Optimal HPLC Condition for Simultaneous Determination of Anthocyanins in Black Soybean Seed Coats (검정콩 함유 안토시아닌의 동시분석을 위한 최적 HPLC 분석 조건)

  • Black soybean has been widely utilized as foods and oriental medicinal materials. The pigmentation in the seed coat of black soybean is due to accumulate anthocyanins in the epidermis palisade layer. The anthocyanins of black soybean seed coat are considered as a parameter of quality evaluation of black soybean. Therefore, the purpose of this study was to investigate the most suitable HPLC condition for simultaneous determination of anthocyanins in black soybean seed coats extracts. The efficient HPLC analytical condition of D3G, C3G, and Pt3G contained extracts of black soybean seed coats was developed. The gradient elution employed a $250\;mm\;{\times}\;4.6\;mm$ i.d. YMC-pak ODS-AM 303 column. The gradient system was used two mobile phases. A gradient elution was performed with mobile phase A, consisting of 5% aqueous formic acid, and mobile phase B, comprising 5% formic acid - acetonitrile, and delivered at a flow rate of 0.7 mL/min as follows: $0{\sim}35\;min$, $90%\;A{\sim}60%\;A$; 36 min, 90% A; 46 min, 90% A. The UV-VIS. detection wavelength was set at 520 nm. The limit of detection (LOD) for D3G, C3G, and Pt3G were under 10 ng/mL.

The Genetic Variations of Pleurotus spp. on Subculture (느타리버섯 속(Pleurotus spp.)의 계대배양에 따른 유전적 변이)

  • The genetic variations and the rate of mycelial growth in the dikaryon and the monokaryon stages of Pleurotus spp. were surveyed during their subcultures. The highest growth rate was observed on both the 3rd and the 4th subcultures. The remarkably rapid growth rate was detected in P. ostreatus dikaryon. Genetic similarities in the dikaryon and the monokaryon of P. ostreatus were more than 57.5% and 85.7%, respectively, and those of P. eryngii were more than 85.2% and 84.8%, respectively. The genetic similarities of monokaryotic P. ostreatus were higher than those of dikaryotic. The topology of phylogenetic trees showed that the divergence and the clustering patterns of branch did not correlated with the number of subcultures. This suggests that genetic variations occur very randomly on mycelial cultures. These results suggest that the monokaryotic mycelia is genetically more stable than the dikaryotic in subcultures, and that it is very useful to stock monokaryotic mycelia for making spawns and breeding of Pleurotus spp.

Analysis of the Electromagnetic Scattering by a Tapered Resistive Strip Grating with Zero Resistivity at the Strip-Edges On a Grounded Dielectric Plane (접지된 유전체층 위에 저항띠 양끝에서 0으로 변하는 저항율을 갖는 저항띠 격자구조에서의 전자파 산란 해석)

  • In this paper, Electromagnetic scattering problems by a resistive strip grating with tapered resistivity on a grounded dielectric plane according as strip width and spacing, relative permittivity and thickness of dielectric layers, and incident angles of a electric wave are analyzed by applying the FGMM(Fourier-Galerkin Moment Method) Known as a numerical procedure. The scattered electromagnetic fields are expanded in a series of floguet mode functions. The boundary conditions are applied to obtain the unknown field coefficients and the resistive boundary condition is used for the relationship between the tangential electric field and the electric current density on the strip. The tapered resistivity of resistive strips varies zero resistivity at strip edges. Then the induced surface current density on the resistive strip is expanded in a series of Chebyshev polynomials of the second kind. The numerical results of the geometrically in this paper are compared with those for the existing uniform resistivity and perfectly conducting strip. The numerical results of the normalized reflected power for conductive strips case with zero resistivity in this paper show in good agreement with those of existing paper.

Effect of Storage Conditions and Varietal Mixture on Quality of Malting Barley (저장조건 및 품종혼합가공에 따른 맥주맥의 품질변이)

  • Improvement in production, post-harvest processing, quality grading, storage and malting process of malting barley is apparantly necessary to upgrade the beer quality and to reduce the losses in malt production. To establish the bases for the improvement, the present study examined the effect of storage conditions on the germinability of malting barley by quality grade of barley at production, and feasibility of mixing different varieties and different quality grades. Germinability of malting barley decreased significantly after 13 months of storage in the off-grade grain storage house, and after 15 months in the 1st grade storage house under the climate of Korea. Malting barley that was off-grade at harvest deteriorated faster than the 1st grade during 5 years storage under ordinary conditions. Mixing of varieites, different grain-quality grades or differently stored barleys seems to increase variability in germinating forces and germinability, resulting in uncontrollable malting processes and poor quality of malts and beer.

  • A method is developed for solving the natural frequencies and mode shapes of linearly variable tapered beams. The governing differential equation for the tapered beam is derived. Three kinds of cross sectional shape are considered in differential equation. The Runge-Kutta method and the determinant search method are used to perform the integration of the differential equation and to determine the natural frequencies, respectively. The hinged-hinged, hinged-clamped, damped-clamped and free-damped end constraints are investigated in numerical examples. The lowest four nondimensional natural frequencies are obtained as functions of $d_b/d_a$. ratio. The effects of end constraints and cross sectional shapes on frequencies are analyzed and typical mode shapes are also presented.

  • This investigation was carried out to study the effect of heating treatment on the saponin fractions in ginseng extract and crude saponin. The changes of saponin patterns and amounts were investigated using HPLC and compared with peak area of each fraction. Shape of crude saponin was changed more easy to compare with in ginseng extract. The more extracting temperature risen and treated time longer, the more changes of saponin amounts and patterns were shown, expecially at $100^{\circ}C$ over. All of the saponin fraction except ginsenoside-Rd were relatively unstable in heating treatment. Suitable extracting condition was extracted at $80^{\circ}C$ for 40 hours with $H_2O$.

  • Alkali digestion value of rice kernels was increased with delayed planting date and decreased with temperature during ripening. Varietal difference in average digestion value between low and high groups of rice varieties was the greatest under the conditions of 1.4 percent solution of potassium hydroxide, early planting and day/night temperature of 30/22$^{\circ}C$ Segregation ratio of alkali digestion value in $F_2$ generation was varied with crosses showing 3:1 for nine crosses, 1:3 for one cross, 9:7 and 13:3 for two crosses respectively and non-segregation for one cross of 15 crosses between low and high varieties in the digestion value.
