This study was conducted to investigate the effects of brown seaweed waste (BSW) fermented with DS-01 microbe on in vitro rumen microbial fermentation. In in vitro trial, three different diets supplemented with 2%, 4%, 6% BSW fermented with DS-01 either for one month or two months was tested at 3 h, 6 h, 9 h, 12 h, and 24 h incubation. The chemical composition (CP, EE, CF, and ash) between brown seaweed waste (BSW) and fermented BSW (FBSW) were not different. The contamination of pathogenic microbes was not detected in FBSW. The pH value tended to be higher with 6% level of supplementation of FBSW for one month than other treatments. The pH at 24 h was significantly higher in FBSW than that of treatments without FBSW (p<0.05). In FBSW for two months, the pH value in 6% FBSW at 3 h in vitro fermentation tended to be higher than 2% or 4% FBSW treatments (p=0.0540), but there were no differences in other fermentation times. Although the concentration of $NH_3$-N of BSW fermented for one month was higher than control at 3 h (p<0.05), the volatile fatty acid values were significantly increased in 4 and 6% FBSW fermented for one month at 6 h incubation (p<0.05). In BSW fermented for two months, the volatile fatty acid values were significantly decreased in 6% treatment at 9 h (p<0.05). As a result of in vitro trial, it was recommended that the 2~4% supplementation level of brown seaweed waste fermented with DS-01 microbe for two months could be utilized for in vivo trial in ruminants.
This study was conducted to evaluate effects of mixed microbes addition on physico-chemical, fermentative and microbial parameters of sawdust-based spent mushroom substrates(SMS). The SMS was inoculated with mixed microbes(Enterobacter ludwigii, Bacillus cereus, 2 strains of Bacillus subtillis, Saccharomyces cerevisiae and Lactobacillus plantarum) at 1% level(wet basis) and anaerobically fermented during the different periods(up to 8th week). Compared with the SMS before ensiling, the ensiled one had higher CP, NDF and ADF percentages and lower DM and NFC percentages. However, levels of change were very low. The in situ ruminal disappearance of SMS DM and NDF decreased with the ensiling period prolonged. For fermentative parameters, pH reduced and lactic acid contents increased after ensiling, compared with those after ensiling. At 8th week of ensiling, pH increased and lactic acid contents reduced again, compared with those at 4th week of ensiling; however, the silage still showed favorable fermentation status. Lactic acid bacteria counts did not change throughout 8 weeks of ensiling. Counts of total microbes and yeast reduced after 4th week of ensiling period. Counts of lactic acid bacteria and yeast at 8th week of ensiling were in the levels of 108cfu/g. These results indicate that anaerobic fermentation with microbial addition could be an effective way for the long term(8 weeks) storage of the SMS.

The study was designed to estimate the in vitro rumen by-pass rate of both chromium methionine chelate as an organic supplement and $ClCl_3$ as an inorganic supplement. Rumen by-pass rates of the supplements were evaluted by comparing ruminal metabolites in rumen fluid and Cr and methionine contents in the body of ruminal microorganism. For in vitro digestion examination, basic nutrients for ruminal microbes were supplied with 7g(DM) of feed, 2g of rice straw, and 2g of corn silage per each incubation jar. Three treatments including Control(no supplementation of Cr), T1(1000ppb supplementation of $ClCl_3$) and T2(chromium methionine chelate supplementation equivalent to 1000ppb of Cr content) were prepared with five replications per each treatment. pH of T2 was lower than that of Control and T1 regardless of incubation time. Ammonia content was higher in T2 than in Control and T1 during first 6 hours of incubation. However, the ammonia content in Control was remained low after 6 hours. Total volatile fatty acids(VFA) content in control was increased constantly as incubation time was extended. Therefore, VFA content in T1 and T2 were significantly lower (P<0.05) than those of Control. Dry matter recovery rate by ruminal microorganism was the lowest in T1, however ruminal microbial population was increased most efficiently in T2 during 12 hours of in vitro incubation. Cr concentrations in the body of ruminal microbes were not different(P>0.05) between Control and T2, but it was significantly high in T1(P<0.05). Contents of methionine and cystine in ruminal microbes also were not different between Control and T2(P>0.05), but it was relatively low in T1. Based on the above results, the chromium methionine chelate was believed to by-pass rumen and could remain intact until it reaches small intestine compared to inorganic chromium. This results implies that chromium methionine chelate could be more effective to function in the small intestine of ruminant animals.
The objective of this study was to investigate the effect of different feed inoculation method on rumen fermentation in an in vitro. Three experimental treatments were used: control (CON, direct dispersion of feed (2 g) in rumen fluid), combinations of direct dispersion (1 g) and nylon bag (DNB, pore size: 50 ㎛, 1 g), and nylon bag (NB, 2 g). An in vitro fermentation experiment was carried out using strained rumen fluid for 48 h incubation time and timothy was used as a substrate. At the end of the incubation, in vitro dry matter digestibility (IVDMD), in vitro neutral detergent fiber digestibility (IVNDFD), pH, volatile fatty acids (VFA), ammonia nitrogen (NH3-N), and microbial community were evaluated and gas production was estimated at 3, 6, 12, 24, 48 h incubation periods. Gas production was higher in CON than DNB and NB at 6 and 12 h incubation time (p<0.01). There were no differences in final gas production, pH, NH3-N concentration, total VFA production, and VFA profiles among treatments. The IVDMD was lowest in CON (p<0.01) but the IVNDFD was not differed by feed distribution methods. There were no significant differences in general bacteria and fungi. Protozoa count was highest in NB treatment among treatments (p<0.01). The abundance of cellulolytic bacteria, Ruminococcus flavefaciens and Fibrobacter succinogenes, was highest in the CON among treatments (p<0.01).
This study was conducted to investigate effects of the brown seaweed residues supplementation on in vitro fermentation, and milk yield and milk composition of dairy cows. Therefore, two experiments consisting of an in vitro and an in vivo growth trial were used. In in vitro experiment, brown seaweed residues(BSR) was supplemented in basal diet with 0, 1, 2 and 4% respectively, and incubated for 3, 6, 9, 12, and 24 h. The pH value, ammonia-N and VFA were investigated. The pH value tended to increase with increasing BSR during the incubation. Particularly, pH was significantly higher in BSR treatments compared with control at 9 h(p < 0.05). While, ammonia-N concentration was not significantly different across treatments during the whole incubation. BSR supplementation did not affect total VFA production, but acetate was linearly increased in BSR treatments compared with control at 12 h(p < 0.05), and its concentration was highest(92.70 mM) in 4% BSR among treatments. The concentration of iso-butyrate tended to increase in BSR treatments in comparison to control during the incubation. In addition, the concentration of iso-valerate was higher in BSR treatments compared with control at 12 and 24 h. In growth trial, BSR was added(800 g/d/animaI) to diets of dairy cow. Dry matter intake was not affected by BSR supplementation, but daily milk yield(kg) significantly increased in BSR treatment compared with control(p < 0.05). However, milk composition(%) and milk yield(kg) were not significantly different between treatments. Milk fat(% and kg/d) tended to slightly decrease in BSR treatment compared with control(3.59% and 1.06 kg/d vs. 3.32% and 1.01 kg/d), The contents of C16:0 and C20:4 in milk significantly increased in BSR treatment compared with control reflecting from dietary fatty acid composition. The content of C18:0 in milk which is end product of biohydrogenation of CI8 unsaturated fatty acids in the rumen significantly increased in BSR treatment compared with control(p < 0.05). C18:2 content in milk tended to decrease, but tended to increase trans-II C18:l and CLA contents in milk in BSR treatment compared with control. In conclusion, it could be summarized that BSR may stabilize rumen pH, and it could improve milk yield and CIA content in milk with more than 4% of diet. Therefore, BSR could be beneficially used in dairy diets as a feed additive.
This study was conducted to utilize the wastes produced from thinning the forest as a roughage source for ruminants. Four ruminally cannulated Korean steers were used to investigate the ruminal fermentation characteristics and blood concentrations of stress-related hormones. Treatments were composed of the rice straw only (Control) and the 30% pine silage plus 70% rice straw(Pine silage) as roughages. The experiment was conducted with four replicates by a double tum over design. For sampling of blood under high temperature steers were accommodated in respiratory chamber. Concentrations of ruminal pH, volatile fatty acids and ammonia were not affected by substitution of pine silage. Mean concentration of blood thyroxine was significantly(P < 0.01) lower in the pine silage than the control. However, blood cortisol concentration was not affected by substitution of pine silage, even though it was significantly(P < 0.05) decreased after exposure for 6 hrs in high temperature. It was concluded that the pine silage using the wastes produced from thinning the forest could be use as a substitute roughage for reduction of heat stress in ruminant.
The objective of this study is to investigate the effects of Cynanchum wilfordii (CW) on cell viability, anti-oxidant activity, volatile fatty acid (VFA) production and methane gas production. Collected rumen fluid incubated with CW powder (1% w/v) for 12 and 24 hours were analyzed for pH, VFAs and methane. Alamar blue assay showed no significant difference on the viability of 3T3-L1 and C2C12 cells treated with CW for 24 hours. TBARS data showed a dose dependent increase on the antioxidant activity of CW. VFAs increased in the CW-treated groups compared to the control group. In addition, propionate increased more than other VFAs by the treatment with CW. There was a significant decrease in methane gas production in batch culture treated with CW in 12hrs. In conclusion, it was suggested that Cynanchum wilfordii could manipulate rumen fermentation considered by increasing VFA production and inhibition of methanogenesis.
This study was aimed at investigating the effect of fat supplementation with divalent ions such as MgO and $CaCl_2$ on 1) in vitro ruminal fermentation characteristics and insoluble fatty acid formation, and on 2) animal performance in finishing Hanwoo bulls. In in vitro trial, five different types of diets based on supplementation sources of fat and divalent ions, i.e. T=basal diet+4% tallow, T-Ca=T+0.5% $CaCl_2$, T-Mg=TA+0.5% MgO, T-MgCa = T +0.5% $CaCl_2$+0.5% MgO, T-caS =4% Ca salt tallow, were tested. Higher pH values were observed at 6 hr incubation(P<0.01) while higher amount of VFA were produced in diets 4 and 5 at 12 hr incubation(P<0.05). Nutrients(DM, OM, Crude protein and NDF) degradation tended to increase in divalent ions or Ca-salts treated tallow treatments compared with tallow treatment after 12 h. The amount of insoluble fatty acid increased by adding MgO or $CaCl_2$ to tallow or Ca soap tallow during incubation(P<0.05). In in vivo trial, thirty finishing Hanwoo(average BW 460kg) were divided into three groups based on fat sources and divalent ions, i.e. Control(EE 2.40), T-MgCa = control + tallow + $CaCl_2$ + MgO, T-CaS = control + Ca soap tallow (EE 5.30%). After feeding each diet for 80 days, average daily weight gain showed 0.89, 1.02, 1.17kg in diets 1, 2 and 3, respectively. The highest feed efficiency was observed(0.12) in diet 2 group, followed by diet 3 (0.10) and 1 groups(0.08; P < 0.05). In conclusion, the present results could be sununarized that the performance of Hanwoo bulls was improved by tallow with divalent ions without any negative effect on rumen fermentation.
This study was conducted to evaluate the effect of different processing of rice on rumen fermentation in in vitro and in situ experiments. Different processing treatments (extruding, roasting, and steaming) were used in this study and all treatments were ground through a cyclone mill (Foss, Hillerød, Denmark) fitted with a 1 mm screen. Non-treated rice was considered to a control substrate. Then, all treatments were used in in vitro and in situ experiments. Total gas production and dry matter digestibility in control were lower than any other treatment at all incubation times (P<0.01). The lowest ammonia nitrogen ($NH_3-N$) concentration was observed in control among treatments at 6, 12, and 24 h incubation (P<0.01). Extruding had a highest total volatile fatty acids (VFA) concentration at 6, 12 h incubation (P<0.01) and Steaming exhibited a highest total VFA at 24 h (P<0.01). The lowest total VFA concentration was observed in control at 6, 12, and 24 h (P<0.01). In an in situ, The highest value of soluble fraction, degradation rates, effective degradability was observed in extruding (P<0.01). It was considered that feed processing increased dry matter digestibility, total VFA concentration, and decreased pH as well as $NH_3-N$ concentration indicating that processing may increase nutrient degradation of rice in the rumen.
This study, consisting of two experiments, was conducted to determine the effects of feeding heat treated protein and mineral complex (HPM) on milk production and composition, and ruminal fermentation of Holstein dairy cows. In in vitro experiment, HPM levels were 0, 0.2, 1 and 2%, and Timothy hay, which was substrate, was milled as 1 mm size, and the effects of HPM on pH, ammonia and VFA were analyzed after incubation times of 0, 6, 12, 24 and 48 h, respectively. The pH and ammonia production were not significantly different between treatments during the incubation. In addition, generally, total VFA and individual VFA were not affected by HPM on 0, 6 and 24 h. While, total VFA and individual VFA were increased in 0.2% and 1% of HPM supplemented treatments, but decreased in 2% of HPM treatment compared with control on 12 h. On 48 h, total VFA and individual VFA were increased in HPM treatments compared to control (P<0.05). However, A/P ratio was not affected by HPM supplementation. Gas production was higher in HPM treatment compared to control on 24 h (P<0.05) and 48 h (P<0.05). In lactating experiment, fourteen lactating Holstein cows were used for 4 months in a cross over experimental design. There were two treatments; no added HPM as a control and 0.2% of HPM added as a test treatment. Daily milk yield (P<0.001), 4% FCM (P<0.001), milk protein (P<0.05) and SNF (solid not fat; P<0.05) were increased in HPM treatment compared to control. While, milk fat, MUN (milk urea nitrogen) and SCC (somatic cell count) were not significantly different between treatments.
