THE EXPRESSION PATTERN OF BMPS AND THEIR RECEPTORS IN CALVARIAL SUTURE DEVELOPMENT (두개봉합부의 초기형태발생과정에서 BMP와 그 수용체의 발현 양상)

  • Bone morphogenetic proteins(BMPs) are secretory signal molecules which have a variety of regulatory functions during morphogenesis and cell differentiation. To evaluate roles of BMPs and their receptors on mouse sagittal suture development, we have examined their expression patterns in serial sections of sagittal sutures by in situ hybridization during embryonic stages(E15-E18). BMP-2 and BMP-3 were expressed in the osteogenic front and parietal bone on embryonic 15day, from E16 in hair follicle. BMP-4 was strongly expressed in the osteogenic front and weakly expressed in the mesenchyme and parietal bone. BMP-S was expressed in the hair follicles. BMP-6 was not expressed in this study. BMP-7 was expressed in parietal bone during embryonic stage. BMPR-IB was expressed in the osteogenic front, but BMPR-IA was not. From these datas, we suggest that the BMP-4 regulates the early commitment of mesenchymal cells to the osteogenic lineages, the BMP-2 and BMP-3 may be involved in regulating the differentiation of osteoblast precursor cells. BMP-7 was involved in maintenance of differentiated osteoblasts. BMPs were key signaling molecules that regulate early calvarial bone morphogenesis, mediated by BMPR-IB.

Correlation Between the Expression of Epidermal Growth Factor Receptor and MR Features in Glioma (신경교종에서 표피성장인자수용체의 발현도와 자기공명영상 소견의 상관관계)

  • Purpose: The aim of this study was to find correlation between the expression of epidermal growth factor receptor (EGFR) and MR findings in the brain glioma. Materials and Methods: MR features including edema, margin, necrosis, heterogeneity, hemorrhage and contrast enhancement were retrospectively analyzed with preoperative MR images in 41 patients with proven brain gliomas (8 low grade astrocytomas, 12 anaplastic astrocytomas, 21 glioblastoma multiformes). Immunohistochemical study of EGFR was done and their expressions were graded by both stained distribution and intensity. Correlation analysis between the MR features and EGFR expressions was done. Results: Peritumoral edema was correlated with both distribution (r=0.71, p=0.00) and stain intensity (r=0.69, p=0.00) of EGFR expression. Other MR features showed no statistical correlation with EGFR expression. Conclusion: MRI is useful in evaluation of brain glioma, and peritumoral edema is useful finding that suggests EGFR expression as well as malignant histopathologic grade of the tumor.

Differential Expressions of Adhesion Receptor Genes in the Rat Uterus Associated with Ovarian Steroid Hormone (흰쥐 자궁에서 난소 스테로이드 호르몬에 의한 Adhesion 수용체 유전자 발현조절에 대한 연구)

  • This report aimed at investigating the differential gene expressions of the adhesion receptors between ovariectomized (OVX) and estrus stage rat uteri (OVX vs. estrus pair) using the cDNA expression away analysis. In addition, this report aimed at confirming of the differential gene expressions of the adhesion receptors between OVX and progesterone (P$_4$) injected OVX rat uteri (OVX vs. OVX+P$_4$ pair). RNA samples were extracted from the uterus and reverse-transcribed in the presence of [$\alpha$$^{32}$ P]-dATP. Membrane sets of Rat Atlas array 1.2 II (Clontech) were hybridized with CDNA probe sets. RT-PCR was employed to validate the relative gene expression patterns obtained by the cDNA array. The results were well consistent to cDNA array analysis data except the fold changes of gene expression. Among a total of 1176 cDNAs, 5 genes of adhesion receotor including embigin protein, activated leukocyte cell adhesion molecule, afadin, neuroligin 2, semaphorin Z showed significant (more than 2-fold) changes in the OVX vs. late estrus pair. All of these genes were up regulated in estrus stage than OVX rat uterus. In the OVX vs. OVX+P$_4$ pair, 4 genes including osteonectin, afadin, neuroligin 2, semaphorin Z showed significant changes. All of these genes were also up regulated in OVX+P$_4$ injected rat uterus than OVX control. Three genes including afadin, neuroligin 2, semaphorin Z which were up regulated in estrus and OVX+P$_4$ injected rat uteri of both experimental pairs than OVX rat uteri. These genes seem to be under the control of P$_4$.

Restriction of Ca2+ deficiency-like symptoms by co-expressing a Ca2+ transporter and a Ca2+-binding protein in tomato (토마토에서 칼슘수송체와 칼슘결합단백질 공동발현에 의한 칼슘결핍유사증상의 완화)

  • Here we focused on tip-burn and blossom-end rot (BER) symptoms in the tomato plants expressing the constitutively active form of $Ca^{2+}/H^+$ antiporter (sCAX1) and/or a Ca-binding protein (calreticulin, CRT) genes during their whole growth period. Conclusively we demonstrated that CRT is able to suppress the tip-burn and BER symptoms that were induced by sCAX1. Under poor nutrition condition, tomato plants overexpressing sCAX1 showed severe necrotic collapses in both roots and shoot polar tissues, which are in accordance with $Ca^{2+}$ deficient symptoms frequently observed in an agricultural cultivation of tomato. Reciprocal grafting trials using sCAX1 and wild type plants revealed that the tip-burn symptom by sCAX1 overexpression is not caused by hindrance of $Ca^{2+}$ uptake from soil. We constructed CRT overexpressing transgenic tomatoes, and crossed them with sCAX1 transgenic plants to investigate the effects of CRT on the symptoms of sCAX1 transgenic plants. Co-expression of sCAX1 and CRT significantly suppressed the $Ca^{2+}$ deficient symptoms of sCAX1 transgenic plants. Those results suggest the model that $Ca^{2+}$ homeostasis disturbed by the overexpression of sCAX1 may be suppressed by the co-expression of CRT.

Analysis of Toxicity in Escherichia coli from the Expression of Human Purinergic Receptor $P2X_4$ (인간 퓨린수용체 $P2X_4$를 발현시킬 때 나타나는 대장균 독성의 원인분석)

  • In general, expression of membrane protein in Escherichia coli is very toxic to the host organism, but the mechanism for the toxicity is not clear yet. Expression of human purinergic receptor $P2X_4$ was found to be extremely toxic to the host E. coli. We examined this toxicity by isolation and analysis of less toxic mutant proteins. We could isolate 30 less toxic mutants of $P2X_4$ after hydroxylamine mutagenesis. Western blot showed that all of them produced proteins smaller than the wild type $P2X_4$. DNA sequencing of two largest mutant proteins showed that they were lost its second transmembrane domain. Localization analysis of these mutant proteins showed that they are not in cytoplasmic membrane, but in inclusion bodies. These data showed that inactive truncated $P2X_4$ is not toxic to E. coli and membrane integration and functionality of $P2X_4$ may be needed to show host toxicity.

Expression Analysis of OsCPK11 by ND0001 oscpk11 Mutants of Oryza sativa L. under Salt, Cold and Drought Stress Conditions (염분, 저온 및 가뭄 스트레스 조건에서 벼 ND0001 oscpk11 돌연변이체의 OsCPK11 발현 분석)

  • Calcium-dependent protein kinases (CDPKs) are known to be involved in regulating plant responses to abiotic stresses such as salinity, cold temperature and dehydration,. Although CDPKs constitute a large multigene family consisting of 31 genes in rice, only a few rice CDPKs' functions have been identified. Therefore, in order to elucidate the functions of OsCPK11 in rice, this study was intended to focus on the expression pattern analysis of OsCPK11 in wild type and ND0001 oscpk11 mutant plants under these abiotic stresses. For the salt, cold and drought stress treatment, seedlings were exposed to 200 mM NaCl, 4℃ and 20% PEG 6,000, respectively. RT-PCR and quantitative real-time PCR were performed to determine the expression patterns of OsCPK11 in wild type and ND0001 mutant plants. RT-PCR results showed that OsCPK11 transcripts in the wild type and heterozygous mutant were detected, but not in the homozygous mutant. Real-time PCR results showed that relative expression of OsCPK11 of wild type plants was increased and reached to the highest level at 24 hr, at 6 hr and at 24 hr under salt, cold and drought stress conditions, respectively. Relative expression of OsCPK11 of ND0001 homozygous plant was significantly reduced compared to that of wild type. These results suggested that oscpk11 homozygous mutant knocks out OsCPK11 and OsCPK11 might be involved in salt, cold and drought stress signaling by regulating its gene expression.

  • 생물체의 cDNA를 유리기판위에 고밀도로 첨착 시킨 유전자 칩과 정량적인 방법으로 개별 유전자 발현을 진단 가능한 Real- time PCR (실시간 고분자중합연쇄반응 기술) 기법은 첨단 의학분야와 신약개발 및 독성유전체 연구분야에 활발히 도입되고 있는 기술이다. 본 발표의 첫 번째 부분에서는 유전자칩 에서 얻어진 유전자 발현패턴분석에 기반한 바이오마커 선정 및 real time PCR에 의한 확증 관련 기술 과 유전자칩에서 얻어지는 수많은 데이터를 재정렬 및 다양한 분석기법과 display기술을 활용하여 광범위한 화학물질에 대한 독성효과 분석을 가능하게 해주며, 특정 독성물질에 대한 관련유전자 그룹 발견 및 독성영향에 따른 분류방법에 관한 결과를 발표할 것이다. 또한 바이오마커 활용의 하나로 박테리아세포 기반 바이오센서 제작및 세포칩 개발등에 대한 결과도 추가될 것이다. 두 번째 부분에서는 non-model organism(유전체정보가 확보되지 않은 생물체)인 송사리를 이용하여 새로운 2K 유전자칩을 개발하고, 여기서 각종 화학물질에 대하여 얻어진 수많은 유전자칩 분석 데이타를 활용하여 각각의 화학물질이 보여주는 독성효과를 매우 효과적이고 쉽게 이해할 수 있는 display기술을 개발, 적용함으로써 유전자칩 발현에 기반한 화학물질 독성 screening 및 specificity discrimination을 가능케 하는 예가 발표될 것이다. 이 연구에서 개발한 송사리 유전자칩은 간조직의 RNA를 직접 cDNA화 하는 방식을 취하고 있어 전체 송사리의 유전정보를 필요로 하지 않아 비용 및 효율에서 전체 송사리의 유전정보를 얻는 비용과 노력을 취하지 않고 간에서 발생하는 독성학적 영향 및 유전자의 발현정도를 정밀하고 효율적인 방법으로 얻어 낸다. 현재 2000여개의 cDNA유전자중 50%이상의 유전자가 17베타에스트라디올, 페놀, 노닐페놀, 비스페놀, 감마레이조사, 잔류약품중 이보프란, 다이클로펜악, 농약중의 파라???R, 돌연변이 유발물질 중의 이티비알, 금속류중의 카드뮴을 통해 발현양상과 특정 캐미칼별 발현 특이성이 조사되었고, 이들 유전자는 염기서열 분석을 통해 염기서열이 분석되었으며, 미국 NCBI의 유전자 은행과의 비교를 통해 일부유전자는 새로운 유전자로 밝혀지고 있다. 또한 이 발표에서는 소염진통제계열 의약품인 dichlofenac 이 송사리의 각종 조직에 미치는 독성영향을 Real-time PCR을 이용하여 대표적 스트레스 유전자의 발현에 미치는 영향에 대한 분석 예가 발표될 것이다.

The Gene Expression Level Differences associated with Exercise in the Mouse Brain exposed to Radiofrequency Radiation (전자파에 노출된 생쥐에서 운동량에 따른 뇌의 유전자 변화)

  • The effect of radiofrequency radiation (RF) exposure on mouse associated with the exercise was investigated in the brain at the molecular level. The expression of tyrosine hydroxylase(TH), FoxO3a, AMPKα and mRNA was investigated by real-time RT-PCR in striatum and the hypothalamus. In the striatum, TH mRNA expression was decreased in the exercise and RF exposure group. FoxO3a mRNA expression was significantly increased in the spontaneous exercise group and a significant decrease was observed in the RF exposure and spontaneous exercise group. In the hypothalamus, TH mRNA expression was significantly decreased in the RF exposure and spontaneous exercise group. But, FoxO3a mRNA expression was significantly increased in the RF exposure and spontaneous exercise group. We will further investigate the expression of protein molecules in the hippocampus of the brain to reveal the effects of RF radiation on memory.

Expression of Calponin 3 in the Striatum Following 3-Nitropropionic Acid-induced Neurotoxicity (선조체에서 3-nitropropionic acid 투여 후 calponin 3의 발현 연구)

  • Calponin 3 is an F-actin-binding protein and plays a key role in regulating spine plasticity and synaptic activity in neurons. Unlike the other subtypes, calponin 1 and 2, which are expressed in smooth and cardiac muscle cells, calponin 3 is highly expressed in the brain. The goal of this study was to elucidate the spatiotemporal expression pattern of calponin 3 following repeated administration of 3-nitropropionic acid in mice. The repeated administration of 3-nitropropionic acid generated necrotic neuronal cell death in the striatum. Calponin 3 was up-regulated in the neuroprotective penimbral region from 1.5 days after the last injection and thereafter. Double immunofluorescence study revealed that calponin 3 was induced in GFAP-positive astrocytes. These results suggest that calponin 3 induction in the neuroprotective penumbral area following 3-nitropropionic acid intoxication may play a key role in reactive astrogliosis in the striatum.
