Modulation of Chemical Stability and Cytotoxic Effects of Epigallocatechin-3-gallate by Different Types of Antioxidants (Epigallocatechin-3-gallate의 화학안정성 및 세포독성에 미치는 각종 항산화제의 영향)

  • Epigallocatechin-3-gallate (EGCG) is a polyphenolic compound frequently found in green tea, and its physiological actions have been extensively investigated. In the present study, changes in chemical stability and cytotoxic properties of EGCG in the presence of different types of antioxidants were investigated. The antioxidants used modulated the chemical stability of EGCG. Superoxide dismutase (SOD) significantly increased EGCG stability; EGCG was less stable in the presence of catalase. Ascorbic acid, N-acetylcysteine (NAC), and glutathione (GSH) stabilized EGCG concentration dependently. The $H_2O_2$ level generated from EGCG was decreased by catalase, SOD, and NAC but not by GSH. The cytotoxic effects of EGCG also decreased in the presence of NAC, catalase, and SOD. GSH, however, showed a complicated modulatory pattern according to the EGCG and GSH concentrations, and ascorbic acid rather enhanced EGCG toxicity. The results suggest that certain antioxidants could modulate the cytotoxic properties of EGCG in a cell culture system not only by removing reactive oxygen species but by modulating chemical stability and other factors, which should be considered carefully when studying reactive oxygen species-dependent mechanisms of EGCG.

Stability of Protein Colloids in the Mixture of Cheese Whey and Soy Milk (유청(乳淸) 및 두유(豆乳) 혼합액에서의 단백질 콜로이드 안정성)

  • To investigate effectors on the colloidal stability of whey and soybean proteins, characteristics of tofu-gel formation, effects of heat treatment and salt composition on the colloidal stability, and effects of heat treatment on storage stability were analyzed. When experimental tofus were made from the mixture of whey and soybean, the calcium in the whey precipitated the soy proteins, and disrupted the gel formation, which resulted in the curd of poor texture. In the heat treatment at $60{\sim}100^{\circ}C$, whey and the whey proteins dialyzed against distilled water were readily preciptated at over $70^{\circ}C$, but the mixture of whey and soy extract as well as soy extract were stable at the range of temperature. The proteins of soy extract, whey dialyzed against sodium phosphate buffer, and the mixture were stable at the same heat treatment, and this suggested that phosphates in the soy extract stabilize specialty the whey proteins. Soy proteins were easily destabilized by adding $CaCl_2(0.05{\sim}0.07M)$ at $80{\circ}C$ and $70{\sim}85%$ of the proteins in soy extract and the mixture were preciptated, while only $30{\sim}55%$ of the proteins in whey dialyzed against distilled water were destabilized at the same conditions. Storage stability at $4^{\circ}C$ of the mixture was increased when the mixture was treated at $63^{\circ}C$ and lower temperature.

  • STO-3G level computations were performed on n-propylamine, n-propylamine radical and cis-and trans-ethylene diamines in order to investigate structural contributions of (n${\pi}$/m) and (n-${\sigma}^*$) structures to the energy variations accompanying the conformational changes. It was found that (5${\pi}$/5) and (4${\pi}$/4) structures had attractive and repulsive nonbonded interactions, respectively, which were approximately additive. anti(n-${\sigma}^*$) structures had more stabilzing hyperconjugative interactions than syn(n-${\sigma}^*$) structures, but due to the large internuclear repulsion the net effect was destabilizing inthe former in contrast with the net stabilizing contribution in the latter. Moreover it was found that the stabilizing ${\pi}$-nonbond structure, (5${\pi}$/5) was always cooperatively reinforced by the more stabilizing anti(n-${\sigma}^*$) interaction, whereas the destabilizing (4${\pi}$/4) structure was accompanied by the less stabilizing syn(n-${\sigma}^*$) interaction. This type of cooperativity was found general through-bond interaction of the terminal lone pair lobes split the energy levels into two, $n_+ = \frac{1}{\sqrt{2}}(n_1 + n_2)$ and $n_- = \frac{1}{\sqrt{2}}(n_1 - n_2)$, the latter being the lower level, which can be shown using simple overlap patterns of the two lobes with a common vicinal ${\sigma}^*$ orbital.

The Effects of Ethanol on Nano-emulsions Containing Quercetin Prepared by Emulsion Inversion Point Method (에멀젼 반전법으로 제조된 쿼세틴을 함유하는 나노에멀젼에 대한 에탄올의 영향)

  • The objective of this study was to find out the stable formulation of nano-emulsion containing high concentration of quercetin and to investigate the effect of an ethanol on the nano-emulsion prepared by POE (30) hydrogenated castor oil (HCO-30)/oil/quercetin/ethanol/water system. Nano-emulsion was prepared using emulsion inversion point (EIP) method as low-energy method plus homogenizer as high-energy method. To evaluate effect of ethanol and other components on the nano-emulsion, physical properties such as droplet size, morphology, and size distribution were determined. The optimal quercetin concentration was 0.2 % on the nano-emulsion. The droplet diameter was below 300 nm at the HCO-30 concentration below 2.00 %. Nano-emmulsion containing 4.75 % HCO-30 was the most stable and its mean droplet size was 172.40 nm. Finally, the size of nano-emulsion containing 4.00 % ethanol was 128.15 nm and size distribution was also narrow. The results showed that the breakdown process of this nano-emulsion could be attributed to Ostwald ripening. This study about effect of ethanol on the nano-emulsion showed that loading capacity of drug could be increased by using a small amount of ethanol. As prepared stable nano-emulsion, this study showed that these results could be applied to pharmaceutics, cosmetic including skin-care products, perfume and etc.

Factors Affecting Hemodynamic Instability in Patients with Pelvic Bone Fracture (골반골 골절 환자의 혈역학적 불안정화에 미치는 영향요소)

  • Purpose: Treatment and prognosis in patients with pelvic bone fracture depend on the characteristics of the fracture and the stability of the pelvic ring. The purpose of this study is to analyze the characteristics of and the relationships between fracture patterns, injury mechanisms, clinical courses, and prognoses according to the hemodynamic pattern. Methods: Between January 2004 and September 2006, 89 patients under diagnosis of pelvic bone fracture were retrospectively analyzed on the basis of medical records and radiologic examinations. Patients with confirmed hemorragic shock with a systolic pressure of less than 90 mmHg were defined as the shock group. Young's classification was used to characterize fracture patterns. Factors relating to the clinical manifestation and to treatments such as transfusion and surgery were analytically compared. Results: The mean age of the patients was $48.8{\pm}18.7$, among which 49 (55.1%) were male. The numbers of shock and non-shock patients were 35 (39.3%) and 54 (60.7%) respectively. Eighteen (51.4%) of the shock patient had injuries resulting from pedestrian accidents (p=0.008). According to Young's classification, lateral impact fractures amounted to 20 and 33, front-rear impact fractures to 9 and 20, and multiple fractures to 6 and 1 among the shock and non-shock patients, respectively (p=0.027). Thirty-nine (39) cases in non-shock injuries were conservatively managed while 18 cases in shock injuries were surgically treated. In the shock group, the liver and the kidney were often damaged, as well. Among the shock patients, the average admission period was $7.5{\pm}8.7days$ in intensive care and $55.1{\pm}47.9days$ in total, which were longer than the corresponding numbers of days for the non-shock patients (p<0.05). No deaths occurred in the non-shock group while 5 deaths (14.2%) occurred in the shock group (p=0.007). Conclusion: The factors affecting hemodynamic instability in patients with pelvic bone fracture are injury mechanism, classification of fracture, and associated injuries.

Inventory Investment and Business Cycle: Asymmetric Dynamics of Inventory Investment over the Business Cycle Phases (재고투자와 경기변동: 재고투자 동학의 경기국면별 비대칭성)

  • When it comes to explaining the relationship between inventory investment and business fluctuations, the production smoothing theory and the stock-out avoidance theory take contradictory stances. Decision-making related to inventory investments of corporations is thought to be influenced by both motives, but the relative sizes or directions of their respective influences can differ depending upon the phase of the business cycle. Against this backdrop, this paper differs from existing studies in that it theoretically tests the relative significances of the production smoothing and stock-out avoidance motives in the inventory investment dynamics, while placing its analytical focus on determining the existence and patterns of the asymmetric dynamics of inventory investment over the business cycle phases. To this end this paper sets up a non-linear model that is expanded from the existing linear inventory investment model, and checks whether its predictive power is better than that of the existing model. The results of analysis confirm the nature of the asymmetric dynamics of inventory investment over the business cycle phases. A stock-out avoidance motive appears but there is no significant production smoothing motive in boom times. In downturns, in contrast, the stock-out avoidance motive is insignificant, but a quality of asymmetric dynamics in which changes in inventory cause the deepening of recessions, due to the non-convexity of production costs proposed by Ramey (1991), is detected. This paper confirms that a model considering the asymmetric dynamics of inventory investment can have better predictive power than one that does not consider it, through within-sample and out-of-sample predictions and various predictive power tests. These research results are expected to be useful for economic forecasting, through their enhancement of the understandings of the inventory investment dynamics and of the nature of its business cycle destabilization.

Fabrication of a Nano/Microfiber Hybrid Mat for Control of Mechanical Properties and Porosity (기계적 특성 및 공극률 조절을 위한 나노/마이크로섬유 하이브리드 매트 제작)

  • Fine polymeric fibers have been gaining interest from the energy harvesting/storage, tissue, and bioengineering industries because of advantages such as the small diameter, high porosity, permeability, and similarities to a natural extracellular matrix. Electrospinning is one of the most popular methods used to fabricate polymeric fibers because it is not as limited in regards to the materials selection, and it does not require expensive or complex equipment. However, electrospun fibers have a severe aerodynamic instability because the small diameter fibers are able to pass through the atmospheric layer when there is a high electric field. As a result, electrospun fibrous mats have serious difficulties with controlling its shape and geometric properties. In this study, a hybrid nano/microfibrous mat is presented that is fabricated using electrospinning with two different solvent-based PCL solutions. This provides control of the fiber diameter, mat porosity, and mechanical properties. Various hybrid fibrous mats were fabricated after an experimental investigation of the effects of solvent on fiber diameter. It was then demonstrated that the mechanical properties and porosity of the fabricated various hybrid mats could be successfully controlled.

A Review Study in Treatment for Anxiety Disorder in Traditional Chinese Medicine (불안장애 치료에 관한 최근 임상연구 동향)

  • Objectives : This study is set out to discover more about the treatment of anxiety disorder in Traditional Chinese Medicine. Methods : We searched the database-China Academic Journals(CAJ). For papers published since 2007-2011, which are classified in categories. Results : We have finally selected 23 articles, and the results were as follows: 1. Stages of Anxiety disorder are discussed in various aspects. 2. Diagnostic criteria and evaluation criteria were the same, especially, in the CCMD and HAMA was used most frequently. Adverse effects were reported using TESS. 3. There are various ways, such as Herbal medicine, acupuncture, electroacupuncture stimulation, acupoints injection, and abdominal massage therapy to treat Anxiety disorders in TCM. 4. Zizyphus jujuba, Poligala tenuifolia, Poria cocos are mainly used in herbal medicine. Sinmun(HT7), Sameumgyo(SP6), Naegwan(PC6) are mainly used in acupuncture points. 5. The treatment group, cured by herbal medicine or acupuncture, was similar in its therapeutic efficiency to that of the control group, which was cured by western medicine. In some cases, the treatment group was better off than the control group. Herbal medicines have been reported that they have less side effects and drug addiction than the western medicines. We predict that herbal medicine in the treatment for anxiety disorders will have an advantage. Conclusions : There are a wide range of approaches in the treatments for Anxiety disorders in TCM. In some aspects, herbal medicine is more effective than that of the western medicine. According to this study, we predict that herbal medicine in the treatment for anxiety disorders, has some advantage. Therefore, there is a need for more clinical research for this disorder in Korean traditional medicine.

Interim Report about The Comparative Clinical Study of Efficacy of Gamisoyo-san(Jiaweixiaoyaosan) on Anxiety of Generalized Anxiety disorder (범불안장애의 불안증상에 대한 가미소요산의 제형별 효능 비교 임상연구의 중간보고)

  • Objectives : The purpose of this research is to examine the efficacy of Gamisoyosan on anxiety of generalized anxiety disorder according to dosage form. Methods : In this randomized, double blinded, placebo-controlled study, we planned to give Gamisoyosan simple extract mixture or Gamisoyosan compound extract or controlled medication for major symptoms of generalized anxiety disorder. As preparatory research, Hamilton rating Scale for Anxiety(HAM-A) was measured as the 1st evaluative instrument, and Korean State-Trait Anxiety Inventory(STAI-K), Penn State Worry Questionnaire(PSWQ) Korean Beck's Depression Inventory (BDI-K), Symptom Checklist-90-Revised(SCL-90R), WHO Quality of Life Abbreviated(WHOQOL-BREF) and Heart Rate Variability(HRV) were also measured as the 2nd evaluative instrument at the before treatment. Results : Demographic characteristics showed that there are Clinical characteristics-vital signs are within the normal range. The characteristics of disease-chief complaint, pattern Identification and etiological factor of the highest frequency number were worry, heart deficiency with timidity(心膽虛怯), family matters. The average period of disease in subjects is 6.31years. The results of Chest PA, EKG and clinicopathologic examination are within the reference range. The Scores of HAM-A, STAI-K, PSWQ were measured above the cutoff point. There are significant positive correlations among HAM-A, STAI-K, BDI-K and among HAM-A, STAI-K, PSWQ. There are no significant correlations between PSWQ and BDI-K. Conclusions : We considered that selection of subjects in this research is appropriately accomplished And this methodology is expected to be applied to the subsequent research. And also, we hope to make up for this study through various study and discussion.

The Effect of Destabilization Heat Treatment on the Carbide and Matrix Microstructures of 3%C-18%Cr-1%Mo-2%Ni-1%Mn High Chromium Cast Iron (3%C-18%Cr-1%Mo-2%Ni-1%Mn 고크롬백주철의 탄화물 및 기지조직에 미치는 불안정화열처리의 영향)

  • 3%C-18%Cr-1%Mo-2%Ni-1%Mn high chromium cast iron was casted and destabilized at temperatures of 900, 1000 and $1100^{\circ}C$ for 1, 2, 4 and 8hr under $N_2$atmosphere to observe the effect of destabilization temperature and time on the carbide and matrix structures. In as-cast condition, the microstructure consisted of $M_{ 7}$$C_3$ carbides and matrix structures which were composed of 91.50% austenite and 8.50% martensite. Numerous fine secondary carbides were observed in the specimens destabilized at $900^{\circ}C$ for 1, 2, 4 and 8hr. But, the number of secondary carbides were much reduced with the increased destabilization temperature. More austenite was formed in the matrix with the higher destabilization temperature. The amounts of austenite in the matrix were 4.23% at $900^{\circ}C$, 29.68% at $1000^{\circ}C$ and 66.51% at$ 1100^{\circ}C$, respectively. However, the effect of destabilization time on the secondary carbide and matrix was very weak compared with that of destabilization temperature. The ranges of the amount of austenite in the matrix from 1hr to 8hr destabilization heat treatment were: 3.95%-4.35% at $900^{\circ}C$, 28.89%-30.15% at $1000^{\circ}C$ and 65.13%-67.12% at $1100^{\circ}C$, respectively. The variation ranges were very narrow. The equilibrium concentration of C and Cr in austenite was already reached within 1hr during destabilization heat treatment. After an attainment of the equilibrium concentration of C and Cr in austenite, no more secondary carbide was precipitated from the matrix.
