Induction of ${\beta}$-carotene by Ozone and Hydrogen Peroxide and Extraction Using Vegetable Oil from Microalga Dunaliella bardawil (미세조류 Dunaliella bardawil에서 오존과 과산화수소에 의한 ${\beta}$-carotene의 축적과 식용기름을 이용한 추출)

  • Halotolerant microalga Dunaliella bardawil was reported to massively accumulate the ${\beta}$-carotene, which protects cells from excess light intensity. Maximum specific growth rate of 0.168/hr was achieved when cells were cultivated at 1 N NaCl, pH 8.0, light intensity 80 ${\mu}E/m^{2}/s$, agitation 70rpm. For the effectiv accumulation of ${\beta}$-carotene, ozone ro hydrogen peroxide was added to media which was irradiated with white fuorescent lamps with moderate light intensity of 250 ${\mu}E/m^{2}/s$. As a result, maximum volumetric content of ${\beta}$-carotene was 324 ${\mu}$g/㎖. The ${\beta}$-carotene extraction efficiency of vegetable oils was in the order of olive oil, sesame oil, rice brain oil, corn oil, and soy bean oil. Sonication and warming was effective in ${\mu}$-carotene extraction and finally 96.9% of ${\beta}$ could be extracted using olive oil.

ENZYMATIC STUDIES ON VITAMIN B6 METABOLISM

  • Vitamin B6(pyridoxine, pyridoxamine. and pyridoxal) is a dietary requirement in relatively small quantities for growth, health, and function in animals and fish. The metabolically active B6 is pyridoxal-5-phosphate(PLP). It does function as a coenzyme in number of enzymes(PLP-dependent enzymes) in which amino acids are metabolized, including decarboxylases, aminotransferases, sulfhydrases, tryptophanase, and hydroxylases. Vitamin B6 requirement is higher for fish because fish are fed much higher protein diet than land animals. B6 is also involved in metabolism of carbohydrates and lipids and essential for the synthesis of heme and serotonin. Deficiency signs in fish develop quickly, in cluding nervous disorders, convulsions, poor swimming coordination, skin lesions, edema, exophthalmos, and tetany. The conversion of vitamin B6 to metabolically active form(PLP) is catalyzed by pyridoxal kinase and pridoxine(pyridoxamine) oxidase. In this review, we summarized in detail the enzymatic studies on vitamin B6 metabolism and about the mechanisms and properties of a PLP-dependent enzyme.

Effects of Rearing Condition and Species of Microalgae on Growth and Survival of Larvae of the Sunray Surf Clam, Mactra chinensis (사육환경과 먹이종류에 따른 개량조개, Mactra chinensis 유생의 성장과 생존)

  • Rearing condition and species of microalgae on growth and survival of the sunray surf clam, Mactra chinensis larvae were investigated for artificial seedling production. The larvae of M. chinensis on higher temperature showed high growth and low survival and was grown over $230{\mu}m$ in shell length 14 days after hatching, but low growth as $151.1{\mu}m$ at $18^{\circ}C$. The larvae of M. chinensis on salinity showed highest daily growth and survival as $11.3{\mu}m$ and 65.8% at 30 psu, respectively. The optimum of water temperature and salinity for the larval rearing were $23^{\circ}C$ and 30-35 psu over 25 psu at least. The density of larval rearing was below 10 per 1 ml in rearing seawater for elevating the development rate from D-shaped to settled (metamorphosing) stage. The larvae fed the mixed diet of Isochrysis galbana, Pavlova lutheri, Isochrysis sp. (green), Chlorella ellipsoidea showed highest growth and survival. The larvae fed the single diet of I. galbana, Isochrysis sp. (green) showed high growth and survival. But the larvae fed the single diet of P. lutheri and C. ellipsoidea showed low growth and survival. The optimum diet of larvae of M. chinensis was over two species of microalgae included I. galbana, Isochrysis sp. (green) for elevating the high growth and survival.

Seasonal Variation of Microalgae in the Surface Water of Marian Cove, King George Island, the Antarctic 1998/1999 (1998/1999 남극 킹조지섬 마리안소만 표층수에 서식하는 미세조류의 계절적 변동)

  • We investigated seasonal variation of microalgal assemblages, sea water temperature, salinity and suspended solid and the parameters measured daily from January 1998 to October 1999 at a nearshore shallow-water in Marian Cove, Maxwell Bay, King George Island, the Antarctic. Annual mean surface water temperature was -0.3$0^{\circ}C$ and the highest water temperature was 4.53$^{\circ}C$ (22 January 1999) and the lowest water temperature was -2.07$^{\circ}C$ (23 August 1998). Annual mean salinity was 33.38 psu, ranging from 42.80 psu (6 January 1999) to 19.50 psu (6 June 1999). Annual mean suspended solid (SS) during two years was 34.14 mgㆍ1$^{-1}$, ranging from 60.62 mgㆍ1$^{-1}$(7 March 1998) to 12.90 mgㆍ1$^{-1}$ (26 December 1998). Chlorophyll $\alpha$ (Chl $\alpha$) concentrations were measured in order to know seasonal variations of microalgae in the surface seawater. Annual mean of total Chl a concentration was 0.55$\mu\textrm{g}$ㆍ1$^{-1}$, the highest Chl $\alpha$ concentration (12.16$\mu\textrm{g}$ㆍ1$^{-1}$) appeared in 4 October 1998, the lowest Chl $\alpha$ concentration appeared 0.19$\mu\textrm{g}$ㆍ1$^{-1}$, Monthly mean total Chl $\alpha$ concentration was high in October 1998 (1.32$\mu\textrm{g}$ㆍ1$^{-1}$) and low in July on 1998 (0.28$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean nano-sized Chl $\alpha$ concentration was 0.40$\mu\textrm{g}$ㆍ1$^{-1}$, monthly mean nano -sized Chl $\alpha$ concentration was high in November 1998 (0.90$\mu\textrm{g}$ㆍ1$^{-1}$), and low in July 1999 (0.22$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean micro-sized Chl $\alpha$ concentration was 0.15$\mu\textrm{g}$ㆍ1$^{-1}$ monthly mean micro-sized Chl $\alpha$ concentration was high in October 1998 (0.81$\mu\textrm{g}$ㆍ1$^{-1}$), and low July 1998, January, February and September 1999 (0.05$\mu\textrm{g}$ㆍ1$^{-1}$). More than 65% of total Chl $\alpha$ was concentrated during spring and summer time between October and March. Microalgal variation appeared to be due to physical factors of seawater in the Antarctic nearshore from 1998 to 1999. The reason why micro-sized Chl $\alpha$ did not increase during austral summer was the bay had been frozen by decrease of water temperature. We think that total microalgal abundance was decreased because the summer microalgal abundance was determined by variation of water temperature during winter season. Application of Lignocellulosic and Macro-algae Hydrolysates for Culture of Chlorella saccharophila (Chlorella saccharophila 배양을 위한 목질계 및 해조류 바이오매스 가수분해물의 이용)

  • In this study, we investigated the possibility of using hydrolysates of lignocellulosics (rapeseed straw, barley straw, rice straw) and marine macro-algae (Undaria pinnatifida, Laminaria japonica, Enteromorpha intestinalis, and Gracilaria verrucosa) to cultivate Chlorella saccharophila. The growth of C. saccharophila was inhibited by 7 hydrolysates without active carbon treatment. In contrast, hydrolysates treated with active carbon increased the cell growth and product (oil and chlorophyll) formation by C. saccharophila. The oil contents of C. saccharophila treated with each hydrolysate were $41.26{\pm}0.69%$ (glucose), $22.06{\pm}1.21%$ (rapeseed straw), $28.65{\pm}1.08%$ (barley straw), $31.15{\pm}0.76%$ (rice straw), $31.50{\pm}2.12%$ (U. pinnatifida), $31.49{\pm}4.53%$ (L. japonica), $29.63{\pm}3.93%$ (E. intestinalis), and $26.15{\pm}1.99%$ (G. verrucosa), respectively. Lignocellulosics and marine macro-algae may be useful resources for improving the mass cultivation of C. saccharophila.

  • This study was conducted to evaluate the effects of microbial inoculation on sewage sludge composting. The number and species of microorganisms in sewage sludge sampled on February were higher than those sampled on August. The composting of sewage sludge is inhibited by the polyacrylamide cation, which was used as a coagulant and known to repress the growth of microorganisms. The growth of all microorganisms was inhibited by the addition of the polyacrylamide cation at a concentration of more than 0.8%. The species and viable counts of microorganisms were observed to increase during composting sewage sludge by inoculation of the effective microorganisms and addition of the pine tree sawdust as a bulking agent, compared with those without inoculation. A variety of organisms in compost(sewage sludge plus sawdust) were observed after composting for 30 days, such as Fragilaria sp., Proales sp., Vorticella sp., Schizothrix sp., Anabaena sp., Zoothaminium sp., Epstylis sp., Arcella sp., Balantidium sp., Actinophrys sp., Synedra sp., Euglypha sp., Ulothrix sp., Anacystis sp., and Clostium sp.

  • Mycosporine-like amino acids (MAAs) are UV-absorbing substances, and diverse marine organisms have the evolved the capacity to diminished the direct and indirect damaging effects of environmental ultraviolet radiation by synthesis and accumulation of MAAs. In this study, we manufactured a radiofrequency (RF) generation device and applied to microalgal culture. $0.35{\pm}0.05$ mHz of RF was supplied to culture vessel for Chlamydomonas sp. and samples were harvested at the designated time intervals (1, 0.5, 1 and 2 hr). MAAs biosynthetic genes, dehydroquinate synthase homolog (DHQS-like) and nonribosomal peptide synthetase homolog (NRPS-like), were cloned from Chlamydomonas sp. and their gene expressions under the RF exposure were analyzed using qRT-PCR. DHQS-like and NRPS-like gene expressions of Chlamydomonas sp. exposed to RF were increased 1.46 and 1.19 fold at 1 hr, respectively. These results means that DHQS-like and NRPS-like genes can be good biomarker candidates for diagnosis of MAAs biosynthesis in the Chlamydomonas sp.

Lipid Extraction from Spirulina platensis using Supercritical Carbon Dioxide and Analysis of Fatty Acid Compositions in Extracts (초임계 이산화탄소를 이용한 Spirulina platensis로부터 지질추출 및 지방산 조성 분석)

  • This study was performed to obtain information about the extraction conditions of lipids from microalgae, Spirulina platensis, using supercritical fluid $CO_2$. Regardless of extraction temperature conditions, the extracted lipid contents increased as pressure increased, but decreased at 8500 psi on each temperature. The highest yield of extracted lipid content showed in the condition of 5500psi at $50^{\circ}C$, and extracted lipid content was about $20\%$. In same pressure the contents of C18 : 2 and C20 : 0 increased as temperature increased, but fatty acids composition were $60\~75\%$ saturated, $12\~20\%$ monounsaturated and $13\~31\%$ polyunsaturated regardless of extraction conditions. The C18 : 3 was only detected in the condition of 5500psi at $50^{\circ}C$ but the content was very little.

Seasonal Variation in Species Composition and Biomass of Microphytobenthos at Jinsanri, Taean, Korea (한국 태안군 진산리 저서미세조류의 계절별 종조성 및 생물량 변화)

  • Seasonal species composition and biomass of microphytobenthos were examined at the intertidal flats of Jinsanri, Taean, Korea, from April to December 2011. In total, 121 species were identified, including 109 diatoms, 5 blue-green algae, and 5 dinoflagellates. Seasonal variation in species number ranged from 49 to 56 species (minimum in October and maximal in April), while vertical variation across three different shore levels ranged from 64 to 77 species with maximum at high and minimum at low shore. Species diversity ranged from 3.22 to 3.42 seasonally and from 3.50 to 3.17 across vertical shore levels. Seasonal average values of the normalized difference vegetation index and the vegetation index were 0.06 and 1.14, respectively. Chlorophyll a concentrations were $66.00-120.73mg/m^2$, with a seasonal minimum and maximum in July and October, respectively, and a vertical shore maximum of $137.35mg/m^2$ at mid shore. Phaeopigment concentrations peaked in October ($50.78mg/m^2$) and at high shore ($61.58mg/m^2$) and were lowest in December ($22.53mg/m^2$) and at low shore ($15.28mg/m^2$). The number of microphytobenthos ranged from 78,556 to 287,898 cells/$cm^2$, reaching a maximum in April and a minimum in July. The diatoms Paralia sulcata, Navicula sp., and Delphines surirella were dominant at all tidal levels over the study period. Dinoflagellates were dominant in July at low shore, and blue-green algae were abundant between July and October at mid shore. In conclusion, species composition and primary production of microphytobenthos exhibited clear seasonal and vertical patterns.

Preparing Bi-component Dye of Unripe Diospyros kaki THUNB. Fruit and Ecklonia cava and Investigating Its Dyeing Propeties on Fabric (풋감과 감태의 이성분 복합염료 제조와 섬유 염색성 고찰)

  • This paper proposes a bi-component dye, including the unripe fruit of Diospyros kaki THUNB and Ecklonia cava, to substitute for traditional persimmon dyeing because fabrics dyed with persimmon juice become stiffer and natural persimmon is insufficient for dyeing. This study examined the color difference and fabric stiffness depending on the ratio of Ecklonia cava for in a one-bath dye solution with Diospyros kaki THUNB and showed that 6% of Ecklonia cava in the bi-component dye was the optimum for decreasing the fabric stiffness. Based on these results, a bi-component dye constituting of 94% Diospyros kaki THUNB and 6% Ecklonia cava was prepared. The particle size was found to be smaller than both single dyes and it maintained a similar amount of Catechin to Diospyros kaki THUNB dye. Finally, cotton fabric dyed with a bi-component dye was much improved in terms of the fabric hand and the surface color was similar to that of the traditional persimmon-dyed fabric. These results could help to develop the natural persimmon dyeing industry.
