Structure Determination of Macrolactin Compounds with Antibacterial Activities Isolated from Bacillus polyfermenticus KJS-2 (Bacillus polyfermenticus KJS-2에서 분리된 항생물질 마크로락틴류의 구조결정)

  • In this study, we isolated five macrolactin compounds from a fermentation broth of Bacillus polyfermenticus KJS-2. The macrolactin compounds were structurally identified as macrolactin A (MA), 7-O-malonyl macrolactin A (MMA), 7-O-succinyl macrolactin A (SMA), macrolactin E (ME) and macrolactin F (MF) via a variety of NMR techniques, COZY, DEPT, HMQC and HMBC, and mass and specific rotation assays. The three macrolactin compounds, MA, MMA and SMA, profoundly inhibited the growth of both vancomycin-resistant Enterococci (VREs) and methicillin-resistant Staphylococcus aureus (MRSA), the inhibition of which were estimated via a disc agar diffusion bioassay. MA, MMA, and SMA exhibited antibacterial activities superior to those of vancomycin and teicoplanin.

Biocontrol of Leaf Mustard Powdery Mildew Caused by Erysiphe cruciferarm using Bacillus velezensis YP2 (Bacillus velezensis YP2의 겨자채 흰가루병의 생물적 방제)

  • Bacillus velezensis YP2 inhibited the mycelial growth of several plant pathogens including Cercespora spp., Septoria sp., Phoma sp., Botrytis cinerea and Sclerotinia scleotiorum occurring in leafy vegetables. Control efficacy for powdery mildew caused by Erysiphe cruciferarm on red leaf mustard and cheong mustard by treatment of spraying with 10-fold diluted Luria-Bertani (LB) broth of B. velezensis YP2 was 91.8% and 80.9%, respectively. When B. velezensis YP2 was treated four times with five-day interval, three times at seven-day interval and two times at ten day interval in the greenhouse test, the control effect of red leaf mustard powdery mildew was 70.6%, 65.0% and 40.9%, respectively. Also B. velezensis YP2 could promote the seed germination and plant growth of led leaf mustard. The results showed that the culture broth of B. velezensis YP2 was very effective to control the powdery mildew of leaf mustard.

Analysis of Microbial Contamination in Commercial Saengshik Products (유통 생식제품의 미생물 오염 분석)

  • This study was performed to assess the presence of contaminated microorganisms of Escherichia coli, Clostridium perfringens, and Bacillus cereus in the 112 commercial Saengshik products. E. coli was not detected in all the samples, but C. perfringens was detected in 11 products (9.8%). The number of the bacteria was less than 100 CFU/g, which was satisfactory to KFDA microbiological requirement. B. cereus was detected less than $10^2{\sim}10^3$ CFU/g in 7 products and $10^3{\sim}10^4$CFU/g in 13 products out of 25 products. Those detected bacteria from tryptose sulphite cycloserine agar and mannitol egg yolk polymyxin agar showed the typical characteristics of Gram positive and contained lecithinase, which can decompose egg-yolks layers in the biochemical test. Therefore, much more attention must be applied to satisfy the B. cereus requirement for Saengshik products sold in the market.

Removal Characteristics of Total Coliforms in a Rotating Activated Bacillus Contactor Process (회전식 부착 바실러스를 이용한 하수고도처리 공정에서의 총대장균군 제거 특성)

  • This study was performed to examine the disinfection capability of a Rotating Activated Bacillus Contactor (RABC) system, in which the predominant species, Bacillus sp. was expected to have a removal or inactivation effect of total coliforms. In a settling test with mixtures of E. coli and Bacillus sp., a high removal of E. coli was observed at $20{\sim}40^{\circ}C$, while insignificant removal at $10^{\circ}C$. In a batch test, a 4.5% addition of Bacillus sp. to activated sludge considerably enhanced the removal effect of total coliforms, indicating Bacillus sp. played an important role in improving the settlability of the sludge and coliforms. In a pilot scale RABC system, the concentration of total colifroms reduced remarkably in the settling tank, suggesting that total coliforms in the RABC process were eliminated through coagulation and precipitation, probably due to extracellular polymeric substance (EPS) of Bacillus sp. The fraction of Bacillus sp. in the total cell count in the RABC process was in the range of 4.5%~6.3%. The majority (75%) of the Bacillus sp. in the RABC process was Bacillus subtilis which is known to enhance coagulation and precipitation by producing EPS. Hence, an adoption of a RABC process might be able to eliminate the disinfection unit process from a wastewater treatment system.

Degrading and Flocculating Property of A Bacterium Isolated from the Extract of Earthworm (지렁이로부터 분리한 Bacillus pumilus JS-01 균주의 유기물 분해능 및 응집능)

  • To develop the microbial agents for the environmentally-friendly treatment and recycling of food waste, useful microorganisms, which showed higher degradation activities to various organic compounds and possessed flocculating activities, were isolated from the earthworm. One of the isolated strains, named JS-01, was further selected due to its higher flocculating activity against 0.5% Kaolin clay. JS-01 was identified as Bacillus pumilus sp. by 16s rDNA analysis. The optimal temperature and pH for the growth of JS-01 to express the flocculating activity was found to be at $37^{\circ}C$ in pH 7.0 of EPS broth medium. JS-01 also expressed good degrading activity against cellulose, which is one of the representative organic materials in food waste. We propose that JS-01 will be a good candidate for the efficient treatment of food waste and leachate due to the property to degrade cellulose and flocculating activity.

Analysis of Microbiological Contamination in Ready-to-eat Foods (즉석 섭취 식품에 대한 미생물 오염 분석)

  • This study was carried out to examine microbiological contamination of ready-to-eat foods (kimbab, sushi, salad, sandwich, sashimi) and to prove hazard of ready-to-eat foods by microbiological analysis. Collection of 440 samples of ready- to-eat foods were obtained from department stores, discount stores, super-markets in Seoul, Gyeonggi, Chungcheong, Gyeongsang, Honam areas. Sushi showed the highest detection rate of micro organism with value of20.4%, and then detection rates of kimbab, sashimi and salad were 13.0%, 12.5% and 6.9%, respectively. S. aureus was the most detected microorganism with value of 6.6%, and then Coliform and L. monocytogenes were detected 2.7% and 2.3%.

Purification and risk assessment of Bacillus thuringiensis Vip3Aa protein against Apis mellifera (Bacillus thuringiensis 유래 Vip3Aa 단백질 순수분리 및 꿀벌 (Apis mellifera)에 대한 위해성평가)

  • Most insect-resistant LMOs have been produced by applying Cry and Vip3Aa proteins. Vip3Aa protein is activated during the vegetative stage of Bacillus thuringensis (Bt) and the inhibitory activity of the Vip3Aa protein against pathogenic attacks from lepidopteran insect species is well known. However, a risk assessment of the Vip3Aa protein compared to the Cry protein has not been conducted in South Korea. This study demonstrates a possible risk assessment method for Vip3Aa protein against honeybees (Apis mellifera). For the risk assessment of the protein, we purified the recombinant Vip3Aa protein in Escherichia coli. The survival rate and symptoms of general intoxication of 4 months honeybees were measured after Vip3Aa exposure. These results indicated that there was no significant difference in the survival rate and the symptom between Vip3Aa and the control buffer. In this study, we established standard methods of Vip3Aa protein purification and oral adult toxicity test using A. mellifera as an LMO risk assessment technique for preserving the natural ecosystem of South Korea.

The Effect of Bacillus-Fermented Scutellariae Radix Acupuncture Solution on Chemokine and Growth Factor Production in Mouse Macrophage Stimulated by Lipopolysaccharide (바실러스균 발효황금약침액이 마우스 대식세포의 케모카인 및 생성인자 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of Bacillus-fermented Scutellariae Radix acupuncture solution (SB) on chemokine and growth factor production in RAW 264.7 mouse macrophages stimulated by lipopolysaccharide (LPS). Methods : Productions of chemokine and growth factor were measured by High-throughput Multiplex Bead based Assay with Bio-plex Suspension Array System based on xMAP$^{(R)}$ technology. Firstly, cell culture supernatant was obtained after treatment with LPS (1 ${\mu}g$/mL) and SB for 24 hours. Then, it was incubated with the antibody-conjugated beads for 30 minutes. Detection antibody was then added and incubated for 30 minutes. After incubated for 30 minutes, strepavidin-conjugated phycoerythrin (SAPE) was added. After another 30 minutes incubation, the level of SAPE fluorescence was analyzed in Bio-plex Suspension Array System. Results : The results of the experiment are as follows. 1. SB significantly inhibited the LPS-induced production of vascular endothelial growth factor (VEGF), interferon-inducible protein (IP)-10, and granulocyte-colony stimulating factor (G-CSF) at the concentration of 25, 50, 100, and 200 ${\mu}g$/mL in RAW 264.7 cells (P < 0.05). 2. SB significantly inhibited the LPS-induced production of Eotaxin at the concentration of 25, 100, and 200 ${\mu}g$/mL in RAW 264.7 cells (P < 0.05). 3. SB significantly inhibited the LPS-induced production of MIP-$1\alpha$ at the concentration of 25 and 100 ${\mu}g$/mL in RAW 264.7 cells (P < 0.05). Conclusions : These results suggest that SB has immuno-modulatory property related with its inhibition of VEGF, IP-10, G-CSF, and Eotaxin production in macrophages.

Shipboard sewage treatment using Membrane Sequence Batch Reactor (MSBR을 이용한 크루즈선 오·폐수 처리 장치)

  • Lab scale experiment study was carried out for biological process development on cruise. SBR(Sequence Batch Reactor), MBR(Membrane Bioreactor), and MSBR(Membrane Sequence Batch Reactor) system were investigated for practical application on shipboard sewage treatment. From the results it was suggested that MSBR system might be suitable process for cruise in terms of pollutant removal efficiency, maintenance and special environmental conditions of cruise. About 99% of BOD, 98% of COD and 99% of SS were removed in MSBR system. In addition, about 76% of total nitrogen was reduced and the total phosphorus reduction averaged 59%.

Isolation and Characterization of a Novel Bacterium, Bacillus subtilis HR-1019, with Insoluble Phosphates Solubilizing Activity (인산가용화 활성을 갖는 바실러스 서브틸리스 HR-1019 분리와 특성)

  • The objective of this study was to develop a mineral phosphate-solubilizing bacterium as a biofertilizer. A mineral phosphate-solubilizing bacterium HR-1019 was isolated from cultivated soils. It was identified as Bacillus subtilis by 16S rDNA analysis. The phosphate-solubilizing activities of the HR-1019 strain against three types of insoluble phosphate, hydroxyapatite, tri-calcium phosphate, and aluminum phosphate were quantitatively determined. When 5% of glucose concentration was used as a carbon source, the strain showed marked mineral phosphate-solubilizing activity. Mineral phosphate solubilization was directly related to pH drop in the culture solution of the strain. The pathogenic activity and antifungal effects of the HR-1019 strain were measured inclear zones formed in PDA media.
