  • An experiment was conducted to evaluate condensed molasses solubles(CMS, a by-product from monosodium glutamate production) as a source of nitrogen for ruminant with particular reference to its effects on microbial protein synthesis. Four non-lactating dairy cows fitted with rumen cannulas were used in a 4 ${\times}$ 4 Latin square with 14-day periods. The four treatments were (1) basal diet consisting of barley straw ad libitum and 3 kg/d of rolled barley, (2) basal diet plus 200 gld molasses and 300 g/d water, (3) basal diet plus 200 g/d molasses, 100 g/d CMS and 200 g/d water, (4) basal diet plus 200 g/d molasses, 200 g/d CMS and 100 g/d water. Ruminal pH remained at high levels and showed little variation during the day between treatments. The concentration of total and individual VFA in the rumen was similar between treatments. There was no difference in the concentration of ammonia in the rumen between treatments, although the intake of nitrogen in molassesl CMS mixture treatments was higher than that of control and molasses treatment. But there was a suggestion of an increased synthesis of microbial protein with the higher level of inclusion of CMS when the allantoin/creatinine ratio was used as an index of microbial protein production(P <0.10).

  • Four Holstein cows were used in a 4 ${\times}$ 4 Latin square experiment to study effects of fat sources on rumen metabolism and digestibility of nutrients. All cows were fed a total mixed diets containing 60% silage and 40% concentrate. The four concentrates were formulated to contain either Megalac(MEG), formaldehyde-treated whole linseed(LIN), a mixture (50 : 50, oil basis) of fish oil and formaldehyde-treated whole linseed(MIX), or no fat source in the concentrate but 500g per day of linseed oil being infused into the duodemm (OIL). The rumen pH was lowest in OIL among the treatments(P < 0.05), but ammonia N concentration in the rumen was not significantly different among the treatments. The differences of total VFA, acetate, propionate, iso-butyrate and iso-valerate concentrations were not significant among the treatments. While, butyrate and valerate were highest in OIL and lowest in MEG(P < 0.05 and P <0.01, respectively). In addition, A:P ratio was also highest in OIL and lowest in MEG(P < 0.05). As expected, intake of nutrients(DM, OM, NDF and ADF) was lowest in OIL among the treatments(P < 0.01). However, all nutrients flow to the duodenum, and digestion in the rumen and total tract were not significantly different among the treatments. Intake of N was highest in MEG, but lowest in OIL treatment(P < 0.01). Duodenal flow of total N, nonammonia N and microbial N was not significantly different across the treatments. In addition, microbial synthesis and ammonia N and total N digestibility were not affected by different dietary fat sources. The present results show that fermentative characteristic and microbial efficiency in the rumen, and nutrients digestibility in the rumen and total tract were not depressed by supplementation of as much as 6% dietary fat sources.

  • These study were conducted to determine the effects of a whole or steam-flaked corn based diet on rumen microbial fermentation in vitro and ruminal metabolism in the Korean Native Goat(KNG) in vivo. The experiments consisted of two dietary treatments: control, steam-flaked corn(SFC) based diet(80%) + rice straw mixed(20%)(SFCR); 100% whole corn based diet(WC). The first experiment was conducted to investigate the effect of whole corn on ruminal metabolism in vitro for 0 to 48 h. pH values were optimally maintained during incubation time, and were not significantly different between treatments. Gas production of SFCR was significantly higher than WC(p<0.01). $NH_3$-N concentration tended to increase for WC, but not significantly different between treatments. The mean value of total volatile fatty acid concentration of WC was significantly lower than SFCR(p<0.01), but SFCR and WC linearly increased as the time of incubation approached 48 h. Mean value of acetate concentration of SFCR was significantly higher than WC(p<0.01). Propionate concentration of WC for the total incubation time was significantly higher than SFCR(p<0.01). The digestibility of dry matter was not significantly different between treatments, but SFCR was somewhat higher than WC. The second experiment was conducted to effect of whole shelled corn based diet on rumen metabolism in KNG. pH values tended to decrease through all treatments. There was not a significantly difference between treatments. Microbial protein yield of SFCR was significantly higher than WC(p<0.01). $NH_3$-N concentration of WC was significantly (p<0.01) higher than SFCR. Total VFA and propionate concentration of WC was significantly higher than SFCR(p<0.01), but acetate concentrate of WC was not significantly higher than SFCR. The mean value of total lactate concentration was significantly(p<0.01) different but the value of SFCR and WC were lower than the average concentration of acidosis. In sacco DM disappearance rate of SFC was significantly(p<0.01) higher than WC.

  • A series of four in vitro experiments were conducted to evaluate condensed molasses solubles (CMS) as a source of nitrogen for ruminal microbes. In experiment 1, as compared with urea, the value of CMS as a nitrogen source was examined. In experiment 2, to determine the time needed for maximal response of microbial synthesis, the treatments were incubated for increasing times (from 6 h to 16 h). Because a sediment that was assumed to cause nitrogen loss was found after incubation in experiments 1 and 2, it was decided to avoid formation of sediment using sugar instead of molasses or a shorter time incubation (experiments 3 and 4). Furthermore, in experiment 4, because the extent to which ammonia nitrogen is released from CMS and urea before 6 h of incubation was uncertain, it was decided to examine the peaks of concentrations of ammonia nitrogen released from CMS and urea by sampling after 2 h incubation. There was no significant difference in the concentration of microbial-N between molasses/CMS and molasses/ urea treatments in experiment 1, although there were greater decreases in ammonia concentration with the molasses/CMS treatment. The microbial protein synthesis was increased progressively until 10 h for both treatments (experiment 2). Although ingredients that were completely soluble (sucrose, urea) were used in experiment 3, the sediment was still evident suggesting that the sediment was largely of microbial not feed origin. Ammonia release from CMS was much faster than from urea during 2 h incubation. In conclusion, the results of the present studies suggest that the feed value of CMS as a source of nitrogen for ruminal bacteria was similar to that of urea when it was estimated in vitro.

  • The present study was conducted to investigate effective starter culture to improve biological activity of Asarum sieboldii. Antibacterial activity, antioxidant activity and reduction of enteric rumen methane production were used as criterions for biological activity. Ground A. sieboldii was added in MRS broth at 10% (w/v) and fermented by different starter cultures. Weissella confusa NJ28, Weissella cibaria NJ33, Lactobacillus curvatus NJ40, Lactobacillus brevis NJ42, Lactobacillus plantarum NJ45 and Lactobacillus sakei NJ48 were used for starter culture strains. Each starter culture was inoculated with 1% (v/v) ratio and fermentation was performed at $30^{\circ}C$ with agitation (150 rpm) for 48 h. MRS broth for the control was employed without starter culture. Then the fermentation growth was dried and extracted using ethyl alcohol. The growth of starter culture was detected at NJ40, NJ42, NJ45 and NJ48. And the highest cell growth was found in NJ40. Antibacterial activity against to Staphylococcus aureus, Listeria monocytogens, Mannheimia haemolytica and Salmonella gallinarum were observed in the extract fermented by NJ40 and NJ45. All treatments showed antioxidant activities, however, there were no significant differences (p>0.05). In in vitro rumen fermentation, negative control (NC) and positive control (PC) were assigned to without extract and with non-fermented A. sieboldii extract. Significant suppression of gas productions were detected in positive control and treatments compared to negative control (p<0.05). However, total volatile fatty acid production was not suppressed. Significant methane reduction per total volatile fatty acid productions were found in positive control and NJ45 treatment (p<0.05). The present study suggested a fermentation of A. sieboldii using NJ45 strain could improve its biological activity and make possible for its use in bio additive for enteric rumen methane mitigation without suppression of animal productivity.

  • This study was conducted to evaluate effects of halogenated compounds on in vitro rumen fermentation characteristics and methane emissions. A fistulated Holstein cow of 650 kg body weight was used as a donor of rumen fluid. Five kinds of halogenated compounds (bromochloromethane (BCM), 2-bromoethane sulfonic acid (BES), 3-bromopropanesulfonic acid (BPS), chloroform (CLF), and pyromellitic diimide (PMDI) known to inhibit methyl-coenzyme M reductase activity were added to an in vitro fermentation incubated with rumen fluid. The microbial population including bacteria, protozoa, and fungi were enumerated, and gas production including methane and fermentation characteristics were observed in vitro. The pH values ranged from 6.25 to 6.72 in all the treatments, and these showed a similar level at 48 hr. The total gas production in the treatments showed a similar pattern with C at 48 hr, whereas methane production in the treatments was lower (p<0.05) than C. Concentrations of total volatile fatty acids (VFAs) and propionic acid were higher (p<0.05) in the treatments than in C at 12 hr. Therefore, halogenated compounds (BCM, BES, BPS, CLF, and PMDI) inhibited in vitro methane emissions by inhibiting methanogens in the rumen. Further studies on safety are needed.

  • The objective of this study was to evaluate the in vitro effects of organic acids on methane emission and ruminal fermentation characteristics. We expected our methodology to result in a decrease of methanogens attached to the surface of rumen ciliate protozoa by addition of organic acids and in particular a decrease in methane emission. A fistulated Holstein cow of 650 kg body weight was used as a donor of rumen fluid. Organic acids (aspartic acid, fumaric acid, lactic acid, malic acid, and succinic acid) known to be propionate enhancers were added to an in vitro fermentation system and incubated with rumen fluid. The microbial population, including bacteria, protozoa, and fungi, were enumerated, and gas production, including methane and fermentation characteristics, were observed in vitro. Organic acids appeared to affect the rumen protozoan community. The rumen protozoal popuation decreased with the addition of aspartic acid, fumaric acid, lactic acid, and malic acid. In particular, the methane emission was reduced by addition of lactic acid. The concentration of propionate with all organic acids that were added appeared to be higher than that of the control at 12 h incubation. Addition of organic acids significantly affected rumen bacteria and microbial growth. The bacteria in added fumaric acid and malic acid was significantly higher (p<0.05) and protozoa was significantly lower (p<0.05) than that of the control. Microbial growth with the addition of organic acids was greater than the control after 48 h incubation.

  • Two experiments were conducted to observe the effects of direct fed microbials on metabolic characteristics in sheep and milking performance in dairy cows. A metabolic trial with four ruminally cannulated sheep(60±6kg) was conducted in a 4×4 Latin square design to investigate the supplementation effects of Saccharomyces cerevisiae, Clostridium butyricum or mixed microbes of S. cerevisiae and C. butyricum on ruminal fermentation characteristics and whole tract digestibility. Sheep were fed 1.25 kg of total mixed ration(TMR, DM basis) supplemented with S. cerevisiae (2.5g/day), C. butyricum (1.0g/day) or its mixture(S. cerevisiae 1.25g/day+C. butyricum 1g/day), twice daily in an equal volume. But control sheep were fed only TMR. A feeding trial with 28 lactating Holstein cattle was also conducted for 12 weeks to investigate the effects of the same microbial supplements as for the metabolic trial on milking performance. The cows were fed the TMR(control), and fed S. cerevisiae(50g/day), C. butyricum(15g/day) or its mixture (S. cerevisiae 25g/day + C. butyricum 7.5g/day) with upper layer dressing method. Total VFA concentration and the digestibility of whole digestive tract in the sheep increased by supplementation of S. cerevisiae, C. butyricum or their combined microbials compare to control group. The proportion of propionic acid at 1h(P<0.039) and 3h(P<0.022) decreased by supplementation of S. cerevisiae while tended to increase acetic acid proportion at the same times. Daily dry matter intake(DMI) was not influenced by the microbial treatments, but milk yield(P<0.031) and feed efficiency(milk yield/DMI, P<0.043) were higher for the cow received C. butyricum than those for other treatments. The milk fat content was higher (P<0.085) when cows fed S. cerevisiae(4.11%) than that fed the control (4.08%), the diets with C. butyricum (3.85%) and the microbial mixture. Based on the results obtained from the current experiments, supplementation of C. butyricum or mixture with S. cerevisiae might be increased milk fat content and milk productivity of lactating daily cows. • The comparative in vitro and in situ analysis were conducted to evaluate the rumen degradability and physical structure of domestic roughage as rice straw, timothy, alfalfa, wormwood and green tea. The feedstuffs incubated with rumen fluid and was used to determine gas production, microbial growth rate and pH changes in an in vitro experiment. The gas production was increased during incubation times and was significantly(p<0.05) lower in green tea than other feedstuffs. The microbial growth rate in the feedstuffs was increased during incubation times. However, microbial growth rate was significantly(p<0.05) lower in wormwood and green tea than other feedstuffs. Ruminal pH was decreased during incubation times, and timothy was the lowest, and rice straw was the highest among feedstuffs. The disappearance rate of dry matter(DM) and crude protein(CP) in all feedstuffs were increased during incubation times and green tea was the highest(p<0.05) compared with other feedstuffs. In effective degradability, when rumen out-flow rate was assigned to 4%, wormwood showed the highest in DM, and alfalfa was the highest in CP. Whereas, green tea was the highest in both in situ DM and CP degradability. Many cilia on the surface and stoma of wormwood and stoma in green tea were observed by scanning electron microscopy. Microbes breaked down the cilia at the beginning and then degraded the surface in wormwood. In case of green tea, microbes attached to stoma. Therefore, wormwood and green tea have a potential value as ruminal feed stuffs.

  • A study was conducted to improve the biological activity of two medicinal plants, Eucommia ulmoides Oliv. and Glycyrrhiza uralensis, by fermentation. The biological activity was assessed by determining antibacterial, antioxidant and antimethanogenic properties. Fermentation was achieved by adding the plant materials in MRS broth at 10% (w/v) and different starter cultures at 1% (v/v). Condition for fermentation were incubation temperature of $30^{\circ}C$ and agitation at 150 rpm for 48 h. Six starter cultures, Weissella confusa NJ28 (Genbank accession number KJ914897), Weissella cibaria NJ33 (Genbank accession number KJ914898), Lactobacillus curvatus NJ40 (Genbank accession number KJ914899), Lactobacillus brevis NJ42 (Genbank accession number KJ914900), Lactobacillus plantarum NJ45 (Genbank accession number KJ914901) and Lactobacillus sakei NJ48 (Genbank accession number KJ914902) were used. Antibacterial activity was observed in L. curvatus NJ40 and L. plantarum NJ45 only as opposed to other treatments, including the non-fermented groups, which showed no antibacterial activity. Both plants showed antioxidant activity, although E. ulmoides Oliv. had lower activity than G. uralensis. However, fermentation by all strains significantly improved (p<0.05), antioxidant activity in both plants compared to non-fermented treatment. Six treatments were based on antibacterial activity results, selected for in vitro rumen fermentation; 1) non-fermented E. ulmoides, 2) fermented E. ulmoides NJ40, 3) fermented E. ulmoides NJ45, 4) non-fermented G. uralensis, 5) fermented G. uralensis NJ40, 6) fermented G. uralensis NJ45. A negative control was also added, making a total of 7 treatments for the in vitro experiment. Medicinal plant-based treatments significantly improved (p<0.05) total volatile fatty acid (VFA) concentration. Significant methane reduction per mol of VFA were observed in G. uralensis (p<0.05). Based on the present study, fermentation improves the biological activity of E. ulmoides Oliv. and G. uralensis. Fermented G. uralensis could also be applied as an enteric methane mitigating agent in ruminant animals.
