Effect of Nuruk protease activity on the quality of anchovy sauce (누룩의 protease 활성이 멸치액젓의 품질에 미치는 영향)

  • This study investigated the quality characteristics of anchovy sauce fermented using Nuruk to maintain a unique flavor, reduce fishy smell, and improve the fermentation rate. Six kinds of fermented fish sauces, including the control, fermentation using traditional Nuruk; SH Koji (Fs-A), JJ Koji (Fs-B), GJ Koji (Fs-C); and fermentation with improved Nuruk; Aspergillus luchuensis (Fs-D) and Aspergillus oryzae (Fs-E), were prepared. Samples were collected at 15 days intervals with 10% Nuruk added to raw anchovy and fermented at 25o C for 60 days. The free amino acids, especially glutamic acid content and amino nitrogen, were the highest in Fs-C, reflecting the high protease activity of Nuruk C (GJ). Regarding overall sensory evaluation, the control was the lowest, whereas Fs-C was highly evaluated among the sample groups. The addition of Nuruk not only shortened the fermentation period, but also increased the overall sensory level by adding umami and reducing fishy odor.

Distillation and Quality Characteristics of Medicinal Herb Wines (약용주의 증류와 품질특성)

  • Korean general medicinal herbs-sasam, gilkyung, jakyak, danggwi, hwangki, and chunkung-were added In the normal brewing procedure as a raw material or in the distilling procedure as a packing material. The distillates from the former procedure and those from the latter procedure were compared in quality and distillation properties. As distillation proceeded, pH of the medicinal herb wine distillate and the control(not added herbs) distillates were decreased, whereas that of the herb packing distillate was increased slowely of $0.05{\sim}0.97$ during $1{\sim}4$ fractions and decreased remarkably of $0.92{\sim}0.98$ afterward. Average pH was the highest of 5.70 in jakyak and lowest of 4.37 In gilkyung. Absorbances of the herb Packing distillate were decreased rapidly of $0.60{\sim}1.59$ in the $1{\sim}4$ fractions but slowely of $0.19{\sim}0.54$ in the next fractions. During distillation both fractional alcohol concentration of the distillates and distillation rate were decreased. Their values were decreased more slowly than the control. Distillation rates of medicinal herb wine distillate were varied by medicinal herb varieties and alcohol concentration of fermented wine. Danggwi and control showed the highest average distillation rate as $0.12\;m{\ell}/sec$ and gilkyung the lowest value as $0.073\;m{\ell}/sec$. Maximum concentration of index component, paeoniflorin of jakyak was observed as 293 mg% in the 5th fraction of herb packing distillate and decrusin of danggwi as 3514 mg% In the 1st fraction of herb packing distillate. The extraction rate was 41.3% for paeoniflorin and 20.5% for decrusin. From sensory evaluation, the highest overall Qualify was observed in the medicinal herb wine distillate of hwangki added wine, the next in those of danggwi and jakyak added wine.

  • S. cerevisiae mutant(reg1-501, gal1), which cannot use galactose and has alleviated glucose repression level, is used as host for optimizing induction of GAL promoter. The optimum concentration of galactose as inducer for recombinant protein production and the galactose consumption rate have been tested with S. cerevisiae mutant and compared with conventional S. cerevisiae. The extent of glucose repression were investigated for both strain and the degradation pattern of produced foreign protein have been compared in both cases. The effect of pH on foreign protein degradation pattern were studied for both strains. The secetion efficiency of both strains were carried out. Through these experiments, optimum condition of recombinant protein production by GAL promoter using S. cerevisiae mutant (reg1-501, gal1) were found.

  • 필기 단어 인식 방법에는 낱자별 분할 및 낱자 단위 인식을 통해 인식하는 방법과 단어 사전을 이용하여 단어와 영상을 직접 비교하는 방법이 있다. 이 중 후자는 인식 대상이 되는 단어들이 작은 수의 어휘로 제한되었을 대 매우 효과적이다. 본 논문에서는 입력 영상이 주어졌을 때 자모를 순차적으로 탐색하고 그 결과의 최적 조합을 찾아 인식하는 사전을 이용한 필기 한글 단어 인식 방법을 제안한다. 입력 영상은 사전의 각 단어와의 매칭을 통해 인식된다. 단어는 필기 순서로 정렬된 자모열로 표현하고 입력 영상은 획들의 집합으로 표현한다. 단어의 자모들은 입력 영상으로부터 추출된 획들의 집합으로부터 단계적으로 탐색된다. 각 단계에서는 전 단계까지의 매칭 상태와 탐색하려는 자모의 형태로부터 자모가 존재할 것이라고 기대되는 정합 기대 영역을 설정한 후 그 안에서 자모 탐색기를 이용해 자모를 찾는다. 자모 탐색기는 획들의 집합으로 이루어진 복수의 자모 후보와 그 점수를 출력한다. 각 단계마다 생성된 자모 후보들은 최적의 단어 매칭을 찾기 위한 탐색 공간을 이룬다. 본 연구에서는 단어 사전을 trie로 구성하고, 탐색 과정에서 dynamic programming을 이용하여 효과적으로 탐색을 수행하였다. 또한 인식 속도를 향상시키기 위해 산전 축소, 탐색 공간 축소 등 다양한 지식을 이용하였다. 제안하는 방법은 무제약으로 쓰여진 필기 단어도 인식 할 수 있을 뿐 아니라, 동적 사전을 이용하기 때문에 사전의 내용이 변하는 환경에서도 적용할 수 있다. 인식 실험에서는 39개의 단어로 이루어진 사전에 대하여 613개의 단어 영상에 대해 실험한 결과 98.54%의 높은 인식률을 보임으로써 제안하는 방법이 매우 효과적임을 확인하였다. 아니라 곰팡이 균주도 실제 praxis에 적합하게 개발시킬수 있다. 따라서 앞으로 발효육제품제조에 있어 starter culture가 갖는 의미는 매우 중요하며 특히 짧은 숙성기간을 거치는 발효소시지의 제조에 있어서는 필수불가결한 공정의 한 분야로 자리잡게 될 것이다.큰 차이 없었으나 이중포장과 진공포장은 상당히 효과적임을 알 수 있었다.로는 18%에 비하여 22%가 더 적합한 것으로 생각되었다.$0.15{\sim}0.35%$이었다.irc}C$에서 $13.49{\times}10^{-3}$이었다. 이 값들을 Arrhenius식에 대입하여 구한 활성화 에너지는 24.795 kJ/Kmol이었다. 이 값으로부터 결정한 살균 포장약주 명가의 상용 저장 수명은 $10^{\circ}C$에서 2년, $20^{\circ}C$에서 1년 4개월, $25^{\circ}C$에서 1년 2개월 이었다. 서울의 매월 평균 온도를 기준으로 계산할 때 본제품의 상용저장기간은 1년 8개월이었다.로 반죽이 호화되고 가열시간이 그 이상으로 증가할 때도 반죽의 호화가 약간은 진행되지만 $90^{\circ}C$ 이상의 가열온도에서는 가열시간 0.5분 이내에 반죽의 호화가 급속히 일어나고 가열 시간을 증가시켜도 더이상의 호화는 일어나지 않았다. 같은 조건에서는 waxy corn starch 반죽의 호화 속도가 corn starch보다 더 빠른 것으로 나타났다. 대표적으로 52% 수분함량에서 반응속도상수(k)와 가열온도(T)사이의 관계식은 corn starch의 경우 $logk=11.1140-4.1226{\times}10^3(1/T)

Importance of Strain Improvement and Control of Fungal cells Morphology for Enhanced Production of Protein-bound Polysaccharides(β-D-glucan) in Suspended Cultures of Phellinus linteus Mycelia (Phellinus linteus의 균사체 액상배양에서 단백다당체(β-D-glucan)의 생산성 향상을 위한 균주 개량과 배양형태 조절의 중요성)

  • Strain improvement and morphology investigation in bioreactor cultures were undertaken in suspended cultures of Phellinus linteus mycelia for mass production of protein-bound polysaccharides(soluble ${\beta}$-D-glucan), a powerful immuno-stimulating agent. Phellineus sp. screened for this research was identified as Phellinus linteues through ITS rDNA sequencing method and blast search, demonstrating 99.7% similarity to other Phellinus linteus strains. Intensive strain improvement program was carried out by obtaining large amounts of protoplasts for the isolation of single cell colonies. Rapid and large screening of high-yielding producers was possible because large numbers of protoplasts ($1{\times}10^5{\sim}10^6\;protoplasts/ml$) formed using the banding filtration method with the cell wall-disrupting enzymes could be regenerated in relatively high regeneration frequency($10^{-2}{\sim}10^{-3}$) in the newly developed regeneration medium. It was demonstrated that the strains showing high performances in the protoplast regeneration and solid growth medium were able to produce 5.8~6.4%(w/w) of ${\beta}$-D-glucan and 13~15 g/L of biomass in stable manners in suspended shake-flask cultures of P. linteus mycelia. In addition, cell mass increase was observed to be the most important in order to enhance ${\beta}$-D-glucan productivity during the course of strain improvement program, since the amount of ${\beta}$-D-glucan extracted from the cell wall of P. linteus mycelia was almost constant on the unit biomass basis. Therefore we fully investigated the fungal cell morphology, generally known as one of the key factors affecting cell growth extent in the bioreactor cultures of mycelial fungal cells. It was found that, in order to obtain as high cell mass as possible in the final production bioreactor cultures, the producing cells should be proliferated in condensed filamentous forms in the growth cultures, and optimum amounts of these filamentous cells should be transferred as active inoculums to the production bioreactor. In this case, ideal morphologies consisting of compacted pellets less than 0.5mm in diameter were successfully induced in the production cultures, resulting in shorter period of lag phase, 1.5 fold higher specific cell growth rate and 3.3 fold increase in the final biomass production as compared to the parallel bioreactor cultures of different morphological forms. It was concluded that not only the high-yielding but also the good morphological characteristics led to the significantly higher biomass production and ${\beta}$-D-glucan productivity in the final production cultures.

Fermentation Process Characteristics of Phaffia rhodozyma Mutant B76 for Astaxanthin Biosynthesis (Astaxanthin 생합성을 위한 Phaffia rhodoxyma 변이주 B76의 발효공정 특성)

  • Specific carotenoids and astaxanthin biosynthesis power of Phaffia rhodozyma mutant 876, which was obtained after NTG a and UV treatments, was higher than those of the wild type by 40% and 50%, respectively. The mutant strain did not show t the catabolite repression even at 22% (w/v) glucose concentration. The optimum C{N ratio was 2.0, and the optimum t temperature and initial pH were $22^{\circ}C$ and 6.0, respectively. 80th cell growth and astaxanthin formation decreased drastically a as the fermentation temperature was increased over $22^{\circ}C$, whereas they were comparable in the pH range between 5.0 and 7 7.0. Inoculum size did not affect the final cell density nor the carotenoids biosynthesis, and 3%(v/v) was selected as optimal. H Higher dissolved oxygen concentration facilitated astaxanthin biosynthesis, and aeration rate of 1.0 v/0/m and agitation speed of 400 rpm were selected as optimum. The final cell dens때 of 43.3 g/L and the volumetric astaxanthin and carotenoids concentrations of 110.6 mg/L and 149.4 mg/L, respectively, were obtained. The specific carotenoids concentration was 3.45 m mg{g-yeast(dry). Yx/s and Yp/s values of 0.37 and 1.08 were obtained. The result of this study will provide basic information u useful for mass production of astaxanthin from P. rhodozyma fermentation.

Comparison of Ethanol Fermentation Properties between Laboratorial and Industrial Yeast Strains using Cassava Hydrolysate (카사바 당화액을 이용한 실험실용 및 산업용 효모의 에탄올 발효성능 비교)

  • In order to investigate the ethanol fermentation properties of alcohol yeasts a laboratorial strain (CEN.PK2-1D) and two industrial alcohol yeasts (JHS100 and JHS200) of Saccharomyces cerevisiae were cultured in a pure YP medium with 300 g/L glucose and cassava hydrolysate. Spot assay and cell viability tests showed that both the JHS100 and JHS200 strains exhibited higher ethanol tolerance than the CEN.PK2-1D strain. The JHS100 strain demonstrated the highest cell growth, glucose consumption and ethanol production. In particular, an anaerobic batch fermentation of the JHS100 strain using cassava hydrolysate with 250 g/L glucose resulted in a 106.1 g/L ethanol concentration, 0.42 g/g ethanol yield and 3.15 g/L-hr ethanol productivity, which were 53%, 13%, 53% higher than the corresponding values for the CEN.PK2-1D strain. By changing the pure YP medium to cassava hydrolysate, 19% and 17% decreases in ethanol yield and productivity for the CEN.PK2-1D strain were observed, whereas the cultures of the JHS100 and JHS200 stains showed similar ethanol productivities and only an 8% decrease in ethanol yield. Furthermore, the JHS100 and JHS200 stains produced lower levels of glycerol and acetate byproducts than the CEN.PK2-1D strain. Consequently, the outstanding ethanol fermentation performance of the industrial strains might be owing to rapid cell growth, high ethanol tolerance, low nitrogen requirements and the low formation of by-products.

  • Experimental studies on the effects of impeller to provide the microbial transglutaminase derived from Streptoverticillium mobaraense have been conducted. The optimal production medium was determined by latin-square design, and the effects of aeration and agitation were observed by using different sizes and shapes of impellers for the poduction of transglutaminase. The effects of pH and temperature were also evaluated for the production of transglutaminase in flasks. As a result, pH is more effective than temperature for both enzyme production and growth of the microorganism. The peak enzyme activity for transglutaminase in fermenter was 0.7 U/mL, but this was still well below the avereage enzyme activity, 1,3 U/mL, obtained in flask runs.

Growth model for Pichia stipitis growing on sugar mixtures (혼합당에서의 Pichia stipitis의 생육 모델)

  • Low cost fermentation substrates frequently contain a mixture of carbon sources including hexoses, pentoses and disaccharides. Fermentation of such mixtures requires an understanding of how each of these substrates is utilized. During batch culture of Pichia stipitis CBS 5776 on sugar mixtures, glucose causes catabolite repression of xylose and cellobiose utilization. Also, glucose causes a permanent repression of xylose utilization as evidenced by reduced growth rates during the xylose phase of glucose/xylose fermentation. The growth model for multiple substrates is developed based on a cyclic AMP mediated catabolite repression mechanism and this model adequately described the growth and ethanol production from sugar mixtures.

  • From the on-line mass spectrometric analyese of the exhaust gaseous composition of fermentor and the material balance equations for oxygen and carbon dioxide, oxygen uptake rate (OUR) and carbon dioxide evolution rate (CER) were calculate using a personal computer (IBM PC-AT) interfaced to a quadrupole mass spectromter. The calculate OUR and CER were used for the indirect estimation of cell and substrate concentrations during the batch culture of Candida utilis. For the estimation of sustrate concentration, the yield model of Pirt was applied. It was found that the cell and substrate (glucose) concentration could be ssatisfactorily estimataed and the results showed the more accurate estimations of both fermentation state variables when OUR data were used than CER data.
