Antioxidant Activity of Chestnut (Castanea crenata S.et Z.) bur Fermented by Lactobacillus casei (유산균 발효에 의한 밤송이추출물의 항산화 효과)

  • The aim of this study was to show the antioxidant properties of chestnut (Castanea crenata) bur extracts fermented by Lactobacillus casei. The chestnut has been used as a cosmetic material in Korea for many years. This study showed that reactive oxygen species (ROS) were inhibited by the fermentation materials of chestnut bur extracts. The antioxidant activities were analyzed and expressed as EDA, ABTS, reducing power, SOD-like activity, hydrogen peroxide scavenging activity, superoxide anion radical scavenging activity, and nitric oxide scavenging ability. The antioxidant activities of fermentation materials from L. casei of chestnut bur extracts were higher than those of butylated hydroxyanisole (BHA), epigallocatechin gallate (EGCG), and ascorbic acid (AA). Therefore, we expect that fermentation materials from L. casei of chestnut bur extracts are valuable resources as natural antioxidants and functional cosmetics ingredients.

Effect of Methanol Extract of Dryopteris Crassirhizoma in Human Oral Cancer Cells (관중 메탄올추출물이 구강암세포주에 미치는 영향)

  • Dryopteris crassirhizoma is one of the naturally occurring substance wood ferns and is known for having anti-inflammatory, antiviral and anthelmintic activities. However, there is less report about its anticancer effect in human cancer cell lines. In the present study, the effect of methanol extract of dryopteris crassirhizoma (MEDC) on apoptosis in human oral cancer cell lines (MC3 and HN22 cells) was investigated. MEDC inhibited cell viability and induced apoptosis. MEDC significantly increased Bak and truncated Bid proteins in MC3 cells and elevated only truncated Bid compared to the control while other Bcl-2 family proteins were not altered. MEDC has anticancer activity by inducing apoptotic cell death through the regulation of either Bak or Bid. These findings suggest that its extract possibly may be used for treating oral cancer.

  • 본 연구는 1981-1991년 농업과학기술원 라이시미터에서 수집한 결과를 이용하여 집중강우시 경사지 밭토양의 물유출 특성을 구명하였다. $7\~9$월 집중강우시 토양 침투수나 지표 유거수는 농업지역에서 환경으로 물질이 이동하는 주요 경로이며 특히 경사지 밭토양에서 지표 유거수는 토양유실의 주원인 중 하나이기 때문에 이에 대한 이해는 매우 중요하다. 이를 위해 강우량, 지표 유거수량, 지하 침투수량 측정 자료 중 호우주의보가 발령되는 일강우량 80mm이상일 때를 대상으로 하여 토성과 경사도에 따른 강우량과 유거수, 침투수의 관계를 분석하였다. 강우량이 적을 때 강우에 대한 침투수와 유거수의 비율은 강우시 표토의 토양수분함량에 많은 영향을 받는다. 이는 표토의 토양수분함량에 따라 유출 또는 침투 발생 유효강우량이 결정되기 때문이다. 강우량이 적을 때의 유거수량과 침투수량을 판단하기 위해 범용토양유실예측공식(Universal soil loss equation, USLE)에서는 0.5 inch 즉, 12.5 mm 이상의 강우를 유출에 대한 유효강우로 가정하고 있으며 많은 모형에서 토양의 침투속도, 포장용수량, 강우시점의 토양수분함량의 함수로 유출 또는 침투 유효강우량을 산정하고 있다. 그러나 강우량이 클 때는 강우에 대한 침투수와 유거수에 비율에 토양수분함량이 미치는 영향이 비교적 적기 때문에 토양의 수분함량에 대한 고려없이 강우와 침투수, 유거수에 대한 관계를 평가하는 것이 가능하였다. 경사도 $10\%$, 경사장 15m, 피복작물 콩인 양토를 기준으로 할 때 강우량과 침투수의 관계는 $I_{10}(mm)=0.44R(mm)+5.8(r^2=0.55)$이었다. y절이 발생한 이유는 이전 강우에 의해 침투되고 있는 물이 있음을 함축하며 기울기 0.40은 강우의 $40\%$가 지하로 침투하였음을 의미한다. 침투수량은 토성별로 양토를 1.0으로 기준할 때 사양토가 1.12로 가장 컸고, 식양토 0.94, 식토 0.91로 평가되었다. 이는 토성간의 침투속도 및 투수속도의 경향이 반영된 것이다. 경사에 따라서는 경사도가 증가할수록 지수적으로 감소하였으며 $10\% 경사일 때를 기준으로 $I(mm)=I_{10}{\times}1.17{\times}e^{-0.0164s(\%)}$로 나타났다. 같은 조건에서 강우량과 유거수의 관계는 $Ro_{10}(mm)=5.32e^{0.11R(mm)}(r^2=0.69)$로 나타났다. 이는 토양의 투수특성에 따라 강우량 증가에 비례하여 점증하는 침투수와 구분되는 현상이었다. 경사와 토양이 같은 조건에서 나지의 경우 역시 $Ro_{B10}(mm)=20.3e^{0.08R(mm)(r^2=0.84)$로 지수적으로 증가하는 경향을 나타내었다. 유거수량은 토성별로 양토를 1.0으로 기준할 때 사양토가 0.86으로 가장 작았고, 식양토 1.09, 식토 1.15로 평가되어 침투수에 비해 토성별 차이가 크게 나타났다. 이는 토성이 세립질일 수록 유거수의 저항이 작기 때문으로 생각된다. 경사에 따라서는 경사도가 증가할수록 증가하였으며 $10\% 경사일 때를 기준으로 $Ro(mm)=Ro_{10}{\times}0.797{\times}e^{-0.021s(\%)}$로 나타났다.

Evaluation of Dose Distribution Using Gafchromic $EBT^{(R)}$ Film (Gafchromic $EBT^{(R)}$ 필름을 이용한 선량분포의 평가)

  • Dose evaluation for small field such as stereotactic radiosurgery was performed using $Gafchromic^{(R)}$ EBT film. Every film which irradiated 6MV photon beam was scanned and obtained the optical density(OD) by flat bed scanner after 24 hours of irradiation. This study compared dose from diode in water and Gafchromic $EBT^{(R)}$ film in acrylic phantom to verify the reliability of the film, and to evaluate the SRS in clinical dose distributions from calculation and measurement in the region of virtual target in humanoid and cylindrical phantoms were compared. The Gafchromic $EBT^{(R)}$ film was found to be linear up to 9Gy. The $D_{max}$ for 6 MV was measured at 1.5 cm from the surface by both of diode and the film. As the depth is deeper, the error was measured within $2{\sim}3%$ at $10{\sim}20\;cm$ depth. Comparing between distribution from calculation and measurement, we found that there is 5% error at 90% isodose line. We found that given dose could be measured accurately by using the phantoms. It was feasible to use the Gafchromic $EBT^{(R)}$ film in quality assurance of SRS.

  • A long-term R&D program for HLW disposal technology development was launched in 1997 in Korea and Korea Reference disposal System(KRS) for spent fuels had been developed. After then, a recycling process for PWR spent fuels to get the reusable material such as uranium or TRU and to reduce the volume of radioactive waste, called Pyro-process, is being developed. This Pyro-process produces several kinds of wastes including metal waste and ceramic waste. In this study, the characteristics of the waste from Pyro-process and the concepts of a disposal container for the wastes were described. Based on these concepts, thermal analyses were carried out to determine a layout of the disposal area of the ceramic wastes which was classified as a high level waste and to develop the disposal system called A-KRS. The location of the final repository for A-KRS is not determined yet, thus to review the potential repository domains, the possible layout in the geological characteristics of KURT facility site was proposed. These results will be used in developing a repository system design and in performing the safety assessment.

A Study of Frangibility of 9MM Bullet Related to Material Composition and Sinter Condition (합금 조성 및 소결 조건에 따른 9MM 탄자의 파쇄성에 관한 연구)

  • Frangible bullets, which are shredded after impact on a target, reduce the possibility of both ricochet and unexpected injury in shooting training and in mission acts in dams, nuclear power plants, and cultural properties. Reducing the levels of hazardous materials in shooting ranges, such as lead, has become an important agenda for the government and environmental groups. In this study, the shape of a frangible bullet was designed for efficient shredding, and the safety and reliability were confirmed by actual firing under different process conditions. In addition, the physical characteristics, such as compaction pressure, density, and frangibility of each process, were compared by analyzing the microstructure of the sintered frangible bullet. The experiment revealed the smallest fragmentation after impact on the target under the following conditions: Cu-Sn 85:15; sintering temperature, 600℃; sintering time, one hour. Further development of the process conditions and experimental methods will contribute to the performance and environmental improvement of a frangible bullet.

Expression of Mycosporine-like Amino Acids Biosynthetic Genes in the Chlamydomonas sp. Exposed to Radiofrequency (Radiofrequency에 노출된 Chlamydomonas sp.의 mycosporine-like amino acids 생합성 유전자 발현)

  • Mycosporine-like amino acids (MAAs) are UV-absorbing substances, and diverse marine organisms have the evolved the capacity to diminished the direct and indirect damaging effects of environmental ultraviolet radiation by synthesis and accumulation of MAAs. In this study, we manufactured a radiofrequency (RF) generation device and applied to microalgal culture. $0.35{\pm}0.05$ mHz of RF was supplied to culture vessel for Chlamydomonas sp. and samples were harvested at the designated time intervals (1, 0.5, 1 and 2 hr). MAAs biosynthetic genes, dehydroquinate synthase homolog (DHQS-like) and nonribosomal peptide synthetase homolog (NRPS-like), were cloned from Chlamydomonas sp. and their gene expressions under the RF exposure were analyzed using qRT-PCR. DHQS-like and NRPS-like gene expressions of Chlamydomonas sp. exposed to RF were increased 1.46 and 1.19 fold at 1 hr, respectively. These results means that DHQS-like and NRPS-like genes can be good biomarker candidates for diagnosis of MAAs biosynthesis in the Chlamydomonas sp.

Effects of Operating Parameters on Phenol Degradation by Pulsed Corona Discharges in Aqueous Solutions (펄스 코로나 방전에 의한 페놀 분해에 미치는 운전변수의 영향)

  • Effects of operating parameters such as applied voltage, solution conductivity, ferrous ion concentration, electrode material on phenol degradation by pulsed corona discharges were investigated in laboratory scale experiments. The increase of applied voltage enhanced the phenol degradation by generating more energetic electrons. The solution conductivity inversely affected phenol removal rate in the tested ranges because the increase of conductivity decreased the electric field strength through the liquid phase. The addition of ferrous sulfate promoted the phenol degradation through the OH radical production by the Fentonlike reactions between ferrous ion and hydrogen peroxide generated by pulsed corona discharges. Catechol and hydroquinone were detected as primary intermediates of phenol degradation and the decrease of pH and the increase of conductivity were observed probably due to the generation of organic acids. Almost all of the initial phenol was disappeared and 29% of total organic corbon (TOC) was removed in the condition of 0.5 mM of ferrous sulfate after approximately 230 kJ of discharge energy transferred to the reactor.

Investigation of Active Antifungal Compounds of Essential Oil from Chamaecyparis obtusa Against Dermatophytes, Microsporum canis and Trichophyton Mentagrophytes (피부사상균 Microsporum canis 및 Trichophyton mentagrophytes에 대한 편백정유의 항진균활성물질 탐색)

  • The present study was conducted to evaluate the application of Chamaecyparis obtusa and to investigate potential utilization of essential oil from C. obtusa as plant-based medicine. The antifungal activity of essential oil from leaves and twigs of C. obtusa (Sieb. Et Zucc) was determined and the major components of active fractions against Microsporum canis (KCTC6591) and Trichophyton mentagrophytes (KCTC6077) were identified by GC/MS analysis. In treatment of essential oil from C. obtusa, the strain M. canis was more resistant than the other, T. mentagrophytes. In the agar diffusion assay, essential oil from C. obtusa inhibited hyphal growth of M. canis and T. mentagrophytes at the concentration of more than 5,000 ppm. The zones named B and C in the TLC assay of essential oil from C. obtusa showed antifungal activities. Among four sub-fractions of n-hexane extract from B and C zones, named as B-1, B-2, C-1 and C-2, the C-2 showed the highest antifungal activity. Instrumental GC/MS analysis for sub-fractions showed that a major component of C-1 was ${\alpha}$-terpineol as terpene alcohol, while C-2 contained sesquiterpenes such as elemol, cedrol and eudesmol.

The Growth Effects of Creeping Bentgrass by Application of Liquid Fertilizer with Saponin and Liquid Fertilizer with Amino Acid (사포닌과 아미노산 함유비료의 살포가 크리핑벤트그래스의 생육에 미치는 효과)

  • This study was conducted to evaluate to the effect of liquid fertilizer with saponin (SLF) and liquid fertilizer with amino acid (ALF)on the growth of creeping bentgrass. In creeping bentgrass, turf color index, chlorophyll index, dry weight and shoot number were measured. It was hardly affected by SLF and ALF applications in investigation of chemical properties of the soil. By applying SLF and ALF, turf color index and chlorophyll index in 2SLF and 2ALF were increased more than CF, and shoot number and root length in 2SLF, ALF and 2ALF were higher than CF. In correlation coefficient among growth factors of creeping bentgrass, turf quality was significantly different in root length, shoot number, dry weight, and content of N and K in turf tissue (P<0.05), N content of tissue was significantly in root length, shoot number and dry weight (P<0.05), and K content was significantly in shoot number and dry weight (P<0.05). These results suggested that application of functional liquid fertilizers such as SLF and ALF was expected to replace compound fertilizer in turf management and that applied SLF and ALF was stimulated the uptake of N and K into turf so that turf qualities were improved by enhancing growth shoot and root of turf.
