The Antioxidant and Anticancer Effects of Butterbur (Petasites japonicus) Extracts (머위(Petasites japonicus) 추출물의 항산화와 항암활성 효과)

  • The antioxidant activities of the extracts of butterbur (Petasites japonicus) derived from different extraction methods were investigated. SOD (superoxide dismutase)-like activity differed according to the extraction solvents, showing a greater antioxidant effect with ethanol solvent than that of water. Ethanol extracts of butterbur leaves showed higher SOD-like activity of 96.7% than those of water extracts. The contents of polyphenolic compounds were higher in water extracts than those in ethanol extracts. The highest content of polyphenolic compounds was 223mg/g dry weight for butterbur leaves. EDA of butterbur roots was 61.5% in the water extract and EDA in butterbur leaves was 34.9% in the ethanol extract. The anticancer effects with the extracts of butterbur were experimented by fractionations with different solvents. 41.9% of the growth of stomach cancer cells, SNU-719, were inhibited and also 72.7% of the growth of liver cancer cells, Hep3B, were inhibited by the butanol fractions of butterbur, while not affecting the growth of normal cell, DC 2.4.

  • This study was carried out to analyze the major chemical component of the leaf and stem of dried Petasites japonicus S. et Z. Max.. Comparing proximate composition of leaf and stem of Petasites japonicus as dry matter basis, stem contained higher moisture, crude fat, crude ash and carbohydrate, with less crude protein. The main component of free sugar and disaccharide in both leaf and stem were fructose and sucrose, respectively. Total amino acids of leaf and stem were 6,853.32 mg% and 2,350.61 mg% respectively. Although the amino acid composition of leaf and stem were different glutamic acid and aspartic acid were the major amino acids in samples. The major fatty acids of total lipids were linolenic acid in leaf and linoleic acid in stem. The ratios of unsaturated fatty acids to saturated fatty acid were 3.93 in leaf and 3.44 in stem. The unsaturated fatty acid content of samples were 3 times higher than those of saturated fatty acid contents. Oxalic acid was the major organic acids in leaf and stem. The contents of vitamin A, C and E were higher in leaf than in stem. The mined compositions of both leaf and stem were composed in order of K, Mg, Ca, Fe, Na, and Zn.

Cytoprotective Effect of Petasites japonicus Extract on Cadmium-induced Cytotoxicity in HaCaT cell (Cadmium으로부터 손상을 유도한 HaCaT 세포에서 머위(Petasites japonicus) 추출물의 세포보호효과)

  • In this study, we investigated the effect of Petasites japonicus extract on the cytotoxicity and cytoprotective effects against cadmium for cosmetics use. We measured the protein expression of apoptosis regulatory factor (Bcl-2 and procaspase-3) after treatment of Petasites japonicus extract in the cadmium-induced keratinocyte. As a result, high cell viabilities above 98% were observed in the all treated concentrations except at $200{\mu}g/mL$ of Petasites japonicus extract in keratinocytes with cadmium-induced damages. In keratinocytes with cadmium-induced damages, Bcl-2 and procaspase-3 protein expression increased in the experimental group treated with Petasites japonicus extract. Also HaCaT cells resulted in cleavage of PARP protein at 12 h post-cadmium exposure. Western blot analysis and relative density of the bands suggested that pretreatment of cells with Petasites japonicus extract inhibited cadmium-mediated cleavage of PARP. These results suggest that Petasites japonicus extract can be used as the cosmetic ingredients for cytoprotective effect.

Effects of Ethanol Extracts from Petasites japonicus S. et Z. Max. on Hepatic Antioxidative Systems in Alcohol Treated Rats (머위 추출물이 알코올 투여한 흰쥐의 간조직 내 항산화 체계에 미치는 영향)

  • This study investigated the hepatoprotective effects of an ethanol extract of Petasites japonicus S. et. Z. Max. (PJ) on alcohol-induced liver-damaged rats. Sprague-Dawley rats weighing $100{\sim}150\;g$ were divided into 5 groups; normal diet group (NOR), alcohol (35%, 10 mL/kg/day) treated group (CON), PJ 200 mg/kg/day treated group (PJ1), PJ 200 mg/kg/day and alcohol treated group (PJ2), and PJ 400 mg/kg/day and alcohol treated group (PJ3). The growth rate of the control group was higher than that of normal group, whereas the group administered PJ concomitantly was significantly increased. Also, feed efficiency ratio decreased by alcohol administration was gradually increased to the adjacent level of the normal group by administering PJ. The AST activity in serum elevated by alcohol was significantly decreased by administering the high dosage of PJ, but exerted no significant change on serum ALT activity. It was also observed that the hepatic activities of catalase and GSH-Px increased by alcohol were markedly decreased in PJ2 and PJ3, but not in the activities of XO and SOD as compared with the control group. The depleted content of GSH by alcohol was increased to the level of normal group by administering PJ in a dose-dependant manner. In conclusion, these results suggest that PJ may have a possible protective effect on liver function in hepatotoxicity-induced rat by alcohol administration.

Investigation of the Dormant Characteristics for Early Production of Young Leaf in Butterbur(Petasites japonicas MAX.) (머위 유엽 조기생산을 위한 휴면특성 조사)

  • In order to produce young leaf of butterbur(Petasites japoflicus MAX.) in early spring, the planting date and relationship between abscisic acid(ABA) content and dormancy were investigated. Under open field condition, the dormancy of rootstock was initiated in the beginning of October, was the deepest in the middle of November and was completely broken in the end of December. When those periods were converted by the low accumulation hour below 5$^{\circ}C$, 900 hours were required approximately. This means that the rootstock needs for dormant breaking necessitates under the low temperature. In relationship between growing period and ABA content, the ABA in root-stock did not exist during maximum growing period, from April to September. This means that the ABA together with other substances in rootstock can be transferred to the shoot part with sprouting. While shoot part withered by decreacing the open field temperature since October, the ABA intiated to exist in rootstock. This means that the ABA in the shoot part can be transferred to the rootstock part. Therefore, it was concluded that the ABA which has been known that inhibiting growth and inducing dormancy was closely related with dormancy of rootstock.

Skin barrier and anti-inflammatory effect of petasites japonicus (머위(Petasites japonicus)의 피부장벽과 항염증 효과)

  • In this study, to investigate the possibility of using ethanol extract of Petasites japonicus (PJE) as a functional material, we investigated the activity of improving skin barrier and inflammation through UVB-induced human keratinocyte (HaCaT cell). As a result of confirming the antioxidant effect through DPPH radical scavenging activity, ABTS+ radical scavenging activity, and hydrogen peroxide scavenging activity, it was confirmed that it had an antioxidant effect similar to that of ascorbic acid, a control, at a concentration of 1 mg/ml. As a result of confirming the mRNA expression of the production ability of filaggrin and aquaporin-3 in HaCaT cells induced by UVB, it was confirmed that the reduced expression level by UVB stimulation increased in a concentration-dependent manner when the PJE was treated. It was confirmed that the mRNA expression of TNF-𝛼 and IL-1𝛽 were increased by UVB stimulation and decreased when the PJE was treated. As a result of the migration assay, it was confirmed that the proliferation of skin keratinocytes and the recovery rate of wounds were increased in a concentration-dependent manner. Based on the experimental results, it suggests that Petasites japonicus can be used as a functional cosmetic product that can improve skin moisturizing and skin barrier function.

Anti-allergic Effects of Petasites japonicum (머위(Petasites japonicum) 추출물의 항알레르기 효과)

  • It is well known that the Petasites japonicum have been used for a long time in traditional medicine for the treatment of allergic diseases such as lacquer poisoning and asthma. Anti-allergic actions of Petasites japonicum extracts were asessed by testing their effects on the degranulation of mast cells. For this, hexosaminidase released (degranulation marker) from RBL-2H3 cells(mast cell line) was used. At the concentration of 300 $\mu\textrm{g}$/mL of the methanol, ethylacetate and hot water extract, the degranulation of RBL-2H3 cells were inhibited 83.33, 69.75 and 35.4%, respectively. These results suggest that the Petasites japonicum could be provide a effective resource for the control of allergic diseases.

  • The leaf of Petasites japonicus was extracted with 80% aqueous MeOH and solvent fractionated with EtOAc, n-BuOH and water successively. From the EtOAc fractions, two triterpenoids were isolated through the repeated silica gel and ODS column chromatographies. The chemical structures of the isolated terpenoids were determined as rosamutin (1) and peduncloside (2) through the interpretation of several spectral data including 2D-NMR such as $^1H-{^1H}$ gCOSY, gHSQC and gHMBC.

  • Antioxidative and anti-inflammatory activities of Petasites japonicus extract were evaluated. P. japonicus extract showed 70.1% inhibition on peroxidation of linoleic acid. In the experiment using the cell permeable dye, 2',7'- dichlorofluorescein diacetate (DCFDA) as an indicator of reactive oxygen species (ROS) generation, intracellular oxidative stress in UVB-irradiated keratinocytes was shown to be decreased by P. japonicus extract. Also, UVB-induced production of interleukin-$1{\alpha}$ and prostaglandin $E_2$ in human HaCaT keratinocytes was reduced in a dose-dependent manner by treatment with P. japonicus extract. All these results suggest that P. japonicus extract can be effectively used for prevention of UV-induced adverse skin reactions such as radical production and inflammation.
