Application of Silver Ion for Clinical Mastitis in Holstein Cows (임상형 유방염을 가진 홀스테인(Holstein) 소에 대한 은 이온의 적용 연구)

  • Mastitis is an inflammation of the mammary gland that develops as a response to pathogenic microorganisms. Somatic cell count (SCC) and bacteriological analysis are the most accepted tools to determine udder health. This study evaluated the effects of intra-mammary gland infusion of silver ion against clinical mastitis in Holstein cows. Silver ion (20 ${\mu}g$/ml) was infused through the intra-mammary gland in quarters having clinical mastitis and milk was collected to determine SCC, and levels of lactoferrin and bovine serum albumin (BSA). Silver ion infusion decreased udder inflammation, firmness and swelling, and reduced clots, BSA, lactoferrin and SCC in milk. However, milk yield and circulating blood cells remained unchanged. The silver ion-mediated reductions of BSA and SCC indicate reduced inflammation and bacterial activity in silver ion-treated mammary glands in Holsteins with mastitis, which may be exploited in a curative strategy.

A Study on Characteristics of Natural Organic Matter using XAD and FTIR in Yeongsan River System (XAD 및 FT-IR을 이용한 영산강수계 광주시 유역 자연유기물질의 분포특성 연구)

  • This study investigated the characteristics of natural organic matter(NOM) with tXAD resin and FT-IR in the Yeongsan river system of Gwangju region. NOM fractionation by XAD 8/4 resins was used to classify hydrophobic and hydrophilic substances. FTIR was applied to classify functional groups in the structure of NOM. In the XAD investigation, most of the four site-samples were mainly hydrophilic substances. In March, hydrophobic substances were dominant in the Gwangju 1 site (GJ-1), while hydrophilic substances were dominant for the other sites. In May, samples of all four sites were hydrophilic with a vigorous activity of microorganism due to increasing temperatures. The October results were very similar with those from March. In the FT-IR investigation, most of the broad and large peaks were assigned to the aliphatic group, particularly the OH group, C-H, $C-H_2$, $C-H_3$, and C-O alcohol group. All were related to hydrophilic substances. Other peaks showed the aromatic group, particularly the C=O (Ketone) Group. As a result, there is an identification of NOM in the Yeongsan river system composing mainly of hydrophilic substances and functional groups (OH, C-H etc.) of the aliphatic compound.

Safety Evaluation of Korean Mistletoe Extract (한국산 겨우살이 추출물의 안전성 평가)

  • Mistlero C was shown to be non-genotoxic in a series of genotoxicity tests, including a bacterial reverse mutation test and a combined in vivo mammalian erythrocyte micronucleus test. In a bacterial reverse mutation assay, no significant increases in the number of revertant colonies, compared to the negative control, was detected in $5,000{\mu}g/plate$ of Mistlero C. In addition, with Mistlero C, no changes were shown in the number of micronucleated polychromatic erythrocytes (MNPCE) among 2,000 polychromatic erythrocytes compared to the negative control. Mistlero C was administered orally in rats to investigate acute toxicity. The $LD_{50}$ values in rats were above 2,000 mg/kg. In a repeated dose, 13-week, oral toxicity study conducted in rats, no compound-related adverse effects were shown at doses of Mistlero C of up to 1,000 mg/kg body weight/day. The results of these studies support the safe use of Mistlero C in food for human consumption.

Changes of Quality Properties of Pork Loaves with Chemical-free Sweet Persimmon Powder during Chilled Storage at 5℃ (무 농약 단감분말로 제조한 미트로프의 5℃ 냉장 저장 중 품질 변화)

  • Four different pork loaves were made with two levels, HP/FP-3% and HP/FP-6%, containing 3.0 and 6.0% HP(hot air dried sweet persimmon powder)/FP(freezer dried sweet persimmon powder), respectively. HP and FP(3% and 6%) were added into ground pork loin separately. A control without persimmon powder were used for comparison. Patties were cooked to 74℃ in electric oven, after cooling, the aerobic packaged meat loaves were stored at 5℃. The moisture and crude ash were higher in the FP-6%, and the crude protein and crude fat were no different among the samples. The pH in control sample was significantly higher(p<0.05) than those of treatment group, however no significant difference was found among the 4 treatments. The TBARS(thiobarbituric reactive substance) values of all samples showed a tendency of increasing value along the storage days. The TBARS and a* value increased(p<0.05) as the addition ratio of HP/FP increased, respectively. With regard to microorganisms, the number of total aerobic bacteria were lower than 3.11 log10 CFU/g. In sensory evaluation, meat loaves containing persimmon powder resulted in a high overall acceptability, although they were not significantly different in overall acceptability.

Clinical Laboratory Aspect of Carbapenem-Resistant Enterobacteriaceae (카바페넴내성장내세균속균종의 임상검사 측면)

  • The correct distinction of carbapenem-resistant Enterobacteriaceae (CRE) and ccarbapenemase producing Enterobacteriaceae (CPE) and the rapid detection of CPE are important for instituting the correct treatment and management of clinical infections. Screening protocols are mainly based on cultures of rectal swab specimens on selective media followed by phenotypic tests to confirm a carbapenem-hydrolyzing activity, the rapid carbapenem inactivation method, lateral flow immunoassay, the matrix-assisted laser desorption ionization-time-of-flight test and molecular methods. The CPE is accurate for detection, and is essential for the clinical treatment and prevention of infections. A variety of phenotypic methods and gene-based methods are available for the rapid detection of carbapenemases, and these are expected to be routinely used in clinical microbiology laboratories. Therefore, to control the spread of carbapenemase, many laboratories around the world will need to use reliable, fast, high efficiency, simple and low cost methods. Optimal effects in patient applications would require rapid testing of CRE to provide reproducible support for antimicrobial management interventions or the treatment by various types of clinicians. For the optimal test method, it is necessary to combine complementary test methods to discriminate between various resistant bacterial species and to discover the genetic diversity of various types of carbapenemase for arriving at the best infection control strategy.

Cleaning Effects of Environmental-Friendly Washing Detergent on Milking Installation in Dairy Farms (젖소농가의 착유시설에 따른 친환경세제의 세척에 미치는 영향)

  • This study was conducted to determine cleaning effects of environmental-friendly washing detergent on milking installation in dairy farms. Milking machine, cooling machine and milking liner were washed by alkaline detergent and acid detergent for first three weeks and alkaline detergent and environmental~friendly detergent for next three weeks and the results get through microorganism and physico-chemistry analysis at the five district dairy farms different from environment. E-coli, coliform, fecal coliform, staphylococcus aureus, fecal streptococcus, prsudomonas aeruginosa and yersinia were not found in all of the dairy farms, and total colony counts were no difference compared with chemical detergent. The water for washing exceeded the determining acceptable level of nitrate nitrogen in the Anseong, Onyang and Cheonan and the remaining substance of washed water was also high levels. The cause of the result was that the level of nitrate nitrogen of water is basically high. Therefore, this study indicated that washing effect is no differences between environmental-friendly detergent and chemical detergent by washing for milking machine and cooling machine. Above all, producing high quality milk is to manage the water quality as well as using detergent.

The Effect of Portulaca oleracea on the Pathogens of Gastroenteritis in Infants (쇠비름 추출물이 신생아 장염 유발 미생물에 미치는 영향)

  • Diarrheal diseases constitute one of the major causes of morbidity and mortality in infants and young children globally. One of the main microorganisms causing diarrheal diseases is Campylobacter jejuni. For treatment of these diseases, Portulaca oleracea has been widely used as a folk remedy for a long time. This study was performed to investigate the antimicrobial activity of P. oleracea against gastroenteritis pathogens including C. jejuni. P. oleracea was extracted with petroleum ether, chloroform, ethylacetate, methanol, and hot water. The antimicrobial activity of the P. oleracea extracts was determined using the paper disc method, minimum inhibitory concentration, and the liquid culture method. The 10 $mg/m{\ell}$ ethylacetate extract showed the strongest antimicrobial activity against Salmonella typhimurium, Salmonella enteriditis, and Shigella spp.. The hot water extract from P. oleracea showed the highest anti-microbial activity against C. jejuni at 10~20 $mg/m{\ell}$. The hot water extract of P. oleracea retarded the growth of C. jejuni for 36 hr at $42^{\circ}C$.

Improved Quality Properties of Low-Fat Meat Patties Containing Sweet Persimmon Powder during Freeze Storage (단감분말을 함유한 저지방 돈육 미트패티의 냉동 저장 중 품질 변화)

  • Pork patties were made containing hot air dried sweet persimmon powder (T1: 3%, T2: 6%) and freezer dried sweet persimmon powder (T3: 3%, T4: 6%). The control (C: no addition) and each treatment were stored for 40 days at $-18^{\circ}C$. The pH value decreased (p<0.05) in all products over 30 days of storage but slightly increased thereafter. There was no significant difference in WHC between the control and treatment groups. Cooking loss and meat color (CIE L, a, b) increased significantly (p<0.05) as storage increased. The cooking loss of meat patties containing sweet persimmon powder were significantly (p<0.05) higher than the control. The L value of the control increased significantly (p<0.05) relative to the treatment groups, however the a and b values were slightly higher in meat patties containing sweet persimmon powder. In all products, the reduction ratio of diameter increased (p<0.05) with storage time. With regard to microorganisms, all samples were in good condition, containing below 4.20 log CFU/g based on total plate counts through 40 days, though total plate counts and coliform plate counts of the control were lower (p<0.05) than T3 and T4. The VBN values of T4 was significantly higher than those of other products at 15, 30, and 40 days of storage. The TBARS values of meat patties containing sweet persimmon powder were significantly (p<0.05) higher than those of the conlIol. Sensory panels determined that meat patties containing sweet persimmon powder had a higher (p>0.05) preference score for overall acceptability.

Comparison of Single-stage Thermophilic and Mesophilic Anaerobic Sewage Sludge Digestion (단상 고온 및 중온 혐기성 하수 슬러지의 소화 공정 비교)

  • In this study, single-stage continuous anaerobic reactors to treat sewage sludge were operated under different temperature (55 and $35^{\circ}C$; $R_{TAD}$ and $R_{MAD}$) to evaluate the reactor stability and performance of the thermophilic and mesophilic anaerobic digestion. During the overall digestion, both anaerobic reactors maintained quite stable and constant pH and total alkalinity (TA) values in the range of 6.5-8.0 and 3-4 g $CaCO_3/L$, respectively. After the start-up period, $R_{TAD}$ showed 10% higher VS removal efficiency than that of $R_{MAD}$ ($R_{TAD}$; 43.3%; $R_{MAD}$ : 33.6%). Although organic acids such as acetic and propionic acid were detected in both anaerobic reactors at the start-up period, all organic acids in $R_{TAD}$ and $R_{MAD}$ were consumed at the steady state condition. Also $R_{TAD}$ showed 31.4 % higher methane production rate (MPR) than that of $R_{MAD}$ at the steady state condition ($R_{TAD}$; 243 mL $CH_4/L/d$; $R_{MAD}$ : 185 mL $CH_4/L/d$). Meanwhile, the experimental results indicated similar methane yield between $R_{TAD}$ and $R_{MAD}$.

Preparation and Characteristics of Mixed Fruit and Vegetable Juices (혼합과채(果菜)쥬스의 제조와 제조방법에 따른 품질특성)

  • An experiment to make mixed juices carrying the freshness and other specific characteristics of vegetables and fruits which are useful for the prevention and treatment of various diseases was attempted on the emphases of pretreatment methods, combination of fruits and vegetables, and elimination of microorganisms. Blanch in boiling water prior to extraction for green vegetables, addition of ascorbic acid during extraction for tomatoes and apples, or addition of ascorbic acid after blanch in 3% acetic acid for carrots was effective to keep colors and suspended solids in liquid extract. On the basis of sensory evaluation the extracts from tomatoes, apples. carrots. mallows, watercreses+pine needles, Angelica keiskei Koiz, jujubes and lemons were selected and mixed at the ratio of 3 : 3 : 3 : 1/2 : 1/2 : 1/2 : 1/2 : 1/5. The mixed extracts were pasteurized for 15sec at $96^{\circ}C$ or filtered through a ultramembrane filter. While the centrifuge precipitation and retentates on the membrane filter were autoclaved and combined with ultrafiltrates. The mixed juices showed $pH\;4.07{\sim}4.10$ titratable acidity $66.35{\sim}84.08$, soluble solid $7{\sim}9^0Brix$, reducing sugar $5.42{\sim}6.97%$, glucose $1.96{\sim}2.30%$, fructose $3.46{\sim}4.14%$ and high content of K, Mg and Ca. Ultrafiltration showed better quality scores in color, juice. Peroxidase and microorganisms were inactivated by thermal treatment and ultrafiltration.
