Radioprotective Effects of Cordyceps sinensis Extracts on ${\gamma}-Irradiated$ Mice (동충하초 추출물이 감마선에 조사된 생쥐에 미치는 방사선방호효과)

  • Effect of single intraperitoneal administration of Cordyceps sinensis (Cs) extract at 24 hour before whole-body ${\gamma}$ - irradiation on the survival ratio, body weight, organ weight changes and serum metabolites in the irradiated mice were investigated. The single pre-administration of Cs extract increased the 40-day survival ratio of irradiated mice from 66.7% to 83.4%. The administration of Cs extract completely prevented weight reductions of spleen and thymus produced by ${\gamma}$ - irradiation (P<0.05, P<0.01). Similar but somewhat less radioprotective effect was also found in the testis of the Cs treated mice. The administration of Cs inhibited the serum hyperglycemia produced by irradiation on the day 7th(P<0.01). However, it did not influence the serum cholesterol and protein levels on the days examined. The present study is the first report regarding Cs which was tested and found to be radioprotective.

Radioprotective Effect of Propolis on the Blood Corpuscle of a Mouse by SEM after X-irradiation on the Whole Body (전신 X-선 조사 후 마우스의 대동맥 혈구에서 SEM을 이용한 프로폴리스의 방사선방어효과 연구)

  • After x-ray 5Gy radiation on the whole body of a mouse using a linear accelerator, its leucocyte, erythrocyte, and platelet were observed by SEM. Also, after injecting propolis into the abdominal cavity, the radio-protective effect of blood corpuscles was studied. The observation of micromorphology in blood corpuscles revealed that the number of leukocyte, erythrocyte, and thrombocyte decreased in the experimental group and the lump got together in blood corpuscles after 10 and 20 days. In RBC, crack or break on the surface and poikilosperocytes were observed. In the irradiation group, the size of leucocytes was smaller than that in control group and the number of villus at the verge substantially decreased. The blood corpuscles in the propolis group, however, had the similar results to control group.

Synergistic Effect of Green Tea EGCG Treatment with Gamma Radiation in Ieukemia Cell Necrosis (방사선조사와 병행 처리한 녹차 EGCG의 혈구암세포 사멸촉진 효과)

  • During cancer therapy, gamma-ray irradiation and treatment of anti-cancer chemicals destroy the normal cells as well as cancer cells. In this study, we investigated the effect of epigallocathechin-gallate(EGCG) extracted from green tea, which is known to have anti-cancer and anti-oxident activities, in order to find out the feasible method to protect the normal cells and to kill the cancer cells efficiently. We investigated the effect of EGCG on the leukemia cell growth and cell necrosis, especially when treated along with gamma radiation. The EGCG inhibited the leukemia cell, HL-60, growth at the appropriate concentration while it exhibited no influence on the normal cell growth. More significantly, it enhanced leukemia cell necrosis when its treatment was combined with gamma irradiation. Simultaneous treatment of EGCG and gamma radiation increased leukemia cell necrosis up to 35% compared with separate treatments. These results suggest that drinking of green tea or co-treatment of EGCG during gamma irradiation therapy may result in better prognosis through enhancement of the tumor cell necrosis and protection of the normal cells.

Modification of Gamma-radiation Response in Mice by Green Tea and Diethyldithiocarbamate (마우스에서 방사선 영향에 대한 녹차와 Diethyldithiocarbamate의 조절효과)

  • We performed this study to determine the effect of green tea on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of gammairradiation. The radioprotective effect of green tea was compared with the effect of diethyldithiocarbamate (DDC). Jejunal crypts were protected by pretreatment of green tea (p<0.01). Green tea administration before irradiation resulted in an increase of the formation of endogenous spleen colony (p<0.05). The frequency of radiation-induced apoptosis in intestinal crypt cells was also reduced by pretreatment of green tea (p<0.05). The radioprotective effect on jejunal crypts and apoptosis in the DDC treated group appeared similar to those in the green tea treated groups. Treatment with DDC showed no significant modifying effects on the formation of endogenous spleen colony. These results indicated that green tea might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the promotion nature of green tea and its components.

The Protective Effect of Selenium on Radiation in Rat (흰쥐에 있어 방사선에 대한 셀레늄의 보호효과)

  • In this study, the radiation protection effects of selenium, known as a powerful antioxidant, were studied. After 14 days of oral injection of selenium into the rat, a full body irradiation of 10 Gy was carried out. And after 1 day, 3.5 day, 7 day and 21 day, we observed blood cell components and superoxide dismutase(SOD) and small intestine changes. Compared to irradiation groups, there was a significant protection effect of reducing the hematopoietic immune system damage in the irradiated group after selenium administration(p<0.05). Then, selenium is a valid ingredient that increases the activity of the superoxide Dismutase(SOD), and it is confirmed that it has an effect of inhibiting apoptosis expression of small intestinal cells by irradiation. Based on these results, selenium is considered to be an essential ingredient for protecting living things from radiation.

Sequence and Time Interval in Combination of Irradiation and Cis-Diamminedichloroplatinum in C3H Mouse Fibrosarcoma (C3H 마우스 섬유육종에 있어서 방사선 조사와 Cis-diamminedichloroplatinum의 병용시 순서 및 시간간격의 영향)

  • Experiments have been carried out with C3H mouse fibrosarcoma (FSa II) to determine the effect of different sequence and time intervals between irradiation and administration of cis-diammihedichloroplatinum (cis-DDP) with gross tumors (6 mm in diameter), microscopic tumors (3 days after transplantation of $10^3$ cells) and cells in culture. The drug was administered either 24, 12, 8, 4, 2, 1, 0.5 hour before irradiation, immediately before irradiation, or 0.5, 1, 2, 4, 8, 12, 24 hours after irradiation. In case of in vivo studies, tumor growth delay was used as an end point. Clonogenic cell surviving fraction was used for in vitro studies. Tumor growth delay for gross tumor after 10 Gy radiation plus 10 mg/kg cis-DDP ranged from 6.3 to 10.66 days and the enhancement ratio ranged from 1.37 to 2.23. The most effective combination was when cis-DDP was given 4 hours before irradiation. Tumor growth delay for microscopic tumor after 5 Gy of radiation and 5 mg/kg of cis-DDP ranged from 3.55 to 11.98 days with enhancement ratio from 2.05 to 6.92. Microscopic tumors showed response significantly greater than additive in every time interval and the most effective treatments were when cis-DDP was given 2 and 1 hour before irradiation. In in vitro experiment, the surviving fraction after 6 Gy of radiation and 1 hour exposure to 4 ${\mu}M$ cis-DDP fluctuated as a function of time between treatments, but the difference between maximum and minimum surviving fractions was very small. According to the above results the sequence and time interval between irradiation and chemotherapy is very critical especially for the management of microscopic tumors as in the case of postoperative adjuvant treatment.

방사선에 조사된 생쥐의 간 및 신장에서 홍삼 추출물의 방어효과

  • This study was prepared to observe the charge of tissues weight, glutamate oxaloacetate transminase(GOT) and glutamate pyruvate transminase(GPT) activities in the liver and kidney treated with red ginseng extract injection in the gamma ray(5.0 Gy) irradiated mice groups were divied in to control, red ginseng extract injection, irradiation after saline injection and irradiation after red ginseng extract injection. The GOT activity showed earlier recovery at irradiation after red ginseng extract injection group than irradiation after saling injection group in the liver and kidney. The GPT activity showed earlier recovery at irradiation after red ginseng extract injection group than irradiation after saling injection group in the liver and kidney. The above result suggest that red ginseng extract have the protection effect on the change of GOT and GPT activity after radiation injury in the liver and kidney.

  • 무독성 혼합안정제인 Zn/Ca-stearate의 방사선 안정화 효과를 증진시키기 위해서 안정화 조제로 알코올들을 PVC에 배합하여 방사선 조사하였을 때 PVC의 색차변화를 측정하여 이들 안정화 조제의 방사선 안정화 효과를 비교 검토하였다. 2가알코올에 의한 PVC 안정화 효과는 2가알코올의 주쇄길이에 비례하여 나타나고 있으나 sorbitol을 제외한 다가알코올에 의한 PVC 안정화 효과는 거의 없는 것으로 나타났다. 이들 알코올에 의한 PVC의 방사선 안정화는 2가알코올이 PVC에서 발생된 고분자 라디칼에 대한 라디칼 포착제로서 작용하기 때문인 것으로 판단된다.

Histomorphologic Change of Radiation Pneumonitis in Rat Lungs : Captopril Reduces Rat Lung Injury Induced by Irradiation (X-선 조사로 생긴 흰쥐 폐장 상해의 형태학적 변화: Captopril에 의한 폐장 상해의 경감 효과)

  • Purpose : To assess the histomorphologic changes in the rat lung injury induced by radiation, to determine whether captopril reduces the rat lung injury and to evaluate change in TNF-${\alpha}$ and TGF-${\beta}$ in rat lung damage by radiation and captopril Methods and material : Right lungs in male Sprague-Dawley rats were divided irradiation alone (10, 20, 30 Gy) or radiation (same dose with radiation alone group) with captopril (500 mg/L). Radiation alone group were sacrificed at twelve hours and eleven weeks after radiation and radiation with captopril group (captopril group) were sacrificed at eleven weeks after radiation with captopril. We examined the light microscope and electron microscopic features in the groups. Results : In radiation alone group, there were patch parenchymal collapse and consolidation at twelve hours after radiation. The increase of radiation dose shows more prominent the severity and broader the affected areas. Eleven weeks after radiation, the severity and areas of fibrosis had increased in proportion to radiation dose given in the radiation alone group. There was notable decrease of lung fibrosis in captopril group than in radiation alone group. The number of mast cells rapidly increased with increase of radiation dose in radiation alone group and the degree of increase of mast cell number and severity of collagen accumulation more decreased in captopril group than in radiation alone group. In radiation alone group, expression of TNF-${\alpha}$ and TGF-${\beta}$ increased according to increase of radiation dose at twelve hours after radiation in both group. At eleven weeks after radiation, expression of TGF-${\beta}$ increased according to increase of radiation dose in radiation group but somewhat decreased in captopril group. In the captopril group the collagen deposition increased but less dense than those of radiation alone group. The severity of perivascular thickening, capillary change, the number and degranulation of mast cells more decreased in the captopril group than in the radiation alone group. Conclusion : It is concluded that the effect of captopril in the rat lungs after radiation was considered to be due to its effect on inhibition of mast cells and reduction of collagen deposition, and captopril may be protect in lung damage after radiation. We observed expression of TNF-${\alpha}$ and TGF-${\beta}$ increased at the early phase after radiation and expression of TGF-${\beta}$ increased in proportion to increase of radiation dose at the chronic phase after radiation. This results will contribute to future investigation in reduction mechanism of captopril in lung damage after radiation.

In vivo Radioprotective Effects of Basic Fibroblast Growth Factor in C3H Mice (Basic Fibroblast Growth Factor (bFGF)의 방사선보호작용에 대한 실험적 연구)

  • Purpose : In order to understand in vivo radiation damage modifying of bFGF on jejunal mucosa, bone marrow and the effect of bFGF on the growth of transplanted mouse sarcoma 180 tumor in mice. Materials and Methods : Mice were treated with $6\;{\mu}g$ of bFGF at 24 hours and 4 hours before exposing to 600 cGy, 800 cGy and 1,000 cGy total body irradiation (TBI), and then exposed to 3,000 cGy local radiation therapy on the tumor bearing thigh. Survival and tumor growth curve were plotted in radiation alone group and combined group of bFGF and irradiation (RT). Histologic examination was performed in another experimental group. Experimental groups consisted of normal control, tumor control, RT (radiation therapy) alone, $6\;{\mu}g$ bFGF alone, combined group of $3\;{\mu}g$ bFGF and irradiation (RT), combined group of $6\;{\mu}g$ bFGF and irradiation (RT). Histologic examination was peformed with H-E staining in marrow, jejunal mucosa, lung and sarcoma 180 bearing tumor. Radiation induced apoptosis was determined in each group with the DNA terminal transferase nick-end labeling method ($ApopTag^{\circledR}$ S7100-kit, Intergen Co.) Results : The results were as follows 1) $6\;{\mu}g$ bFGF given before TBI significantly improved the survival of lethally irradiated mice. bFGF would protect against lethal bone marrow syndrome. 2) $6\;{\mu}g$ bFGF treated group showed a significant higher crypt depth and microvilli length than RT alone group (p<0.05). 3) The bone marrow of bFGF treated group showed less hypocellularity than radiation alone group on day 7 and 14 after TBI (p<0.05), and this protective effect was more evident in $6\;{\mu}g$ bFGF treated group than that of $3\;{\mu}g$ bFGF treated group. 4) bFGF protected against early radiation induced apoptosis in intestinal crypt cell but might have had no antiapoptotic effect in bone marrow stem cell and pulmonary endothelial cells. 5) There was no significant differences in tumor growth rate between tumor control and bFGF alone groups (p>0.05). 6) There were no significant differences in histopathologic findings of lung and mouse sarcoma 180 tumor between radiation alone group and bFGF treated group. Conclusions : Our results suggest that bFGF protects small bowel and bone marrow from acute radiation damage without promoting the inoculated tumor growth in C3H mice. Improved recovery of early responding normal tissue and reduced number of radiation induced apoptosis may be possible mechanism of radioprotective effect of bFGF.
