  • We have researched on methods which can reduce fruit-body malformations of king oyster mushroom(Pleurotus eryngii). We collected many pathogens from diseased fruit-body or malformated fruit-body and identified with chemicobiological test and microscope. The factors of fruit-body malformations and increment of contamination during the pin-heading induction time were researched with ventilation amounts in growing room. When the pathogens having high pathogenicity were inoculated in spawn running bottle and at pin-heading induction time, symptom appeared or not appeared in according to air ventilation amounts in growing room. During the pin-heading induction time, humidity degree in growing room have kept of high level and air ventilation amounts were so little that fruit-body malformations ratio was high. But, even though pathogens were inoculated at the surface of bottle factitiously, if air ventilation amounts were enough, fruit-body malformation ratio was low.

In vitro Fruit Assay for the Evaluation of Fungicide Activity Against Pepper Anthracnose (살균제 효과 검정을 위한 고추 탄저병의 실내 열매 검정법)

  • This study was performed to investigate the effect of conidial density, wetness period and temperature on conidial germination, appressoria formation and disease incidence. While there was not significantly correlated between conidial density and temperature, and conidial germination and appressoria formation, there was a significant correlation between those factors and disease incidence. The longer wetness period was, the higher the ratio of conidial germination, appressoria formation and the disease incidence was. The optimum conidial density, temperature and wetness period was $1{\times}10^6$ conidia $mL^{-1}$, $30^{\circ}C$ and 5 days, respectively. In case the wetness period was more than 5 days, the typical symptom was not found on pepper fruits because of the overgrowth of mycelia. Using this fruit assay method, which the pepper anthracnose pathogens were inoculated by spraying spore suspension on non-wounded or wounded pepper fruits, control effect of three fungicides were evaluated against pepper anthracnose by the protective and/or the curative application. Propineb showed high protective control activity, while it showed curative control activity on unwounded fruits, but did not showed curative control activity on wounded fruits. Tebuconazole, one of curative fungicide, showed higher control activity in non-wound inoculation than wound inoculation. Trifloxystrobin, one of strobilurin group, showed high both protective and control activity against anthracnose. In conclusion, we supposed that the newly developed in vitro pepper fruit assay can be used to evaluate antifungal activity of control agents against pepper anthracnose.

Isolation of Antifungal Bacterial Strain Bacillus sp. against Gray Mold infected in Kiwi Fruits and its Disease Control (참다래 잿빛곰팡이 병원균에 대한 길항균 Bacillus sp. 분리와 병해 억제 작용)

  • This study was carried out to identity the effects of antifungal bacteria isolated from the soil grown kiwi fruit plants on the growth inhibition of Botrytis cinerea causing gray mold in kiwi fruit plants in the southern districts of Jeonnam. Two hundred and fifty antagonistic microorganisms were isolated and examined into the antifungal activity against Botrytis cinerea. We screened and isolated four bacterial strains which strongly inhibited Botrytis cinerea from the soil grown kiwi fruit plants. And the best antifungal bacterial strain which called CHO 163 was finally selected. Antagonistic microorganism CHO 163 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. CHO 163 showed 86.9% of antifungal activity against Botrytis cinerea. By the bacterialization of culture broth and heated filtrates of culture broth, Bacillus sp. CHO 163 showed almost all of antagonistic activity against Botrytis cinerea. And we also confirmed that in vitro the treatment of Bacillus sp. CHO 163 cultured by SD+B+P broth efficiently controled the growth of Botrytis cinerea causing gray mold in kiwi fruit plants.

Effect of Gamma Irradiation and Its Convergent Treatments on Lily Leaf Blight Pathogen, Botrytis elliptica, and the Disease Development (감마선 및 화학 대체제 융복합 처리가 백합 잎마름병균 및 잎 절편 감염에 미치는 영향)

  • Gamma irradiation and its convergence with nano-silver particles and sodium dichloroisocyanurate (NaDCC) were investigated to inhibit germination and mycelial growth of Botrytis elliptica, the pathogen of lily leaf blight. In addition, the same treatments were studied on the process of disease development with detached leaf of lily cv. Siberia. Spray inoculation, which is closer to natural infection than wound inoculation, can be a way to investigate infection ability of the treated pathogen. The irradiating dose required to reduce the population by 90%, $D_{10}$, was 526 Gy irradiating with 0-2000 Gy gamma ray on the conidial suspension as well as the growing mycelia. Even at 2000 Gy, the mycelium was not killed but just delayed its growth at 1-2 days behind. Convergent treatment with 40 mg/l of NaDCC just before 200 Gy gamma irradiation was the best way to decrease the conidial germination about 1/1000 times. The control values of gamma irradiation were 23% and 19.5% at wound inoculation and spray inoculation, respectively. On wound-inoculation, the control value of NaDCC only was 89%, and that of NaDCC convergent with 200 Gy gamma irradiation was 32%. On sprayinoculation, the highest control value was NaDCC at 50%, and that of NaDCC convergent with gamma irradiation was 24%.

Studies on the Relationship between Inoculum Concentration and Disease Development in Bacterial Leaf Blight of Rice (벼흰빛잎마름병에 있어서 병원균의 접종 농도가 병의 발전속도에 미치는 영향)

  • The study has been carried out for the development of an inoculation method in screening resistant varieties and/or lines to bacterial leaf blight of rice with special consideration on plant ages and inoculum concentrations. A higher incidence of the disease was found on younger plants than on the older ones when the plants were inoculated with the same concentration of inoculum by clipping method under the same circumstances. Applications of extremly high concentration of inoculum resulted the death of younger seedlings from all varieties within short period after inoculation, and the inoculum that was lower than optimum concentration failed in distinguishing varietal characteristics in regard of resistance. The proper inoculum size for the screening of resistance among the varieties and/or lines depended on the age of plants. The optimum concentration of inoculum for the plant age of 14, 37, 48 and 58days was $10^6,\;10^7,\;10^7-10^{-9}\;and\; 10^9 cells/ml.,$ respectively.

  • Pathogenic isolates of Alternaria mali produced host-specific toxin(AM-toxin) in liquid culture. The toxin was also released by germinating spores of the fungus. The physiological event of apple leaves induced by germinating spores was an increased loss of electrolytes from susceptible leaves. This reaction was evident soon after spore inoculation, indicating that the leakage was caused by AM-toxin from germinating spores. Typical symptoms were developed only in susceptible leaves of apple within 48hr after inoculation with pathogenic spores. Similar symptoms occurred on susceptible leaves when non-pathogenic isolates plus AM-toxin were used.

Effect of Inoculum Concentration of Rhizoctonia solani and Pythium ultimum Causing Damping-off of Pepper and Cucumber on the Efficacy of the Mixture of Etridiazole and Thiophanate-methyl (Rhizoctonia solani와 Pythium ultimum의 접종 농도가 etridiazole과 thiophanate-methyl 합제의 모잘록병 방제 효과에 미치는 영향)

  • This study was performed to investigate the inhibiting activity of etridiazole and thiophanate-mthyl on mycelial growth of Rhizoctonia solani and Pythium ultimum and the effect of inoculum density of each pathogen on the control efficacy of the mixture of etridiazole and thiophanate-methyl in a seedling assay test. In mycelial growth inhibition test, $EC_{50}$ values of etridiazole and thiophanate-methyl against R. solani were 15.87 and 9.34 ${\mu}g\;mL^{-1}$, while those were 0.2 and more than $500\;{\mu}g\;mL^{-1}$ against P. ultimum, respectively. Controlling activity of the mixture of etridiazole and thiophanate-methyl against damping-off of pepper and cucumber, caused by R. solani and P. ultimum was tested in a greenhouse. With the inoculation of R. solani, disease incidences of seedling in pepper were 73% and 95% at 0.5% and 1.0% of inoculum concentration, while in cucumber those were 55% and 62% at 1.0% and 2.0%. When P. ultimum was inoculated into soil by 2.0% of inoculum concentration, those in pepper and cucumber were 66.7% and 96.8%, respectively. The efficacy of the mixture was somehow affected by the concentration of R. solani. While each control value of the mixture was 94.4% and 90.7% in pepper and cucumber at low inoculum concentration (0.05%), the efficacy of the fungicide decreased in pepper and cucumber by 70.7% and 72.9% at high concentration of R. solani (0.1% in pepper and 0.5% in cucumber). However, the control value of the mixture was 100% in pepper and cucumber, irrespective of the inoculum concentration of P. ultimum, however, the increase of inoculum concentration in soil did not result in the decrease of the fungicide efficacy.

Characteristics of Flexibacter columnaris Isolated from Tilapia (Tilapia sp.) (틸라피아(Tilapia sp.)에서 분리한 Flexibacter columnaris의 성상)

  • The characteristics of a pathogen isolated from infected tilapia, which showed the external symptoms of fin erosion and body deceleration, were examined. The strain of isolated pathogen was identified as Flexibactor columnaris, The normal tilapia dipped into suspensions of the isolated pathogens has developed the infective symptoms. The strain of Flexibactor columnaris was sensitive to kanamycin, tetracycline, and amikacine, but resistant against sulfa drugs. This strain could not grow in the cytophaga media containing over $2\%$ NaCl or over $50\%$ sea water. In cytophaga broth containing more than $50\%$ or sea water instead of distilled water, the number of Flexibacter columnaris decreased from about $10^6/ml\;to\;10^4ml$ within one hour after inoculation. On the other hand, in cytorhaga broth containing more than $5\%$ of NaCl, the number rapidely reached less than 10/ml from about $10^5/ml$ within one hour after inoculation.

Development of Nested-PCR Assay to Detect Acidovorax citrulli, a Causal Agent of Bacterial Fruit Blotch at Cucurbitaceae (박과 작물에 과일썩음병을 일으키는 Acidovorax citrulli 검출을 위한 nested-PCR 검사법 개발)

  • The specific and sensitive nested-PCR method to detect Acidovorax citrulli, a causal agent of bacterial fruit blotch on cucurbitaceae, was developed. PCR primers were designed from the draft genome sequence which was obtained with the Next Generation Sequencing of A. citrulli KACC10651, and the nested-PCR primer set (Ac-ORF 21F/Ac-ORF 21R) were selected by checking of specificity to A. citrulli with PCR assays. The selected nested-PCR primer amplified the 140 bp DNA only from A. citrulli strains, and detection sensitivity of the nested PCR increased 10,000 times of $1^{st}$ PCR detection limit (10 ng genomic DNA/PCR). The nested PCR detected A. citrulli from the all samples of seed surface wash (external seed detection) of the artificially inoculated watermelon seeds with $10^1cfu/ml$ and above population of A. citrulli while the nested PCR could not detected A. citrulli from the mashed seed suspension (internal seed detection) of the all artificially inoculated watermelon seeds. When the naturally infested watermelon seeds (10% seed infested rate with grow-out test) used, the nested PCR detected A. citrulli from 2 seed samples out of 10 replication samples externally and 5 seed samples out of 10 replication samples internally. We believe that the nested-PCR developed in this study will be useful method to detect A. citrulli from the Cucurbitaceae seeds.

Biological control of crown gall disease on rose by Agrobacterium radiobacter K84 (Agrobacterium radiobacter K84에 의한 장미 뿌리혹병의 생물적 방제)

  • Severe crown gall disease was occurred in green house cultivating rose in Jinchen, Chungbuk recently. Although it causes problem on rose cultivation, the growers do not have many choices of control measures for the disease now. Agrobacterium radiobacter K84 has been known as a strong antagonist against A. tumefaciens, a pathogen causing crown gall disease, and used as a biopesticide for crown gall in many countries since it had been introduced in 1972. We tested control activity of A. radiobacter K84 for the crown gall disease on rose. Spray of A. radiobacter K84 suspension on above ground of rose either before or after pathogen spray reduced size and fresh weight of galls significantly. Size and fresh weight of galls on roses inoculated with pathogen either before A. radiobacter K84 spray (pathogen-K84 treatment) or after A. radiobacter K84 spray(K84-pathogen treatment) were 4 to 5% of those of galls on roses inoculated pathogen only. Disease incidence of plants inoculated pathogen only was 85% whereas disease incidence of pathogen-K84 or K84-pathogen treatments were 6.7% and 5.0% respectively. Dipping of roots of rose in suspension of A. radiobacter K84 was also reduced size of galls and diseased rate significantly. These results indicate that A. radiobacter K84 is effective in the prevention of gall formation by A. tumefaciens and it can be used to control of crown gall disease of rose.
