우리나라에서는 패류에 있어서 마비성패류독 함유량이 가식부 100g당 saxitoxin으로서 80$\mu\textrm{g}$이하로 기준을 정하고 국립수산과학원에서는 패류독의 발생 여부를 년중 감시하고 있으며, 특히 독소가 주로 발생하는 봄철에는 집중적으로 검사하여 패류독이 기준치를 초과하는 경우에는 채취 및 유통을 금지하여 패류의 안전성을 확보하고 있다. (중략)

AOAC Mouse Bioassay Analysis (MBA) has been the gold standard for the analysis of paralytic shellfish poisoning toxin (PSP toxin) for more than 50 years. However, this method has inaccurate limit of quantification and cannot be used to determine toxic profiles. An HPLC method (PCOX) was optimized for Korean shellfish to establish an alternative or supplementary method for PSP analysis and was intended to be used for the official monitoring and regulation of food. The recovery rate of the PCOX method was 83.5-112.1% and the limit of quantification for total toxin was about $8.6{\mu}g$/100 g. A long-term comparison study showed a good correlation of the PCOX results with the AOAC MBA results: the correlation factors were 0.9534 and 0.9109 for oyster and mussel matrices, respectively. The PCOX method may be used as an alternative or supplementary method for AOAC MBA to monitor the occurrence of PSP and to analyze PSP toxin profile in oysters and mussels.
Identification of species within the toxin-producing genus Alexandrium is vital for biotoxin monitoring and mitigation decisions regarding shellfish industry. In particular, the discrimination of resting cysts of only A. tamarense from that of Alexandrium spp. is considerable important to fundamentally monitor and predict this species before vegetative cells occur in the nature. Fluorescent cTAM-F1 DNA probe was responsible to not only binding the activity of the vegetative cells in A. tamarense, but also to the resting cysts, which was treated with methanol after fixation and stained by primuline on the surface The location of fluorescence in cultured vegetative cells and resting cysts was almost at tile bottom of the nucleus. The optimal incubation temperature and time using in situ hybridization were 50-$54^{\circ}C$ and 40-60 min, respectively, to penetrate the DNA probe into cell.
Paralytic shellfish poison (PSP) occurrence and variation in th\ulcorner different bivalve species including oyster,Crassostrea gigas and mussel, Mytilus edulis Jinhae bay Korea from January to December in 1997 were surveyed. And also compositional characteristics of PSP ingredients in the different bivalve species were investigated. PSP in shellfish was detected from late February and increased continuously until the middle of April in Jinhae bay. And after April PSP level had been decreased gradually and the toxicity was not detected by mouse bioassay in the early of June. Of the examined bivalve species, PSP content in the mussel exhibited the highest value and the PSP content in the mussel in the middle of April, PSP high season in Jinhae bay, was 6 times higher than that in the oyster. Gonyautokin (GTX) 1~4 group occupied 59.0~78.8% of whole PSP contents and identified as dominant ingredient in the examined bivalves except oyster. And it is also identified that the PSP toxicity in the tested species were derived from the GTX group. And the dominant ingredient of PSP in the oyster was carbamoyl-N-sulfo-11$\alpha$-hydroxysaxitoxin sulfate(Cl)(37.9%) and neosaxitoxin(neoSTX)(26.2%). But the toxicity of Cl in the tested oyster could be ignorable and most toxicity (80.0%) was derived from saxitoxin (STX) group.

To evaluate bacteriological and toxicological safety hygienic indicator bacterium and paralytic and diarrhetic shellfish toxins in the shellfish produced in Hansan Geojeman 2013-2017 were investigated. Fecal coliforms were < 18~330 MPN/100 g in 404 oyster samples. But all samples tested, did not exceed 230 E. coli MPN/100 g. Geometric mean of E. coli for oyster samples collected during major shellfish production period was 24.3 MPN/100 g, considerde stable results. Bacteriological quality of oysters collected from Hansan Geojeman meets the standard value based on shellfish hygiene of the Food Sanitation Act of Korea and also meets Grade A, according to classification of shellfish harvesting areas of the European Union. For toxicological evaluation of Hansan Geojeman, 532 oyster samples and 268 mussel samples as an indicator, were analyzed. Paralytic shellfish toxins were detected in the range of 0.42~2.29 mg/kg in eight mussel samples, and exceeded criteria in three samples from early to late April 2013. Diarrhetic shellfish toxin was detected in three of 120 samples, but it was revealed to be under regulation value (0.16 mg Okadaic Acid equ./kg). As a result of toxicological evaluation, paralytic and diarrhetic shellfish toxins were not detected in oyster samples, but it was found that mussel as an indicator species, exceeded the threshold value of paralytic shellfish toxin. Accordingly, sanitary surveys were continuously requested for food safety management of shellfish.

To evaluate bacteriological and toxicological safety, the hygienic indicator bacterium and paralytic and diarrhetic shellfish toxins in the shellfish produced in the Kamakman Area from 2012 to 2016 were investigated. Fecal coliforms and E. coli of all 194 oyster samples tested did not exceed 230 MPN/100 g. The geometric mean of the fecal coliform analyzed with the oyster samples of harvesting period was 19.6 MPN/100 g, which was more stable than the non-harvesting period (26.5 MPN/100 g). For the toxicological evaluation of the Kamakman Area, 77 oyster samples and 350 mussel samples as an indicator were analyzed. Paralytic shellfish toxins were detected very low in the range of $40{\sim}46{\mu}g$/100 g in 13 mussel samples during late April and early June, but not in oyster samples. Diarrhetic shellfish toxin was detected in 2 of 180 samples, but it was found to be below the regulation value (0.16 mg OA equ./kg). Based on the bacteriological studies, it was confirmed that the shellfish produced in Kamakman area meets the standard of shellfish hygiene of the Food Sanitation Act and meets the Grade A of the shellfish production area of EU. As the results of the paralytic and diarrhetic shellfish toxin evaluation, it was confirmed that the Kamakman Area is also toxicologically safe for shellfish production.
In order to investigate the distribution of paralytic shellfish poison, we examined the toxicity during from February to October in 2000. Of 591 shellfish samples, 17(2.88%) samples were detected. Scapgarca broughtonii was highest collected 14.29%(2/14). In the monthly detection rate of PSP, April was hitest 13.3%(8/60), in the regional collecting rate, Cheon-nam coastal area was highest 3.82%(10/262), and in cases of imported area, China was 8.3%(1/12). Imported area as well as domestic area samples should be strengthen to examine enduringly.

We monitored paralytic shellfish poison (PSP) by HPLC method in shellfishes including a sea squirt, and its products produced in domestic and imported, total 35 species, 850 samples, collected at 9 cities such as Seoul, Busan, Daegu, Ulsan, Pohang, Masan, Tongyeong, Geoje and Sacheon, 10 times by month (2 times in April and May) from March to October, 2004. PSPs were detected in a few raw samples on March, and it showed highest toxic ratio $(72.9\%)$ on third week of April (51 samples toxic in total 70 samples) and decreased suddenly to $2.4\%$ on third week of May. After then, PSPs were detected in a few samples, sporadically, and disappeared after August. Most raw shellfishes in the market were safe from PSP, showing the toxicity below 1 MU/g (quarantine level 4.0 MU/g), except oyster collected on April at Masan (1.9 MU/g), blue mussel on April (1.8 MU/g) and purplish washington clam on May at Sacheon (2.1 MU/g). However, it was strongly suggested to survey and under control for the imported scallops and ark shells which showed highly toxic in the quarantine level on May, July, even September. PSP were not detectable in the all shellfish products collected on May, July and September, except 2 boiled and dried mussels contained trace amount (0.01 MU/g), and all those products were safe as below quarantine toxin level (4.0 MU/g).
The purpose of this study was to determine the kinetics of paralytic shellfish poison (PSP) destruction at various temperature. The toxic digestive gland homogenate of blue mussel (Mytilus edulis), PSP crude toxin, gonyautoxin group and saxitoxin group were heated at temperature ranging from 90 to $120^{\circ}C$, and then the toxicities were measured in samples heated for various time intervals. The rate constant (k) of the toxic digestive gland homogenate, PSP crude toxin, gonyautoxin group and saxitoxin group were $3.28{\times}10^{-2},\;1.20{\times}10^{-2},\;5.88{\times}10^{-2}\;and\;2.58{\times}10^{-2}\;at\;120^{\circ}C$, respectively. The decimal reduction time (D-value) of the toxic digestive gland homogenate, PSP crude toxin, gonyautoxin group and saxitoxin group were 70, 192, 39 and 89 at $120^{\circ}C$, respectively. These results indicate that PSP crude toxin is most heat-stable of 4 types of PSP toxins and PSP toxin are more heat-stable than food poisoning bacteria and spores. The retorting condition to reduce PSP toxicity below quarantine limit ($80\;\mu\textrm{g}/100\;g$ in Korea and America, 4 MU/g in Japan) could be calculated by rate constant. For example, the digestive gland homogenate having a initial toxicity of $200\;\mu\textrm{g}/100\;g$ could have toxicity below quarantine limit when heated at $90^{\circ}C$ for 129 min., $100^{\circ}C$ for 82 min., $110^{\circ}C$ for 48 min. and $120^{\circ}C$ for 28 min. These results suggest that commercial retorting condition ($115^{\circ}C$ for 70 min) in Korea is enough to reduce toxicity below quarantine limit from initial toxicity of $200\;\mu\textrm{g}/100\;g$. From these results, the quarantine limit of PSP-infested shellfish for canning can be level up to raw score of $200\;\mu\textrm{g}/100\;g$.

Optimum temperature for paralytic shellfish poison (PSP) detoxofication of Enterobacter sp. CW-6 isolated from sea water and changes of contents and ingredients composition of PSP during bacterial detoxification process were investigated. Enterobacter sp. CW-6 detoxicated $61.5{\~}67.7{\%}\;and\;87.4{\~}96.8{\%}$ of initial PSP toxicity ($25.0{\~}28.5\;nmole/g$) after $5{\~}12$ days at 30 and $35^{\circ}C$, identified as optimal growth temperature, respectively. The detoxification rate of Enterobacter sp. CW-6 for crude PSP with initial concentration of 38.2 nmole/g after 8 and 12 days at $30^{\circ}C$ in the Marine broth was 88.4 and $92.7{\%}$, respectively. During bacterial detoxification process using crude toxin solution, temporary increasement of STX group was detected and identified that was derived from GTX2, 3 group. The detoxification rate of Enterobaoter sp. CW-6 on purified GTX1 and 4 with initial concentration 47 nmole/g and 37 nmole/g were more than $90{\%}$ after 12 days in the marine broth at $30^{\circ}C$. Enterobacter sp. CW-6 also showed a detoxification activity on purified GTX2 and 3, and the detoxification rate for the initial concentration 25.6 nmole/g after 12 days was $66.4{\%}$.
