Optimization of Precipitation Process for the Recovery of Lactic Acid (Lactic acid 회수를 위한 침전공정 최적화)

  • In this study, precipitation process was developed for the recovery of the lactic acid from calcium lactate fermentation broth. Calcium lactate yield was improved by decreasing the solubility of calcium lactate through the addition of ethanol (25%, v/v) as a co-precipitant. The optimal lime type, lime concentration, stirrer speed, precipitation time, temperature, and solvent amount for $Ca(LA)_2$ precipitation were CaO, 0.0175 g/mL, 220 rpm, 24 h, $5^{\circ}C$, ethanol 25% (v/v), respectively. Lactic acid was easily and efficiently recovered from precipitated $Ca(LA)_2$ by adding sulfuric acid ($Ca(LA)_2/H_2SO_4$ molar ratio=1:1). In the model solution of organic acids and fermentation broth, the overall yields of recovered lactic acid were 62% and 55%, respectively, under the aforementioned optimal conditions.

Retardation of Kimchi Fermentation by the Extracts of Allium tuberosum and Growth Inhibition of Related Microorganisms (부추추출물의 김치발효 지연 및 관련 미생물 증식억제)

  • The effect of retarding the fermentation of Kimchi by the extract of leek(Allium tuberosum) were tested by measuring the changes in pH, acidity and total cell number as well as number of microorganisms involved in Kimchi fermentation such as Lactobacillus, Leuconostoc, Pediococcus, Streptococcus and yeasts. The changes of pH and acidity of Kimchi stored at $25^{\circ}C$ indicated that the shelf-life of Kimchi with leek extract was retarded by 1.5 days compared with Kimchi without leek extract. Growth of Lactobacillus, Leuconostoc, Pediococcus and yeasts in Kimchi were remarkably inhibited by adding the leek extract at the initial and the 1st day of fermentation. This result suggested the methanol extract of leek can be sucessfully used for the extension of shelf-life of Kimchi.

Sustained Cell Growth and Improved Cyclosporin A Production Capablity of Immobilized Tolypocladium Inflatum Cells (고정상 Tolypocladium inflatum균의 세포성장 지속성과 Cyclosporin A 생산성 향상)

  • In batch bioreactor fermentations for cyclosporin A (CyA) production, good potential for bioprocess improvement was demonstrated in the immobilized cell system, providing appreciably better utilization of the catalytic activity of the biomass than the freely suspended cells, especially during the exponential phase. When concentrated nutrient medium was added pulsely during the exponential phase of cell growth(at hour 139 of fermentation), reactivation and regermination in both immobilized and suspended cell cultures were observed to contribute to the longevity of CyA production, maintaining maximum CyA titre until 250 hours of fermentation. Contrarily, simple batch fermentations without any supplement of medium in both systems showed repid decrease in CyA concentrations during the late stationary phase. Notably, the CyA yield coefficient $(Y_p/x)$ for the immobilized cell system was maintained quite high even after the pulse addition of the concentrated full medium, reaching almost 80% of the level attained during the exponential phase. This is in sharp contrast when compared with the corresponding value of 58% in the case of parallel-suspended cells. This pattern of CyA production resulted in considerably enhanced CyA production in the immobilized cell system, reaching almost 2 time higher maximum CyA production in comparison with the free cell system.

  • Optimized fermentation medium and cultural conditions for the production or exopolysaccharide KS-1 with Bacillus polymyxa KS-1 was following as; 30g g1ucose, 2.59g yeast extract, $2.5g KH_2PO_4, 0.5g NaCl, 0.3g MgSO_4.7H_20, 0.1g CaC0_3 0.05g, FeSO_4.7H_2O, and 0.05g MnS0_4 . 4H_20in 1 liter distilled water. The exopolysaccharide production was influenced by the by the temperature and pH, the optimal conditions for the production of exopolysaccharide KS-1 seemed to be $30^{\circ}C$ and pH 7.0, respectively. About $10.3g/\ell$ of maximum exopolysaccharide was obtained al the initial pH 7.0, $30^{\circ}C$, 2vvm of aeration rate and 400 rpm of impeller speed in a jar fermentor.

Effect of biogenic amine forming and degrading bacteria on quality characteristics of Kimchi (바이오제닉 아민 생성균과 분해균이 김치의 품질 특성에 미치는 영향)

  • The purpose of this study was to investigate the quality characteristics of kimchi prepared with a single starter culture of biogenic amines (BA)-forming lactic acid bacteria (LAB) or a combined starter cultures composed of BA-forming and BA-degrading LAB. As the fermentation proceeded, the lactic acid bacterial count, titratable acidity, and BA content in kimchi prepared with myeolchi-aekjeot were slightly higher than those of kimchi prepared with saeu-jeot. The amount and type of BA produced by LAB were mostly strain dependent rather than species specific. Among all of the isolated LAB strains, the highest levels of cadaverine, histamine, putrescine and tyramine were produced by Leuconostoc mesenteroides MBK32, Lactobacillus brevis MBK34, Lactobacillus curvatus MBK31 and Enterococcus faecalis SBK31, respectively. BA-forming and BA-degrading starter cultures played an important role in the growth rate and organic acid-producing ability of LAB in kimchi. Interestingly, BA contents in kimchi increased by adding single BA-forming LAB starter were effectively lowered by the mixed cultures with BA-degrading LAB.

Identification and Fermentation Characteristics of Lactic Acid Bacteria Isolated from Dongchimi as Starter for Radish Juice (무 쥬스 제조를 위한 starter로써 동치미에서 분리한 유산균의 동정 및 발효 특성)

  • This study was aimed at the investigation of the possibility of the addition of lactic acid bacteria as "starter"for the preparation of radish juice. Forty strains of lactic acid bacteria were isolated from dongchimi that was fermented by a traditional method. The isolates were assorted into 5groups, Leuconostoc mesenteroides subsp. mesenteroides (J-9), Lacobacillus brevis (J-12), Lactobacillus fermentum (J-7), Lactobacillus sake (J-20), and Lactobacillus plantarum (J-39). Leuconostoc mesenteroides was predominated in the sample of dongchimi with frequency of 52.5%. Each of the strain, which exhibited the beat growth in the species, was selected in the 5species, and investigation of the fermentation characteristcis was carried out. The fermentation were performed for 9 days at 25${\circ}C$ after the inoculation of 0.3% ($10^{6}$ cfu/㎖) to each ultra-filtrated radish juice. The pH, total acidity, content of non-volatile organic acids were examined during the fermentation period. Lactobacillus plantarum showed the highest growth rate and the growth rate of Lactobacillus sake was the lowest. The pH (6.3-6.36) and total acidity (0.09-1.0 %) fo the ultrafiltrated radish juice before fermentation were changed to 3.2-4.3 and 0.65-1.2% after 9days, respectively. The changes of the pH and total acidity were related with the growth of the lactic acid bacteria; the better growth of lactic acid bacteria, the more rapid decrease of pH and increase of the total acidity. when the amount of non-volatile organic acids were estimated during fermentation, citric acid, malic acid, malonic acid, and succinic acid were decreased in all cases. However, the content of lactic acid increased with the progression of fermentation. L. mesenteroides (J-9), L. brevis (J-12) and L. fermentum (J-7) were chosen for the candidates of the starter for the lactic fermentation of radish juice based on the biochemical analysis and sensory evaluation.

Physiochemical and Quality Characteristics of Young Radish (Yulmoo) Kimchi Cultivated by Organic Farming (유기농법에 의해 재배된 열무김치의 품질 및 기능적 특성)

  • This study compared and analyzed the physiochemical and quality characteristics of young radish kimchi made with YR-FNC or YR-GC cultivated by organic farming as well as kimchi made with young radish cultivated by chemical composting/general farming (YR-Control). YR-Control showed higher contents of water and crude protein than YR-FNC and YR-GC kimchi but lower contents of crude ash, dietary fiber, and vitamin C. YR-FNC and YR-GC kimchi also showed higher contents of P and Cu than YR-Control but lower contents of N, K, Ca, Mg, Fe, and Zn. YR-Control fermentation progressed faster than that of YR-FNC or YR-GC after 24 days, and YR-Control more rapidly reached an appropriate pH. On the other hand, YR-FNC and YR-GC kimchi fermentation progressed slow and did not reach a pH level below 5.3. For period of fermentation, YR-Control and YR-FNC kimchi showed no significant difference in reducing sugar content, whereas that of YR-GC kimchi decreased significantly after 24 days of fermentation (P<0.002). YR-FNC and YR-GC kimchi showed 1.5~3 times higher total polyphenol and flavonoid contents than YR-Control at the beginning of fermentation, whereas YR-Control and YR-FNC kimchi showed a significant increase in total polyphenol and flavonoid contents during the fermentation period. By day 7 of fermentation, YR-FNC kimchi showed higher contents of lactic acid bacteria and total microbes than YR-Control. In the sensory evaluation, YR-FNC and YR-GC kimchi showed higher preference values than YR-Control. Therefore, kimchi made from young radish cultivated by organic farming has a longer freshness period than control kimchi and is characterized by excellent sensory quality, increased physiological contents, and improved beneficial health effects.

Anti-Obesity Effect and Fermentation Characteristics of American Preferred Kimchi Added to Garcinia Cambogia Extracts (Hydroxy Citric Acid) (가르시니아 캄보지아 추출물(Hydroxy Citric Acid) 첨가 미국인 선호 김치의 발효특성 및 항비만 효과)

  • To develop functional kimchi which had anti-obesity effect, garcinia cambogia extract containing $51.46\%$ hydroxy citric acid (HCA) was used as a sub-ingredient of American preferred kimchi (APK). The APK added to garcinia cambogia extracts of 0.5$\%,\;1.0\%,\;1.5\%\;and\;2.0\%$ were prepared, and fermentation characteristics and anti-obesity effect of those kimchi were investigated. The pH of APK added to garcinia cambogia extracts (APKH) was low as the amount of garcinia cambogia extract increased at initial stage of fermentation but the pH of those kimchi showed similar values after optimum ripened stage. The number of Lactobacillus sp. and Leuconostoc sp. were small as the amount of garcinia cambogia extract increased while the period was delayed that the number of Lactobacillus sp. and Leuconostoc sp. attained to maximum. In Hunter's color values of APKH, lightness and redness decreased as the amount of garcinia cambogia extracts increased while yellowness increased. Sensory scores in overall acceptance, taste and texture of APKH evaluated by Americans as sensory panels were similar until the addition amount of garcinia cambogia extract was $1.5\%$, therefore the garcinia cambogia extract of $1.5\%$ was determined as tile amount adding to APK. The secretions of glycerol and leptin as a key signalling factor for anti-obesity effect were examined in APK and APKH added to garcinia cambogia extract of $1.5\%$. There were no significant differences in the glycerol secretion when adipocytes were treated with APK and APKH extracts. However, leptin secretion in the adipocytes treated with APKH extract was significantly decreased compared to that of control (p<0.05).

Study on the Relationships between Rice Straw Degradation and Changes of Fibrolytic Bacteria Population by in Vitro Rumen Fermentation (In Vitro 반추위 발효를 통한 볏짚 분해와 섬유소 박테리아 군집 변화의 관계 연구)

  • This study was to research the relationships between rice straw degradation and changes of fibrolytic bacteria population during the in vitro rumen fermentation. Dry matter(DM) digestion of rice straw and population of fibrolytic bacteria were measured at the 0. 4, 8, 12 and 48 hours during the incubation. The populations of F. succinogenes. R. albus and R. flavefaciens were defined as log copy number of 16S rDNA by technical method of Quantitative real-time PCR. Total population of F. succinogenes, R. flavefaciens and R. albus was sum of bactera attached on rice straw and suspended in medium. It's population was increased with incubation, reached top level of 29.0 Log copy No at the 24 hour and then decreased. In the meantime, DM digestion of rice straw showed the higher increasement from the 8 hour to the 24 hour than from the 0 hour to the 8 hour, and then a slowdown in increasing trend of digestibility. Attachments of F. succinogenes, R. flavefaciens and R. albus were detected immediately after start of in vitro rumen incubation. At the same time, the colonized bacterial share were respectively 34.5%, 84.4% and 67.9% in total population. All of them was reached the highest colonized bacterial share above 94.7% at the 4 hour incubation. However population of attached bacteria was shown the highest level at the 12 hour or the 24 hour incubation. Kinetics of colonization were formed area of top speed from the 12 hour to the 24 hour and respectively reached 10.33, 9.28 및 8.30 Log copy No/h/g DM at the 24 hour by F. succinogenes, R. flavefaciens and R. albus. The kinetics of rice straw degradation was formed top level of 0.95% DM/h at the 24 hour. The present results gave clear evidence that degradation of rice straw was increased with the development of total fibrolytic bacteria in process of rumen fermentation. Also, their attachment was largely occurred immediately after insertion of rice straw, the colonized bacteria was actively proliferated, and then degradation of rice straw was maximized.

Relationship among Physical & Chemical Properties of Supports and Performance of Methane Fermentation in Anaerobic Fluidized-Bed Reactor (혐기성 유동층 반응기에서 지지체의 물리.화학적 특성과 메탄 발효 성능 사이의 관계)

  • Active carbon which has the smallest bulk and wet density was found as the best support media among 4 different kinds of materials(celite, natural zeolite, Pusuk stone, active carbon) to make a proper fluidized-bed with small energy consumption. Its minimum and optimum fluidization velocity were found as 0.03cm/sec and 0.25cm/sec, respectively. As organic loading rate for methane fermentation was increased, CODcr removal efficiencies of all the media were decreased. But, CODcr, removal efficiencies of active carbon was maintained more than 90% in this experimental range of the organic loading rate. Larger amount of microorganism was adsorbed on the active carbon which has very high specific surface area. At the organic loading rate of 16g CODcr,/l day, its adsorbed cell mass was 157mg/g. Comparing natural zeolite with roast celite, adsorbed cell mass did not increase in proportion to specific surface area of the media. Even though roast celite has the same specific surface area as the Pusuk stone, its organic removal ability was superior to that of the Pusuk stone, which explains that the relatively great surface roughness and the positive surface charge are important for cell adsorption. It was concluded that the support media for anaerobic fluidized reactor should have small wet density and small fuidization velocity, if possible, in order to increase cell adsorption by reducing the fluid shear stress.
