Effects of Fermented Red Ginseng Supplementation on Blood Glucose and Insulin Resistance in Type 2 Diabetic Patients (발효홍삼이 제2형 당뇨병 환자의 혈당 및 인슐린저항성에 미치는 영향)

  • We performed a randomized placebo-controlled trial to determine whether or not fermented red ginseng supplementation modulates blood glucose and insulin resistance in type 2 diabetic patients. A total of 38 patients were randomized to either a fermented red ginseng group or placebo group. The patients in the experimental or placebo group consumed 780 mg of fermented red ginseng or cellulose supplement per day for 12 weeks, respectively. Lifestyle factors and dietary intakes of the patients were not altered during the 12-weeks period. In the fermented red ginseng group after 12 weeks, the fasting blood glucose levels were significantly decreased ($136.29{\pm}16.45$ mg/dL to $127.71{\pm}17.74$ mg/dL) and $HbA_1c$ was also decreased. Especially, high HbA1c (HbA1c $\geq$8%, $8.45{\pm}0.56%$ to $7.82{\pm}0.53%$) was significantly decreased compared to low HbA1c (HbA1c <8%, $6.71{\pm}0.85%$ to $6.44{\pm}0.49%$) in the fermented red ginseng group. Serum low-density lipoprotein was slightly decreased in the fermented red ginseng group compared to the placebo group. Homeostasis model assessment-insulin resistance was significantly reduced in the fermented red ginseng group compared to the placebo group. These results suggest that fermented red ginseng supplementation could be helpful to reduce blood glucose by improving insulin resistance in type 2 diabetic patients.

Effect of Some Herbal Extracts on Entomopathogenic Nematodes, Silkworm and Ground Beetles (몇 가지 한약재 추출물이 곤충병원성선충과 누에 및 먼지벌레에 미치는 영향)

  • Effect of four nematicidal herbal extracts (Daphne genkwa, Eugenia caryophyllata, Quisqualis indica and Zingiber officinale) and 3 acricidal herbal extracts (Pharbitis nil, Xanthium strumarium, and Desmodium caudatum) on entomopathobenic nematodes [Steinernema carpocapsae Pocheon strain (ScP) and Heterorhabditis sp. Gyeongsan strain (HG)], silkworm (Bombyx mori), and ground beetles (Synuchus sp.) were investigated in the laboratory and field. D. genkwa was highly toxic to SCP and HG (100% mortality) at the concentration of 5,000 ppm in X-plate. All the infective juveniles of HG were dead after 3 days by E. caryophyllata and Q. indica. The mortality of ScP and HG was below 10% by D. genkwa, D. caudatum, E. caryophyllata, Q. indica and Z. officinale at the concentration of 1,000 ppm two days after treatment while mortality of HG was 62.8% by D. genkwa at the concentration of 1,000 ppm in X-plate. However, 1,000 ppm had not effect on nematode survival and pathogenicity of ScP in sand column. On the contrary, E. caryophyllata had effect on pathogenicity of HG. Mean number of dead Galleria mellonella larva of HG was 0.5 in E. caryophyllata treatment. Q. indica did not effect silkworm reared on mulberry leaves at the treatment of 1,000 ppm in 10 days after treatment. However, there were 20.0 and 100% mortalities in the treatment of D. genkwa 3 and 10 days after treatment, respectively. The weight of silkworm was low in D. genkwa and did not pupate. The weight of pupa and cocoon were not different in E. caryophyllata, P. nil, Q. indica, X. strumarium and Z. officinale. D. genkwa, E. caryophyllata, P. nil, Q. indica and Z. officinale had no effect on ground beetles, Synuchus sp. in forest soil.

Effects of Bosaengtang Administration in Pregnant Rats and Fetuses (보생탕이 랫드의 모체와 태자에 미치는 영향에 대한 연구)

  • The experiments were undertaken to evaluate the effects of Bosaengtang in pregnant rats and fetuses. Female Sprague-Dawley rats were orally administered with Bosaengtang at the dose of 5mg/kg/day for 20 days. Pregnant rats were sacrificed at the 20th day of gestation, and observed internal and reproductive organs. Fetuses were randomly selected and fixed in 95% ethanol. Fetuses were stained with alcian blue and alizarin red S, and observed skeletal malformations. The results obtained were as follows : Bosaengtang administered group showed higher maternal body weight than the control group, but both groups showed increase in weight. Bosaengtang administered group showed lower than the control group, and higher liver and kidney weight than the control group, but the differences were minimal. There were no significant changes between the control and treated group in blood chemistry values and hematological values but all the groups were within in normal ranges. There were no significant changes in the number of corpus luteum, implantation, live fetus and implantation rate, delivery rate, late resorption rate, sex ratio, but Bosaengtang administered group showed higher early resorption rate than control group. comparing the control and Bosaengtang group, neonatal body weight and the number of fetuses were increased in Bosaengtang group. The fetuses of dams treated with Oriental medicine didn't showed external malformation. Vertebral and sternal variations were observed in Bosaengtang group, but the differences were not apparent compared to the control group. The number of ribs, cervical, thoracic and lumbar vertebrae were normal. The number of sacral was similar and the number of caudal was increased. Fetuses showed significant difference in the number of caudal vertebrae. (P<0.01) From these results, we can carefully conclude that Bosaengtang showed beneficial effects on maternal body weight, early resorption rate, number of live fetus. There were no significant changes in organ weight, hematoscopy, reproduction organs. External malformation wasn't visible. Skeletal variations were showed in vertebrae and sternum but compared to the control group, these variations weren't much different.

Long-term (2002~2017) Eutropication Characteristics, Empirical Model Analysis in Hapcheon Reservoir, and the Spatio-temporal Variabilities Depending on the Intensity of the Monsoon (합천호의 장기간 (2002~2017) 부영양화 특성, 경험적 모델 분석 및 몬순강도에 따른 시공간적 이화학적 수질 변이)

  • The objective of this study was to analyze eutrophication characteristics, empirical model analysis, and variation of water quality according to monsoon intensity in Hapcheon Reservoir for 16 years from 2002 to 2017. Long-term annual water quality analysis showed that Hapcheon Reservoir was in a meso-nutrition to eutrophic condition, and the eutrophic state intensified after the summer monsoon. Annual rainfall volume (high vs. low rainfall) and the seasonal intensity in each year were the key factors that regulate the long-term water quality variation provided that there is no significant change of the point- and non-point source in the watershed. Dry years and wet years showed significant differences in the concentrations of TP, TN, BOD, and conductivity, indicating that precipitation had the most direct influence on nutrients and organic matter dynamics. Nutrient indicators (TP, TN), organic pollution indicators (BOD, COD), total suspended solids, and chlorophyll-a (Chl-a), which was an estimator of primary productivity, had significant positive relations (p<0.05) with precipitation. The Chl-a concentration, which is an indicator of green algae, was highly correlated with TP, TN, and BOD, which differed from other lakes that showed the lower Chl-a concentration when nutrients increased excessively. Empirical model analysis of log-transformed TN, TP, and Chl-a indicated that the Chl-a concentration was linearly regulated by phosphorus concentration, but not by nitrogen concentration. Spatial regression analysis of the riverine, transition, and lacustrine zones of $log_{10}TN$, $log_{10}TP$, and $log_{10}CHL$ showed that TN and Chl-a had significant relations (p<0.005) while TN and Chl-a had p > 0.05, indicating that phosphorus had a key role in the algal growth. Moreover, the higher correlation of both $log_{10}TP$ and $log_{10}TN$ to $log_{10}CHL$ in the riverine zone than the lacustrine zone indicated that there was little impact of inorganic suspended solids on the light limitation in the riverine zone.

Development and Validation of Analytical Method for Wogonin, Quercetin, and Quercetin-3-O-glucuronide in Extracts of Nelumbo nucifera, Morus alba L., and Raphanus sativus Mixture (연잎, 상엽, 건조 무 혼합 추출물의 지표성분 wogonin, quercetin 및 quercetin-3-O-glucuronide의 분석법 개발 및 검증)

  • The aim of this study was to develop and validate an analytical method for determining the presence of wogonin, quercetin, and quercetin-3-O-glucuronide in extracts of Nelumbo nucifera, Morus alba L., and Raphanus sativus mixtures. We evaluated the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) of analytical methods for wogonin, quercetin, and quercetin-3-O-glucuronide using high performance liquid chromatography. Our result showed that the correlation coefficients of the calibration curve for wogonin, quercetin, and quercetin-3-O-glucuronide were 0.9999. The LOD for wogonin, quercetin, and quercetin-3-O-glucuronide ranged from 0.09 to 0.16 and those for the LOQ ranged from 0.26 to $0.48{\mu}g/mL$. The inter-day and intra-day precision values of wogonin, quercetin, and quercetin-3-O-glucuronide ranged from 0.74 to 1.87 and from 0.28 to 1.12%, respectively. The inter-day and intra-day accuracies were 99.96~115.88% and 99.73~114.81%, respectively. Therefore, the analytical method was validated for the detection of wogonin, quercetin, and quercetin-3-O-glucuronide in extracts of Nelumbo nucifera, Morus alba L., and Raphanus sativus mixtures.

Changes of Cold Tolerance and it Mechanisms at Young Microspore Stage caused by Different Pre-growing Conditions in Rice (벼 수잉기내냉성의 전역조건에 따른 변동과 기구)

  • It was proved that cold tolerance of rice plants at the young microspore stage was affected by water temperature and nitrogen application from the spikelet differentiation stage to the young microspore stage, and this mechanism could be explained in the point of view of pollen developmental physiology. The cold tolerance of rice plants at the young microspore stage was severely affected by water temperature (Previous water temperature) and nitrogen application(Previous nitrogen application) from the spikelet differentiation stage to the spikelet differentiation stage. Although the duration is only 10 days or so from the spikelet differentiation stage to the young microspore stage, these days are very important period to confirm the cold tolerance of rice plants at the young microspore stage. The higher previous water temperature up to $25^{\circ}C$ and the deeper previous water depth up to 10cm caused the more cold tolerance of rice plants. Water irrigation of 10cm before the cretical stage showed lower cool injury than that of water irrigation of 20cm during the critical stage. The preventive effect of cool injury by combined treatment of the deep water irrigation before and during the critical stage was not additive but synergistic. The cold tolerance of rice plants grown in previous heavy nitrogen level was rapidly decreased when nitrogen content of leaf blade at the young microspore stage was excessive over the critical nitrogen level. Nitrogen content of leaf blade at the changing point of cold tolerance was estimated as about 3.5% for Japonica cultivars and about 2.5% for Indica x Japonica cultuvars. It is considered that these critical nitrogen contents of leaf blade can be used as a index of the safe critical nitrogen level for the preventive practices to cool injury. It was summarized that increase of engorged pollens per anther by high previous water temperature resulted from the increase of number of differentiated microspores per anther, otherwise, the increase of engorged pollens by the decrease of previous nitrogen level was caused by the decrease of the number of aborted microspores per anther.

Thirteen-week Repeated Oral Dose Toxicity Study of WK-38 in Rats (랫트에서 WK-38에 대한 13주 반복경구투여 독성에 관한 연구)

  • The subcronic toxicity of WK-38, a herbal preparation for the atherosclerosis, was examined in male and female Sprague-Dawley rats. WK-38 is composed of Rhei Rhizoma, Magonoliae Cortx, Moutan Cortex Radicis. Rats were treated with the test substance at a dose of 5 mg/kg, 50 mg/kg and 500 mg/kg intragastrically for 13 weeks. Control rats were treated with vehicle only. No death and abnormal clinical signs were observed throughout the administration period. Also there was no difference in net body weight gain, food and water consumption, organ weight, gross pathological findings, and urine analysis among the groups of rats treated with different doses of the WK-38. Hematological findings and biochemical examination revealed no evidence of specific toxicity related to WK-38. The results suggested that under the condition employed in this study no observation effect level (NOEL) of WK-38 would be 500 mg/kg/day.

Protective Effect of Green Tea Extract, Catechin on UVB-Induced Skin Damage (녹차추출물 성분 catechin이 자외선에 의해 손상된 피부에 미치는 영향)

  • The main constituent of green tea, catechins have been reported to have numberous biological anti-vites including antimutagenic, antibacterial, hypocholesterolemic, antioxidant and antitumor properties. In the present study, we examined the protective effect of catechin on UVB-induced skin damage. Catechin (3 mg/mouse) was topically treated to dorsal area of SHK-1 hairless mouse daily for 2 weeks. UVB (100 mJ/$\textrm{cm}^2$) was also treated soon after application of catechin alone or with catechin for 2 weeks. Catechin reduced UVB-induced infiltration of inflammatory cells, fibrosis of cells and collagen-fiber formation. In addition, catechin also prevented UVB-induced DNA fragmentation and apoptosis cell number, but not changed p53 level. Furthermore catechin inhibited UVB-induced cell proliferation. There results showed that catechin have preventive effect aganinst UVB-induced skin damages. and these effects could contribute to the antitumor promoters activity.

  • Ground subsidence can result in the formation of sinkholes, potholes, settlement of structures, and road subsidence. Road subsidence is described as the sudden collapse of the road surface into subsurface cavities caused by the loss of bearing capacity in the ground, such as the dissolution of limestone by fluid flow in the surface causing the formation of voids leading to subsidence at the surface. Road subsidence occurs about 665 times annually, and this incidence has been increasing until 2013. Damaged underground facilities, management negligence, and lowering of the ground water table have been the causes of road subsidence in Seoul. Seoul metropolitan government announced special management counter plans to relieve the anxieties and make the roads safe for passing. Construction sites, such as excavation works, need to be managed properly because they have strong potential to induce road subsidence. The aim of this study was to identify the main causes of road subsidence and suggest management plans. First, life cycle cost analysis revealed the daytime construction to be more appropriate than nighttime. In addition, by analyzing the limitations of using sand as a backfill material, it is proposed to use a flowable backfill material instead of sand. Finally, to reduce the blind spots, which is a problem in surveying the road pavement conditions of local governments, the road to be managed is divided into several zones, and a specialized agency is selected for each zone and a method of surveying the blind spots through collaboration is suggested.

Effects of HACCP System Implementation on Medicine Use and Productivity of Large Scale Swine Farms (HACCP 시스템 적용이 대규모 양돈장의 동물용 의약품 사용 및 생산성에 미치는 영향)

  • The objective of this study was to examine the effect of HACCP implementation on the medicine use, antibiotic utilization in each feeding stage and productivity of large scale swine farms (over 5,000 pigs) in Korea. Data were collected from ten swine farms before and after implementation of a HACCP system. Total number of piglets, number of initial weaning piglets, number of weaning piglets, survival rate of piglets, date of weaning piglets, number of piglets per sow per year and price for medicine used per month on HACCP implemented swine farms had a tendency to increase without any significant difference. However, parturition rate, market pig per sow per year, number of antibiotic used and farm number of feeds containing antibiotics at fattening stage added at the swine farm were significantly increased after HACCP implementation (p<0.05). Due to increase in the feed cost recently, production cost for shipment in HACCP system implemented swine farms had increased in this study. In conclusion, our results indicated that the implementation of HACCP system might have many effects including reduction of medical expenses and improvement of productivity of the pig farms.
