${\alpha}-Tocopherol$ as a Fluorescent Probe -< I > Its spectroscopic characterization and use as a probe of liposome- (형광성 탐색자(探索者)로서 ${\alpha}-Tocopherol$ -< I > 분자분광학적 특성과 Liposome Probe로서의 이용-)

  • ${\alpha}-Tocopherol$ was found to be a fluorescent probe in determination of the phasetransition temperature of liposome. Since this was a discovery of a new aspect of ${\alpha}-tocopherol$ as an important biochemical molecule, its molecular spectroscopic characterization was carried out in order to obtain some informations on its spectral and, structural properties in various media, anticipating that the compound may entertain a wide applications in biochemical systems as a spectroscopic probe. Two species of α${\alpha}-tocopherol$, monomer and dimer, were found to exist in organic media, especially in solvents of nonhydrogen bonding ability. Monomer with maximum UV-absorption around $(291{\sim}294nm)$ is highly fluorescent, while dimer which is formed by intermolecular hydrogen bonds and absorbes with spectral peak at 298nm is nonfluorescent. ${\alpha}-tocopherol$ incorporated to liposome exhibits emission property quite different from that in various organic media showing broad and red-shifted fluorescence excitation and emission spectra. This spectral abnormality is to be interpreted to arise from chromanolate-type ion, H-dissociated ${\alpha}-tocopherol$.

Sieving the Polymer Chains through Anodic Aluminum Oxide Membranes (Anodic Aluminum Oxide Membrane을 통한 고분자 사슬의 선택적 투과)

  • Techniques for selectively separating molecules of gas and liquid states using various separation membranes have been widely used in variety of applications such as chemical, biological, pharmaceutical, and petrochemical industries. As the nanochannel diameter, inter-channel distance and length of the nanochannel of the anodic aluminum oxide (AAO) membranes can be precisely controlled, various studies to effectively separate mixture of various molecules using AAO membrane have been widely carried out. In this study, we fabricated AAO membranes of cylindrical nanochannels of various diameter sizes and of through-hole structure, that is, nanochannels of which both ends of each nanochannel are open. Using those AAO membranes of through-hole nanochannel structure, we studied the selective permeation polymer chains dissolved in a solvent based on hydraulic volume of the polymer chains. We found a precise, quantitative relationship between the radius of gyration of polymer chains that permeated through nanochannels inside AAO membrane and the diameter of nanochannels. In addition, we demonstrate that the behavior of the polymer solution flowing through nanochannel of the AAO membrane can be successfully described with the Hagen-Poiseuille relationship. It is, therefore, possible to theoretically interpret the nanoflow of the solution flowing inside the cylindrical nanochannel.

Inhibition of Xanthine Oxidase by Seaweed Extracts (해조류 추출물의 Xanthine Oxidase 저해작용)

  • Inhibition of xanthine oxidase by seaweed extracts obtained from Undaria pinnatifida, Ecklonia stolonifera, Ecklonia cava, Laminaria japonica, Sargassum, Codiumfragile, Enteromorpha compressa and Porphyra tenera were investigated. Extracts of E. stolonifera and E. mua remarkably inhibited xanthine oxidase activity compared to those of other seaweed. The xanthine oxidase inhibitory activity of E. cava was higher than that of E. stolonifera. Diethyl ether extract from E. cava was more effective in the inhibition of xanthine oxidase than other solvent extracts. Two xanthine oxidase inhibitors(A-1 and A-2) from diethyl ether extract were isolated and purified by silica gel column chromatography, thin layer chromatography and high performance liquid chromatography. Xanthine oxidase inhibitory activities of these compounds were 27.8 and 48.1% per 0.4mg, respectively. The active compound A-2 had absorption peak at 420nm, 456nm and 467nm, which can be considered as siphonaxanthine.

Anticoagulant Activities of Brown Seaweed Extracts in Korea (국내산 주요 갈조류 추출물의 항혈액응고 활성)

  • Effects of extraction conditions and molecular fractionation on anticoagulant activities of major brown seaweeds in Korea were investigated. Hot water extracts of C. costata, U. pinnatifida (Sporophyte), L. japonica, K. crassifolia, E. stolonifera, E. bicyclis, S. horneri, and E. kurome increaced activated partial thromboplastin time (APTT) over 190 seconds, which may be related to intrinsic pathway of blood coagulation. Hot water extract of E. Kurome (EKJ) was further fractionated by ethanol precipitation. EKJ-eim, ethanol-insoluble material of EKJ, showed higher anticoagulant activity than EKJ. EKJ-eim was further fractioned with ultrafiltration. EKJ-eim 1, (over 100 kDa) fraction showed higher APTT activity than EKJ-eim. A EKJ-eim 1 was sulfated polysaccharide consisting of fucose, xylose, mannose, galactose, glucose and, sulfate at molar ratio of 1 : 0.05 : 0.10 : 0.15 : 0.17 : 1.46. The anticoagulant activity increased as sulfate content and molecular weight increased.

Synthesis and Properties of Semi-Flexible Aromatic Polyesters Containing Pentamethylene Group in Main Chain (주사슬에 펜타메틸렌기를 가지고 있는 반 유연성 방향족 폴리에스터의 합성 및 성질)

  • Semi flexible polyesters containing aromatic rings and pentamethylene groups in the main chain were synthesized by direct polycondensation reaction. The structures of these polymers were investigated by $^1H$-NMR and FT-IR and the phase transition behavior was characterized with DSC, TGA and crossed polarizing microscope. Inherent viscosities ($\eta_{inh}$) of polymers measured in phenol/p-chlorophenol/1,1,2,2-tetrachloroethane were between 0.46 and 1.30 dL/g. As increasing the linearlity of rigid moieties in polyster, melting transition temperatures ($T_m$) increased and solubilities in organic solvents decreased. P-H, P-mH and P-4H of the polymers formed turbid melts that showed stir-opalescence and nematic phase at the broad anisotropic region, However, P-R, P-C and P-2B did not exhibit any textures related to the liquid crystallinity.

  • In this study, the pretreatment of Helianthus tuberosus residue had been performed. The two-stage pretreatment on flow-through process were applied in the interests of increase of sugar production yield on enzymatic saccharification. The delignification by aqueous ammonia and the fractionation of hemicellulose by sulfuric acid solution as pretreatment solution were confirmed for effects of enzymatic saccharification. Two-stage pretreatment process was performed using aqueous ammonia and sulfuric acid. The first step was performed with aqueous ammonia for 40 min at $163.2^{\circ}C$ and the second step was performed with sulfuric acid solution for 20 min at $169.7^{\circ}C$. And then, the first step was performed with sulfuric acid solution and the second step was pretreated with aqueous ammonia. At this time, the glucose production was 30.7 g and the glucose yield was 72.4% in the first step process with aqueous ammonia. And, the glucose production was 20.9 g and the glucose yield was 49.3% in the first step process with sulfuric acid solution.

High $T_c$ Superconductors $LnBa_2Cu_3O_{7-\delta}$ from 2-Ethylhexanoate Precursors (2-Ethylhexanoate 화합물에서 제조한 고온초전도체 $LnBa_2Cu_3O_{7-\delta}$)

  • 2-Ethylhexanoate compounds which contained Y, Er, Yb, Ba, and Cu were prepared separately. These metallo-organic compounds were dissolved together in the common solvent and the solution was homogeneous. From these solution, high $T_c$ superconductors $YBa_2Cu_3O_{7-\delta}(Y_{123})$, $ErBa_2Cu_3O_{7-\delta}(Er_{123})$, and $YbBa_2Cu_3O_{7-\delta}(Yb_{123})$ were prepared. XRD analysis showed that these superconductors were not single phase and contained some impurity phases including $YBa_2Cu_4O_8(Y_{124})$. This was also confirmed from the magnetic susceptibility measurement. Generally, critical temperatures were higher than that of the superconductors prepared by ceramic method. Volume diamagnetic susceptibility showed little variation compared to the specimen made by ceramic method.

Effect of nitridation of sapphire in $NH_3$ ambient on GaN grown by MOCVD

  • Wide band gap을 갖는 III-V족 반도체인 GaN는 파란색에서 자외선영역에 이르는 발광소자용으로, 그리고 최근에는 전자소자로도 가장 유망한 반도체 중의 하나이다. 하지만 격자상수가 일치하는 적당한 기판이 존재하지 않아 성장된 GaN 박막 내에는 많은 결함들이 존재하게 된다. 일반적으로 가장 널리 쓰이는 기판은 사파이어 기판이 주로 이용되고 있는데 사파이어는 GaN와 격자상수 불일치가 16%에 이르므로 고품질의 GaN 박막을 성장시키기 위해서는 격자상수 불일치를 어느 정도 완화시키면서 초기성장과정의 컨트롤이 매우 중요하다. 이러한 방법들로는 GaN박막 성장 전에 사파이어 기판 질화처리를 하거나 buffer 층을 도입하는 것인데, 이에 관한 많은 연구들이 보고되고 있다. 하지만 각각 두 공정에 관한 연구는 많이 되어 있지만 두 공정사이를 연결해 주는 공정처리법에 관한 연구는 보고되고 있지 않다. 따라서 본 연구에서는 사파이어기판 질화처리를 한 후 buffer 층을 성장시키기 전까지 chamer 내부의 분위기 가스가 GaN 박막성장 거동에 어떤 영향을 주는지에 관해 연구하였다. 질화처리 후 chamber 내부의 분위기 가스가 GaN 박막 성장 거동에 미치는 영향을 연구하기 위하여 두 개의 시편 A,B를 준비하였다. 시편 A는 먼저 사파이어 기판을 유기용매를 이용하여 cleaning 한 후 장비에 장입되었다. 수소분위기하에서 10nsrks 104$0^{\circ}C$에서 가열한 후 30초간 암모니아 유속을 900sccm으로 유지하며 사파이어 기판 질화처리를 수행하였다. GaN buffer 층을 성장하기 위하여 104$0^{\circ}C$에서 56$0^{\circ}C$로 온도를 내리는 과정중 질화처리를 위하여 흘려주었던 암모니아 유속을 차단한 채 수소분위기에서만 온도를 내렸다. 56$0^{\circ}C$에서 GaN buffer 층을 300 성장시킨 후 102$0^{\circ}C$의 고온에서 2$\mu\textrm{m}$ 두께로 GaN 박막을 성장하였다. 시편 B는 질화처리 후 단계부터 GaN 박막성장 단계에 이르기까지 AFM을 이용하여 두 시편의 성장거동을 비교 분석하였다. 두 시편의 표면을 관찰한 결과 시편 A는 2차원적 성장을 하며 매우 매끄러운 표면을 갖는데 반해, 시편 B는 3차원적 성장을 하며 매우 거친 표면을 보였다. 또한 두 시편 A, B를 XRD, PL, Hal 측정으로 분석한 결과 시편 A가 시편 B보다 우수한 구조적, 광학적, 전기적 특성을 보였다.

Solvent Resistance and Gas Permeation Property of PEI-PDMS Hollow Fiber Composite Membrane for Separation and Recovery of VOCs (VOCs의 분리 및 회수를 위한 PEI-PDMS 중공사 복합막의 내용매성 및 기체 투과 특성)

  • To separate and recover of VOCs, supporting membranes using PEI were prepared by phase separation method and it was coated with PDMS to prepare PEI-PDMS hollow composite membrane. To investigate characteristic of prepared membrane, pure gas permeability was measured using oxygen and nitrogen, the stage cut and permeance property with feed concentration were evaluated using xylene, ethyl benzene, toluene and cyclohexane. Also, to check solvent resistance on VOCs, stress-strain property of membrane with immersion time in solvent were measured by DMA. The permeance value of $O_2$ and $N_2$ showed 63 GPU and 30 GPU respectively. Permeated VOCs concentration was decreased with increasing stage cut. But, conversely, recovery efficiency that was increased with increasing stage cut. As a result of DMA test, the stress and strain were 11.93 MPa and 13.52%, respectively.

Comparison of Radical Scavenging and Immunomodulatory Activities Exhibited by an Aqueous Extract of Diospyros kaki Thunb. Fruit (Persimmon) (청도반시 추출물에 의한 라디칼소거 활성과 면역조절 활성의 상호 비교)

  • To assess the relationship between anti-oxidant and immunomodulatory activities of Diospyros kaki Thunb. fruit, we harvested persimmons on the first Friday of every month from July to October, and conducted a DPPH assay, a T-bet promoter assay, and an ELISA for IL-4 determination. Anti-oxidant activity increased as fruit weight rose. When the fruit was extracted with five different solvents, and the extracts examined for radical scavenging activity, such activity exhibited a pattern similar to that seen when anti-oxidant activity was assessed. T-bet promoter activity decreased on ripening, whereas IL-4 expression increased, as tested in a DNFB-induced animal model. Collectively, the results suggest that inflammation-inhibitory activity, valuable in treatment of some immune diseases, rises as persimmons ripen.
