Development of a Species Identification Method for the Egg and Fry of the Three Korean Bitterling Fishes (Pisces: Acheilognathinae) using RFLP (Restriction Fragment Length Polymorphism) Markers (제한절편 길이 다형성(RFLP) 분자마커를 이용한 납자루아과 담수어류 3종의 난과 치어 종 동정 기법 개발)

  • This study aimed to develop a species identification method for the egg and fry of the three Korean bitterling fishes (Pisces: Acheilognathinae), including Acheilognathus signifer, Acheilognathus yamatsutae and Rhodeus uyekii based on the PCR-based Restriction Fragment Length Polymorphism (RFLP) markers. We conducted a field survey on the Deokchicheon River from the North Han River basin, where the three Acheilognathinae species co-occur, and also analyzed the existing sequence dataset available from the GenBank. We found coexistence of the three species at the study site. The egg and fry were obtained from the host mussels (Unio douglasiae sinuolatus) by hand from May to June 2015 and in May 2017. To develop PCR-based RFLP markers for species identification of the three Acheilognathinae fish species, restriction enzymes pinpointing species-specific single nucleotide variation (SNV) sites in mitochondrial DNA COI (cytochrome oxidase I) and cyt b (cytochrome b) genes were determined. Genomic DNA was extracted from the egg and fry and RFLP experiments were carried out using restriction enzymes Apal I, Stu I and EcoR V for A. signifer, A. yamatsutae and R. uyekii, respectively. Consequently, unambiguous discrimination of the three species was possible, as could be seen in DNA band patterns from gel electrophoresis. Our developed PCR-based RFLP markers will be useful for the determination of the three species for the young and would assist in studying the spawning patterns and reproductive ecology of Acheilognathinae fishes. Furthermore, we believe the obtained information will be of importance for future maintenance, management and conservation of these natural and endangered species.

  • 3종 복합소재인 양모와 나일론, 레이온의 염색공정은 먼저 알칼리 욕에서 반응성염료로 레이온을 염색한 후, 산성욕에서 산성염료로 양모와 나일론을 염색하는 것이 일반적이다. 그런데 양모, 나일론, 레이온은 알칼리에 민감하므로 면 염색 시 적용되는 강알칼리인 수산화나트륨을 사용하면 섬유의 취화로 인하여 강도와 촉감저하 등의 문제가 발생하게 된다. 따라서 본 연구에서는 알칼리 중 탄산나트륨과 중탄산나트륨을 사용하여 염료를 투입하지 않고 염색공정을 거친 후 백도와 인장강도를 측정하여 황변 및 강도 변화 여부를 알아보았다. 레이온 70%, 양모 19%, 나일론 11%의 혼용율을 가지는 전처리 된 편물(130$g/m^2$, 32 inch, 18 gauge)에 탄산나트륨(시약 1급) 20g/l과 중탄산나트륨(시약 1급) 20g/l 각각을 투입하고 반응성염색공정($60^{\circ}C$, 60 min)으로 처리한 경우와 반응성 염색 후 산성염색공정($98^{\circ}C$, 60 min)으로 처리한 경우로 나누어 측색을 통해 백도와 L, a, b값을 측정하고 KS K 0521에 따라 인장강도 시험을 실시하였다. 그 결과, 탄산나트륨 투입시 백도 값이 중탄산나트륨에 비해 10% 정도 낮아지고 L, a, b값도 상대적으로 yellow 방향으로 이동해 있는 것으로 나타났다. 이것은 황변이 일어남을 의미하며 육안으로 확인하였다. 또한 인장강도 측정결과를 통해 소다회 처리 시 중조에 비해 30% 정도의 강도 저하가 일어남을 확인할 수 있었다. 그러나 반응성염색 후 산성염색을 거치게 되면 황변과 강도 저하 현상이 회복되는 경향을 나타내었다. 즉, 천연/나일론 편물을 반응성염색 시 알칼리로 탄산나트륨을 사용하면 염색공정 상에서 중탄산나트륨에 비해 황변이 일어나 염료 고유의 색상 발현에 영향을 줄 수 있고 강도 또한 30% 정도 저하되지만, 후에 산성염색 공정을 거치면 산 조건과 욕중 효과를 통해 일부 개선됨을 확인하였다. 이와 별도로 이번에는 시판되는 반응성염료 5종과 산성염료 3종을 조합하여 탄산나트륨과 중탄산나트륨 투입에 따른 염색실험을 실시하고 측색(DataColor SF600 광원D65, Strength)을 통해 염착량을 비교하였다. 그 결과, 중탄산나트륨으로 염색하면 탄산나트륨에 비해 모두 염착량이 저하되었으며, 염료의 구조적 차이와 컬러별로도 그 차이는 다양하였다. 그 중 저온에너지형 반응성염료는 탄산나트륨 투입에 비해 47~60% 정도로 가장 양호하였으며, 일부 반응성염료는 20%까지 떨어지는 값을 나타내었다. 이것은 탄산나트륨보다 중탄산나트륨의 알칼리 정도가 낮으므로 반응성염료의 염착이 적은 것으로 생각되며, 저에너지형 반응성염료의 경우에는 낮은 온도나 알칼리 조건에서도 상대적으로 높은 고착률을 나타내므로 적절한 반응성염료의 선택을 통해 그 차이를 극복할 수 있을 것으로 사료된다. 이상의 결과를 통해 탄산나트륨과 중탄산트륨의 알칼리 정도가 강도와 염착량에 미치는 영향의 차이를 고려하여 최적의 현장 처방을 선정해야 함을 알 수 있었다.

Mitochondrial DNA Analysis of the Fleshy Prawn (Penaeus chinensis) for Stock Discrimination in the Yellow Sea (황해산 대하(Penaeus chinensis)의 계군분석을 위한 미토콘드리아 DNA 분석)

  • The mitochondrial DNA (mtDNA) restriction fragment length polymorphi는 (RFLPs) of five populations were analyzed to delineate the stocks of Penaeus chinensis (Osbeck) in the Yellow Sea. Comparison of P. chinensis with P. japonicus to clarify the nucleotide divergence between two species was also carried out. Based on the fragment patterns, three composite haplotypes were analyzed in P. chinensis mtDNA as four naplotypes were in P. japonicus. Most individuals of each P. chinensis population are shared by one dominant haplotype. Another two haplotypes haying variations at the C/a I and hull sites were also distributed evenly in the Korean and Chinese populations. It is suggested that the gene exchange occurring between populations in the Yellow Sea is frequent. Average length of the mtDNA molecule was estimated to be about 16.44 kb in P. chinensis and 16.31 kb in P. japonicus, Sequence divergence (p) of mtDNA between two species estimated by using Upholt's (1977) fomula was $13.7\%$.

Antimutagenicity of Korean Sweet Potato (Ipomoea batatas L.) Cultivars (한국산 고구마의 품종별 항돌연변이 효과)

  • Polyphenolic content and antimutagenicity of the methanol extracts prepared from 22 cultivars of sweet potato with different flesh colors were investigated using Folin-Ciocalteu's phenol reagent method and Ames test, respectively. There was a remarkable cultivar difference in the polyphenolic content of sweet potato. Su, Hayanmi and Shinhwangmi among 17 cultivars of non-purple sweet potato had higher polyphenolic contents of 21.4, 21.5 and $20.3{\mu}g$ (GAE/g dried sweet potato), respectively, whereas Manami and Yeonhwangmi were very much lower at 4.6 and $4.8{\mu}g$. Mokpo No.62, Borami, Sinjami, Jami and Ayamurasaki had much higher polyphenolic contents of 67.7, 76.9, 44.9, 128.3 and $93.2{\mu}g$, respectively, than non-purple sweet potato. The methanol extract from the sweet potato effectively inhibited the reverse mutation induced by 1-NP, daunomycin, Trp-P-1, Trp-P-2 and 2-AA on S. Typhimurium TA 98, and by 1-NP on S. Typhimurium TA 100. These results suggest that the antimutagencity properties may be influenced by the tested mutagen and strain rather than the polyphenolic content of non-purple and purple sweet potato. However, in the purple sweet potatoes, a high polyphenolic content may influence the antimutagencity properties.

Responses on the Agronomic Characteristics for Different Sowing Times with Perilla(Perilla ocimoides L.) (파종기 이동이 들깨 생태변이에 미치는 영향)

  • Four varieties of perilla(Perilla ocimoids L.) were tested to investigate the ecological adaptation of the crop to variations in sowing time extending from April 25 to July 25 with an interval of 30 days between each of the four sowing. As sowing time was delayed, the plant height, the number of first branches and cluster, the weight of fresh and dried stems, the day to germination and flowering decreased, showing a highly negative correlation between the showing dates and these characteristics, but a highly positive correlation between days to flowering and the required accumulated temperature. With early sowing under low temperature and long day conditions, it took about 132 days from sowing to flowering due to the long period of vegetative growth, but as sowing was delayed, the days to flowering decreased with a minimum period of 57 days. The yield of seeds in each case varied with each variety. Type A(Nonsan var.) and type B(Jinchon var.) had higher yields when sowed earlier. But type C(Namyang var. and Wooljin var.) had its highest yield in the plot sowed on May 25 and the 1000 grain weight showed a tendency to increase as sowing was delayed. In conclusion, the perilla was preyed to be a short-day plant that flowers from the begining to the middle of Sep. regardless of sowing time, so that the sowing time should be decided after due consideration of the length of the vegetative growth periods of the varieties.

Genetic variation and population structure of Asarum misandrum (Aristolochiaceae) in Korea (각시족도리풀(Asarum misandrum)의 유전적 다양성 및 집단 구조)

  • Genetic variation in Asaum misandrum, a woodland herb in Korea, was investigated based on allozyme analysis with starch gel electrophoresis. All of previously reported populations in Korea were sampled and seven loci from six enzymes were analyzed. Overall genetic variation of A. misandrum population showed considerably high levels of genetic variation within the species (A = 2.05, P = 71.4, $H_E$ = 0.294). A positive $F_{IS}$ value of A. misandrum indicated overall deficiency of heterozygotes, and a low $F_{ST}$ value (0.112) meant very little differentiation among populations. Factors contributing to the high levels of genetic diversity found within populations of A. misandrum include population maintenance via wide distribution range from Korea to Japan and primarily outcrossing breeding system. Although it showed moderate genetic diversity level, most habitats of the species were scattered and discontinuous. Besides, low numbers of individuals were found in the most habitats and individuals are collected frequently from the wild due to the unique shape of the flowers as well as the rarity of the species. Thus, there is a need to set up a reasonable conservation strategies including the maintenance mechanism of genetic diversity of A. misandrum.

Foliar Characteristics and Photosynthetic Efficiency of Three Species of Schisandraceae Trees Distributed in Korea (우리나라에 분포하는 오미자과 2속 3종의 잎 형질 및 광합성 특성)

  • Foliar and physiological characteristics of Schisandraceae plants distributed in Korea were investigated to elucidate differences in growth and the variations of inter and intra species of the plants. For this study, three species of Schisandraceae plants were used; i) Schisandra chinensis, decidious broad-leaved tree, ii) S. nigra, decidious broad-leaved tree, including monoecious, male and female types, ii) Kadsura japonica, evergreen broad-leaved tree. There were no significant differences in leaf length and width among three species, but leaves of Schisandra sp. plants were shorter and wider relative to those of K. japonica. The length of petiole of Schisandra sp. plants was greater than that of K. japonica and the difference was evident between female type of S. chinensis and K. japonica as longer as two times. Schisandra sp. trees showed larger size of stomata, but fewer numbers of stomata, compared with K. japonica. Although substantial changes in leaf morphology were not found in the Schisandraceae plants grown in different environmental conditions, the foliar length of K. japonica was seemed to be increased with increasing an altitude. Chlorophyll contents were also variable in inter and intra species and in, general K. japonica showed higher levels of chlorophyll contents relative to Schisandra sp. plants. K. japonica also showed higher light compensation point(LCP), light saturation point(LSP) and $CO_2$ assimilation rates at LSP, compared with Schisandra sp. trees. However, Schisandra sp. trees showed lower respiration per photosynthesis rates at LSP. The finding suggested that Schisandra sp. trees were acclimated to the growth conditions of lower light intensity with more effective photosynthetic activity. This was also confirmed by the fact that Schisandra sp. plants grown in shading places of a forest demonstrated the elevated rates of net $CO_2$ assimilation under the conditions of low light intensity.

Analysis of ITS DNA Sequences of the Viola albida Complex (태백제비꽃군 ITS DNA 염기서열 분석)

  • ITS DNA sequences from five individuals, representative of five groups designated according to the degree of leaf teeth and lobes from simple to palmate compound leaf in the Viola albida complex, established and further analysed in order to solve the taxonomic difficulty. A total 702 bp was sequenced at the 5.8S ribosomal DNA and internal transcribed spacer 1 and 2. The 5.8S coding region is 163 bp, and has no sequence variations. The ITS1 and ITS2 noncoding regions have a little bit sequence variations, and those were further analysed by the methods of the analysis of variance (ANOVA), the analysis of sequence divergence and the phylogenetic analysis. The result of ANOVA showed no significant differences among individuals investigated. The analysis of sequence divergence with Kimura 2-parameter distance revealed that in-groups showed much less than 0.05 in absolute value among individuals, while two out groups more than 0.05, V. grypoceras and V. orientalis. This result appeared that the sequence divergence among in-groups was not yet occurred in the species level but situated at somewhere below the species level. In the phylogenetic analysis, two outgroups formed the basal clades in order. Five individuals in-groups formed a clade. The clade was, however, not very robust as around 50% in bootstrap value, suggesting that this result was not meaningful in the phylogenetic point of views.

  • The 165 collections originated from domestic and foreign countries were tested to evaluate the fatty acid composition of sesame for using as a basic research for oil quality improvement. The Korean and American varieties were lower content in saturated fatty acid and higher especially in unsaturated fatty acids (Oleic and Linoleic acid) than those of other regions. Varieties from temperate areas were higher in unsaturated fatty acid than that of varieties from tropical areas. On the other hand, the varieties which were late matured and had yellow seed coat color and set three capsules showed higher in unsaturated fatty acids than those of early matured, white seed coat and one capsules. Since there was significantly negative correlation between unsaturated fatty acid and saturated fatty acid contents, it was possible to improve the fatty acid composition of sesame.

Development and Application of a Cleaved Amplified Polymorphic Sequence Marker for Discriminating A Mating Type Alleles of Lentinula edodes (표고 A 교배형 구분을 위한 CAPS 마커의 개발 및 적용)

  • Lentinula edodes is one of the most widely consumed edible mushrooms in Korea. Mating in L. edodes is regulated by a tetrapolar system, and two unlinked genetic loci, A and B, are known to be major determinants of the mating types, as reported in other heterothallic basidiomycetes. The A locus of L. edodes encodes a pair of homeodomain (HD) transcription factors. The highly variable N-termini of these HD transcription factors contribute to the diversity among the A mating types. In this study, we developed a cleaved amplified polymorphic sequence (CAPS) marker to discriminate 11 different A mating type alleles predominant among both cultivated and wild strains. Amplification of the variable region of the A locus followed by digestion with HaeIII and EcoRI restriction enzymes enabled successful discrimination among the 11 A mating type alleles. We also evaluated the applicability of this method in the identification of two A mating types of a dikaryotic strain.
