Identification of Growth Inhibitory Substance on Food-borne Microorganisms from Commiphora molmol Engl. and Its Application to Food Products (몰약(Commiphora molmol Engl.)의 식중독 미생물 증식 억제 물질의 구조동정 및 식품적용)

  • The ethanol extract and n-hexane fraction of Commiphora molmol Engl. showed minimum inhibitory concentration of 50 ppm and 25 ppm, respectively, on 5 strains of Listeria monocytogenes at $32^{\circ}C$. The purified substance, C3-3-2 fraction, was isolated by silica gel column and preparative thin layer chromatography from n-hexane fraction of Commiphora molmol Engl. The C3-3-2 fraction showed a strong bactericidal activity on 5 strains of L. monocytogenes at the concentration of 10 ppm in tryptic soy broth medium. At that concentration, the viable count was reduced $5{\sim}6$ log cycle from initial cell number. The n-hexane fraction of Commiphora molmol Engl. showed strong growth inhibition at the concentration of 25 ppm on Bacillus cereus and Staphylococcus aureus, at 50 ppm in broth on Salmonella enteritidis, and at 500 ppm on Vibrio parahaemolyticus. The purified antimicrobial substance, the C3-3-2 fraction, was identified as m-nonylphenol by on the basis of the $^1H-,\;^{13}C-NMR$ and EI/MS data. For the application test, the C3-3-2 fraction which was purely isolated from Commiphora molmol Engl. at 100 ppm were applied to minced Alaska pollack and ground beef at $32^{\circ}C$ and $5^{\circ}C$. The antimicrobial substances did not reduce L. monocytogenes ATCC 19113 at $32^{\circ}C$, while they reduced L. monocytogenes ATCC 19113 in viable number at $5^{\circ}C$. However, the antimicrobial effect of C3-3-2 fraction in food system was lower than that of broth condition.

Effects of Some Crude Drug Extracts on the Brain Neurotransmitters in the Ethanol-Treated Rats (수종의 생약 추출물이 에탄올 투여 흰쥐의 뇌 부위별 신경전달물질에 미치는 영향)

  • The concentration of neurotransmitters in rat brain was determined by HPLC-ECD (electrochemical detection) method and the effects of methanol extracts of some crude drugs, such as Polygala Radix, Myristicae Semen, Zizyphi Semen, Acori graminei Rhizoma, Visci Herba, Liriopsis Tuber, Myrrha on the concentration of neurotransmitters in the ethanol-treated rat brain were investigated. By the administration of ethanol, dopamine (DA), 3, 4-dihydroxyphenyl acetic acid (DOPAC) and serotonin (5-HT) levels in frontal cortex and 5-HT level in hippocampus were significantly increased compared with the neurotransmitter levels in the brain of saline-treated rats. The ${\gamma}$-aminobutyric acid (GABA) level in frontal cortex was decreased by the same treatment. There was a tendency that the DA level in frontal cortex and striatum of ethanol-treated rats were increased by the administration of crude drug extracts. Especially, Myrrha and Visci Herba significantly increased the DA level of frontal cortex in ethanol-treated rats, while they significantly decreased the 5-HT level in the same region of the brain. GABA level in striatum of ethanol-treated rats was significantly decreased by Myristicae Semen, Visci Herba and Myrrha. These results suggest that the tested crude drug extracts have selective interaction with neurotransmitters in specified region of central nervous system.

Effects of Myrrha Water Extract on Rat Myocardial Cells in Cultures (몰약 전탕액이 배양 심근세포에 미치는 영향)

  • Objectives and Methods : In order to elucidate toxic mechanism of myocardial damage and protective effect of myrrha water extract against cytotoxic effect of xanthine oxidase/hypoxanthine(XO/HX), cardioprotective effect of myrrha water extract was examined by MTT assay, LDH (Lactate Dehydrogenase) activity and heart beating rate after cultured myocardial cells derived from neonatal mouse were treated with various concentration of XO/HX, a free radical. Results : XO/HX induced a decrease of cell viability, an increase in the amount of LDH, and a decrease of heart beating rate on cultured myocardial cells in a dose-dependent manner. In cardioprotective effect of myrrha water extract, it showed a decrease in the amount of LDH and an increase of heart beating rate on cultured myocardial cells damaged by XO/HX. Conclusions : From the above results, it is suggested that XO/HX showed toxic effect in cultured myocardial cells derived from neonatal mouse and that myrrha water extract is very effective in the prevention of XO/HX-induced cardiotoxicity.

Effect of antibacterial effects of myrrh, rhatany, chamomomilla against to oral microorganisms (몰약, 라타니아, 카모밀레 등의 구강 내 병원균에 대한 항균작용)

  • Even though there exist a lot of study about antibacterial effects and reactions of extracted materials from plant, few study exist about oral pathogenic bacteria. Therefore we tried to recognize about the suppression effect to the periodontal pathogenic bacteria and halitosis, when add some kinds of plant extracted materials, myrrh, rhatany, chamomolilla to saliva. We used Crude drug : Myrrh tincture (100mg/ml), Ratanhia tincture (100mg/ml), Chamomile tincture(100mg/ml). We inspected about the cariogenic bateriae, S. mutans GS5 and S. sobrinus 6715, periodontal pathogenic bacteria, P. gingivalis 2561, P. intermedia ATCC 25611, Candida albicans ATCC 18804, and E. feacalis ATCC 4083, then the result follow. The plant extracted material, myrrh, rhatany, chamomomilla, which have convergence effect, bacteriocidal effect and anti-inflammation effect, show an antibacterial effect and reaction to the oral pathogenic bacteria. And with treating rhatany that have the most strong antibacterial effect, through transmission electron microscopy we could see a severe morphologic change of bacteria. This means with the plant extracted material, we can suppress the oral harmful bacteria and prevent periodontal diseases, caries, halitosis and oral inflammations. And within the future studies for the improvement of oral hygiene, our result might be a clinical evidence.

Effects of Commiphora Myrrha (CM) on the Monosodium Urate (MSU)-induced Gout Model in Rats. (몰약이 Monosodium Urate로 유발된 백서의 통풍에 미치는 영향)

  • Objective : To identify the inhibitory effects of Commiphora Myrrha (CM) on monosodium urate (MSU)-induced gout model in rats. Materials and Methods: After pretreatment with CM-I (125mg/kg) or CM-II (50mg/kg) for 7 days followed by ones injection of MSU solution. the various indicators related to gout were measured on hematological and serum level including joint inflammation, Also, it was studied whether FBM directly inhibits the activity of xanthine oxidase in vitro. Results : As a result of this study, CM didn't show cytotoxicity in Jurkat cells, but it showed significant inhibition of activity of xanthine oxidase in vitro. CM slightly inhibited joint inflammation induced by MSU though not with statistical significance. CM partially decreased MSU-induced albumin, globulin, AST, ALT, BUN, creatinine. WBC, platelet count and ESR level and significantly decreased MSU-induced uric acid in serum. Conclusion : These results suggest that CM has therapeutic effects that are applicable to prevention and treatment of gout, and should be further investigated.

Ameliorating Effects of HPM-1 on Scopolamine-induced Memory Impairments in Mice (몰약 당귀 오미자 혼합제제 HPM-1의 Scopolamine에 의해 유도된 기억력 감퇴에 대한 개선 효과)

  • Myrrh, Angelicae Radix, and Schisandrae Fructus have been used to treat diverse diseases including neurological disorder in the traditional Korean medicine. In the present study, we investigated the ameliorating effects of HPM-1, a combined extract of Myrrh, Angelicae Radix and Schisandrae Fructus, on scopolamine-induced memory impairments in mice. First, we assessed HPM-1, HPM-2 and HPM-3, in which Myrrh was extracted with water, 30% ethanol, and 30% ethanol/3% vinegar, respectively. The oral administration of HPM-1, HPM-2, or HPM-3 significantly reversed scopolamine-induced reduction of spontaneous alternation in the Y-maze task. In the passive avoidance task, HPM-1 or HPM-3 restored the decreased latency time of the retention trial by scopolamine treatment, but in terms of efficacy, HPM-1 showed more beneficial effects than HPM-3. In addition, HPM-1 administration reversed scopolamine-induced reduction of spontaneous alternation in the Y-maze task and scopolamine-decreased latency time in the passive avoidance in a dose-dependent manner. These results suggest that HPM-1 has the therapeutic potential in memory impairments.

The Study on anti-obesity of Myrrh ethanol extract (몰약(沒藥) 에탄올 추출물의 항비만에 관한 연구)

  • Objectives : The objective of this study was to investigate the effect of Myrrh 80% ethanol extract on adipocyte differentiation and adipogenesis in 3T3-L1 cell.Methods : Myrrh was prepared by extracting with 80% ethanol. Cell viability was assessed by MTT assay using 3T3-L1 cells. Anti-obesity activity was measured in lipid droplets and triglyceride (TG) accumulation in 3T3-L1 cells. We also analyzed the expression of C/EBPβ, C/EBPα, PPARγ, SREBP1c, and aP2 by reverse transcriptase polymerase chain reaction (RT-PCR). In addition, we observed the production of fatty acid, acetyl-CoA carboxylase and Oil-red O stainingResults : No cytotoxicity from Myrrh 80% ethanol extracts was observed at the concentration of 1, 10, 100 (㎍/㎖) in 3T3-L1 cells. Treatment with Myrrh significantly suppressed the terminal differentiation of 3T3-L1 in a dose-dependent manner, as confirmed by a decrease in triglyceride and Fatty acid and Acetyl-CoA carboxylase. Also, Myrrh exhibited potential adipogenesis inhibition and downregulated the expression of pro-adipogenic transcription factors, such as sterol regulatory element binding protein-1c (SREBP-1c), peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancer binding proteins α (C/EBPα) and C/EBPβ, and adipocyte expressed genes, such as adipocyte fatty acid binding protein (aP2) and Fas. In addition, lipid accumulation determined by Oil-red O staining showed that Myrrh extract had inhibitory effects on lipid accumulation in 3T3-L1 cells.Conclusions : These results suggest that Myrrh suppresses obesity factors in 3T3-L1 cells. Myrrh may be a useful medical herbs for attenuating metabolic diseases such as obesity.

Anti-inflammatory Effects of $1{\beta}$,$6{\alpha}$-Dihydroxyeudesm-4(15)-ene Isolated from Myrrh on LPS-induced Neuroinflammation in BV2 cells (몰약으로부터 분리된 $1{\beta}$,$6{\alpha}$-dihydroxyeudesm-4(15)-ene의 LPS로 유도된 BV2 미세아교세포에서의 항염증효과)

  • Myrrh is a resinous substance obtained from Commiphora trees, which has long been used as an antiseptic agent. A sesquiterpene, $1{\beta}$, $6{\alpha}$-dihydroxyeudesm-4(15)-ene (DE), was isolated from the hot water extract of Myrrh. In the present study, we found that DE attenuates the lipopolysaccharide (LPS)-induced inflammation in BV2 microglial cells. DE significantly inhibited LPS-induced production of pro-inflammatory mediators such as nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in BV2 microglia in a concentration-dependent manner without cytotoxic effect. Furthermore, DE dose-dependently suppressed the protein expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). These results suggest that DE may be a good candidate to regulate LPS-induced inflammatory response.
