Effect of Nutrient Solution for Hydroponics of Liliaceae Leaf Vegetables on the Amount of Ascorbic Acid in Chinese Chive (백합과 전용배양액이 부추의 Ascorbic acid의 함량에 미치는 영향)

  • In this study, nutrient solution for Liliaceae (Chinese chive) leafy vegetable was developed. The various strength (1/2, 1 and 3/2) of nutrient solution recommended by National Horticultural Research Institute (NSH) was applied to the crops in deep flow technique (DFT) system for 42 days. The growth of Chinese chive were highest in the treatment of 1/2 strength. The proper constitution of nutrient solution developed for liliaceae crops (NSL) was N 12, P 2.5, K 7, Ca 4 and Mg 2 met. The crops were grown two times in March and September to examine the appropriateness of the NSC. As a result, the relative growth rate and the amount of chlorophyll (SPAD value)of Chinese chive treated with NSL were increased 1.11 times. The most important nutrient factor of ascorbic acid were also increased 1.16 times.

  • To enhance the productivity of recombinant protein, ${\beta} -glucuronidase(GUS)$, by transgenic Nicotiana suspensions, perfusion culture was carried out in a 5-L stirred tank bioreactor. Maximum cell density of 20.0 gDCW/L was obtained and GUS activity was noticeably affected by medium composition, such as salt concentration. Effect of headspace purging by $CO_2-enriched$ air was also investigated. Aeration of $CO_2(5%)-enriched$ air with 0.2 vvm significantly promoted the cell growth in lag pahse and increased volumetric GUS activity from 0.69 U/mL to 5.76 U/mL. In contrast, aeration of $CO_2(5%)-enriched$ air with 0.1 vvm did not affect the cell growth but enhanced the GUS acitivity from 3.24 to 5.25 U/mL at the 4th day.

  • Salt-tolerant transgenic Panax ginseng plants were produced by introducing the SAL1 geue (3'(2'), 5'-bis-phosphate nucleotidase) that confers tolerance to the salts through Agrobacterium tumefaciens co-cultivation. Cotyledon explants of immature ginseng zygotic embryos cultured on Murashige and Skoog medium lacking growth regulators formed somatic embryos directly with below 10%, but the 74% tranformation rate were observed at the treatment of phytohormone with 1.0 mg/l 2,4-D and 0.5 mg/l kinetin. Somatic embryos were initially cultured on MS medium supplemented with 250 mg/l cefotaxime for 3 weeks and subsequently subcultured five times to a medium containing 100 mg/l kanamycin and 250 mg/l cefotaxime. Upon development into the cotyledonary stage, these somatic embryos were transferred to on the medium containing 50 mg/l kanamycin and 10 mg/l gibberellic acid to induce germination and strong selection. Integration of the transgene into the plants was confirmed by polymerase chain reaction with specific primers. The ginseng transformants with well-developed shoots and roots were successfully acclimatized in a greenhouse when they were planted in soil.

  • Screening, Identification and some cultural characteristics of ALA$({\delta}-aminolevulinic\;acid)$-producing photosynthetic bacteria were carried out for the optimal production of ALA, one of the bioherbicides. Among photosynthetic bacteria isolated from soil, marsh, pond, etc., KK-10 was the best producer of ALA and identified to be Rhodobacter capsulatus belonging to a typical group of nonsulfur purple bacteria. By addition of 15 mM LA (levulinic acid), an inhibitor of ALA dehydrase in cyclic tetrapyrrole biosynthesis, into culture broth at middle log phase of cell growths, ALA production was considerably increased to about 20-fold (28 mg/l). The combined supplementation of glycine and succinate, each with a concentration of 30 mM also enhanced production of ALA and activity of ALA synthase to about 50-fold (73 mg/l) and 2-fold, respectively. The isolated strain was able to produce upto 80 mg/l under the cultural condition optimized by addition 15 mM LA into the synthetic medium at four different points starting middle log phase.

  • This study aimed to analyze crude protein, amino acid and bioactive changes in Phellinus linteus HN00K9 cultured oat. The crude protein content of P. linteus cultured oat (PCO) was 12.9%, which was higher than that of uncultured oats (UCO) as control at 11.26%. The total free amino acid contents of PCO and UCO were 11,4 mg/100 g and 9,686.205 mg/100 g, respectively. Glutamic acid accounted for 21% of the total amino acids of PCO, and the histidine content was increased by more than 51% in PCO compared to UCO. The total polyphenol content of PCO was 275 mg GAE/g, which was more than twice that of UCO (135 mg). The DPPH radical scavenging activity was 15.5% in PCO, which was more than five times that of UCO (3.5%). The β-glucan content of PCO was 12.5 g/100 g, which was more than five times that of UCO (3.2 g/100 g). Therefore, it is believed that PCO can be utilized as a material for various functional foods.

  • This experiments were carried out to determine the optimum concentrations of plant growth regulators for the direct embryogenesis from the cotyledon culture of Paeonia lactiflora Pall. Zygotic embryos rescued from true seeds were developed abnormally in the medium containing ABA. But somatic embryogenesis from cotyledons of abnormal seedlings was induced more effectively. Also the somatic embryogenesis from cotyledons was promoted in the medium containing ABA. The frequency of embryogenesis was maximum(59.9%) from the cotyledons cultured on MS medium containing 0.5 mg/L ABA on which the frequency of somatic embryos with two cotyledons was 22.6%.

Bacillus subtilis EKll was most effective for PPase Production For increasing of PPase productivity effects of glucose concentrations, pHs and aeration rates were studied in batch culture The most proper concentra- tion of glucose pH and air condition for PPase Production were 1% 8.0 and lvvm respectively In these condi- tion PPase productivity was $84,364 UL^{-1}$ $h^{-1}$ and increased about 15.6 times than flask fermentation.

  • To investigate a simple and rapid screening method for photosynthetic inhibitory herbicides, responses of tobacco(Nicotiana tabacum L.) and liverwort(Marchantia polymorpha L.) PA(photoautotrophic) cells to various commercial herbicides with different modes of action and leaf extracts of four weed species were compared. PET (photosynthetic electron transport) inhibitory type of herbicides has greater inhibitory effect in liverwort photoautotrophic cells than the photomixotrophic and heterotrophic cultured cells. Similary, PET inhibitory type of herbicides inhibited the oxygen evolution more in liverwort PA cells than the other type of herbicides. Based on oxygen evolution, 60% inhibition was observed by the 10% aqueous extracts of Polygonum hydropiper, while there was 100% inhibition by the 10% methanol extracts of Polygonum hydropiper. This assay gave well correlated results to the Hill reaction data using isolated thylakoids. Thus liverwort photoautotrophic cells might be suitable materials for rapid screening method for photosynthetic inhibitory herbicides.

${\gamma}$-PGA Production by Cultures of Alkalophilic Alcaligenes sp. (호알카리성 Alcaligenes sp.의 배양에 의한 ${\gamma}$-PGA의 생산)

  • Methods for production of ${\gamma}$-Polyglutamic acid( ${\gamma}$-PGA ) by an alkalophilic Alcaligenes sP. were investigated for batch and continuous culture processes. Both quantity and productivity of ${\gamma}$-PGA by Alcaligenes sp. in batch culture were gradually increased with the increase of glucose concentration up to 50g/l , but the maximal production yield of 63% was obtained at 10g/l of glucose concentration. The highest specific growth rate was about $0.25hr^{-1}$ at 50un of glucose concentration, and substrate inhibition was observed at above 50g1f of glucose concentration. The highest ${\gamma}$-PGA formation about 11g/l in a batch system was obtained at 31'C, pH 10.0 and 87rpn Productivity of 2.80g/l/hr for continuous cultivation was 9 times higher than the productivity for batch cultivation.

  • A defensin is an inducible antibacterial peptide from a fleshfly and contains 40 residues basic peptide with six cysteines. For the consiruction of recombinant S cerevisiae expressing defensin, the structural gene coding for active defensin was chemically synthesized and fused in fiam to GAP promoter, MFul preprosequence and the GAL7 transcription terminator, generating a recombinant plasnlid pGMD18. S. ce~evisine 2805 Gells were transror~ned to uracil prototroph by the pGMDl8 arid the transformed cells showing antibacterial activity against 111. luteus TAM1056 were selected by growth inhibition zone assay. The optimal culture conditions for the unprovement of the defensin production of a selected tmdonnant were investigated. The optirmzed medium containing 0.4% yeast extract, 2% corn steep liquor, 2.5% glucose and 0.05% $C_2CO_3$, could be determined and the optimum lemperature. and initial pH could be detennnied as $28^{\circ}C$ and pH 3, ~mpectively. The optimized conditioiis revealed the trvofold Increase in the cell growth and the fourfold in the antibaclerial activity. coinpar-ed with tllc Yl'D medium.
