Development of Optical Molecular Imaging System for the Acquisition of Bioluminescence Signals from Small Animals (소동물 발광영상 측정을 위한 광학분자영상기기의 개발)

  • Purpose: Optical imaging is providing great advance and improvement in genetic and molecular imaging of animals and humans. Optical imaging system consists of optical imaging devices, which carry out major function for monitoring, tracing, and imaging in most of molecular in-vivo researches. In bio-luminescent imaging, small animals containing luciferase gene locally irradiate light, and emitted photons transmitted through skin of the small animals are imaged by using a high sensitive charged coupled device (CCD) camera. In this paper, we introduced optical imaging system for the image acquisition of bio-luminescent signals emitted from small animals. Materials and Methods: In the system, Nikon lens and four LED light sources were mounted at the inside of a dark box. A cooled CCD camera equipped with a control module was used. Results: We tested the performance of the optical imaging system using effendorf tube and light emitting bacteria which injected intravenously into CT26 tumor bearing nude mouse. The performance of implemented optical imaging system for bio-luminescence imaging was demonstrated and the feasibility of the system in small animal imaging application was proved. Conclusion: We anticipate this system could be a useful tool for the molecular imaging of small animals adaptable for various experimental conditions in future.

Highly sensitive and selective detection of cyanide in aqueous solutions using a surface acoustic wave chemical sensor (표면음향파 화학센서를 이용한 수용액 중 시안화이온의 선택적인 고감도 검출)

  • We report a highly selective and sensitive 200 MHz Surface Acoustic Wave (SAW) sensor that can detect cyanide ion in aqueous solution using surface immobilized thioester molecules in combination with gold nanoparticles (AuNPs). To construct the sensor device, a monolayer of thioester compound was immobilized on the SAW sensor surface. At the sensor surface, hydrolysis of thioester group by nucleophilic addition of cyanide occurred and the resulting free thiol unit bound to AuNP to form thiol-AuNP conjugate. For the signal enhancement, gold staining signal amplification process was introduced subsequently with gold (III) chloride trihydrate and reducing agent, hydroxylamine hydrochloride. The SAW sensor showed a detection ability of $17.7{\mu}M$ for cyanide in aqueous solution and demonstrated a saturation behavior between the frequency shift and the concentration of cyanide ion. On the other hand, our SAW sensor had no activities for other anions such as fluoride ion, acetate ion and sulfate ion, moreover, no significant interference observed by other anions. Finally, all the experiments were carried out in-house developed sensor and fluidics modules to obtain highly reproducible results.

  • Polydiacetylene(PDA)-based sensors possess a number of properties that can be successfully applied for label-free detection system. PDA is one of the most attractive color-generating materials, with growing applications as sensors. Here we introduce various PDA-based devices, used as biosensor, chemosensor, thermosensor, and optoelectronics sensor. In general, PDA liposomes and films are closely packed and properly designed for polymerization via 1,4-addition reaction to form an ene-yne alternating polymer chain. PDA-based two/three dimensional structures have been used for colorimetric or fluorescent devices, sensing biological as well as chemical components. This color-generating material also present a very high charge carrier mobility, allowing its application as field-effect transistor (FET). The immobilized PDA structures or films have distinct advantages for the detection of low concentration target molecules over the aqueous solution-based detection systems. In the present review, reported detection methods by using various PDA structures are summarized with updated references.

Effect of Sequence Variation in Bovine Mitochondrial DNA D-Ioop Region on ~ilk Production for Hanwoo (한우 산유량에 미치는 Mitochondrial DNA D-loop영역의 염기서열 변이효과)

  • This study was performed to analyze the sequence variations of mtDNA D-loop and their effects on milk in Hanwoo(Korean cattle). The resulting sequences were compared with previously published sequences for other cattle breeds (GenBank JOI394). The Polymerase Chain Reaction was performed to amplify a total of 964 bp between nucleotide 15758 and 383 within D-loop region of mtDNA using specific primers. Twenty polymorphic sites by nucleotide substitution were found in mtDNA D-loop region of Hanwoo. The frequencies of positions at 8, 169, 16042, 16051, 16057, 16093, 16119, 16122, 16209, 16255 and 16302 nt with high levels of sequence polymorphism were 0.150, 0.950, 0.085, 0.138, 0.106, 0.085, 0.138, 0.212, 0.085, 0.148 and 0.180, respectively. The substitution effect at 16119(p<0.1) and 16185(p< 0.05) nt was found significant on milk production. Polymorphism of mtDNA sequence in D-Ioop region could be useful for the analysis of cytoplasmic genetic variation and associations with the other economically important traits and maternal lineage analysis in Hanwoo.

Development of Ultra-Rapid Reverse-Transcription PCR for the Rapid Detection against Slow Bee Paralysis Virus (SBPV) (Slow Bee Paralysis Virus (SBPV) 신속 검출을 위한 초고속 역전사 중합효소 연쇄반응법의 개발)

  • Slow Bee Paralysis Virus (SBPV) is a pathogenic virus against honeybee and bumblebee, causes the death of adult bee by paralyzing the fore-leg of bee. In this study, for rapid detection of SBPV from bumblebee, SBPV-specific Ultra-rapid Reverse transcription PCR was developed. After optimizing of SBPV-specific Ultra-rapid PCR, the existence of $1.0{\times}10^8$ SBPV-specific DNA molecules could be recognized in 3 minute and 35 seconds. Even $1.0{\times}10^1$ molecules of SBPV-specific DNA could be measured with quantitative manner. Meanwhile, from both imported bumblebee and bumblebee produced in Korea, SBPV were detected using proposed method. In the laboratory as well as in the field, SBPV-specific Ultra-rapid Reverse transcription PCR would be applied and might be expected as useful tools at production of bumblebee or inspection for the import and export system of bumblebee.

  • 수 MV급 탄뎀형 가속기와 정전형, 자장형 질량분석기를 결합함으로써 존재비가 극히 작은 동위원소 측정이 가능해진다. 수 MeV로 가속이 되면 분해에 방해가 되는 모든 분자이온들이 제거되며, 탄뎀형 가속기에서는 음이온으로부터 가속이 개시되므로 몇몇 음이온을 형성하지 못하는 동중원소의 간섭으로부터 자유로워지기 때문에 고감도분석이 실현될 수 있다. 이외에 음이온을 형성하는 동중원소들은 주로 이온함인 최종 검출기에서의 유효 전하에 따른 에너지손실 차이를 이용하여 효과적으로 제거할 수 있다. 현재는 주로 장반감기 방사성동위원소인 $^{10}Be$, $^{14}C$, $^{26}Al$, $^{36}Cl$$^{129}I$ 등의 측정법이 확립되어 천연시료 중에서 동위원소 존재비 $10^{-12}$에서 $10^{-15}$까지의 정량이 가능하며, 원자수로 환산한 검출하한을 $10^5$개가 된다. 또한 해당 원소를 기준으로 소요 시료량은 대부분 mg 정도로 충분하다. 지금까지는 불가능했던 이러한 특징으로 인해 지난 수년간 AMS(Accelerator Mass Spectrometry)가 활용되어 온 연구분야는 지구과학(기후학, 우주화학, 빙하학, 수문학, 해양학, 퇴적학, 화산학 및 광물탐사), 인류 및 고고학(연대측정), 그리고 물리학(천체물리, 핵 및 입자물리) 등으로 다양하다. 이외에 생의학 및 재료과학 분야에서도 AMS를 활용하고자 하는 노력이 계속되고 있다. 본 해설에서는 가속기질량분석기술의 특징, 원리, 장치 및 활용분야 등을 소개하고자 하며, 이로써 관련 분야의 연구 활성화에 기여하고자 한다.

Thermo-Field emission in silicon nanomembrane ion detector for mass spectrometry (실리콘 나노 박막의 열-전계 방출효과를 이용한 분자 질량분석)

  • This paper describes the characteristics of thermo-field emission in a freestanding silicon nanomembrane under ion bombardment with various thermal and field conditions. The thermal effect and field effect in thermo-field emission in silicon nanomembrane are investigated by varying kinetic energy of ions and electric field applied to the silicon nanomembrane surface, respectively. We found that thermo-field emission increases linearly as the electric field increases, when the electric field intensity is lower than the threshold. The thermo-field emission (schottky effect) increases proportionally to the power of temperature, which agree well with the predictions of a thermo-field emission model.

  • Determination of xanthan gum as watersoluble-polymer in commercial cosmetic samples was carried out by High Perfomance Liquid Chromatography(HPLC). An $C_{18}$ reversed-phase column and the selected ELSD detector was applied. The 25mM ammonium acetate/acetonitrile was used for the mobile phase of gradient conditions. The analysis results of HPLC showed good linearity with correlation coefficient of $r^2=0.9993$ in the rage of $50.3{\sim}604.1{\mu}g/ml$ and detection limit of $12.0{\mu}g/ml$.

Molecular Characterization of Iridovirus in Cultured Turbot, Scophthazmus maximus (양식산 Tubot, Scophthalmus maximus 감염원으로 확인된 Iridovirus의 분자생물학적 연구)

  • The high mortality of cultured turbot, Scophthalmus maximus was occurred from Gochang in June, 2003. Infect fish with iridovirus showed a lower feed intake and lethargic. These fish exhibited pale body color, extended abdominal and exophthalmus. Histopathological studies showed basophilic enlarged cells from kidney, spleen, gill, heart, muscle, stomach, intestine, liver and pancreas of these fish. In PCR amplification with red sea bream iridovirus (RSIV) specific primer set of a DNA fragment of 286 bp was obtained from infect turbot with the virus. The strain showed the high homologies with RSIV, LBIV (largemouth bass iridovirus), GSIV (giant seaperch iridovirus) and SBIV (sea bass iridovirus).

Interface Engineering in Superconducting Ultra-thin Film of Ga (Ga 극초박막의 계면특성과 초전도 물성제어에 대한 연구)

  • Spin polarized tunneling studies were carried out with Al-Ga bilayer as a spin detector, by Meservey-Tedrow technique. The superconductor (SC)/Insulator (I)/Ferromagnet (FM) tunnel junctions were provided by ultra high vacuum molecular beam epitaxy (UHV-MBE) technique. The analysis of interfacial properties in the Al-Ga bilayer was also carried out by Auger electron spectroscopy. It was observed that the superconducting transition temperature and energy gap were raised in comparison with that of bulk Ga and pure ultrathin Al films. Current studies clearly show how one can modify the material properties at the interface just with a few monolayers.
