Assessment of Mycobacterial Viability by Fluorospectrophotometry (형광분광측정법에 의한 항산균의 생명력 평가)

  • Viable potential of Mycobacterium smegmatis, a slow grower in vitro cultivation and of M. leprae, an obligate intracellular parasitic bacterium, which can not be cultured yet in vitro was assessed by fluorospectrophotometry. Bacterial cells in different numbers and under various physiological status were incubater with fluorescein diacetate(FDA). After an incubation of the bacterial preparations with FDA at specified conditions, amount of fluorescein inside bacteria was measured by a fluorospectrophotometer at 470nm and 510nm of excitation and emission wavelengths, respectively. Fluorounit given by such bacteria showed a correlation with assessment of viability of the same preparations made by other methods, such as optical density and colony forming units of M. smegmatis and intracellular ATP content of M. leprae. The possible use of fluorospectrophotometry in assessing viability or physiological potential of bacteria, particularly intracellular parasites and fastidious organisms to culture in vitro is discussed in relation to other methods.

  • Rare earth elements including Y and Th in the monazite were separated and determined by X-ray fluorescence spectrometry. The matrix effects among the rare earth elements were simultaneously corrected by means of empirical coefficient method. The values of the coefficients were quite dependent on the number of the standards. However, the different set of coefficients led to the same results. The analytical results corrected by the present method agreed with those by the inductively coupled plasma spectrometry.

  • 본 연구는 양전자 소멸 측정 분광법을 통하여 BaSrFBr:Eu 형광물질의 결함 농도 분석을 시도하였다. LABO를 이용한 동시계수 도플러 방법과 Fast - Slow - Coincidence 시스템으로 구성한 양전자 수명 분광법을 통하여 에너지의 변화에 따른 양성자 조사에 의한 시료 결함에 따른 동시 계수 도플러법과 양전자 수명의 변화를 측정 하였으며, SRIM 시뮬레이션을 통한 에너지에 따른 양성자 투과 깊이의 변화를 연구하였다.

Measurement of Fluorescence Correlation Function by Using Size and Concentration of Fluorescence Particles (형광입자들의 크기와 농도에 따른 형광 상관 분광함수 측정)

  • The concentration and hydrodynamic radius of nano-sized fluorescence particles diffusing in solution were compared by using fluorescence correlation spectroscopy (FCS), which can measure the variation of the correlation function of a fluorescence signal by size and number of particles. The used nano-sized fluorescence particles are Alex Fluor 647, quantum dots, and fluorescence beads, and three kinds of sample solutions with different concentrations were prepared by dilution to 1/10 and 1/100 with distilled water for each kind of particles. The effective focal volumes were calculated by using the known diffusion coefficient of Alexa Fluor 647 particles, and the diffusion time, number of particles in focal volume, and variation of concentration according to the dilution could be measured by the FCS system. Through this study, we determined that the concentrations of arbitrarily diluted sample solutions can be measured by a home-built FCS setup in the range of 0.1 nM ~ 10 nM and that the diffusion coefficient of the quantum dot was $27{\pm}1{\mu}m^2/s$.

Analysis on Red-colored Dyeing by using Non-destructive UV-visible and Fluorescence Spectrophotometry (비파괴 자외-가시 및 형광 분광 분석법을 이용한 적색계 전통 염료 분석)

  • This study examined non-destructive UV-Vis spectrophotometry as well as 3-D fluorescence spectrophotometry of textile that made use of red dye such as Sappan wood, madder, Safflower, Gromwell. The authors produced two textile specimen that were dyed by not only two kinds of textile (cotton and silk) but also three kinds of mordanting (no-mordanting, alumen and iron), and they investigated effects of each dye material upon investigation results. At analysis with UV-Vis spectrophotometry of dyed textile specimen, dyeing made by sappan wood, madder and gromwell had significant difference depending upon mardant regardless of kinds of textile, and safflower had no significant difference depending upon textile and mordant. At analysis with 3D-fluorescence spectrophotometry, specimen dyed with sappan wood had difference with mordants, and with madder, there were difference with textiles, and safflower had inherent fluorescence spectrum regardless of textiles and mordants, while gromwell had no fluorescence spectrum.

  • Membrane fouling control in water treatment may be the main obstacle for wider implementation and lower cost. A novel fluorescent spectroscope sensor device for membrane fouling integrity monitoring has been developed and evaluated in this study. PSf membranes for water treatment has been fabricated with three types of organic fluorescent materials, OB, FP, KCB. The fluorescent signal from membrane surface was analyzed throughout the filtration process. It was found that the fluorescent signal due to the membrane fouling decreased and the developed device is reliable for membrane fouling monitoring.

  • Czochralski 방법으로 KMg $F_3$:S $m^{2+}$ 결정을 성장시키고 고 분해 레이저 분광법을 이용하여, 형광특성을 조사하였다. S $m^{2+}$ 이온이 KMg $F_3$ 결정에서 $C_{4v}$, $C_{2v}$, $C_{3v}$$O_{h}$ 형광 방출 사이트를 가진다는 것을 확인하였고, 아울러 각 사이트의 에너지 준위를 구하였다. 구하였다.

  • 저전압용 형광체로 주목받고 있는 SrTiO$_3$:Pr,Al 형광체를 고상반응법으로 제조하였다. 부활성체 Al이 23 mol% 첨가되었을 때 SrTiO$_3$:Pr 형광체는 최대 발광 강도를 보여주었다. 최대 발광 강도를 보인 형광체에 대해 제한시야회절상과 투과전자현미경을 이용하여 SrTiO$_3$:Pr,Al 적색 형광체내 2차상 형성에 대한 연구를 행하였고 또한 에너지 분산 분광 분석을 행하여 SrTiO$_3$:Pr,Al 형광체내 주입된 부활성제 Al의 함량을 결정하였다. 제한시야회절상의 결과에 의하면 SrTiO$_3$:Pr,Al 적색 형광체내에 단사정 SrAl$_2$O$_4$, 삼사정 Ti$_4$O$_{7}$, 육방정 SrAl$_{12}$O$_{19}$가 2차상으로 존재함을 보여 주었다. 에너지 분산 분광 분석 결과 부활성제 Al의 함량 중 일부는 SrTiO$_3$:Pr,Al 적색 형광체 격자에 고용되고 일부 Al은 SrAl$_{12}$O$_{19}$와 SrAl$_2$O$_4$의 2차상을 형성하는데 소모됨을 보여주었다.주었다.
