS) and Bisphenol F (BPF) was also assessed, where the selected phage peptide showed a higher affinity to BPA over BPS and BPF.
