Study of Quality Control of Traditional Wine Using IT Sensing Technology (IT 센싱 기술을 이용한 전통주 발효의 품질관리 연구)

  • The objective of this study was to investigate the quality characteristics of traditional wine using an radio-frequency identification (RFID) system annexed to a fermenter. In this study, we proposed an RFID-based data transmission scheme for monitoring fermentation of traditional alcoholic beverages. The pH, total acidity, total sugar, soluble sugar, free sugar, alcohol content, and organic acids of were investigated and subjected to fermentation of traditional alcoholic beverages three times. The pH ranged from 7.98, 7.95, and 7.68 at day 0, decreased drastically to 3.31~2.96 at day 2, and then slowly increased to the end point, finally reaching 3.34 at day 20. Acidity tended to increase quickly with time, especially for all samples after day 2. The fermentation environment induced a sudden increase acidity in reactants and indicated a low pH. The total sugars during fermentation quickly decreased to the range of 20.3, 22.43, and 19.2% at day 2, and the slope of reduction steadily decreased to 5.1, 6.1, and 4.8% at day 10. On the other hand, the alcohol content showed the reverse trend as total sugars. The alcohol content also showed the same pattern as total acids, showing the highest alcohol content of 17.3% (v/v) on day 20. In this study on traditional wine fermentation using an RFID system, we showed that pH, soluble sugar, and alcohol content can be adopted as key indicators for quality control and standardization of traditional wine manufacturing.

Physicochemical and functional properties of Gochujang with fermented tomato products (토마토 발효액을 이용한 고추장의 이화학적 및 기능적 특성)

  • This study was conducted to investigate the physicochemical and functional properties of Gochujang in the presence of fermented tomato products. To accomplish this aim, tomatoes were fermented using Bacillus subtilis SRCM100333, and the products were used with Gochujang. As a control, non-fermented tomato liquid was used with Gochujang. There were no significant differences in quality characteristics except for amino nitrogen in Gochujang. The functional properties of tomato liquid (TL), fermented tomato product (FTP), Gochujang made with tomato liquid (TLG), and Gochujang made with fermented tomato products (FTG) were investigated. There were significant differences in the pancreatic lipase inhibition (PLI) and superoxide scavenging activity between FTP (87% and 28%, respectively) and TL (77% and 14%, respectively). Nitric oxide scavenging activity of FTG (52%) was higher than that of other groups (TL, FTP, and TLG). Overall, the results indicate that the fermented tomato can be used to develop functional Gochujang.

  • 우리나라의 발효식품 산업의 규모는 2010년 약 4조 원으로 식품 산업 전체의 4.0%정도이며, 이 중 전통 발효식품이 50.0% 수준으로 전체 식품 산업 대비 비중은 2.0%에 불과하지만, 발효제품의 세계 시장 규모는 2000년 152억 달러, 2010년 730억 달러로 급속한 성장으로 확대되고 있다. 국내 소금시장의 규모는 2010년 기준 1,300억 원이며, 전 세계적으로 나트륨 대체 염미제의 시장수요는 절실히 필요한 실정으로, 최근 2,000억 원대 규모를 유지하고 있는 간장 시장에서 염도를 낮춘 '저염 간장'매출 성장세를 보이고 있어 소금을 줄이는 식생활 개선에 대한 교육 홍보와 더불어 '나트륨 저감화 식품 개발의 필요성'이 국민건강증진, 경제성 측면에서 날로 커지고 있다. 또한, 2007년부터 웰빙 열풍으로 자연조미료 제품의 비중이 크게 늘어나는 추세이며, 100% 천연재료로 사용하여 소비자용 제품 출시 등 다양한 변화를 가져온 결과, 자연조미료 시장은 2008년 151억 3,200만 원의 매출규모에서 215억 2,600만원으로 무려 42.3%를 차지했으며, 저염을 위하여 활용하는 저염 소금도 연령대가 높아질수록 선호도가 높아지는 있다는 보고가 있다. 식품의약안전처의 '나트륨 저감화 사업'은 2012년부터 연차별로 가공식품의 나트륨 저감화 가이드라인을 개발하여 가공식품 생산업체들의 참여를 유도하며, 다양한 홍보를 펼쳐 2014년 6월 기준으로 165개 제품의 나트륨이 저감화 되었으며, 2015년에는 95종의 면류제품의 나트륨 함량이 약 20% 이상 저감되는 실적을 보여주었다. 여러 지역자체 단체들은 지역의 특성을 최대한 살려 다양한 발효산업 육성을 진행하고 있음에 영남권의 장류 업체를 대상으로 나트륨 섭취량을 줄일 수 있도록 '경북 발효식품 수출농업기술지원단'은 식품의약품안전처와 연계하여 지속적으로 가공식품 중 나트륨 저감화에 노력을 할 것이며 이에 대기업과 함께 중소기업들의 참여도를 높이기 위해 현장 컨설팅을 지역적으로 실시할 계획으로 각 지역별 업체들의 참여도를 높일 계획이다. 이를 통해 전통 발효식품이 기능성을 유지하면서 나트륨 함량을 줄인 건강식품으로 우리나라 식품산업 발전에 기여하고자 노력할 것이다. 향후 발효산업 중 한류의 매운맛, 김치 등을 시작으로 고성장이 기대되는 세계화 전략 품목이며 지역 농산물을 활용한 발효식품 세계화를 위하여 나트륨 저감화는 반드시 선행되어야 할 것으로 생각된다.

Quality characteristics of rice and rice starch-based Yakju (쌀 및 쌀 전분을 활용한 약주의 품질특성)

  • Yakju, a Korean traditional alcoholic beverage, is made from rice and Nuruk. In this study, the fermentation characteristics of Yakju was investigated using rice and rice starch. Ingredients was classified into raw material (rice, rice starch) and starter (enzyme supplements, modified Nuruk, traditional Nuruk, and yellow rice koji) for fermentation. The crude protein content of rice, rice starch, and starter were determined as follows (%): rice 6.69, rice starch 0.44, enzyme supplements 7.84, modified Nuruk 15.29, traditional Nuruk 14.28, and yellow rice koji 7.28. The alcohol content of rice with traditional Nuruk ($20.13{\pm}0.12%$) was higher than other Yakju. The concentration of organic acids of rice starch-based Yakju (389.83~538.34 mg%) was higher than that of rice Yakju (259.27~357.70 mg%). The concentration of nitrogen compound of rice Yakju (498.38~5976.93 ppm) was higher than that of rice starch-based Yakju (600.43~4463.79 ppm). In line with these findings, further studies will be necessary for the quality analysis of the rice, rice starch and fermented starter (enzyme supplements, modified Nuruk, traditional Nuruk and yellow rice koji).

Optimal Lactic Acid Fermentation Conditions and Quality Properties for Rubus coreanus Miquel (Bokbunja) and Chlorella Mixtures (복분자와 클로렐라 혼합물의 젖산발효 최적조건 및 품질 특성)

  • To develop a functional fermentation food from Rubus coreanus Miquel (Bokbunja) and chlorella mixtures, optimal lactic acid fermentation conditions were established, and quality properties based on physicochemical evaluation such as chemical compositions, free sugars, organic acids, and antibacterial activities were investigated. Regarding optimal fermentation strain selection, formation of lactic acid was best in Lactobacillus plantarum among the experimental strains (10 kinds), and the optimal fermentation temperature was $37^{\circ}C$. In addition, overall acceptability in the sensory evaluation was highest in the 5% chlorella mixture sample. Therefore, quality properties of the prepared sample under the established optimal fermentation conditions were investigated. Moisture, total sugar (dry basis), crude fiber (dry basis), and pH of fermented Rubus coreanus Miquel juice (RCM) with 5% chlorella mixture (RCM-C5) were reduced by 4.90%, 14.15%, and 0.32%, respectively, as compared with non-fermented RCM. Meanwhile, crude protein, crude fat, and crude ash (dry basis) of RCM-C5 were elevated by 13.75%, 0.18%, and 0.73%, respectively, as compared with RCM. The yellowness (b value) of color values was greater in RCM-C5 compared to RCM. The free sugar and organic acid contents of RCM-C5 were elevated by 0.97% and 616.30 mg%, respectively, as compared with RCM. In addition, the gram positive bacterium Staphylococcus aureus was elevated by 5.83% while gram negative bacteria Escherichia coli and Salmonella Typhimurium were elevated by 2.94% and 4.67%, respectively, as compared with RCM. In conclusion, the quality properties of RCM and chlorella lactic acid fermentation mixtures were improved compared with the general RCM product. Consequently, it is possible to apply fermented RCM as a functional fermentation food.

Effects of Fermented Diets Including Liquid By-products on Nutrient Digestibility and Nitrogen Balance in Growing Pigs (착즙부산물을 이용한 발효사료가 육성돈의 영양소 소화율 및 질소균형에 미치는 영향)

  • This study was conducted to evaluate the effects of fermented diets including liquid by-products on nutrient digestibility and nitrogen balance in growing pigs. Treatments were 1) CON (basal diet), 2) F (fermented diet with basal diet), 3) KF (fermented diet with basal diet including 30% kale pomace), 4) AF (fermented diet with basal diet including 30% angelica keiskei pomace), 5) CF (fermented diet with basal diet including 30% carrot pomace) and 6) OF (fermented diet with basal diet including 30% grape pomace). A total of 24 pigs (41.74kg average initial body weight, Landrace $\times$ Yorkshire $\times$ Duroc), were assigned to 6 treatments, 4 replicates and 1 pig per metabolic cage in a randomized complete block (RCB) design. Pigs were housed in $0.5\times1.3m$ metabolic cage in a 17d digestibility trial. During the entire experimental period, Digestibility of dry matter (p<0.05) of treatment CON, F and CF were higher than other treatments. In crude protein digestibility, treatment F was higher than treatment AF and GF (p<0.05). Treatment GF showed the lowest digestibility of crude fiber among all treatments (p<0.05). In ether extract digestibility, treatment AF and CF showed higher than other treatments (p<0.05) except KF treatment. CF treatment showed the best digestibility of ash among all treatments (p<0.05). Whereas, For Ca and P digestibility, CF and OF treatments were improved than other treatments (p<0.05). Energy digestibility (p<0.05) of CON, F and CF treatments were higher than KF, AF and GF treatments. In total essential amino acid digestibility, F treatment was improved than AF, CF and GF treatments (p<0.05). In total non-essential amino acid digestibility, F treatment was higher than CON, AF and GF treatments (p<0.05). In total amino acid digestibility, F treatment was higher than AF and CF treatments (p<0.05) and GF treatment showed the lowest digestibility (p<0.05). In fecal nitrogen excretion ratio, GF treatment was greatest among all treatments (p<0.05) and F treatment was decreased than other treatments (p<0.05). In urinary nitrogen excretion ratio, CON and GF treatments showed the lowest among all treatments (p<0.05). In nitrogen retention ratio, CON treatment showed the high and KF treatment showed the lost among all treatments (p<0.05). Therefore, this experiment suggested that fermented diet could improve nutrient and amino acid digestibilities of growing pigs.

  • Chemical composition, dry matter and cellulose digestibilities and fermentation rates of alfalfa and orchard grass cut at different time were estimated in vitro method and the results summurized as follows: 1. Crude protein, crude fiber cwc and cellulose content of Alfalfa were decreassed as advancing cutting time. Crude protein, crude fiber and cwc content of Orchard grass were decreased as advancing cutting time up to 3 cutting, but crude protein was slightly increased at 4th cutting, crude fiber and cellulose content were higher at 2nd cutting, but decreased thereafter. 2. DM digestibility of Alfalfa was 51.80, 51.86, 52.92 and 59.52% at 1, 2, 3 and 4th cutting time, respectibly, thus slightly increased as advancing cutting time, and cellulose digestibility of Alfalfa was not much different with different cutting time. DM digestibility of Orchard grass was 62.21, 66.10, 60.95 and 66.32% at 1, 2, 3 and 4th cutting time, respectibly, and cellulose digestibility of Orchard grass was slightly increased at 3rd cutting time and then increased at 4th cutting time. 3. Fermentation rate of DM of alfalfa was the highest 1st cutting time (0.83%/hr.) and was not different at 3 and 4th cutting time. Cellulose fermentation rate of Alfalfa was the highest at 1st cutting time (1.29%/hr.), decreased at 2 and 3th and then increased at 4th cutting time. Fermentation rate of DM of Orchard grass was 1.42, 1.58, 1.60 and 1.57%/hr. and of cellulose was the highest at 2nd cutting time (1.77%/hr.)

Optimization of Medium for $\beta$-Mannanase Production by Aspergillus oryzae (Aspergillus oryzae에 의한 $\beta$-Mannanase 생산배지의 최적화)

  • Medium optimization for ${\beta}$-mannanase production by Aspergillus oryzae ATCC 2114 was performed. Effect of carbon source (locust bean gum) concentration on ${\beta}$-mannanase production was investigated. Above 20 g/L locust bean gum, a lag time for ${\beta}$-mannanase production was appeared because high concentration of locust bean gum caused high viscosity which made the mixing of medium poor. As the locust bean gum concentration in the medium increased, ${\beta}$-mannanase activity and cell growth increased proportionally. Effect of various nitrogen sources on ${\beta}$-mannanase production was also studied. (NH4)2SO4 and malt extract were the most effective for ${\beta}$-mannanase production among the inorganic nitrogenous compounds and organic nitrogen nutrients. Inorganic compounds such as KH2SO4, NaCl, Na2CO3, and MgSO4, on ${\beta}$-mannanase production were optimized for ${\beta}$-mannanase production. Locust bean gum of 10 g/L, malt extract of 3 g/L, (NH4)2SO4 of 2 g/L, KH2SO4, of 10 g/L were selected as the optimal medium. Culture in a fermentor by using the optimal medium was carried out. Lag time of ${\beta}$-mannanase production was shorter due to the better mixing of the fermentor. The maximum ${\beta}$- mannanase activity of 9.7 unit/mL and specific ${\beta}$-mannanase activity of 1.9 unit/mg-cell could be obtained at 27 hours and the productivity of ${\beta}$-mannanase was 0.36 unit/mL$.$h.

Effects of Supplementing Coated Vitamin C+E with Cottonseed on Meat Composition, Meat Color and Carcass Grade in Hanwoo Steers Fed Brewers Grain and Cracked Corn-Based Fermented Feedstuff (맥주박과 옥수수 위주의 발효사료에 대한 Coated Vitamin C+E 및 면실의 첨가가 거세한우의 육조성분, 육색 및 도체등급에 미치는 영향)

  • Twenty four Hanwoo steers(average body weight=550.4±42.8kg) were used in the experiment to determine the effects of supplementing coated vitamin C+E with cottonseed on meat quality, meat color and carcass grade in Hanwoo steers fed brewers grain and cracked corn-based fermented feedstuff. Steers were randomly assigned to feeding groups of fermented feedstuff without any supplements(T1) and fermented feedstuff supplemented with coated vitamin C+E and cottonseed(T2). Ether extract content of longissimus dorsi was higher in T2 than in T1(p<0.05). Yellowness and hue-angle among surface colors of longissimus dorsi was lower(p<0.05) in T2 than in T1 but lightness was higher(p<0.05) in T2 than in T1. Ratio of yield grade ‘A’ was higher from T2 group than from T1 group. Marbling score was higher in T2 than in T1(p<0.05). Present results indicate that supplementation of coated vitamin C+E and cottonseed to brewers grain and cracked corn-based fermented feedstuff has positive effects on ether extract content, color, marbling score of longissimus dorsi in Hanwoo steers.

진탕배양에 의한 Monascus sp.가 생산하는 적색색소에 관한 연구 (제2보) 적색조 색소의 생산과 물리적 성질 및 생리적 성질에 대한 검토

  • 이미 발표한바 있는 Monascous sp.의 색소 배양액으로부터 식품응용에 적용하기 위한 기초적 자료를 얻기 위하여 조색소 분말의 생산, 물리적 성상과 생리적 성질을 검토하였다. 본 실험은 발효액으로부터 60% alcohol과 pe-troleum ether로 추출하여 alcohol층에서 적색계, petroleum ether층에서 황색계 색소를 얻었으며, 생산된 적색계 조색소는 5종의 색소물질이 복합되어 있었고 pH 3에서 9까지 안정하였으며 O.D 495∼500nm에서 흡수극대를 나타내었다. 적색 조색소분말(색소 1000mcg/mg 기준 때)과 색소액은 시험 시료농도에서 bio-activity는 없었으며 급성 독소시험은 positive 용혈반응 negative였다.
