Effect of Attachment Substrate Size on the Growth of a Benthic Microalgae Nitzschia sp. in Culture Condition (실내 배양시 부착기질 크기에 따른 저서성 미세조류 Nitzschia sp.의 성장 특성)

  • To understand the effect of attachment substrate on the growth of benthic microalgae, we experimentally investigated the growth of benthic microalgae Nitzschia sp. (Jinhae Bay strain) with additions of glass beads in different sizes. The glass beads used in this study are 0.09-0.15 mm (G1), 0.25-0.50 mm (G2), 0.75-1.00 mm (G3) and 1.25-1.65 mm (G4). No addition of glass beads used as controls. Highest specific growth rate (0.37/day) and maximum cell density ($9,232{\pm}840$ cells/mL) of Nitzschia sp. showed at the smallest glass beads (G1), and the specific growth rate and maximum cell density were decreasing with increasing size of glass beads (specific growth rate and maximum cell density of G4 was 0.24/day and $6,397{\pm}524$ cells/mL, respectively). Moreover, specific growth rate of the control experiment (0.23/day) was significantly lower than their of G1 to G3 experiment. The results indicated that the attachment substrate for benthic microalgae as Nitzschia sp. is important factor which affecting the growth rate as well as cell density. Therefore, the physiological experiment of benthic microalgae seems to be necessary to preliminary experiment, which is addition or not of the attachment suitable substrate and the grain size for the target species of benthic microalgae.

Relative Immigration Activity of Epilithic Diatom in Sum River : Comparison of Natural and Artificial Substrate (섬강수계 부착규조의 유입능 특성: 자연기질과 인공기질의 비교)

  • Effects of substrates on the relative immigration activities (RIAs) of epilithic diatoms were examined in Sum River, a tributary of South Han River, Korea. Two hundreds of tile substrates coated with 5% agar were deposited at seven study stations (30 tiles per site) for one month from March 7 to April 7, 2008. Water sampling, physico-chemical factor measurement, and diatom analysis were performed in the field and laboratory. Over the study, major epilithic diatom species were Nitzschia amphibia, Navicula subminuscula, Cymbella minuta in both the natural and artificial substrates. Two dominant species, Nitzschia amphibia and Cymbella minuta often found at seven stations. Among all observed taxa, Nav. subminuscula and Caloneis silicula showed the highest RIA, with the value over 15 throughout the study. Regarding the algal morphology, the biraphe type species showed higher RIA than any other morph, while they comprised over 55% of total biomass, indicating a density effect. Additionally, a significant relationship between RIA and 20-day deposited substrate (p<0.05) showed in two diatom species, Nitzschia fonticola, Gomphonema quadripunctatum, which showed relatively high RIAs. These results indicate that the relative immigration activities of epilithic diatom species can be influenced by algal density in the water and substrate, nutrients, and deposited time at least 20 days.

The Microalgal Attachment and its Growth on the Artificial Surfaces Immersed in Seawater: I. Attachment and Micro-succession (해수에 잠긴 인공기질 표면에서 미세조류의 부착과 성장: I. 부착 및 천이)

  • To understand the attachment of micro algae and their subsequent growths on artificial surfaces immersed in seawater, the relationship between attachment of diatoms on the immersed artificial substrates and species pool in the surrounding water was investigated. We used acryl slides for the study of diatom attachment and examined the surrounding water samples collected in Incheon Harbour from July 1995 to February 1997. Variations of species composition and abundances by exposure time in seawater were investigated during the early phase of biofilm formation on various substrates, e.g. glass, acryl, titanium, copper and antifouling paint-treated slides. Immigration rates of diatoms to acryl slides during spring and winter were significantly correlated with the abundance of benthic diatoms in surrounding water ($r^2$=0.78, p<0.01, n=42), suggesting that immigration rates were affected by variations of benthic diatom abundances in surrounding water. Immigration coefficient of monoraphid diatoms was 5 times higher than that of biraphid diatoms, but relative abundance of monoraphid diatoms was 3 times lower than that of biraphid diatoms on acryl slides in spring. In winter, immigration coefficient and relative abundance of centric diatoms were higher compared to other raphe forms. These results suggest that the attachment of diatoms seems to be caused by the abundance and immigration coefficients of benthic diatoms in surrounding water. Pennate diatoms predominantly attached to all artificial surfaces throughout all experimental periods. Interestingly, centric diatoms predominantly attached to all artificial surfaces in winter. Hantzschia virgata, Licmophora abbreviata and Melosira nummuloides appeared dominantly on antifouling paint-treated slides, probably being tolerant of the antifouling paint. During incubations, the abundance of attached diatoms increased exponentially on glass, titanium and acryl slides with exposure time. The maximum abundance was highest on glass slide, followed by acryl, titanium, copper and antifouling paint-treated slides. The growth rates of attached diatom community on all artificial surfaces were higher at temperature of $24-25^{\circ}C$ than that of $2-3^{\circ}C$. The growth rate of attached diatoms on glass slide was generally higher compared to other slides during the study period. Dominant morphotypes of observed species with exposure time in seawater were prostrate form Amphora coffeaeformis, fan shape Synedra tabulata, stalk type Licmophora paradoxa and chain type M. nummuloides. A micro-succession in the attached microalgal community was observed. The composition of dominant species seems to be the result of species-specific response to gradually limited space with development of microalgal film.

  • The extracellular matrix (ECM) is a key factor affecting cell growth and adhesion to the culture surface, and it is also important for maintaining the innate characteristics of cells. Here, we describe the effects of the ECM on cardiomyocyte (HL-1 cell line) growth, viability, phenotype, and contractile ability. Five different ECM materials were investigated to analyze their effects on the cell growth. The physical morphology of the ECM-coated surfaces was scanned with an atomic force microscope (AFM), and the attachment, growth, proliferation, viability, and phenotype of the cells were analyzed using fluorescence immunostaining and an inverted phase contrast microscope.

An Effect of Pre-attached Bacteria on Attachment of Diatoms to Artificial Surf aces Immersed in Seawater (해수에 잠긴 인공기질 표면에 먼저 부착된 박테리아가 규조류의 부착에 미치는 영향)

  • Controversial observations on the effect of pre-attached bacteria on the attachment of diatoms to artificial surfaces immersed in seawater have been made. Furthermore, it is not known whether or not pre-attached bacteria on artificial surfaces influence attachment of diatoms in natural seawater. In this study, we used various surfaces to which marine bacteria were pre-attached for different incubation periods. In the first experiment, glass slides were initially attached by marine natural bacteria with different exposure time (0-133 hr) and then immersed into seawater for a certain time period. The attachment of diatoms was not affected by the abundance of pre-attached bacteria (p > 0.05). The maximum abundance of attached diatoms was found on control surfaces, and the minimum abundance on surfaces where attached bacterial abundance was highest. In the second experiment, glass slides and acryl slides, either attached by marine natural bacteria for 6 days or coated by agar, were immersed in seawater. Untreated slides were also employed. On the surfaces of acryl slides with the most abundant attached bacteria ($5.4{\pm}0.02{\times}10^5\;cells\;cm^{-2}$), abundances of attached diatoms were less than those on untreated slides. On the surfaces of glass slides with bacterial abundance of $2.5{\pm}1.0{\times}10^5\;cells\;cm^{-2}$, however, abundances of attached diatoms were not different from those of untreated slides. On the agar-coated slides, the immigration rate and immigration coefficient were on average > 2 folds compared to other surfaces, indicating high rates of diatom attachment on mucilage simulated surfaces. Therefore, it seems that pre-attachment of bacteria is not prerequisite for the attachment of diatoms on artificial surfaces.

  • Kinetic parameters of various substrates and inhibitors were measured to elucidate the binding requirements of the active site of intracellular adenosine deaminase (ADA) in Aspergillus oryzae. 3'-Deoxyadenosine was the best substrate according to the value of relative kcat/$K_m$. Purine riboside was found to be the strongest inhibitor with the $K_i$ value of $3.7{\times}10^{-5}$M. Adenine acted neither as a substrate nor as an inhibitor, suggesting the presence of ribose at N-9 of adenosine was crucial to binding. ADA also catalyzed the dechlorination of 6-chloropurine riboside, generating inosine and chloride ions. Substrate specificity of 6-chloropurine riboside was 0.86% of adenosine. Purine riboside, a competitive inhibitor of ADA, inhibit the dechlorination with similar $K_i$ value, suggesting that the same binding site was involved in deamination and dechlorination. Among the sulfhydryl group reagents, mercurials, pchloromercuribenzoate (PCMB), mersalyl acid and $HgCl_2$ inactivated the enzyme. Mersalyl acid-inactivated ADA was reactivated by thiol reagents, but PCMB-inactivated enzyme was not. When ADA was treated with the mercurial reagents, the inhibition constants and inhibition patterns were determined. Each inhibition was competitive with substrate. The $K_i$ values of these mercurial reagents were lower in 10 mM phosphate buffer than in 100 mM phosphate buffer, showing phosphate dependency.

Inhibitory Effects of Asadisulphide on the TEA-induced Adherence of HL-60 Cells (TPA로 야기된 HL-60 세포의 기질부착에 대한 Asadisulphide의 억제 효과)

  • Asadisulphide were purified from Ferrula assafoetida by organic solvent extraction and chromatography. Its inhibitory effects on the TPA-induced adherence of HL-60 cells was analyzed. Since ethyl acetate extracts of F. assafoetida has the strongest inhibitory effect on adherence of HL-60 cells, it was re-extracted with ethyl acetate, hexane, and ethyl ether and chromatographed three times to isolate asadisulphide. At the minimum concentration of 2 $\mu\textrm{g}$/ml, asadisulphide inhibited adherence of 98% of HL-60 cells that have been treated with TPA. It also showed anti-cancer effect with no cytotoxity in the ED$_{50}$ value of 20 $\mu\textrm{g}$/ml.

Tissue Engineered Bone Formation Using Porous Chitosan and Chitosan/Tricalcium Phosphate Matrices (Chitosan 및 Chitosan/Tricalcium Phosphate 다공성 기질을 이용한 조직공학적 골형성에 관한 연구)

  • chitosan은 골치유증진 및 골세포의 분화를 촉진하는 것으로 알려진 천연의 생분해성 고분자이다. 이연구에서는 chitosan 및 chitosan/tricalcium phosphate(TCP) 다공성 기질을 제조하여 골이식재 및 조직공학적 골형성을 위한 3차원적 세포배양 지지체로서의 가능성을 평가하고자 하였다. chitosan 용액 및 TCP가 포함된 chitosan 용액을 동결건조함으로써 소공의 크기가 $100-200{\mu}m$인 스폰지형태의 chitosan 및 chitosan/TCP 다공성 기질을 제작하였다. 골이식재로서의 효과를 평가하기 위하여 백서의 두개골 결손부에 제작된 chiosan 및 chitosan/TCP 다공성 기질을 각각 이식하고 2주 및 4주 후에 동물을 희생하여 조직학적으로 치유양상을 관찰하였다. 조직공학적 골형성을 위한 세포배양 지지체로서의 가능성을 평가하기 위하여 백서 태자의 두개골에서 분리된 골아세포를 chitosan 및 chitosan/TCP 다공성 기질에 각각 접종하고 56일간 배양하면서 각 기간 별로 세포수, 염기성 인산효소 활성, 축적된 calcium의 양을 측정하였고 배양된 세포-기질 혼합체를 광학현미경 및 주사전자현 미경하에서 조직학적 관찰을 시행하였다. 백서 두개골결손부에 이식된 chitosan 및 chiosan/TCP 다공성 기질은 별다른 이물반응 없이 자연 분해되면서 신생골조직 내에 매립되었으며 이식하지 않은 대조군에 비해 유의하게 높은 신생골형성 효과를 나타내어 우수한 골전도성이 있음이 확인되었다. 신생골형성 양상이나 형성된 양에 있어서 두 가지 기질간의 유의한 차이는 없었다. 골아세포-기질 혼합체의 배양결과, 접종후 배양 28일 경과 시까지 골아세포수는 지속적으로 증가하다가 이후에는 5 8일까지 성장정도가 둔화되었다. 염기성 인산효소의 활성 및 calcium 축적량은 접종후 배양시간경과에 따라 56일까지 지속적으로 증가하였다. 세포수 및 염기성 인산효소의 활성에서 두 기질간의 유의한 차이는 없었고, calcium 축적량에 있어서는 chitosan/TCP 기질에서 유의하게 높았고 증가속도도 컸다. 배양된 골아세포가 접종된 다공성 기질의 조직학적 관찰결과, 골아세포는 다공성 기질에 잘 부착하여 중층의 형태로 성장하면서 광화된 골기질을 형성함이 관찰되었다. 배양 14일부터 작은 골편형태의 골형성이 기질 표면에 부착되어 관찰되었고, 배양기간이 길어짐에 따라 성장하여 배양 56일째에는 상당한 양의 광화된 골질이 형성됨이 관찰되었다. 배양 56일 경과후의 광화된 골질의 양은 chitosan/TCP 기질에서 더 많았다. 이 연구의 결과, chitosan 및 chitosan/TCP 다공성 기질이 골이식재로서 뿐만 아니라, 조직공학적 골형성에 적용되는 골아세포의 배양을 위한 3차원구조의 세포지지체로 이용되어 골재생술식에 유용한 생체재료로 활용될 수 있음이 확인되었다.

  • In order to investigate the occurrence of Pectinatella magnifica in Nakdong River, extensive series of sampling was conducted through July to November of 2014. Results revealed that these species show preference to attach themselves on natural substrates over artificial substrates. P. magnifica does not show preference for specified substrates, but itappearthat availability of substrates determines their specific distribution. Considering that most commonly found substrates in Nakdong River were natural substrates such as dead twig, woody plants or aquatic plants, it is possible that high availability of substrates is one of the principal factors which increase the rates of growth and distribution of P. magnifica.
