Isolation and Characterization of Oil Degrading Bacteria from Southern Sea of Korea (남해안 해수로부터 원유 분해 세균의 분리 및 특성)

  • A marine bacterium having a high oil-degrading activity was isolated form the oil-polluted southern sea of Korea, and was identified as Pseudomonas aeruginosa and was named Pseudomonas aeruginosa BYK-2. The optimal tmeperatur, culture time, pH and NaCl concentration for biosurfactant production and cell growth showed $25^{\circ}C$, 48h, 7.0 and 0%(w/v), respectively. After cultivation at $25^{\circ}C$, 180 rpm in 250 mL erlenmeyer flask for 7days, 1%(w/v) arabian light crude oil and bunker C oil which are considered to be hardly degradable compounds were degraded 92.1%(w/w) and 76%(w/w) respectively. And then, cell adherence was measured on various carbon sources. The cell adherence indicated over 80% on hydrocarbons(arabian light crude oil, kuwait curde oil, bunker C oil, n-paraffine, n-hexadecane, n-tetradecane) as carbon sources. Lecithin among fatty acids(oleic acid, olive oil, lecithin) showed highest cell adherence of 91.5%. The cell adherence of sugars(arabinose, trehalose, dextrose, galactose, lactose, fructose, maltose, sorbitol, sucrose) observed to be less than 70% except for arabinose, galactose, sorbitol and sucrose.

A Study on the Multi-sensor Toilet (다중센서 양변기에 관한 연구)

  • In this paper, a flushing system with automatic opening and closing function of the toilet cover was studied. It works by an infrared sensor attached to the cover is detected or not. When the infrared sensor of the open function detects it, the servomotor is driven on the toilet cover to raise the cover, and when the infrared sensor with the close function detects the infrared sensor, the cover is set to be lowered. A tilt sensor is attached to the inside of the cover to operate when the cover goes down, and when this is activated, the servomotor connected to the toilet lever opens the stopper of the supply port and sends water down. In addition, we minimized the inconveniences for the next user when they use the toilet by using the non-contact water level sensor and LED function to notice the water is clogged during the flushing process. Also, we implemented UV-LED function to prevent bacterial growth while the toilet is closed.

Evaluation of Dental Hygienist Job Validity according to Judgment Standard of Medical Practice in Medical Law (의료행위 기준에 따른 치과위생사 직무 타당도 평가)

  • The purpose of this study was to evaluate the validity of dental hygienist job according to judgment standard of medical practice in medical law. In this study, we conducted a self-administered questionnaire survey to evaluate the validity of dental hygienist job for 12 professors of dental college in Gangneung-Wonju National University from November 10 to 20, 2017. We investigated whether the dental hygienist job conforms to the three criteria of medical practice such as disease prevention and treatment, patient care, and health hazard. The response rates were scored and classified into four categories according to the final score. As a result of this study, dental hygienist job are classified into four categories according to judgment standard of medical practice. The higher the level of the category, the higher the degree of difficulty, and the higher the level of expertise and skills required. More than 50% of respondents answered that measuring the gingival pocket, bleeding on probing, professional tooth cleaning, oral health education, counseling after dental treatment are all three criteria for medical treatment. And these were classified into Level 4 group which requires the difficulty and expertise in the final score 4.3. It is necessary to develop and utilize standardized guidelines on the level of knowledge, education, and qualification standards required for dental practice in order to effectively allocate work among the dental personnel while ensuring the health rights of patients in the dental clinic field in Korea. In addition, there is a need to evaluate the various aspects of cost effectiveness, dental health service productivity, and health promotion contribution to dental hygienist jobs, And based on this evidence, it is necessary to continue to expand and adjust the dental hygienist job and to reorganize the dental workforce system.

Prevalence of Microbiological Hazard on Nursery School Children's Hands and Effect of Hand Washing Education (어린이집 유아 손의 미생물학적 위해 평가 및 손 씻기 교육의 효과)

  • This study was conducted to evaluate the microbiological hazard on nursery school children's hands and to investigate the reduction effect of hand washing education. A total of 59 nursery school children's hands were tested. The average number of total aerobic bacteria was $3.72{\pm}0.38log\;CFU/hand$. Five children's hands(2 male and 3 female) were positive(14.3%) for the coliform bacteria. These results showed that hand washing education are required repetitively. Among the pathogenic bacteria tested in this study, $Staphylococcus$ aureus and $Bacillus$ $cereus$ were detected in 9(25.7%) and 16(45.7%) out of 35 their hands, respectively. Twelve out of sixteen $B.$ $cereus$ isolates(70.0%) produced enterotoxin. The results indicate that the hand hygiene of nursery school children needs to be improved. Comparing before and after hand washing in educated and non-educated group, the reduction effect of total aerobic bacteria on their hands was 0.42 and 0.60 log CFU/hand, respectively. The educated group showed 0.18 log CFU/hand higher reduction effect than non-educated group but microorganism did not eliminate perfectly. From the results, using a hand sanitizer after washing with soap and the continuous hand washing education are required to control the contaminated bacteria on nursery school children's hands.

Monitoring of Biosafety of Agricultural Products from Urban Community Gardens and Roof Gardens in Korea (도시텃밭과 옥상정원에서 재배되는 농산물의 생물학적 안전성 조사)

  • Urban farming supplies emotional stability and fresh vegetables to participating persons, however, no information regarding the biosafety of agricultural products from urban farming is available. Here, we collected 260 samples of Chinese cabbages and lettuce from 4 urban community gardens and 6 roof gardens in Seoul from September through October 2012, and monitored the microbiological and parasitological contamination quantitatively and/or qualitatively. The mean counts of total aerobic bacteria and coliforms were $6.1{\pm}0.8\;log\;CFU{\cdot}g^{-1}$ ( range, $5.4{\pm}0.6{\sim}7.1{\pm}0.8\;log\;CFU{\cdot}g^{-1}$) and $4.0{\pm}0.7\;log\;CFU{\cdot}g^{-1}$ (range, $2.3{\pm}0.6{\sim}6.1{\pm}0.9\;log\;CFU{\cdot}g^{-1}$), respectively. Coliforms were detected on 59.6% among 260 vegetable samples. There were no significant differences in the contamination levels of total aerobic bacteria and coliforms between the Chinese cabbages and lettuce, whereas both levels of vegetables from urban community gardens were higher than those of roof gardens (p > 0.05). Escherichia coli was isolated at 3.1% among whole vegetables, and contamination level was $1.5{\pm}0.2\;log\;CFU{\cdot}g^{-1}$. Among foodborne pathogens, Staphylococcus aureus was detected in 1.5%, however, Salmonella spp., Listeria monocytogenes and E. coli O157:H7 were not detected on any of the vegetable samples. We also found undefined parasite eggs from two samples of Chinese cabbages (0.8% of total vegetables we tested). From these study, we found the presence of microbial contamination of agricultural products from urban farming, thus we need further concern to improve the biosafety during production of agricultural products.

DETECTION OF SALIVARY STREPTOCOCCUS MUTANS LEVELS USING MONOCLONAL ANTIBODIES (단클론항체를 이용한 타액 내 Streptococcus mutans 수준의 측정)

  • Streptococcus mutans, one of the major causal agents of dental caries, is component of the dental plaque. It produces various organic acids such as lactic acid which is the end-product of glycolysis, and this leads to dental caries. A new system using species-specific monoclonal antibodies was developed to detect Streptococcus mutans in saliva. The system quickly detects salivary Streptococcus mutans in 30min and classifies the result into two levels. The purpose of this study was to investigate correlation between monoclonal antibody-based detecting system and selective medium-based detecting methods. Children's deft indices were also compared with Streptococcus mutans counts in MSB agar plate. Subjects consisted of 15 children in the age of 3 to 6 years. They were assigned to three groups : Group I(deft index = 3), Group II(deft index $\leqq$3), Group III(deft index $\geqq$4). The results are as follows : 1. The rate of children with positive response was 13.3% and with negative response was 86.7% in the result of Saliva-checkTM Mutans test kit. 2. There was a positive correlation between monoclonal antibody-based detecting system and selective medium-based detecting methods(p<0.05). 3. Streptococcus mutans counts in MSB agar plate were irrelevant to deft of children(p=0.34).

Structure-activity Analysis of Benzylideneacetone for Effective Control of Plant Pests (벤질리덴아세톤 화학구조 변이에 따른 생리활성 변화 분석 및 식물 병해충 방제 효과)

  • Benzylideneacetone (BZA) is a compound derived from culture broth of an entomopathogenic bacterium, Xenorhabdus nematophila (Xn). Its immunosuppressive activity is caused by its inhibitory activity against eicosanoid biosynthesis. This BZA is being developed as an additive to enhance control efficacy of other commercial microbial insecticides. This study was focused on the enhancement of the immunosuppressive activity of BZA by generating its chemical derivatives toward decrease of its hydrophobicity. Two hydroxylated BZA and one sugar-conjugated BZA were chemically synthesized. All derivatives had the inhibitory activities of BZA against phospholipase $A_2$ ($PLA_2$) and phenoloxidase (PO) of the diamondback moth, Plutella xylostella, but BZA was the most potent. Mixtures of any BZA derivative with Bacillus thuringiensis (Bt) significantly increased pathogenicity of Bt. BZA also inhibited colony growth of four plant pathogenic fungi. However, BZA derivatives (especially the sugar-conjugated BZA) lost the antifungal activity. These results indicated that BZA and its derivatives inhibited catalytic activities of two immune-associated enzymes ($PLA_2$ and PO) of P. xylostella and enhanced Bt pathogenicity. We suggest its use to control plant pathogenic fungi.

EFFECT OF SURFACE ROUGHNESS OF ACRYLIC RESIN ON THE ADHESION OF BACTERIA (Acrylic resin 표면의 거칠기에 따른 세균부착 비교)

  • Statement of problem: The microbial adhesion on the surface of materials used in prosthodontics and restorative dentistry significantly influences microbial infection. Purpose: The purpose of this study was to evaluate the effect of how the degree of surface roughness of acrlyic resin affect the adhesion of bacteria. Material and methods: Resins were finished with $50{\mu}m$ and $250{\mu}m$ aluminium oxide particles by using sandblaster, by using stone point, and high polished with $Opa^{(R)}$ and Lace $motor^{(R)}$. The surface of acrylic resin attached by bacteria was directly touched on the surface of BHI agar, which was incubated. Bacteria colonies formed on BHI agar were counted in accordance with the degree of the surface roughness. Results: 1. The viable cell number of Streptococcus mutans increased on the acrylic resins incubated in BHI broth than in PBS. 2. The viable cell number of Streptococcus mutans increased on the acrylic resins incubated without agitation than with agitation, washed three times than six times, and incubated in broth added with 5% sucrose than without sucrose. 3. When Streptococcus mutans incubated in BHI broth, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins finished with $250{\mu}m$ aluminium oxide particle using sandblaster. But when incubated in BHI broth containing sucrose, the number of colonies formed on that was the largest on the acrylic resins high polished using $Opal^{(R)}$ and Lace $motor^{(R)}$. 4. When Streptococcus sanguis was incubated in BHI broth with or without sucrose, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins finished with $250{\mu}m$ aluminium oxide particle using sandblaster. 5. When Actinomyces viscosus was incubated in BHI broth with or without sucrose, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins high polished using $Opal^{(R)}$ and Lace $motor^{(R)}$. Conclusion: These results indicated that when acrylic resins attached by bacteria were touched on the surface of BHI agar, the number of bacterial colonies formed on the agar was dependent on the bacterial species. Also, the result of this study was showed that increase in the surface roughness and the addition of sucrose increased retention of microbial cells.

Cloning and expression of Streptococcus mutans GS-5 glucosyltransferase (Streptococcus mutans GS-5 Glucosyltransferase의 클로닝과 발현)

  • Dental caries is an infectious disease caused by mutans streptococci, and is a primary etiologic agent of dental caries in humans. The molecular pathogenesis of mutans streptococcal-associated dental caries occurs in three phases. Firstly, S. mutans attaches to tooth surface via a cell surface adhesion termed antigen I/II. In the second phase, the glucosyltransferase(GTFs) synthesize polymers like glucans in the presence of sucrose. In the third phase, the multivalent glucans interacts with glucan binding proteins (GBPs) and they make dental plaque and accumulation of microorganisms. Many studies and clinical trials have indicated that a mucosal immune response to these antigens(Ag I/II, GTFs, GBPs) of S. mutans can influence the pathogenesis of dental caries. So these antigens can be important vaccine candidates for immunologic intervention against dental caries. In this study, we cloned the genes for GTFb, GTFc, GTFd from S. mutans GS-5 and did the nucleotide sequence analysis. And the recombinant proteins of GTFd and N-terminus of GTFd were expressed. Intact GTF which we get from this experiment can be used for antibody production specific for any GTF activity domain through animal experiment.

Pathogenesis of Inflammation in H. pylori Infection

  • 위의 parietal cell 혹은 대식세포와 유사한 세포 내부에서 H. pylori가 발견된다는 보고가 있기는 하나 일반적으로 H. pylori는 Shigella와 같은 침습성 세균은 아닌 것으로 알려져 있다. 그럼에도 불구하고 H. pylori에 감염된 위점막에는 많은 수의 호중구를 위시한 염증세포의 침윤이 관찰되는데 H. pylori가 위상피세포에 부착 할 경우 위상피세포를 자극하여 interleukin-8을 위시한 cytokine 을 발현케하고 이에 의하여 호중구 등의 염증세포가 몰려들게 된다. 한편 고유층에 몰려든 호중구에서는 다시 interleukin-8을 위시한 일련의 호중구 활성화 chemokine을 분비하여 염증반응을 증폭해 나갈 것이다. 호중구에서 발현되는 myeloperoxidase나 활성 산소 등도 위점막의 조직 손상에 기여할 것이다. 위상피세포를 덮고 있는 점액층은 위상피세포를 보호한다고 알려져 있으나 H. pylori 감염의 경우 점액층에 의하여 H. pylori의 운동성이 증가하고 이것이 위상피세포로부터의 cytokine 발현을 자극하여 염증반응을 증폭하는데 관여할 가능성도 있다. H. pylori는 위상피세포에 대하여 apoptosis를 유도함과 동시에 고유층에 몰려든 호중구에 대하여는 apoptosis를 억제케하여 궁극적으로 염증반응을 증폭 및 지속시켜 나가는 쪽으로 작용한다. 한편 H. pylori는 위상피세포로부터 COX-2의 발현을 증가시키는데 이는 위상피세포의 apoptosis를 억제하는 방향으로 작용한다. 이외에 H. pylori의 urease에 의하여 발생한 암모니아나 H. pylori 자신이 분비하는 세포독소가 세포 손상을 유발할 가능성도 있다. 상술한 여러 독성 인자들 중 어느 하나가 단독으로 작용하기보다는 여러 인자가 같이 동시에 또는 시차를 두고 작용할 가능성이 많다고 생각된다.(\gamma-FeOOH)$, 침철광$(\alpha-FeOOH)$, 적금광$(\beta-FeOOH)$, 그리고 자철광$(Fe_3O_4)$이다. 인위적 부식에서는 전부 인철광의 부식물이 생성되었고 자연적 부식에서는 모두 침철광의 부식물이 생성되었다. 특히 철제 표면에 자연적으로 생성된 공식 녹을 XRD 분석한 결과 적금광으로 동정되었다. 이런 모든 시편들을 각 탈염방법에 따라 탈염처리한 후 XRD와 SEM-EDS으로 분석한 결과 인철광과 침철광은 어떠한 변화도 보이지 않았고, 다만 적금광으로 동정된 시편만이 잔존하지 않았다. 철기 제작별 $Cl^-$ 이온 추출량과 탈염효과에 대한 비교 실험은 이온 크로마토그래피 분석 결과와 마찬가지로 단조 철제유물이 주조 철제보다 $Cl^-$ 이온을 많이 가지고 있었으며, 탈염 처리 후에는 $Cl^-$ 이온은 전혀 발견되지 않았다. 이상의 결과 $K_2CO_3$와 Sodium 용액은 탈염처리에서 가장 적합한 탈염처리 용액으로 알수가 있었으며 특히 어떠한 탈염 용액으로 유물을 처리한다 해도 철제유물에 생성된 부식물은 제거되지 않는다는 것을 알게 되었다. 따라서 보존처리자는 유물 표면의 부식 상태만을 보고 처리하기 보다는 철기제작물로 고려하여 처리하는 것이 필요하다. 또한 금속에 부식을 야기시키는 $Cl^-$ 이온과 부식물을 완전하게 제거하여 탈염처리를 하는 것이 유물 부식을 최대한 지연시킬 수 있는 것이라 생각된다.TEX>$88\%$)였다.(P=0.063). 결론: 본 연구에서는 MTHFR C/T & T/T 유전자 다형성이 위암의 발생과 그 위치에 대해 관련이 있는 것으로 여겨지고, 흡연력, 음주력과는 관련이 없는 것으로 여겨진다.험이
