Increase of Salt and Low Temperature Tolerance by Overexpressing Glutathione S-Transferase (GST) Gene (염분과 저온에 대한 내성증진을 위한 GST 유전자의 과발현)

  • Cotton Glutathione S-Transferase (GST: EC 2.5.1.18) was cloned and overexpressed in tobacco (Nicotiana tabacum) plants. Northern blot analysis confirmed the successful transformation of cotton gst gene in tobacco plant. Type I and Type ll transcript patterns were identified in transgenic tobacco plants and only Type I transcripts were discussed in this paper, The activity of GST in the type II transgenic plants was about 1.5-fold higher than those of the wild type and non-expresser by using 1-chloro-2,4-dinitrobenzene (CDNB) and reduced glutathione as the substrate. The expression of cotton GST in tobacco plants proved that Gh-5 could be translated into functional protein. Type II transgenic plants produced functional GST in the cells. The effects of cotton GST in the seedlings was evaluated by growing the control and transgenic seedlings at $15^{\circ}C$ in the growth chamber in the light. Overexpressors were grown well compared to the control plants (non-expressors). lo test far tolerance to salinity, seeds of Gh-5 overexpressors and the wild type Xanthi seedlings were grown at 0, 50, 100, 150, and 200 mM NaCl solution. Gh-5 transgenic seedlings showed higher growth rate over control seedlings on 50 and 100 mM NaCl solution. There was no difference in growth rate at 150 and 200mM NaCl concentration.

Insect Pests and Natural Enemies of Hibiscus syriacus in Korea (무궁화 해충의 천적과 종류)

  • Insect pests and their natural enemies of Hibiscus Linne (Malvaceae) were investigated from March 2002 to November 2004. Fourteen insect pest species of 9 families in 5 orders were collected from Hibiscus syriacus: 5 species in Homoptera, 3 species in Lepidoptera, 2 species in Coleoptera, 1 species in Orthoprera, 1species in Hemiptera, 1 spedies in Acarina, and 1 species in Stylommatophora. Especially, Aphis gossypii Glover (Aphididae), Anomis megogona Walker(Noctuidae) and Tetranychus urticae Koch (Tetranychidae) were very important species because of their increasing daminge. The highest donsities were observed from May to June in August in Tetranychus urticae. As the enemies and ento-mopathogens of insect pests on Hibiscus syriacus, 1 species of bacteria, 3 species of fungi, 1 species of fungi, 1 species of Hemiptera, 1 species of Coleoptera, 2 species of Hymenopetera, 2 species of Diptera, and 1 species of Acarina were investigated. As the predators and parasitoids of Aphis gossypii, Aphidoletes aphidoletes aphidimyza Rondani (Cecidomyiidae), Meliscaeva cinctella Zetterstedt (Syrphidae), Harmonia axyridis Pallas (Coccinellidae), and Aphidius gifuensis Ashmead (Braconidae), entomopathogenic fungi, Vericillium lecani naturalis strain (Moniliaceae) and Beauveria bassiana naturalis strain strain (Hypocreaceae) were observed and Bacillus thuringiensis naturalis strain (Bacillaceae), B. bassiana, Metarhizium anisopliae naturalis strain (Hypocreaceae), predators of Tetranychus urticae, Amblyseius sp. (Phytoseiidae), and Orius sp. (Anthocoridae) were observed.

Aphidivorous Activity of a Coccinellid Beetle, Harmonia axyridis Pallas(Coleoptera: Coccinellidae) (포식성천적(捕食性天敵) 무당벌레의 진딧물 포식능력(捕食能力))

  • Some experiments were conducted to evaluate the aphidivorous activity of coccinellid beetle, Harmania axyridis Pallas. Experiments were carried out using with laboratory reared beetles and two aphid species(Aphis gossypii and Myzus persicae). Factors referred in the study of aphidivorous activity were the age of beetle, aphid species, temperatreu and illumination time. The aphidivorous activity was greatly different in relation to aphid species, developmental stages of the beetle, and the temperature. The 3rd and 4th instar larvae consumed more aphids than the 1st and 2nd instar larvae. Though the beetles consumed more number of cotton aphid, total weight of aphids consumed the more or less the same. Aphidivorous activity of the 1st to 4th instar larvae increased with a rise in temperature and adult activity was relativel high at $20^{\circ}C$ and low at $15^{\circ}C\;and\;30^{\circ}C$. The aphidivorous activity of the beetles was not influenced by the illumination period under the conditions given in this experiments, however, seem to require further study.

Effect of Tobacco Smoke on Controlling Aphids in Greenhouse (잎담배 훈연을 이용한 온실 내 진딧물 방제기술)

  • This study was conducted to observe the effects of dried tobacco leaf smoke on controlling aphids in a laboratory and a greenhouse. Insecticidal activity of tobacco smoke against Aulacorthum solani in an acrylic cage was higher when a burley cultivar, rather a flue-cured cultivar, was used. Mortality of A. solani, Aphis gossypii, and Myzus persicae was 63.9%, 94.4%, and 97.2%, respectively, after 50mg of tobacco smoke on their host plants in an acrylic cage. Mortality of M. persicae after tobacco smoke was used was higher in eggplant than in Chinese cabbage. When 100 g and 200 g of flue-cured tobacco were smoked in a $100m^2$ greenhouse for 2 h, the control values against A. solani were 28.9% and 95.4%, respectively; the control value after 14 h of smoking was more than twice the value after 2 h of smoking. The control value against A. gossypii was more than 80% after tobacco smoke was used in a greenhouse in an organic cucumber farm. Tobacco smoke can be an effective control against aphid pests in greenhouses if an appropriate amount of tobacco and smoking time on the basis of the greenhouse conditions are used.

Enhancement of Insecticidal Activity of the Acetamiprid Soluble Concentrate using a Photostabilizer (광안정제를 이용한 acetamiprid 액제의 살충효과 증진)

  • The study was carried out to select the effective adjuvant as a photostabilizer for acetamiprid 5% SL(soluble concentrate) and to find its optimum content by using chemical and biological methods. Adjuvants used for the study were KS1 and KS2. Photodegradation test, insecticidal efficacies and residual analysis for the study were investigated. Although photodegradation rate of acetamiprid SL was decreased by addition of KS1 and KS2, the difference between KS1 and KS2 was not significant. As KS2 showed better efficacies against the cotton aphid (Aphis gossypii) and higher amount of residue on pepper than KS1, KS2 was selected as a photostabilizer for acetamiprid SL. Acetamipid SL with KS2 showed lower photodegradation rate, better efficacies against the cotton aphid (Aphis gossypii) and higher amount of residue on pepper as content of KS2 in acetamiprid SL was decreased. Therefore, it was concluded that the optimum content of KS2 was 0.1%. These results have demonstrated that the selected adjuvant could be used to enhance insecticidal activity and reduce spay dose of insecticide by protecting its photodegradation.

Characteristics and Toxicity of Chrysanthemum sp. Line by Extract Part and Methods against Tetranychus urticae, Nilaparvata lugens, and Aphis gossypii (제충국 계통의 특성과 추출방법에 따른 벼멸구, 점박이응애, 목화진딧물의 살충활성 검정)

  • This study was carried out to evaluate characteristics of Chrysanthemum sp. and to bioassay Tetranychus urticae, Nilaparvata lugens, and Aphis gossypii by different extracts part and method. Flower type of Chrysanthemum sp. line was anemone except 0706 line which was pompon. The flowering date was from April to June but 0721 line kept up November. They could overwinter except 0706 line. The Ethanol extracts of 0718 line of flower was high mortality as 63.8% on Tetranychus urticae. Ethanol mixed emulsifier extracts were the highest mortality as 69.4% at 5 days after. Mortality in water extract was lower than in ethanol and ethanol mixed emulsifier extracts. Ethanol mixed emulsifier extracts were higher mortality as 69.4% at 5 days after in 0718 line on Nilaparvata lugens. Mortality in water extract was lower than in ethanol and ethanol mixed emulsifier extracts. On Aphis gossypii, Chrysanthemum flower extracts could decrease the density of aphids in early days but the density of that was increase as time goes by because aphids produce offspring.

Predatory Response of the Pirate Bug, Orius sauteri Poppius(Heteroptera: Anthocoridae) on Frankliniella occidentalis, Aphid gossypii and Tetranychus urticae (꽃노랑총채벌레 목화진딧물, 점박이응애에 대한 애꽃노린재(Orius sauteri)의 포식반응)

  • Daily predation of female/male O. sauteri during nymphal development were 4.2/3.9, 11.4/9.5, 14.3/10.8, and 14.7/12.5 at 17,22,27, and $32^{\circ}C$, respectively, and consumed 14.3/10.8 thrips, 7.5/7.2 aphids, 45.9/38.8 mites at $27^{\circ}C$, respectively. Adult females ate 42.8 thrips daily during preoviposition period and 63.2 thrips during oviposition period at $27^{\circ}C$. Predation by O. sauteri on F. occidentalis increased as the density of F. occidentalis increased. However, the rate of increase gradually lessened, resembling a Holling's type II functional response. The attack rate of adult female is higher than that of the 5th nymph. Adult female and 5th nymph of O. sauteri appeared to prefer 2nd larva of F. occidentalis.

Monitoring on Insecticide Resistance of Major Insect Pests in Plastic House (시설 재배 작물 주요 해충에 대한 약제저항성 모니터링)

  • This study was carried out for looking into the status of susceptibility of vinyl house insect pests to insecticides. The Thrips (Thrips palmi and Frankliniela occidentalis), Mites (Tetranychus urticae), Aphids (Aphis gossypii) and Whitefly (Trialeurodes vaporariorum) were captured at various areas where the host crop was being cultivated and the susceptibility level of each pest insect was investigated. The susceptibility of each pest insect varied by insect species and areas where they were caught. The tested insecticides showed good control effect to palm thrips in 2000, but in 2003 showed decrease of effect to them. Western flower thrips showed low susceptibility to neonicotinoids, imidacloprid and thiamethoxam, but high to chlorfenapyr, spinosad, emamectinbenzoate and fipronil. Antibiotic insecticides, abamectin and milbemectin, and chlorfenapyr were very effective on mite control and dicofol still had good effectiveness to it despite of long year use. No aphid species showed resistance to pyrethroid and carbamate insecticides. Relatively new insecticides such as imidacloprid, spinosad, pymetrozine were effective to whitefly, but not were organophosphates, carbamates and pyrethroids.

Comparison of Insect Pest Communities on 30 Cultivars of Hibiscus syriacus (나라꽃 무궁화 30품종에서 발생하는 해충상 비교)

  • In this study, insect pest communities and major insect pest species were investigated and compared among 30 cultivars of Hibiscus syriacus. Insects on H. syriacus were observed with the naked eye over 3 years (from 2018 to 2020) in Busan and Suwon. Except for Aphis gossypii (79,059 and 23,654 individuals in Busan and Suwon, respectively), 20 species (1,147 individuals) and 31 species (2,240 individuals) were found in Busan and Suwon, respectively. The number of insect pest species and individuals did not differ among H. syriacus cultivars, but there were differences according to study locations. The dominant insect pest species were A. gossypii, Rehimena surusalis, Rusicada privata, Halyomorpha halys, Haritalodes derogata, Dolycoris baccarum, and Plautia stali; the number of individuals in dominant species differed according to study location and year but not among H. syriacus cultivars. In summary, insect pest communities did not differ among 30 H. syriacus cultivars, but the matrix of surrounding environments where H. syriacus are planted may be more important.

Effect of Vinyl-mulching Culture on Growth and Yield of Cotton( Gosspium hirsutum L.) (비닐피복이 모화의 생육 및 수량에 미치는 영향)

  • This experiment was conducted to explore the cultural method capable of raising of picked cotton ratio and more high-yielding in the middle part of Korea where growth period of cotton is short. By early vinyl-mulching culture, emergence date was earlier, flowering and boll opening were shorter than non-mulching, especially boll opening date was earlier about 8 to 15 days. Plant height was longer by vinyl-mulching, fruiting branches and number of bolls per plant at vinyl-mulching of the 10th April produced more twice than non-mulching of the 10th May. Picked cotton ratio was 79% at non-mulching of the 10th May, while viny-mulching of the 10th April produced most of picked cotton as picked cotton ratio 99%. At vinyl-mulching of the 10th April, lint yield increased more 76-90% than non-mulching. Staple length was some longer and also content of linolenic acid of cotton seed oil was higher than non-mulching.
